      • Selectively crosslinked hyaluronic acid hydrogels for sustained release formulation of erythropoietin

        <P>A novel sustained release formulation of erythropoietin (EPO) was developed using hyaluronic acid (HA) hydrogels. For the preparation of HA hydrogels, adipic acid dihydrazide grafted HA (HA-ADH) was synthesized and analyzed with <SUP>1</SUP>H NMR. The degree of HA-ADH modification was about 69%. EPO was in situ encapsulated into HA-ADH hydrogels through a selective cross-linking reaction of bis(sulfosuccinimidyl) suberate (BS<SUP>3</SUP>) to hydrazide group (pK<SUB>a</SUB> = 3.0) of HA-ADH rather than to amine group (pK<SUB>a</SUB> > 9) of EPO. The denaturation of EPO during HA-ADH hydrogel synthesis was drastically reduced with decreasing pH from 7.4 to 4.8. The specific reactivity of BS<SUP>3</SUP> to hydrazide at pH = 4.8 might be due to its low pK<SUB>a</SUB> compared with that of amine. In vitro release of EPO in phosphate buffered saline at 37°C showed that EPO was released rapidly for 2 days and then slowly up to 4 days from HA-ADH hydrogels. When the hydrogels were dried at 37°C for a day, however, longer release of EPO up to 3 weeks could be demonstrated. According to in vivo release test of EPO from HA-ADH hydrogels in SD rats, elevated EPO concentration higher than 0.1 ng/mL could be maintained from 7 days up to 18 days depending on the preparation methods of HA-ADH hydrogels.       • Students’ Sense of Coherence Before and After Basic Nursing Practice at A University

        Aim: Comparing students’ sense of coherence before and after they experience Basic Nursing Practices 1 and 2. Method: Subjects: A total of 210 university students from A University in Japan, consisting of 106 first-year students who experienced Basic Nursing Practice 1 in March and 104 second-year students who experienced Basic Nursing Practice 2 in February. Survey period: From January to February 2021 Survey contents: The Japanese version of SOC-13 (Yamazaki, 2009) to determine sense of coherence (Antonovsky, 1987/2001) was conducted before and after practical training. Survey method: Self-administered questionnaires that link pre- and post-training through signs. Analysis: Conducted descriptive statistics, t-test, and Wilcoxon signed-rank test using SPSS (Ver. 27). Ethical considerations: This study was approved by the Research Ethics Committee of the researcher"s affiliation (No. 488). Results: 139 (66.2%) responses were deemed valid as they could show connectivity before and after these courses. Sense of coherence was compared by totaling the inclusive sums of comprehensibility, manageability, and meaningfulness before and after the training. The inclusive sums and total of manageability were recognized as normality so that we could analyze them using the t-test. The results revealed that both exhibited significant differences before and after the practical training at P=0.000. The total of comprehensibility and meaningfulness were not recognized as normality, so we conducted the Wilcoxon signed-rank test. The results showed significant difference for both of them at P = 0.000. Discussion: Compared to the pre-training sense of coherence score, the post-training scores were higher. The practical training led students to be self-aware of improvement in their ability to cope with stress. Conclusion: Stress coping ability can be increased through practical training.

        Development of a Two-Way Evacuation Route Database Based on Interviews Conducted with Historic Preservation Area Residents

        It has been determined that two-way evacuation routes that connect houses to evacuation sites should be developed to protect residents of historic preservation areas from future disasters. These routes are required because traditional buildings and other historic spatial elements are located close to each other. It is important to understand residents' perceptions of evacuation routes that connect their houses to temporary safe places to develop evacuation system strategies that include effective two-way evacuation routes. This paper describes a procedure used to construct a two-way evacuation route database designed to preserve two study areas that was based on an interview survey conducted with area residents. The resulting database contained residents' perceptions of evacuation routes. The database contains categorized spatial problems related to these routes that can be used for future research.

        Application of low-crystalline carbonate apatite granules in 2-stage sinus floor augmentation: a prospective clinical trial and histomorphometric evaluation

        Purpose: The purpose of this study was to elucidate the efficacy and safety of carbonate apatite (CO3Ap) granules in 2-stage sinus floor augmentation through the radiographic and histomorphometric assessment of bone biopsy specimens. Methods: Two-stage sinus floor augmentation was performed on 13 patients with a total of 17 implants. Radiographic assessment using panoramic radiographs was performed immediately after augmentation and was also performed 2 additional times, at 7±2 months and 18±2 months post-augmentation, respectively. Bone biopsy specimens taken from planned implant placement sites underwent micro-computed tomography, after which histological sections were prepared. Results: Postoperative healing of the sinus floor augmentation was uneventful in all cases. The mean preoperative residual bone height was 3.5±1.3 mm, and this was increased to 13.3±1.7 mm by augmentation with the CO3Ap granules. The mean height of the augmented site had decreased to 10.7±1.9 mm by 7±2 months after augmentation; however, implants with lengths in the range of 6.5 to 11.5 mm could still be placed. The mean height of the augmented site had decreased to 9.6±1.4 mm by 18±2 months post-augmentation. No implant failure or complications were observed. Few inflammatory cells or foreign body giant cells were observed in the bone biopsy specimens. Although there were individual differences in the amount of new bone detected, new bone was observed to be in direct contact with the CO3Ap granules in all cases, without an intermediate layer of fibrous tissue. The amounts of bone and residual CO3Ap were 33.8%±15.1% and 15.3%±11.9%, respectively. Conclusions: In this first demonstration, low-crystalline CO3Ap granules showed excellent biocompatibility, and bone biopsy showed them to be replaced with bone in humans. CO3Ap granules are a useful and safe bone substitute for two-stage sinus floor augmentation.
