Polyphosphate가 연조직 창상 치유에 미치는 영향에 관한 연구

Polyphosphate has been used as a food additive. It has been approved the use of PolyP to the products as a safe additive. In other study, PolyP seems to has an ability of promoting effect on bone regeneration, possibly by enhancing osteoconductivity of the carrier and/or by increasing osteoinductivity of the damaged bone tissue. So we performed this study in order to evaluate the effect of polyphosphate on the dermal defect healing in critical-sized defect wound. Using welder and blade, a 1㎝ diameter sized burn-defect and rectangular excisional-defect was prepared in the thigh of each Sprague-Dawley rats. Then defect areas were applied topically with distilled water(DW), 0.01% orthoposphate(Pi), or 0.01% polyP75. The application of these materials was continued for seven days. The rats, which had burn wounds, were sacrificed at week 1 and 2. the other goups, which had excisional wounds, were sacrificed at day 3, week 1, 2, and 4. the specimens of the defect were prepared, stained with H&E, and examined under the microscope. Dermal defect healing capacity of polyP75 was also examined by observaing cellular activity of periodontal cells using MTT assay. The results were as follows. PolyP75 at the concentrations of 0.01% increased the MTT activity of periodontal cells by 154.1%. a pathohistologic findings of burn wound with polyP75 showed a faster epidermal healing and more profound maturation in dermis than those of other groups. In excisional wound, compare with other groups, tissue maturity and density of matrix in dermis were much enhanced in the polyP75 groups.

Objectives : Cinnamomi Ramulus (CR), the young twing of Cinnamomi loureirii nees, has been used for treating symptoms related to pain. rheumatic arthritis and inflammation in Korean herb medicine. This study was carried out to investigate the anti-inflammatory effect of CR in vivo and in vitro. Methods : Extracts of CR were prepared and the chemical components of the extracts were examined by gas chromatography-mass spectrometry (GC-MS). The extracts were administrated to the rat paw edema model induced by carrageenan to evaluate the anti-inflammatory effect of CR. The expressions of nitric oxide (NO). prostaglandin E2(PGE2), inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 were also quantified in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages to survey the effect of CR in vitro. The main components were cinnamaldechyde and coumarin. Results : We examined the anti-inflammatory activity of the 80% ethanol extract of Cinnamomi Ramulus in vivo by using carrageenan-induced rat paw edema model. Maximum inhibition of 54.9% was noted at the dose of 1000㎎/㎏ after 2 hours of drug administration in carrageenan-induced rat paw edema and this showed a potent anti-inflammatory effect. Conclusions : The results showed that Cinnamomi Ramulus suppressed dose-dependently LPS-induced NO production in RAW 264.7 macrophages and also decreased iNOS protein expression. Cinnamomi Ramulus also showed a significant inhibitory effect in LPS-induced PGE2 production and COX-2 expression.

Objective : The use of herbal therapy is becoming an increasingly attractive approach for the treatment of various inflammatory disorders. The Alpiniae officinari Rhizoma is popular in Aisa as a traditional herbal medicine. Alpiniae officinari Rhizoma is a species of the ginger family(Zingiberacease). Method : This study was performed to investigate the anti-inflammatory effect of Alpiniae officinari Rhizoma extract by the methods of "carrageenan induced paw edema" and "Lipopolysaccharide-induced inflammatory mediators in mouse macrophage RAW 264.7 cells". Result : We suggest that Alpiniae officinari Rhizoma extract decreased paw volume induced by plantar injection of carrageenan. Also Alpiniae officinari Rhizoma extract inhibited nitric oxide, prostaglandin E₂production and induced nitric oxide synthase, cyclooxygenase-2 protein expression in Mouse macrophage RAW 264.7 cells stimulated with lipopolysaccharide. Conclusion : This study shows that Alpiniae officinari Rhizoma extract seems to have anti-inflammatory effect by inhibition of nitric oxide, prostaglandin E_(2) production and nitric oxide synthase, cyclooxygenase-2 protein expression.

In this study, we made investigation into water permeation, solute separation, concentration and Na^(+), K^(+), Mg^(2+), Ca^(2+), Cl^(-), SO^(2)_(4)-, HCO^(-)_(3) ions separation of seawater by the revers osmosis process using a suitable semipermeable membrane. The different thickness of membranes were prepared. On heat treatment, temperature was 80℃ and thier effects were also investigated. The flat single module system was made to be capable of treating feed solution at 200-2000cc/min and 120 atm. The experimental results can be summerized as follows: 1) As the effective operating pressure increased, permeability of pure water proportionally increased. 2) As the exposure period of the manufactured membrane in air increased, permeation rate increased, but as the thickness of the membrane increased, permeation rate decreased. 3) The capacity for being reproductive of the manufactured membrane (CA: 30wt%, Formamide: 45wt%, Acetone: 25wt%) in their performance was less than 10-30% and agreed with that reported in the literature. 4) We could separated about 85-95% of salt ion in seawater using a flat single module system.

Background: Diabetes is characterized by chronic hyperglycemia, which can increase reactive oxygen species (ROS) production by the mitochondrial electron transport chain. The formation of ROS induces oxidative stress and activates oxidative damage-inducing genes in cells. No research has been published on oxidative damage-related extracellular superoxide dismutase (EC-SOD) protein levels in human diabetic skin. We investigated the expression of EC-SOD in diabetic skin compared with normal skin tissue in vivo. Objectives: To compare the expression of EC-SOD in normal skin versus diabetic skin. Methods: The expression of EC-SOD protein was evaluated by western blotting in 6 diabetic skin tissue samples and 6 normal skin samples. Immunohistochemical staining was also carried out to confirm the EC-SOD expression level in the 6 diabetic skin tissue samples. Results: The western blotting showed significantly lower EC-SOD protein expression in the diabetic skin tissue than in the normal tissue. Immunohistochemical examination of EC-SOD protein expression supported the western blotting analysis. Conclusion: Diabetic skin tissues express a relatively small amount of EC-SOD protein and may not be protected against oxidative stress. We believe that EC-SOD is related to the altered metabolic state in diabetic skin, which elevates ROS production.
An arthroscopic safety zone for the medial compartment of the hip joint.

<P>The purpose of this study was to investigate the safety zone without any neurovascular injury to the medial compartment of hip joint through an anatomical method and describe the relationship of the extra-articular anatomic structures from the surface of the hip joint. Thirty-two fresh specimens from 17 adult Korean cadavers (8 males and 9 females, age range 54-79?years at death) were used for this study. For the measurements, the most superolateral point of the pubic symphysis (PS) and prominent point of the anterior superior iliac spine (ASIS) were identified before dissection. The line connecting the PS and ASIS was defined as a reference line and the PS was a starting point for measurements. All 19 variables measured in this study were related to the femoral head, neck, and surrounding neurovascular structures. The variables were measured according to the x- and y-coordinates in relation to the reference line. The femoral head was generally located 39.5-71.0?mm on the x-coordinate and located 33.5-34.6?mm on the y-coordinate. The junction of the femoral neck and body was located at 52.8?mm on the x-coordinate, and 65.3?mm on the y-coordinate. The junction of the femoral head and neck was located at 47.1?mm on the x-coordinate, and 51.4?mm on the y-coordinate. The location of the medial compartment of the hip joint was located from 38.0 to 43.0?% on the x-coordinate and located from 5.1 to 6.5?cm. These results of this study provide detailed anatomy for arthroscopic hip surgeons.</P>
