      • The P275A Polymorphism in the Macrophage Scavenger Receptor 1 Gene and Prostate Cancer Risk: a Meta-Analysis

        Background: Published data regarding associations between the P275A polymorphism in the macrophage scavenger receptor 1 (MSR1) gene and prostate cancer (PCa) risk are inconclusive. The aim of this study was to comprehensively evaluate the genetic risk of P275A polymorphism in MSR1 gene for PCa. Materials and Methods: A systematic literature search was carried out in Pubmed, Medline (Ovid), Embase, CBM, CNKI, Weipu, and Wanfang databases, covering all available publications (last search was performed on Apr 27, 2015). Statistical analysis was performed using Revman 5.2 and STATA 10.1 software. Results: A total of 5,017 cases and 4,869 controls in 12 case-control studies were included in this meta-analysis. When all groups were pooled, there was no evidence that the P275A polymorphism had a significant association with PCa under dominant (OR=0.93, 95%CI=0.81-1.06, and p=0.28), co-dominant (homogeneous OR=0.97, 95%CI=0.56-1.68, and p=0.92; heterogeneous OR=0.93, 95%CI=0.74-1.15, and p=0.49), recessive (OR=1.10, 95%CI=0.65-1.87, and p=0.73), over-dominant (OR=0.93, 95%CI=0.75-1.15, and p=0.50), and allelic (OR=0.95, 95%CI=0.77-1.16, and p=0.61) genetic models. For stratified analyses by ethnicity and study design, no significant associations were found in the white race, the yellow race, the black race and mixed ethnicity, and the population-based case-control (PCC) and hospital-based case-control (HCC) studies under all genetic models. Conclusions: Based on our meta-analysis, the P275A polymorphism in the MSR1 gene is unlikely to be a risk factor for PCa.

        D-RADA16-RGD-Reinforced Nano-Hydroxyapatite/Polyamide 66 Ternary Biomaterial for Bone Formation

        BACKGROUND: Nano-hydroxyapatite/polyamide 66 (nHA/PA66) is a composite used widely in the repair of bone defects. However, this material is insufficient bioactivity. In contrast, D-RADA16-RGD self-assembling peptide (DRADA16- RGD sequence containing all D-amino acids is Ac-RADARADARADARADARGDS-CONH2) shows admirable bioactivity for both cell culture and bone regeneration. Here, we describe the fabrication of a favorable biomaterial material (nHA/PA66/D-RADA16-RGD). METHODS: Proteinase K and circular dichroism spectroscopy were employed to test the stability and secondary structural properties of peptide D-RADA16-RGD respectively. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were used to characterize the surface of these materials. Confocal laser scanning (CLS), cell counting kit-8 tests (CCK-8), alizarin red S staining, cell immunofluorescence analysis and Western blotting were involved in vitro. Also biosafety and bioactivity of them have been evaluated in vivo. RESULTS: Proteinase K and circular dichroism spectroscopy demonstrated that D-RADA16-RGD in nHA/PA66 was able to form stable-sheet secondary structure. SEM and TEM showed that the D-RADA16-RGD material was 7–33 nm in width and 130–600 nm in length, and the interwoven pore size ranged from 40 to 200 nm. CLS suggests that cells in nHA/PA66/ D-RADA16-RGD group were linked to adjacent cells with more actin filaments. CCK-8 analysis showed that nHA/PA66/ D-RADA16-RGD revealed good biocompatibility. The results of Alizarin-red S staining and Western blotting as well as vivo osteogenesis suggest nHA/PA66/D-RADA16-RGD exhibits better bioactivity. CONCLUSION: This study demonstrates that our nHA/PA66/D-RADA16-RGD composite exhibits reasonable mechanical properties, biocompatibility and bioactivity with promotion of bone formation.

        Clinical and Pathologic Features of Multifocal and Multicentric Breast Cancer in Chinese Women: A Retrospective Cohort Study

        Purpose: This study aims to analyze the clinical-pathological characteristics of multifocal and multicentric breast cancer (MMBC) in Chinese women. Methods: Sixty-seven cases with MMBC were randomly collected and reviewed at seven hospitals in representative districts of China during 1999 to 2008. Results: The incidence of MMBC in breast cancer in China was 1.75%. Compared to those with unifocal breast cancer, women with MMBC were more likely to have larger tumor size, lymph node metastasis (59.70% vs. 45.62%) and stage III to IV (46.26% vs. 21.10%). The peak age at onset of MMBC was 40 to 49 years old and has been gradually increasing during 1999 to 2008. Most of the MMBC women were treated with surgery and adjuvant therapy. Conclusion: In China, the incidence of MMBC in breast cancer is significantly lower than that in Western countries. Compared to unifocal breast cancer, MMBC is biologically more aggressive. Most MMBC women underwent mastectomy, instead of breast conservation surgery.

        Effect of Laser Power on Hybrid Laser-Gas Metal Arc Welding (GMAW) of a 6061 Aluminum Alloy

        The effect of laser power on the geometrical characteristics, microstructure and micro-hardness of the welding joints for 6061 aluminum alloy after hybrid laser-gas metal arc welding (GMAW) was investigated. The results showed that the welding joints from "Bottom" to "Top" were mainly composed of planar crystals, columnar crystals, and equiaxed dendrites. With increasing laser power, the weld depth and width and the grain size increased. When the laser power reached 5 kW, pores could be found in the weld pool region. Micro-hardness measurements showed that the application of higher laser power hardly changed the hardness of the welding joint.

        Pea starch increases the dry matter flow at the distal ileum and reduces the amino acids digestibility in ileal digesta collected after 4 hours postprandial of pigs fed low-protein diets

        Objective: The study was aimed to investigate the rules of postprandial changes in intestine digesta dry matter (DM) flow and amino acid digestibility of growing pigs fed low-protein (LP) diets made of different starch. Methods: Eight barrows (28.8±2.1 kg) with a T-cannula at the distal ileum were randomly allotted to an 8×3 Youden square design. Treatments included: waxy corn starch LP (WLP); corn starch LP (CLP) and pea starch LP (PLP). Diets were given at 08:00 and 20:00. Digesta samples were collected in six 2-h stages from 08:00 to 20:00. Results: The Cr concentrations of ileal digesta increased and then decreased in WLP and CLP, while increased continuously in PLP as time passed after postprandial (p<0.05). Higher average Cr concentrations (0.78% and 0.84% vs 0.70%; p<0.05) and lower average DM flow (181.1 g/kg and 166.3 g/kg vs 240.3 g/kg; p<0.001) were observed in WLP and CLP, compared with PLP. The apparent ileal digestibility coefficient of most amino acids in WLP and CLP increased compared with that in PLP. No difference in lysine or methionine digestibility was observed. When digesta were collected in 2-h periods, the apparent ileal digestibility coefficient of amino acids did not change over time. When digesta was collected in 4-h periods from 16:00 to 20:00 and 6-h periods from 14:00 to 20:00 (p<0.05), WLP and CLP showed markedly higher amino acid digestibility than PLP Conclusion: High-amylose slowly digested starch can increase the DM flow at the distal ileum and reduce the apparent ileal digestibility coefficient of amino acids of pigs fed LP diets. Compared with waxy corn starch and corn starch, pea starch reduced the digestibility of amino acids in digesta collected after 4 h postprandial.         Fabrication technology for light field reconstruction in glasses-free 3D display

        Light field 3D display has attracted great attention as the hardware interface for a virtual 3D application. The design and fabrication of light-modulating structures directly affect the authenticity of the reconstructed 3D image. The critical challenges in 3D display, such as visual fatigue, limited motion parallax, and insufficient light efficiency, are closely related to the inaccurate phase or angular reconstruction of the virtual light field. While the designs of light field modulators are widely introduced in all kinds of studies, the importance of fabrication for 3D display is usually under-estimated. Large format, small feature size, and high precision in shape are the three critical requirements of fabrication for 3D display. In this paper, we focus on the fabrication technologies for light-modulating structures. Fabrication tools for both microstructures and nanostructures are introduced. The fabrication capability, critical challenges and its applications in light field 3D display are discussed. Finally, the future development of 3D display related fabrication technologies are highlighted.

        Effects of Moist Extruded Full-fat Soybeans on Gut Morphology and Mucosal Cell Turnover Time of Weanling Pigs

        Ten barrows, weaned at 28 days (7.2$\pm$0.1 kg BW), were used to evaluate the effects of feeding extruded full-fat soybeans on intestinal morphology and mucosal cell turnover time. All pigs were fed corn-based diets with half of the pigs receiving diets supplemented with 15.5% soybean meal and 3% soybean oil and the remaining pigs fed a diet in which the soybean meal and oil were replaced by 18.5% extruded full-fat soybeans. The pigs were individually placed in $80{\times}150cm$ metabolic cages and fed twice daily an amount approximately equal to their ad libitum intake for a period of 14 days. On day 14, pigs were weighed and then injected intraperitoneally with $^3$H]thymidine ($100{\mu}Ci/kg$ of BW, specific activity 20 Ci/mmol) 6 h after the morning meal. A pig from each treatment was killed 1, 4, 8, 16, or 24 h postinjection and intestinal tissues were collected. Daily gains for pigs fed the soybean diet and extruded full-fat soybean diet were 0.24 and 0.31 kg/day (p=0.05) with feed conversions of 1.58 and 1.39 (p=0.05), respectively. In comparison with pigs fed soybean meal, pigs fed moist extruded full-fat soybeans had a decreased crypt depth in their duodenum and cecum (p<0.1), while the villus height in the mid jejunum and ileum and the total height (villus height plus crypt depth) of the ileum and mid jejunum increased (p<0.05). The villus width in the duodenum and mid jejunum decreased (p<0.05). The number of crypt epithelial cells in the upper jejunum increased but decreased in the ileum, colon and cecum (p<0.05). The number of villus epithelial cells in the ileum and the upper and mid jejunum increased (p<0.05). The time for migration of epithelial cells in the crypt-villus column decreased (p<0.05) in all sites except the upper jejunum, ileum and cecum. The mucosal turnover rate for all intestinal sites except the upper jejunum, colon and cecum decreased (p<0.05). From these data, we conclude that inclusion of moist extruded full-fat soybeans in weanling pig diets can improve the intestinal morphology and slow the migration rate and turnover time of epithelial cells of the small intestine, especially in the mid jejunum compared with soybean meal.

      • Toll-like Receptor 5 Agonist Inhibition of Growth of A549 Lung Cancer Cells in Vivo in a Myd88 Dependent Manner

        The purpose of this study was to examine the effect of a Toll-like receptor 5 (TLR5) agonist, CBLB502, on the growth and radiosensitivity of A549 lung cancer cells in vivo. Expression of myeloid differentiation factor 88 (MyD88) or TLR5 was stably knocked down in human lung cancer cells (A549) using lentivirus expressing short hairpin RNA targeting human MyD88 or TLR5. Lack of MyD88 or TLR5 expression enhanced tumor growth in mouse xenografts of A549 lung cancer cells. CBLB502 inhibited the growth of A549 lung cancer cells, not A549-MyD88-KD cells in vivo in the murine xenograft model. Our results showed that the inhibition of A549 by CBLB502 in vivo was realized through regulating the expression of neutrophil recruiting cytokines and neutrophil infiltration. Finally, we found that activation of TLR5 signaling did not affect the radiosensitivity of tumors in vivo.

      • Simultaneous Estimation of Low- and High-Order Functional Connectivity for Identifying Mild Cognitive Impairment

        <P>Functional connectivity (FC) network has been becoming an increasingly useful tool for understanding the cerebral working mechanism and mining sensitive biomarkers for neural/mental disease diagnosis. Currently, Pearson's Correlation (PC) is the simplest and most commonly used scheme in FC estimation. Despite its empirical effectiveness, PC only encodes the low-order (i.e., second-order) statistics by calculating the pairwise correlations between network nodes (brain regions), which fails to capture the high-order information involved in FC (e.g., the correlations among different edges in a network). To address this issue, we propose a novel FC estimation method based on Matrix Variate Normal Distribution (MVND), which can capture both low- and high-order correlations simultaneously with a clear mathematical interpretability. Specifically, we first generate a set of BOLD subseries by the sliding window scheme, and for each subseries we construct a temporal FC network by PC. Then, we employ the constructed FC networks as samples to estimate the final low- and high-order FC networks by maximizing the likelihood of MVND. To illustrate the effectiveness of the proposed method, we conduct experiments to identify subjects with Mild Cognitive Impairment (MCI) from Normal Controls (NCs). Experimental results show that the fusion of low- and high-order FCs can generally help to improve the final classification performance, even though the high-order FC may contain less discriminative information than its low-order counterpart. Importantly, the proposed method for simultaneous estimation of low- and high-order FCs can achieve better classification performance than the two baseline methods, i.e., the original PC method and a recent high-order FC estimation method.</P>

        Conceptual configuration and seismic performance of high-rise steel braced frame

        Conceptual configuration and seismic performance of high-rise steel frame-brace structure are studied. First, the topology optimization problem of minimum volume based on truss-like material model under earthquake action is presented, which is solved by full-stress method. Further, conceptual configurations of 20-storey and 40-storey steel frame-brace structure are formed. Next, the 40-storeystructure model is developed in Opensees. Two common configurations are utilized for comparison. Last, seismic performance of 40-storey structure is derived using nonlinear static analysis and nonlinear dynamic analysis. Results indicate that structural lateral stiffness and maximum roof displacement can be improved using brace. Meanwhile seismic damage can also be decreased. Moreover, frame-brace structure using topology optimization is most favorable to enhance lateral stiffness and mitigate seismic damage. Thus, topology optimization is an available way to form initial conceptual configuration in high-rise steel frame-brace structure.
