        Co-immobilization of cholesterol esterase, cholesterol oxidase and peroxidase onto alkylamine glass beads for measurement of total cholesterol in serum

        Commercially available cholesterol esterase, cholesterol oxidase and peroxidase have been co-immobilized onto alkylamine glassinitial activity. The co-immobilized enzyme system showed maximum activity at pH 7.0, when incubated at 37.C for 12 min. Amethod was developed for total serum cholesterol determination employing co-immobilized enzymes. There was a linear rela-tionship betweenA520 and cholesteryl acetate concentration ranging from 5mg to 50 mg/dl reaction mixture. The minimum de-tection limit of the method is 50 mg/dl. Within day and between day coecient of variation were< 1.0% and < 6%, respectively. Ar¼ 0:83) was found between the total serum cholesterol obtained by the present method and commercial Enzo-kitmethod employing free enzymes. Among the various serum substances tested at their physiological concentrations; Testosterone, vitD and progesterone caused 59%, 41% and 39% inhibition, while NaCl, KCl, CuSO4, creatinine, NaHCO3, albumin and estrogenhad practically no eect.. 2002 Elsevier Science B.V.         Field larvicidal and oviposition repellent efficacy of three essential oil formulations for the control of filaria vector mosquito Culex quinquefasciatus Say (Diptera: Culicidae): A kill and push concept

        Biological control of larval mosquitoes is in great demand due to the development of resistance against synthetic insecticides, environmental toxicity and the inability to protect habitats from further oviposition. In the present study, three botanical essential oils (BEOs) – citronella, eucalyptus, and pine oils – were formulated for the assessment of larvicidal and oviposition repellent efficacies against Culex quinquefasciatus Say, the filaria vector. The GC–MS profiling of BEOs showed the presence of 16 – 19 compounds covering 87.7–93% of oil composition. The resistance status of Culex quinquefasciatus population was evaluated with temephos (LC 50 = 0.001 ppm, LC 90 = 0.01 ppm). Larval bioassay of emulsifiable concentrate (EC) formulations prepared from eucalyptus and pine oils showed promising efficacy (LC 50 = 22.7 and 23.2 ppm) and LC 90 (63.8 and 62.4 ppm) compared to citronella oil EC (LC 50 = 43.4 ppm and LC 90 = 199.0 ppm). The field trials of eucalyptus + pine (1:1 ratio) EC showed 100% larval mortality for 3 weeks at 300 ppm compared to 2 weeks of individual oils. Further, the oviposition attraction index (OAI) for ECs of eucalyptus, pine, and their combination showed complete protection of breeding habitats from oviposition at 1st week and − 0.9 to − 1.0 OAI at 2nd week with slight reduction to − 0.5 at 3rd week. Citronella EC provided shortest larvicidal and oviposition repellent efficacy under the field con ditions. The promising mosquitocidal activities of EC formulations of eucalyptus and pine or their combination suggest them as potential biocontrol vector control candidates over citronella oil.

        Investigation of thermal performance of SAC variables using fuzzy logic based expert system

        The thermal performance of a solar air collector (SAC) is investigated experimentally under the different climatic conditions of north eastern India using fuzzy logic based expert system (FLES). The FLES based on subtractive clustering (SC) with the fuzzy logic method where here, SC is used for extraction of optimal fuzzy IF-THEN rules while a fuzzy logic is used for modeling of SAC variables. This work considered four input variables [like mass flow rate (m), collector tilt angles (θ), solar radiation (Q), temperature (T)] and the four output variables [i.e. efficiency (η), exergetic efficiency (η II ), temperature rise (∆T), and pressure drop (∆P)]. First, 272 trials of experimentation on SAC are performed by varying m from 0.0078 to 0.0118 kg/s and θ from 30 to 60°, whereas the variation of metrological data is obtained in different working days. Then modeling and parametric analysis is carried out for SAC. Experimental results reveal that the value of η increases with the increase in m, Q, T and θ up to 45°. The higher value of m results in a higher value of ∆P and that reduces the value ofη II . Also, FLES model provides comparable and acceptable values for SAC. At last, validation of the FLES model is done via published data to confirm the results.

        Comparative leaching of spent fluorescent lamp for extracting yttrium and europium: kinetics and optimization studies

        Electronic (e-)waste viz. the spent fluorescent lamps can be a rich source of rare earth elements (REEs) for the secondary supply of them. This article reports a comparative study on mineral acid leaching and, then, compared an alkali diffusion-acid leaching process for higher efficiency of yttrium and europium. Using a direct acid leach process, 2 M HNO3 yielded a better dissolution efficiency of REEs at the moderate temperature of 60°C and in the presence of H2O2in comparison to 91% and 87% leaching efficiency with 2.0 M H2SO4. On the other hand, the alkali-fused phosphor mass leached in 2.0 M H2SO4 without H2O2 addition showed remarkable progress in dissolution efficiency, yielding >99% and >98% of yttrium and europium in the leach liquor. Furthermore, the leaching kinetics of the alkali-diffused phosphor showed the best fit with the logarithmic rate law. The apparent activation energy was calculated to be 43.3 kJ/mol and 35.1 kJ/mol for yttrium and europium, indicating two different mechanisms during the leaching process by following the chemical- and diffusion-controlled reactions, respectively.

        Antihyperlipidemic Effect of Active Principle Isolated from Seed of Eugenia jambolana on Alloxan-Induced Diabetic Rabbits

        Diabetes is accompanied by lipid abnormalities, which contribute significantly to cardiovascular morbidity and mortality in diabetic patients. We previously demonstrated the potent antihyperglycemic activity of the active principle (fraction II from Sephadex LH 20 chromatography [LH II]) isolated from ethanolic seed extract of Eugenia jambolana in diabetic rabbits. In the present study, the efficacy of LH II was evaluated for its hypolipidemic activity in alloxan-induced mildly diabetic (MD) and severely diabetic (SD) rabbits. Phytochemical investigation of LH II by various structural spectra showed the presence of saturated fatty acid, Δ5 lipid, and sterol. Oral administration of LH II (10 mg/kg of body weight) for 21 days resulted in improved glycemic control in both MD and SD rabbits. After treatment with LH II, serum total cholesterol, triglycerides, high-density lipoprotein cholesterol, and the total cholesterol/high-density lipoprotein cholesterol ratio were significantly improved. LH II also resulted in significant (P < .001) improvement in 3-hydroxy-3-methylglutaryl-coenzyme A reductase activity and levels of total lipids and glycogen in both MD and SD rabbits. Thus, the present study demonstrates that LH II possesses potent hypolipidemic activity and efficacy in both MD and SD rabbits.

        Viscometric and FTIR studies of molecular interactions in 2-propanol+hydrocarbons mixtures at 298.15 and 308.15 K

        The deviation in viscosity was coupled with respective excess molar volume data to study the molecular interaction in binary mixtures with one associated component. This approach was applied to the experimentally measured viscosity and excess molar volume data of the 2-propanol+hydrocarbons at 298.15 K and 308.15 K. It was suggested that depolymerization power of aromatic hydrocarbon toward 2-propanol as well as strength of intermolecular interactions (electron-donor-acceptor type) between monomer of 2-propanol and aromatics depend on -electron density of the aromatic hydrocarbon. These interactions were further confirmed by FTIR spectroscopy. The viscosity of these binary mixtures was best predicted by Gruenberg-Nissan correlation among the four correlations applied.

        Optimization of different factors for an Agrobacterium-mediated genetic transformation system using embryo axis explants of chickpea (Cicer arietinum L.)

        In this study, we developed a reliable and efficient Agrobacterium-mediated genetic transformation system by applying sonication and vacuum infiltration to six chickpea cultivars (ICCV2, ICCV10, ICCV92944, ICCV37, JAKI9218, and JG11) using embryo axis explants. Wounded explants were precultured for 3 days in shoot induction medium (SIM) before sonication and vacuum infiltration with an Agrobacterium suspension and co-cultivated for 3 days in co-cultivation medium containing 100 µM/l of acetosyringone and 200 mg/l of L-cysteine. Responsive explants with putatively transformed shoots were selected using a gradual increase in kanamycin from 25 mg/l to 100 mg/l in selection medium to eliminate escapes. Results showed optimal transformation efficiency at a bacterial density of 1.0, an optical density at 600 nm wavelength (OD600), and an infection duration of 30 min. The presence and stable integration of the β-glucuronidase (gusA) gene into the chickpea genome were confirmed using GUS histochemical assay and polymerase chain reaction. A high transformation efficiency was achieved among the different factors tested using embryo axis explants of cv. JAKI 9218. Of the six chickpea cultivars tested, JAKI9218 showed the highest transformation efficiency of 8.6%, followed by JG11 (7.2%), ICCV92944 (6.8%), ICCV37 (5.4%), ICCV2 (4.8%), and ICCV10 (4.6%). These findings showed that the Agrobacterium-mediated genetic transformation system will help transfer novel candidate genes into chickpea.

        A Compact Microstrip Patch Antenna Based on Metamaterials for Wi-Fi and WiMAX Applications

        A low profile asymmetrical fractal boundary patch antenna based on reactive impedance surface (RIS) and a mushroom unit cell (MUC) is proposed and studied for dual band operation. The sides of the square patch antenna are replaced with asymmetrical half circled fractal curves for circular polarization operation at patch mode band. The fractal patch antenna is loaded with MUC for dual band operation. The antenna radiation characteristics are investigated and illustrated with both simulated and experimental results in detail. The 10-dB return loss bandwidth are 8.48% (3.21–3.49 GHz) and 2.59% (2.30–2.36 GHz) at upper and lower resonance frequencies, respectively. The 3-dB axial ratio bandwidth is 4.26% (3.21–3.35 GHz). A close agreement between simulation data with experimental results is observed.

        Enhanced Mechanical Properties of Functionalized Graphene Oxide/linear Low Density Polyethylene Composites Prepared by Melt Mixing

        Graphene oxide (GO) was concurrently reduced and functionalized using long alkyl chain dodecyl amine (DA). The DA functionalized GO (DA-G) was assumed to disperse homogenously in linear low density polyethylene (LLDPE). Subsequently, DA-G was used to fabricate DA-G/LLDPE composites by melt mixing technique. Fourier transform infrared spectra analysis was performed to ascertain the simultaneous reduction and functionlization of GO. Field emission scanning electron microscopy analysis was performed to ensure the homogenous distribution and dispersion of DA-G in LLDPE matrix. The enhanced storage modulus value of the composites validates the homogenous dispersion of DA-G and its good interfacial interaction with LLDPE matrix. An increased in tensile strength value by ~ 64% also confirms the generation of good interface between the two constituents, through which efficient load transfer is possible. However, no significant improvement in glass transition temperature was observed. This simple technique of fabricating LLDPE composites following industrially viable melt mixing procedure could be realizable to developed mechanically strong graphene based LLDPE composites for future applications.

        Influence of formulated organic Plant tissue culture medium in the shoot regeneration study of Brassica juncea (l.) - Indian mustard

        Efficient protocol for plant shoot regeneration of Brassica juncea L. CZERN was established by using organic media components and growth stimulating factors of the vermicompost and coelomic fluids. Formulated organic plant tissue culture media (Vermicompost (30%) extracts supplemented with 20 mL/L coelomic fluid) have shown maximum shoot regeneration when compared with the Murashige and Skoog (MS) medium, which were supplemented with 1 mg/L 6-benzyladenine (BA) and 0.1 mg/L of Naphthaleneacetic acid (NAA). Cotyledon explants produced the highest shoot regeneration frequency from fourday- old germinated seedlings in comparison with non-germinated seedlings. The vermicompost extracts have proved to be the best organic plant growth media to induce shoots from cotyledons compared to the MS media. Statistically significant difference (P = 0.008) for the root length, shoot length (P =0.000350) and the leaves (P=0.375) of the mustard plantlets were analyzed successfully. The survival rate was 98% in the mustard cotyledons on the Vermicompost extract media and 63% on MS media respectively. The coelomic fluid also is much suitable to induce shoots from cotyledons at lower concentrations. It was also shown that the vermicompost extract, which comprised of humic acids along with coelomic fluid, affected shoot regeneration from the cotyledons. An efficient and organic shoot regeneration study was standardized and it can be applicable in the improvement of the economically important crops.
