        Illumina-sequencing based transcriptome study of coat color phenotypes in domestic goats

        This study performed a comprehensive expression profiling of genes expressed in the skin of goats with three different coat colors by Illumina Sequencing. A total of 91 significantly expressed genes were detected when comparing gray skin to white skin library and these included 74 up-regulated and 17 down-regulated genes in gray skin. There were 67 differentially expressed genes between brown skin and white skin libraries, 23 of which were up-regulated and 44 were down-regulated in brown skin. When we compared brown and gray libraries, 154 differentially expressed genes were found, of which 33 showed higher expression and 121 showed lower expression in brown skin. To our surprise, MC1R, MITF, TYR and KIT showed no significant difference in expression between the goats with three skin colors, whereas ASIP was detected in white skin but not in dark skins. In this study, PMEL, TRPM1, TYRP1 and DCT were significantly upregulated in brown goat skin compare with gray and white skins. PMEL showed higher expression in gray goat skin compared with white goat skin, whereas there were no significant differences in the expression of TYRP1, TRPM1 and DCT between gray and white skin samples. In addition, ELOVL3 showed higher expression in gray goat skin than in brown and white skins, whereas there was no significant differences in the expression of ELOVL3 between brown and white skin samples. These results expand our understanding of the complex molecular mechanisms of skin physiology and melanogenesis in goat and provide a foundation for future studies.

        Hyphae and organic acids of Aspergillus Niger promote uranium recovery by destroying the ore surface and increasing the porosity and permeability of ores

        To elucidate the mechanism of damage caused by hyphae and organic acids produced by Aspergillus Niger on the surface and internal structure of uranium ore, direct uranium leaching, indirect uranium leaching and semidirect uranium leaching were conducted, and the surface morphology, strength, mineral crystallinity, porosity, and permeability of the ore were analyzed. The results demonstrated that the combination of biomechanical forces exerted by hyphae and the complexation effects of organic acids led to the dissolution of SiO2 and other substances on the surface of ore, resulting in exfoliation from the exterior to the interior, thereby promoting uranium recovery. Furthermore, the proton exchange involving H+ and the complexation of organic acids resulted in the dissolution of cations within the ore, causing destruction to the crystal lattice structure of minerals and increasing the porosity and permeability inside the ore. The dominant factor contributing to ore damage during recovery was organic acids.

        Novel Position Controller for PMSM Based on State Feedback and Load Torque Feed-Forward

        In this paper, a novel position controller based on state feedback and feed-forward is proposed. Traditional position and speed controllers are replaced by a single controller with the position and speed as state feedbacks, and the position command and load torque as feed-forwards. The feedback and feed-forward gains are obtained by analytic modeling and design. The load torque, rotor speed and position are estimated by an observer based on a Kalman filter (KF) with a low resolution mechanical position sensor. Feed-forward compensation by an estimated load torque is used to improve the dynamic performance during load torque changes.

      • Three-level Inverter Direct Torque Control of Induction Motor Based on Virtual Vectors

        Multilevel inverter has attracted great interest in high-voltage high-power field because of its less distorted output. In this paper, a direct torque control (DTC) technique based on a three-level neutral-point-clamped (NPC) inverter is presented. In order to solve the intrinsic neutral-point voltage-balancing problem and to obtain a high performance DTC, a special vector selection method is introduced and the concept of virtual vector is developed. By using the proposed PWM strategy, a MRAS speed sensor-less DTC drive can be achieved without sensing the neutral-point voltage. The strategy can be verified by simulation and experimental results.<br/>

        Hair follicles transcriptome profiles in Bashang long-tailed chickens with different plumage colors

        Despite the rich variety in plumage color found in nature, genetic studies on how hair follicles affect pigmentation are often limited to animals that have black and white pigment. To test how gene expression influences plumage color, transcriptomes of chicken hair follicles with white, black, hemp, reed catkins, silvery grey, and landscape plumage colors were generated using Illumina sequencing. We generated six RNA-Seq libraries with over 25 million paired-end clean reads per library with percentage of paired-end clean reads ranging from 96.73 to 96.98%. 78% of the reads mapped to the chicken genome, and approximately 70% of the reads were mapped to exons and 6% mapped to introns. Transcriptomes of hair follicles producing hemp and land plumage were similar, but these two showed moderate differences compared with gray and reed colored plumage. The black and white follicle transcriptomes were most divergent from the other colors. We identified several candidate genes, including GPNMB, PMEL, TYRP1, GPR143, OCA2, SOX10, SLC45A2, KRT75, and TYR . All of these genes are known to induce pigment formation in mice. White feathers result from the lack of pigment formation, and our results suggest that the white chickens due to the recessive insertion mutation of TYR . The formation of black area size and color depth may be due to the expression levels of GPNMB, PMEL, TYRP1, GPR143, OCA2, SOX10, SLC45A2, KRT75, and TYR . The GO analysis of the differentially expressed genes (DEGs) revealed that DEGs in our transcriptome analysis were enriched in cytoskeleton and cell structure related pathways. The black plumage transcriptome showed significant differences in melanogenesis, tyrosine metabolism, and riboflavin metabolism compared with transcriptomes of other plumage colors. The transcriptome profiles of the different chicken plumage colors provide a valuable resource to understand how gene expression influences plumage color, and will be an important resource for identifying candidate genes in breeding programs.

      • Research on Model of Free Piston Linear Generator based on Location Domain and Real-Time Motion Control Strategy

        Free Piston Linear Generator (FPLG) is an ideal power resource in Hybrid Electric Vehicle (HEV) for its potential advantages of low friction loss, variable compression ratio, being suitable for various fuels, high thermal efficiency, etc. No FPLG has been successfully applied to an actual vehicle as lack of reliable control. The first part of this paper consists of introduction to FPLG and its research status. Structure and working principle of FPLG are analyzed in the second part. Then the thermodynamic process is analyzed and mathematical models of all kinds of FPLG using real-time motion control strategy based on location domain are established. After these, verifying work of the six different models established before and the realtime control strategy is carried out. Results show that the mathematical models of FPLG based on location domain work well and the control strategy effects with proper linear generator.

        Complete mitochondrial genome of the Pleuronichthys lighti (Pleuronectiformes, Pleuronectidae) with phylogenetic consideration

        Mitochondrial genome has been used to shed light on many fields of both basic and applied research, including the study of molecular evolution. The complete mitochondrial genome sequence of 17368 bp nucleotides from the Pleuronichthys lighti was determined. It was a circular double-stranded DNA molecule with identical set of 22 transfer RNA genes, 2 ribosomal RNA genes, 13 proteincoding genes as well as a non-coding control region. Stand asymmetry in the nucleotide composition was reflected in the codon usage of genes oriented in opposite directions. In the control region, we identified the extended termination associated sequence domain, the central conserved sequence block domain and the conserved sequence block domain, and two complete repeat region. They were “TTA CAA TA” and “TGT TGT AA”, respectively. All known 12 mitochondrial genomes of Pleuronectinae fishes were downloaded and analyzed; there were 5570 variable sites in the consensus sequences of 15241 base pairs, calculation of total sites were 35.5%. The highest sequence divergence was 50% (ATP8) and the Kimura-2-parameter genetic distance was 0.235 (ND6), whereas the COIII had the lowest sequence divergence (28.8%) and genetic distance (0.128); the protein coding genes were mainly acted by purifying selection which was detected by selection tests. Analysis of confidence and the information content for per nucleotide revealed ND5, ATP6, COI and ND4 genes were suitable molecular markers for phylogenetic study of Pleuronectinae fishes. Phylogenetic analysis using Bayesian computational algorithms based on COI genes provided support for the taxonomic status of P. lighti, which was consistent with the traditional taxonomy.

      • A High Step-up Modular Isolated DC-DC Converter for Large Capacity Photovoltaic Generation System integrated into MVDC Grids

        Nowadays, the research and design of the DCDC conversion system has become more and more popular in the application of photovoltaic generation into the medium voltage DC (MVDC) grids. A novel high power modular isolated DC-DC converter with high step-up ratio is proposed as a module of the DC-DC conversion system in this paper. Based on the input parallel and output series (IPOS) connections of the proposed modular converters, the system is easy to achieve high capacity and high voltage. A two-stage structure is employed in the proposed converter. The interleaved boost topology is applied to the front-stage, which realizes MPPT for PV arrays with low input current ripples. The full-bridge LLC topology is employed as the back-stage, which lowers power losses by the implementation of softswitching. The voltage gain of the proposed converter is derived in detail. The theoretical analysis and control strategy are studied in depth. Simulation and experiment verify the feasibility and effectiveness of the proposed converter.

        A Medium-Voltage Matrix Converter Topology for Wind Power Conversion with Medium Frequency Transformers

        A new type of topology with medium-frequency-transformer (MFT) isolation for medium voltage wind power generation systems is proposed in this paper. This type of converter is a high density power conversion system, with high performance features suitable for next generation wind power systems in either on-shore or off-shore applications. The proposed topology employs single-phase cascaded multi-level AC-AC converters on the grid side and three phase matrix converters on the generator side, which are interfaced by medium frequency transformers. This avoids DC-Link electrolytic capacitors and/or resonant L-C components in the power flow path thereby improving the power density and system reliability. Several configurations are given to fit different applications. The modulation and control strategy has been detailed. As two important part of the whole system, a novel single phase AC-AC converter topology with its reliable six-step switching technique and a novel symmetrical 11-segment modulation strategy for two stage matrix converter (TSMC) is proposed at the special situation of medium frequency chopping. The validity of the proposed concept has been verified by simulation results and experiment waveforms from a scaled down laboratory prototype.
