Objective: In the present study, we aimed to investigate the effects of enzymolysis fermentation of Chinese herbal medicines (CHMs) on egg production performance, egg quality, lipid metabolism, serum reproductive hormone levels, and the mRNA expression of the ovarian hormone receptor of laying hens in the late-laying stage. Methods: A total of 360 Hy-Line Brown laying hens (age, 390 days) were randomly categorized into four groups. Hens in the control (C) group were fed a basic diet devoid of CHMs, the crushed CHM (CT), fermented CHM (FC), and enzymatically fermented CHM (EFT) groups received diets containing 2% crushed CHM, 2% fermented CHM, and 2% enzymatically fermented CHM, respectively. Results: Compared with crushed CHM, the acid detergent fiber, total flavonoids, and total saponins contents of fermented CHM showed improvement (p<0.05); furthermore, the neutral and acid detergent fiber, total flavonoids, and total saponins contents of enzymatically fermented CHM improved (p<0.05). At 5 to 8 weeks, hens in the FC and EFT groups showed increased laying rates, haugh unit, albumin height, yolk color, shell thickness, and shell strength compared with those in the C group (p<0.05). Compared with the FC group, the laying rate, albumin height, and Shell thickness in the EFT group was increased (p<0.05). Compared with the C, CT, and FC groups, the EFT group showed reduced serum total cholesterol and increased serum luteinizing hormone levels and mRNA expressions of follicle stimulating hormone receptor and luteinizing hormone receptor (p<0.05). Conclusion: These results indicated that the ETF group improved the laying rate and egg quality and regulated the lipid metabolism in aged hens. The mechanism underlying this effect was likely related to cell wall degradation of CHM and increased serum levels of luteinizing hormone and mRNA expression of the ovarian hormone receptor.
Expression and characterization of RNA-dependent RNA polymerase of Ectropis obliqua virus

Replication of positive-strand RNA virus is mediated by a virus- encoded RNA-dependent RNA polymerase (RdRp). To study the replication of Ectropis obliqua virus (EoV), a newly identified insect virus belonging to the family Iflaviradae, we expressed the RNA polymerase domain in Escherichia coli and purified it on a Ni-chelating HisTrap affinity column. It is demonstrated that EoV RdRp initiated RNA synthesis in a primer- and poly (A)-dependent manner in vitro. Furthermore, the effect of primer concentration, temperature, metal ions (Mg2+, Mn2+, and K+) on enzymatic activity were determined. Cytotoxic anthraquinone dimers from Melandrium firmum

Two new anthraquinone dimers, melrubiellin A(1) and melrubiellin B (2), were isolated from the aerialpart of Melandrium firmum Rohrbach, along with sevenknown compounds (3–9). The structures of these compoundswere elucidated by spectral analyses, including 1Dand 2D NMR (COSY, HMQC, HMBC and NOESY)experiments. Compound 1 and 2 exhibited significantcytotoxicity towards HeLa, NCI-H460, Hep G2, Hep 3Band MKN-28 cell lines with IC50 values ranging from 5.26to 81.16 lM.
Evaluation of the anti-Toxoplasma gondii Activity of Hederagenin in vitro and in vivo

Toxoplasma gondii infection is widespread worldwide, not only posing a serious threat to human food safety and animal husbandry, but also endangering human health. The selectivity index was employed to measure anti-T. gondii activity. Hederagenin (HE) exhibited potent anti-T. gondii activity and low cytotoxicity. For this reason, HE was selected for in vivo experiments. HE showed 64.8%±13.1% inhibition for peritoneal tachyzoites in mice, higher than spiramycin 56.8%±6.0%. Biochemical parameters such as alanine aminotransferase, aspartate aminotransferase, glutathione, and malondialdehyde, illustrated that HE was a good inhibitor of T. gondii in vivo. This compound was also effective in relieving T. gondii-induced liver damage. Collectively, it was demonstrated that HE had potential as an anti-T. gondii agent.

Background: Sorafenib is a promising drug for advanced hepatocellular carcinoma (HCC); however, treatment may be discontinued for multiple reasons, such as progressive disease, adverse events, or the cost of treatment. The consequences of sorafenib discontinuation and continuation are uncertain. Materials and Methods: We retrospectively analyzed 88 HCC patients treated with sorafenib from July 2007 to January 2013. Overall survival (OS), post-disease progression overall survival (pOS), and time to disease progression (TTP) were compared for survival analysis. Cox proportional hazard regression was performed to assess the effect of important factors on OS in the overall patient population and on pOS in patients who continued sorafenib treatment. Results: Sorafenib was discontinued and continued in 24 and 64 patients, respectively. The median OS (355 vs 517 days respectively; p=0.015) and median post-PD OS (260 vs 317 days, respectively; p=0.020) were statistically different between the discontinuation and continuation groups. Neither the median time to first PD nor the time to second PD were significantly different between the 2 groups. In the discontinuation group, 3 of the 24 patients (12.5%) suffered disease outbreaks. In Cox proportional hazard regression analysis after correction for confounding factors, BCLC stage (p=0.002) and PD site (p=0.024) were significantly correlated with pOS in patients who continued sorafenib treatment. Conclusions: Sorafenib discontinuation may cause HCC flares or outbreaks. It is advisable to continue sorafenib treatment after first PD, particularly in patients with Barcelona Clinic Liver Cancer stage B disease or only intrahepatic PD.

The present study was designed to isolate and characterize novel chemical constituents of the stem bark of Juglans mandshurica Maxim. (Juglandaceae).The chemical constituents were isolated and purified by various chromatographic techniques. The structures of the compounds were elucidated on the basis of spectral data (1D and 2D NMR, HR-ESI-MS, CD, UV, and IR) and by the comparisons of spectroscopic data with the reported values in the literatures. Two long chain polyunsaturated fatty acids (1 and 2) were obtained and identified as (S)-(8E, 10E)-12-hydroxy-7-oxo-8, 10-octadecadienoic acid (1) and (S)-(8E, 10E)-12-hydroxy-7-oxo-8,10-octadecadienoic acid methyl ester (2). To the best of our knowledge, this is the first report on the isolation and structural elucidation of the two new conjugated ketonic fatty acids from this genus.

In this paper, bandwidth enhanced design of dielectric resonator antenna fabricated in multi-layer substrate is introduced. The proposed dielectric resonator antenna is operating with fundamental TE101 mode and higher-order TM111 mode. Each resonance frequency is dependent on resonator dimensions. As increasing the height of radiating aperture, the higher-order TM111 mode resonance frequency approach the fundamental TE101 mode resonance frequency and the antenna bandwidth increase by double resonance. Three different aperture height size antennas that operated at 7㎓ are fabricated in FR4 multi-layer substrate. Measured 10 ㏈ matching bandwidth is 8 percent for single resonace antenna and 18 percent for double resonance antenna.
Nickel‑based N/S‑dual doped graphene/carbon nanotubes electrocatalyst for oxygen evolution

Hydrogen energy is a promising source of renewable and clean energy for various industries, such as chemical, automobile, and energy industries. Electrolysis of water is one of the basic methods for the production of hydrogen energy. However, the high overpotential of the oxygen evolution reaction (OER) in water electrolysis has hindered the effective production of hydrogen using this method. Thus, the development of high-efficiency non-precious metal-based electrocatalysts for OER is extremely significant. In this study, we adopted a one-step hydrothermal method to fabricate Ni-based catalysts with N/Sdual doped graphene oxide/carbon nanotube (GO/CNT) supports using thiourea ( CH4N2S) and urea as the S source and the N source. It was observed that the amount of thiourea utilized in the synthesis of the catalyst affected the morphology, composition, and the electrochemical properties of the catalyst. For a GO/CNT-to-thiourea mass ratio of 1:10, the catalyst exhibited the highest activity, where the OER overpotential was 320 mV at a current density of 10 mA/cm2. This was attributed to the high specific surface area, high conductivity, and fast electron transport channels of the N/S-dual doped GO/ CNT composite. Furthermore, sulfurization of the Ni particles to form nickel sulfide played a significant role in enhancing the catalytic performance.
