      • Combined Gene Therapy with Hypoxia-Inducible Factor-1α and Heme Oxygenase-1 for Therapeutic Angiogenesis

        <P>Transfection with either hypoxia-inducible factor-1α (HIF-1α) or heme oxygenase-1 (HO-1) gene can induce neovascularization in ischemic tissues. Although expression of transfected HIF-1α gene occurs rapidly, the expressed HIF-1α protein degrades quickly, limiting its therapeutic efficacy. Meanwhile, expressed HO-1 protein does not rapidly undergo degradation, but gene expression occurs a couple of days after transfection, resulting in apoptosis and a delay in angiogenesis in ischemic tissues at the incipient period of HO-1 gene transfection. We hypothesize that combined delivery of HIF-1α and HO-1 gene will enhance antiapoptosis and neovascularization in ischemic tissue compared with HIF-1α or HO-1 single-gene therapy. To test this hypothesis, ischemic mouse hindlimbs were treated with HIF-1α and/or HO-1 gene therapy. The combined gene therapy proved superior to both single-gene therapies, resulting in rapid expression of HIF-1α gene and long-term maintenance of expressed HO-1 protein. The apoptosis in the ischemic region was significantly less, and angiogenic growth factor secretion and angiogenesis were greater in the combined gene therapy than in either of the single-gene therapies. Our results suggest that a combined gene therapy of HIF-1α and HO-1 enhances the transfection of both genes and improves angiogenesis compared with either single-gene therapy.</P>

        복어중독에 의한 가사 상태에서 소생한 1예

        Tetrodotoxin is a neurotoxin produced by about 90 species of puffer fish and causes paralysis of central nervous system and peripheral nerves by blocking the movement of all monovalent cation. Ingestion of tetrodotoxin produces clinical manifestations such as paresthesias(within 10-45 min), vomiting, lightheadedness, salivation, muscle twitching, dysphagia, difficulty in speaking, convulsion and death that expressed by cardiopulmonary arrest with loss of brain stem reflex sometimes. Tetrodotoxin prevents or delays ischemia induced neuronal death by way of following 3 mechanisms. Firstly, it reduces the energy demand of the brain tissues. Secondly, it delays or even prevents anoxic depolarization. Finally, it deminishes ischemia induced cell swelling and cerebral edema. We report a case of puffer fish poisoning which presented with cardiopulmonary arrest and loss of brain stem reflex, but completely recovered by aggressive cardiopulmonary resuscitation.

        In this study, the influence of energy factors on energy consumption in a selected office building is analyzed by sensitivity analysis. Dominant energy factors of each building and system are determined. Then, the energy management schemes for these factors are suggested. If this method is applied to other office buildings, it may be possible to work out a strategy of energy diagnosis and retrofit more efficiently.

        冷凍冷藏倉庫 前室部 侵氣防止裝置의 適用에 關한 硏究

        This study aims to evaluate the economic performance of infiltration prevention devices for the refrigerated warehouse. It is showed that the cooling loads of refrigerated warehouses is changed by assembling of infiltration prevention devices in a previous study. Space loads and cooling energy costs according to the types of the infiltration prevention devices are analyzed by the DOE-2.1E simulation. Based on the Life Cycle Costs considered the costs of initial construction, repair·replace, cooling energy and operating energy, the probability of reducing energy consumption and application of infiltration prevent devices in vestibule of refrigerated warehouse is founded.

      • 중국 연변 조선족의 의식구조에 관한 조사 연구

        Because we recognized the necessity of a psychological preperation for the unification of Korea. We studied the mental structure of Yanbian Korean in China as a previous work for understanding the mental structure of North Koreans. The data from Yanbian Koreans(n=398) and Seoul National University students(n=308) were analyzed. The survey questionnare was composed of items feflecting the individual's view of value and items requiring evaluation of social issues. The mental structure of Yanbian Koreans was described and analyzed with respect to the classification of Yanbian-IllJung students, Yanbian University students and Yanbian citizen. And the difference between each Yanbian Korean group and the Seoul National University student group were analyzed.

        MPTP 독성물질이 도파민성 신경세포에 선택적으로 작용하여 산화성 손상에 의한 신경세포사를 일으키는 것을 이용하여 파킨슨병의 동물모델을 만들고, 이를 통해서 아폼토시스를 비롯한 포사의 기전에 대한 연구 및 너코틴의 신경세포 보호효과 여부를 판정하는 실험을 병행하고자 하였다. 파킨슨꾐의 동물모델을 MPTf 독성 물질을 이용하여 확립하였으며, MPTP(30mgag, i.p.)를 투여한 후 1, 2,3, 4, 5일째 흑질 조직을 채춰하여 tarm로 박걸하여 tyrosine hydroxylase 면역조직화학염색을 수행하여 cell countif우한 결과, control은 57.635ce11s, 1일째 친.OfDells,2일째 57.9±6cells,3일릴 없.3±죠ells, 4일째 49.0츠3cells, 5일째 39.4±Scells료 4, 3일째 뚜렷한 신경세포 수의 감소를 보였다. 신경세포사 기전 규명을 위한 아폼토시스 분걱에서는 벼PTP 투여 후 1, 2, 3, 4, 5일째 조직을 채취하여 Hoechst staining, TUNEL staining을 수곡하였는데 양성 반응을 보인 신경세포는 관찰되지 않아. 아폼토시스로 인한 세포사가 관찰되지 않았다. bIPTP 파킨슨병 동물모델에서 nicotine 보호효과 탐색에 관한 실험은 nicat푸e 0.2mgAg을 5일 퐁안 투여 후 리『fP(30mgag)를 CS7Bt/6 마은스에 복강 내주사로 nicotine과 병용 투여한 후 1, 2, 3, 4, 5일째 뇌를 적출하땄다. 신경세포사가 뚜렷이 관찰되기 시작하는 4, 5일째의 신경세포 수의 감소 정도를 20. 30% 정도 약화시키는 경향을 보였으나, nicotine 보호효과에 대한 추가 실헝이 현재 수행 중에 있다. The cause of Parkinson's disease (PD) is largely unknown. However, free radical toxicit? may plaf a role ip. the degeneration of substantia nigra, which is the Hajorfocus of pathological damages in PD. Recently, a neuroprotective effect of nicotine in PD has been suggested. Therefore, the mechanism of neurodegenerafion and protective potential o( nicotine in PD were investigated in the experimental modeB of Pll using a neurotoxin, C57BL/6mice were administered with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP, 30 mg/kg,j.p.). The degree of neurodegenerafion was determined by immunohistochemical stainiHB oftyrosine hydroxylase (TH). TH-positive cells on nigral sections were found 56.0 ±4, 57.9 ±6,52.315ce11s, 49.0±3cells, and 39,4±Scells at days 1, 2, 3, 4, 5, respectively (controls : 57.6±Scells). Hoechst and TUNEL staining showed no evidence of apoptosis. The exandnation on themice co-adrunistered with nicotine(0.2mgAg) and MPTP(30mgag) revealed a tendency ofnicotine protective effects. At days 4 and 5, the degree of TH-positive cells was decreased by20-30%, In corclusiffn, the role of apoptosis was not evidenced in this MPTP modeB of PB.The possible proteccon by nicotine should be elucidated with further studies.

In this study, we found a difference of Energy Consumption changes according to corridor style, direction, vertical and horizontal location, and roof-space by using DOE-2.1E energy simulation program. Based on this simulation results, found a probability of reducing energy consumption in educational buildings.

        In the literature study on the cerebral palsy, the results were as follows : 1.Cerebral palsy is defined as a disorder of movement and posture due to a defect or lesion of the immature brain. For practical purposes it is useful to exclude from cerebral palsy those disorders of posture and movement which are of short duration, due to a progressive disease, due solely to mental deficiency. 2.Cerebral Palsy is classified with quadriplegia, diplegia, hemi plegia, triplegia, and monoplegia or spastic CP, athetoid CP, ataxic CP, and combined classifications. 3.Causes of Cerebral Palsy is any damage with prenatal, natal and postnatal causes. 4.Management consists of helping the child achieve maximum potential in growth and development. This should be started as early as possible with identification of the very young child who may have a developmental disorder. Certain medications, surgery, and braces may be used to improve nerve and muscle coordination and prevent dysfunction. 5.The aim of treatment is to encourage children and adults to learn to be as independent as possible. Some children and adults who have midl cerebral palsy will have no problems in achieving independence 6.Oji(五遲),Oyeon(五軟) and Okyeong(五硬) have the simmiar concepts with the cerebral palsy. 7.Oji(五遲) Oyeon(五軟) and Okyeong(五硬) are caused by seoncheon-pumbu-bujok(先天稟賦不足) and related with gan(肝), bi(脾) and sin(腎). 8.The treatment is achieved by the method of bogansin(補肝腎), ganggeungol(强筋骨) and bojungikki(補中益氣). And jihwanghwan(地黃丸) has been used most frequently.

        충청북도 쌀농가의 벤치마킹을 위한 경영진단표 개발

        A new set of benchmarking lists for Korean rice farm is constructed in the following way. First, a set of benchmarking lists is made by a panel of specialists. The panel decides the research items and the scoring method of the lists. Second, a survey on rice farms are made with the lists. Third, by regression analyses based on the data from the survey, insignificant items are dropped out of the lists and the remaining significant items are given new weights.
