        Single Oral Dose Toxicity Evaluation of Leejung-tang, a Korean Traditional Herbal Formula, in Crl:CD (SD) rats

        Objective: Leejung-tang (Rechu-to in Japanese) is a traditional Korean herbal formula used for treatment of gastrointestinal disorders such as vomiting, stomach pain, chronic gastritis and gastrointestinal ulceration. The present study was carried out to investigate the potential acute toxicity of Leejung-tang water extract (LJT) by a single oral dose in Crl:CD (SD) rats in compliance with current guidelines. Methods: In the preliminary study, there were no adverse effects such as death, clinical signs, and body weight changes at dose levels of 500, 1000, and 2000 mg/kg/day body weight. Based on the results, a dose of 2000 mg/kg was selected as the toxicological limited dose. LJT was administered once by gavage to male and female rats at dose levels of 0 and 2000 mg/kg bodyweight. During the study period, mortalities, clinical findings, and body weight changes were observed for 14 days following the administration. On day 14 after the treatment, the animals were sacrificed by carbon dioxide overdose and complete gross postmortem examinations were performed. Results: In present study, no treatment-related deaths were observed. There were no adverse effects on clinical signs and body weight changes. In addition, there were no observed gross findings in all groups except for a kidney cyst in the 2000 mg/kg/day female group. Conclusion: The results indicated that LJT did not induce toxic effects at a dose level up to 2000 mg/kg in rats and its median lethal dose ($LD_{50}$) was considered to be over 2000 mg/kg/day body weight for both genders.

        랫드에서 쌍화탕의 급성독성에 관한 연구

        Objectives: This study was conducted to evaluate the acute toxicity and safety of Ssanghwa-tang (Shuanhetang in Chinese, Sou-wa-to in Japanese) in Crl: CD Sprague-Dawley (SD) rat though the current regulatory guideline. Methods: In this study, 10 rats of each sex were randomly assigned to two groups of 5 rats each and were administrated singly by gavage at dose levels of 0 and 2000 mg/kg/day of ssanghwa-tang water extract (SHT). After single administration of SHT, mortalities, clinical signs, body weight changes, gross findings were observed for the 15-day period. Results: Acute toxicity tests revealed that a single oral administration of SHT at dose levels of 2000 mg/kg did not affect clinical signs, body weight, and gross findings, evaluating the safety of SHT. The SHT treatment did not result in any toxicologically significant changes in mortality, clinical signs, body weight changes. Conclusions: These results showed that the single oral administration of SHT did not cause any toxic effect at the dose levels of 2000 mg/kg/day in rats. In conclusion, the median lethal dose (LD50) of SHT was considered to be over 2000 mg/kg/day body for both sexes.

        폐장 유상피 혈관내피종 1예

        Epithelioid hemangioendothelioma(EH) is a rare vascular tumor of low-grade malignancy. It was previously described as intravascular bronchioloalveolar tumor (IVBAT). But electron microscopic study and immunohistochemical staining results have proved endothelial origin. Patients are usually asymptomatic and the tumors are found incidentally on routine chest X-rays. Most patients show a slowly progressive clinical course, even though some patients occasionally die as results of respiratory failure or extrathoracic complications. Confirmative diagnosis is made through thoracoscopic or open-lung biopsy. There is still no effective therapeutic modality for pulmonary EH. We have experienced a 55-year-old woman whose simple chest x-ray film revealed bilateral multiple small nodules. The nodules were histopathologically and immunohistochemically diagnosed as primary epithelioid hemangioendothelioma of the lung. The possibility of metastatic disease was excluded thorough clinical, laboratory and radiological studies. Comparison of radiographic chest film taken 3 years ago showed no significant progression of the pulmonary nodular lesions. Conservative management was instituted and careful 4 months follow-up showed no significant changes.

      • Simultaneous determination of nine marker compounds in the traditional Korean medicine, Dangguisu-san by high-performance liquid chromatography

        <P><B>Background:</B></P><P>Dangguisu-san (DGSS) has been widely used to treat ecchymosis, blood stagnation and pain resulting from physical shock in Korea.</P><P><B>Objective:</B></P><P>A high-performance liquid chromatography–photodiode array detection (HPLC–PDA) method for simultaneous analysis of nine components, albiflorin (1), paeoniflorin (2), liquiritin (3), nodakenin (4), coumarin (5), liquiritigenin (6), cinnamic acid (7), cinnamaldehyde (8), and glycyrrhizin (9) in DGSS extract has been developed for the first time.</P><P><B>Materials and Methods:</B></P><P>The analytical column for separation of the nine constituents used a Gemini C<SUB>18</SUB> column kept at 40°C by the gradient elution with 1.0% (v/v) acetic acid in water and 1.0% (v/v) acetic acid in acetonitrile as mobile phase. The flow rate was 1.0 mL/min and the injection volume was 10 μL.</P><P><B>Results:</B></P><P>Calibration curves of all compounds showed good linearity (<I>r</I><SUP>2</SUP> ≥ 0.9999) within the test ranges. The limits of detection and quantification for all analytes were 0.01–0.27 μg/mL and 0.04–0.89 μg/mL, respectively. All recoveries of the nine marker compounds were 96.62–102.47% with relative standard deviations (RSD) <1.72%. The RSDs of intra-day and inter-day precision were <1.32% and 1.61%, respectively. The amounts of the nine marker components ranged from 0.10 mg/g to 13.71 mg/g.</P><P><B>Conclusion:</B></P><P>The developed and validated HPLC–PDA method may help for the quality control of DGSS.</P>

        Cytotoxic and DNA Topoisomerases I and II Inhibitory Constituents from the Roots of Aralia cordata

        Bioactivity-guided fractionation, based on the DNA topoisomerase inhibitory activity, lead to the isolation of five compounds (1-5) from the methylene chloride extract of the roots of Aralia cordata Thunb. (Araliaceae). These compounds were identified as ent-pimara-8(14),15-dien-19-oic acid (1), ent-pimara-8(14), 15-dien-18-oic acid (2), $16{\alpha}$-hydrogen-17-isovaleryloxy-ent-kauran-19-oic acid (3), $16{\alpha}$-hydroxy-17-isovaleryloxy-ent-kauran-19-oic acid (4) and dehydrofal-carindiol-8-acetate (5) from their spectral data. Compound 3 was isolated for the first time from this plant, and also showed the strongest inhibition of both DNA topoisomerase I and II activities, with 53 and 96% inhibitions, respectively, at a concentration of $20{\mu}M$. However, all the compounds exhibited either weak or no cytotoxicities against the human colon carcinoma cell line (HT-29), the human breast carcinoma cell line (MCF-7) and human hepato blastoma cell line (HepG-2).

        Simultaneous Determination of Albiflorin, Cinnamaldehyde, Cinnamic Acid, Daidzin, Glycyrrhizin, Liquiritin, Paeoniflorin and Puerarin in Galgeun-tang by HPLC-PDA

        Objectives: We investigated to develop and validate HPLC-PDA methods for simultaneous determination of eight constituents in Galgeun-tang (GGT). Methods: Reverse-phase chromatography using a Gemini C18 column operating at $40^{\circ}C$, and photodiode array (PDA) detection at 230 nm, 254 nm, and 280 nm, were used for quantification of the eight marker components of GGT. The mobile phase using a gradient flow consisted of two solvent systems. Solvent A was 1.0% (v/v) aqueous acetic acid and solvent B was acetonitrile with 1.0% (v/v) acetic acid. Results: Calibration curves were acquired with $r^2$ > 0.9999, and the relative standard deviation (RSD) values (%) for intra- and inter-day precision were less than 3.0%. The recovery rate of each component was in the range of 87.33-101.38%, with an RSD less than 7.0%. The contents of the eight components in GGT were 1.98-12.17 mg/g. Conclusions: The established method will be applied for the quantification of marker components in GGT.

      • Quantitative Analysis and <i>In vitro</i> Anti-inflammatory Effects of Gallic Acid, Ellagic Acid, and Quercetin from Radix Sanguisorbae

        <P><B>Background:</B></P><P>Radix Sanguisorbae has long been used to treat diarrhea, enteritis, duodenal ulcers, and internal hemorrhage.</P><P><B>Objective:</B></P><P>We investigated the <I>in vitro</I> anti-inflammatory effects of Radix Sanguisorbae and performed quantitative analyses of three marker components, namely gallic acid, ellagic acid, and quercetin, using high-performance liquid chromatography coupled with a photodiode array detector.</P><P><B>Materials and Methods:</B></P><P>The three marker components were separated using a reversed-phase Gemini C<SUB>18</SUB> analytical column maintained at 40°C by the gradient elution with two solvent systems. We examined the biological effects of the three marker compounds, gallic acid, ellagic acid, and quercetin, by determining their anti-inflammatory activities in the murine macrophage cell line RAW 264.7.</P><P><B>Results:</B></P><P>All of the marker compounds exhibited inhibitory effects on prostaglandin E<SUB>2</SUB> production in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages, with no cytotoxicity. Particularly, ellagic acid significantly inhibited production of the proinflammatory cytokines tumor necrosis factor alpha and interleukin-6 in LPS-treated RAW 264.7 cells.</P><P><B>Conclusion:</B></P><P>Our results suggest that ellagic acid is the most potent bioactive phytochemical component of radix Sanguisorbae in the treatment of inflammatory diseases.</P><P><B>SUMMARY</B></P><P><P>Established high-performance liquid chromatography method was applied in the quantitative analysis of gallic acid, ellagic acid, and quercetin present in an extract from radix Sanguisorbae</P><P>Among the three compounds, the ellagic acid.(7.65.mg/g) is main component in radix Sanguisorbae</P><P>Ellagic acid significantly inhibited production of the proinflammatory cytokines tumor necrosis factor alpha and interleukin-6 in lipopolysaccharide-treated RAW 264.7 cells.</P></P> >[FIG OMISSION]</BR><P><B>Abbreviations used:</B> HPLC: High-performance liquid chromatography, PDA: Photodiode array, TNF-α: Tumor necrosis factor alpha, IL: Interleukin, LPS: Lipopolysaccharide, PGE<SUB>2</SUB>: Prostaglandin E<SUB>2</SUB>, NSAIDs: Nonsteroidal anti-inflammatory drugs, COX: Cyclooxygenase.</P>
