In this paper, an optimal semi-continuous process for vinegar production from edible alcohol through biotransformation by acetic acid bacteria (AAB) WUST-01 was developed. The optimized medium composition for the starting-up stage was glucose 5.1 g/L, yeast extract 26.2 g/L, and ethanol 11.9 mL/L, and the optimal ethanol for the following semi-continuous stage was 50 mL/L. In the semi-continuous biotransformation process, the optimal withdraw ratio was 50% of working volume with 12 h cycle time. With these conditions, the total acidity could reach to 77.3 g/L and the acidity productivity could reach to 3.0 g/(L h) in a 5 L reactor. Furthermore, it was investigated to strengthen vinegar synthesis through enhancing alcohol dehydrogenase and aldehyde dehydrogenase activity in AAB by ferrous ion and pueraria flower extract as the enzyme regulators. With these regulators, the vinegar synthesis efficiency can be improved 16.3 and 13.2% respectively.

The agricultural residuals buckwheat hulls in the region of Jiaodong, China (BHJC) were employed to adsorb Au(III) ions both in the spiked samples and the gold-plating wastewater samples, and the relevant adsorption kinetics and isotherms were investigated. The investigation on the adsorption selectivity showed that BHJC displayed strong affinity for gold in the aqueous solutions. Furthermore, the process optimization was performed using response surface methodology (RSM), the quantitative relationship between the gold uptake and the different levels of experimental factors was used to work out optimized levels of these parameters.

A lectin gene (plectin) with a high level of expression was previously identified by comparative transcriptome analysis of Pleurotus ostreatus. In this study, we cloned a 733-bp DNA fragment from the start codon of the plectin gene. Sequence analysis showed that the plectin promoter (Plp) region contained several eukaryotic transcription factor binding motifs, such as the TATA-box, four possible CAAT-box, light responsiveness motifs and MeJA-responsiveness motifs. To determine whether the Plp promoter was a light-regulated promoter, we constructed an expression vector with the fused egfp-hph fragment under the control of the Plp promoter and transformed P. ostreatus mycelia via Agrobacterium tumefaciens. PCR and Southern blot analyses confirmed the Plpegfp- hph fragment was integrated into the chromosomal DNA of transformants. qRT-PCR, egfp visualization, and intracellular egfp determination experiments showed the Plp promoter could be a light-induced promoter that may be suitable for P. ostreatus genetic engineering. This study lays the foundation for gene homologous expression in P. ostreatus.

In this study, the influence of γ-irradiation with different dose (0, 4, 8, and 16 kGy) on chemical composition, physicochemical features and bioactivities of polysaccharides extracted from Lentinula edodes (LEP) were investigated. The carbohydrate content (from 59.47 to 70.96%), the solubility, the ⋅OH and DPPH scavenging ability of LEP increased with the increased γ-irradiation dose, while the protein content, the weight-average and number-average molecular weight of LEP were significantly decreased with the increased γ-irradiation dose. Moreover, γ-irradiation treatment caused LEP color changes and surface topography destroyed. γ-Irradiated LEP showed higher hypoglycemic activities in vitro than that of non-irradiated LEP. Moreover, γ-irradiated LEP had better proliferation promoting effects on Lactobacillus rhamnosus and L. plantarum. These results showed that γ-irradiation treatment changes the physicochemical features of LEP, thus affects its antioxidant, hypoglycemic and prebiotic properties, which suggests that γ-irradiated LEP has potential application in the pharmaceutical industries and functional foods.
EVALUATION OF THE ECONOMIC VALUE OF WORLD CULTURAL HERITAGE

This study attempts to assess the economic benefit of the Great Wall of China to tourists via contingent value method (CVM) and travel cost method (TCM). This study found that the economic benefit estimated via indirect observation data and CVM is US$12.49 and the economic benefit estimated via direct observation data and TCM is US$125.31. The recreational economic value for consumers is very high. Therefore, under the concept of sustainable resource development, the responsible agency is encouraged to enhance the core value of the Great Wall through developing its recreational value and preserving its cultural heritage.
An Efficient Multi-PCA Based On-line Monitoring Scheme for Multi-Stages Imperial Smelting Process

Imperial Smelting Process (ISP) is one of the main methods for Zinc and Lead smelting. In this paper, we firstly focus on its unique characteristics that ISP consists of many feeding cycles repeated in a similar way, and the normal process changes and multiple operating conditions would be frequently found in the process which would always lead to false alarms. To this end, we classify each feeding cycles of ISP into initial stage, transition stage, and steady stage, and use standard PCA, first order perturbation (FOP) based recursive dynamic PCA and multi-mode approach to deal with them respectively. The monitoring results using real data show that the proposed scheme can be able to eliminate false alarms and detect faults efficiently.

Tea is a widely consumed drink among elderly people in Chinese population. The consumption is growing in different ages because of the founding of many handy-tea-drink stores in Taiwan. However, determination of tea quality is still mainly based on sensory evaluation, which lacks of objectivity in science. Near infrared (NIR) spectroscopy, a rapid nondestructive inspection method, has been widely applied for evaluation of internal quality of agricultural products. Because an NIR spectrum of a mixture on first approximation is the linear addition of individual spectra of the constituents in the mixture, such a spectrum thus can be regarded as an assembly of ‘blind sources’ as the proportion of constituents in the samples remains unknown. Independent component analysis (ICA), a multiuse statistical approach originally used to implement ‘blind source separation’ in signal processing, is capable of disassembling the mixture’s signals with only a small loss of information and does not require any additional information from the source. To date, ICA has not been applied to analysis of the internal quality of tea. Therefore, the objective of the current study was to examine internal quality of tea in terms of quantitative approaches using NIR spectroscopy combined with ICA technique. The internal quality-related indices of tea, including tea polyphenols (TP), free amino acids (FAA), water, and pH value were evaluated simultaneously. The results show that ICA with NIR has the potential to be adopted as an effective method for evaluating internal quality of tea.
Experimental Behaviors of Steel Staggered-truss System Exposed to Fire under Lateral Force

This paper presents an experimental investigation on structural behaviors of a six-storey steel staggered-truss system model with a scale of 1/10 under lateral force and fire exposure. Pan fire with the fuel of methanol was used to simulate a natural fire. The vertical and lateral loads were applied to tested model using counterweights. Temperatures, deformations and strains of the structure were measured. And probable failure mode of the system was obtained. The results indicate that it is hard to predict the failure of components in fire purely according to the load design of the structure at ambient temperature, due to the complicated redistribution of the internal force and the non-uniform temperature distribution. The mid-span of the top chord and the vertical web members at the center panel at the fire storey presented an obvious deformation during the test, while the components in the truss show a good compatibility. The impact of high temperature on lateral stiffness of the structure mainly concentrated on the storey exposed to fire, but rarely on the adjacent truss and the storey not exposed to fire.
