        Algal bioassay 에 의한 조류생장 제한영양염류 결정

        Determination of Limiting Nutrient for Algal Growth by Algal Bioassay. Oh, Hee-Mock, Seog June Lee, Seong-Bin Kim, Mi- Kyung Park, Byung-Dae Yoon and Do-Han Kim (Environmental Microbiology Research Unit, Korea Research Institute of Bioscience and Biotechnology, Taejon 305-333, Korea and Water Resources Research Institute, Korea Water Resources Corporation, Taejon 305-390, Korea) Water quaiity and algal growth potential (AGP) using Microcystis aeruginosa were investigated from April to September 1997 in Daechung Reservoir. Secchi disk transparency was 1.1 m in August. Chlorophyll-a concentrations were higher than 10㎍/l at all samples. Total phosphorus concentration was in a range of 0.020∼0.027mg/l which is at the approximate value of 0.03 mg/l, the general critical level of eutrophication. These results indicate that Daechung Reservoir is in an eutrophic state except some temporary times and restricted sites. Phosphorus uptake rate of M. aeruginosa was higher in the water from the epilimnion than in the hypolimnion and also higher under light condition than under dark condition. Therefore, phosphorus uptake rate appears to depend on cellular phosphorous content and light condition, a meaning of endergonic process. Phosphorus was the most frequently limiting nutrient: phosphorus was limiting algal gowth in April and May, nitrogen, phosphorus and trace elements were the limiting factors for algae in August, and trace elements in September. If a fast growing alga, M, aeruginosa, is used as a test organism for assaying AGP, in vivo fluorescence instead of dry weight can be measured for algal biomass and incubation period can be reduced from traditional 14 days to 7 days.

      • 불응성 자가면역질환에서의 자가조혈모세포이식

        배경: 기존의 치료에 불응하고 예후가 불량한 자가면역질환 환자들에게 최근 고용량 면역억제 및 조혈모 세포이식이 새로운 치료방법으로 대두되고 있다. 저자들은 다발성 경화증(multiple sclerosis, MS) 및 류마티스 관절염(rheumatoid arthritis, RA) 등 2명의 자가면역질환 환자들에서 자가조혈모세포 이식을 시행하였다. 방법: 말초혈액 조혈모세포 가동화를 위하여 cyclophosphamide (4 g/㎡) 및 granulocyte colony stimulating factor (10 g/kg/day)를 투여하였고, CD34+ 세포를 분리·채집 하였다, 이식 전처치로 MS 환자에서 BEAM 및 antihymocyte globulin (ATG) (3.75 mg/kg), RA 환자에서 fludarabine (180 mg/㎡), ATG (10 mg/kg)와 busulfan (8 mg/kg)을 투여하였다. 결과: 호중구 수가 500/㎕ 이상으로 회복되는 기간은 MS 환자에서 9일, RA 환자에서 15일이었다. 혈소판이 20.000/㎕ 이상으로 회복되는 가간은 RA 환자에서 9일 이었고, MS 환자에서는 혈소판 감소증이 발생하지 않았다. 비혈액학적 독성으로 MS 환자에서 WHO 1도의 오심 및 점막염이 관찰되었다. MS 환자는 이식 6개월 후까지 시력감소가 남아있었으나, 이식전에 관찰되던 감각이상 및 운동장애 등의 신경학적 이상 소견은 더 이상 관찰되지 않았다. RA 환자는 이식 1개월 후 관절 증상 및 검사소견의 호전을 보였다. 결론: 불응성 자가면역질환 환자에서 고용량 면역억제 및 조혈모세포이식은 적은 독성으로 높은 치료효과를 기대할수 있으며, 향후 이 시술의 임상적 의의를 규명하기 위하여 전향적이고 장기적인 연구가 필요할 것으로 사료된다. Background: High-dose immunosuppressive therapy followed by autologous hemathpoietic stem cell transplantation (HSCT) has been proposed as a new approach to treat severe, refractory autoimmune diseases. We describe two patients with refractory autoimmune diseases (one multiple sclerosis 〔MS〕and one rheumatoid arthritis〔RA〕) who underwent T-cell-depleted autologous peripheral bleed stem cell transplantation for the first time in Korea. Methods: We mobilized autologous stem cells with cyclophisphamide (4 g/㎡) and granulocyte colony-stimulating factor (10 ㎍/kg/day). Stem cells were enriched ex vivo using CD34-positive immunoselection and reinfused after high-dose chemotherapy with BEAM and antithymocyte globulin (ATG) (3.75 mg/kg) in MS, or fludarabine (180 mg/㎡), ATG (10 mg/kg) and busulfan (8 mg/kg) in RA. Results: The engraftment with an absolute nerutrophil count greater than 500㎕ occurred on day 9 in MS and 15 in RA, respectively. The time to nontransfused platelet count greater than 2.000/㎕ was 9 day in RA. MS patient did not show ant episode of thrombocytopenia. Regimen-related non-hematopoietic toxicity was minimal. For 6 months since HSCT, them patient with MS had been free from previously existed sensory and motor abnormalities except decreased visual acuity. Then patient with RA and only one tender joint and two mildly swollen joints with improvement in laboratory parameters at one month after HSCT. Conclusion: These results underscore the feasibility and potential efficacy of intensive immunosuppression followed by autologous HSCT for treatment of intractable autoimmune diseases. The durability of remission, however, remains to be clarified.

      • Digital moire 형상측정 시스템의 보정에 관한 연구

        Moire topography method isa well-known non-contacting 3-D measurement method as a fast non-contact test for three-dimension shape measuring method. Recently, it's important to study the automatic three-dimension measurement by moire topography because it is frequently applied to the reverse engineering , the medical, the entertainment fields. Three-dimension measurement using projection of moire topography is very attractive because of its high measuring speed and high sensitivity. In this paper, the classical moire method is computerized-so called digital moire. when a virtual grating pattern is projected on a surface, the captured image by the CCD camera has three-dimension information of the objects. The moire image can be obtained through a simple image processing and a reference grating pattern. and it provides similar results without physical grating pattern. digital projection moire topography turns out to be very effective for the three-dimension measurement of objects. Using different N-bucket algorithm method of digital projection moire topography is tested to measuring object with the 2-ambiguity problem. Experimental results prove that the proposed scheme is capable of finding measurement errors that decreased more by using the four-three step algorithm method instead of the same step in the phase shifting of different pitch.

        Papillary microcarcinoma in comparison with larger papillary thyroid carcinoma in BRAF<sup>V600E</sup> mutation, clinicopathological features, and immunohistochemical findings

        <B>Background.</B><P>Papillary thyroid microcarcinoma (PMC; ≤1 cm) is thought to take a benign course during the lifetime. However, recent studies showed high recurrence rates for PMC.</P><B>Methods.</B><P>We analyzed the clinicopathological features, long-term prognosis, and some molecular characteristics including BRAF<SUP>V600E</SUP> mutation by retrospectively reviewing the records of 1150 patients with papillary thyroid carcinoma (PTC), 278 with PMC, and 868 with PTC >1 cm.</P><B>Results.</B><P>The prevalence of extrathyroidal invasion (52.2%) and initial nodal metastasis (34.9%) in patients with PMC was surprisingly high and almost as high as that for patients with PTC (72.4% and 51.8%, respectively). The rate of recurrent or persistent disease did not differ between patients with PMC and PTC (recurrent or persistent disease, 6.1% vs 14.1%; 53.4- vs 84.2-month follow-up; n = 98 vs 647; corrected p = .112). The frequency of BRAF<SUP>V600E</SUP> mutation was similar in patients with PMC and PTC (65.6% vs 67.2%). Immunohistochemical staining showed no different expression pattern according to the tumor size.</P><B>Conclusion.</B><P>These results suggest that PMC is not an occult cancer and it can act like larger PTC.         High Thermal Conductive Natural Rubber Composites Using Aluminum Nitride and Boron Nitride Hybrid Fillers

        Herein, we investigated the thermal conductivity and thermal stability of natural rubber composite systems con­taining hybrid fillers of boron nitride (BN) and aluminum nitride (AlN). In the hybrid system, the bimodal distribution of polygonal AlN and planar BN particles provided excellent filler-packing efficiency and desired energy path for phonon transfer, resulting in high thermal conductivity of 1.29 W/mK, which could not be achieved by single filler composites. Fur­ther, polyethylene glycol (PEG) was compounded with a commonly used naphthenic oil, which substantially increased ther­mal conductivity to 3.51 W/mK with an excellent thermal stability due to facilitated energy transfer across the filler-filler interface. The resulting PEG-incorporated hybrid composite showed a high thermal degradation temperature (T<sub>2</sub>) of 290℃, a low coefficient of thermal expansion of 26.4 ppm/℃, and a low thermal distortion parameter of 7.53 m/K, which is well over the naphthenic oil compound. Finally, using the Fourier's law of conduction, we suggested a modeling methodology to evaluate the cooling performance in thermal management system.

        Pancreatic Cancer Induced by In Vivo Electroporation-Enhanced Sleeping Beauty Transposon Gene Delivery System in Mouse

        OBJECTIVE: The aim of this study was to establish a pancreatic tumor model of mouse using the electroporation-enhanced Sleeping Beauty (SB) transposon system. METHODS: The SB transposon system was used in conjunction with electroporation to deliver oncogenes, c-Myc and HRAS, and shRNA against p53 into the mouse pancreas to induce tumors. Oncogenes (c-Myc and HRAS) and shRNA against p53 gene were directly injected into the pancreas of the mouse along with in vivo electroporation applied on the injection site. The tumors were identified grossly and confirmed using animal positron emission tomographic imaging. The tumors were then characterized using histological and immunohistochemical techniques. The expression of the targeted genes (c-Myc, HRAS, and p53) was analyzed by a real-time quantitative polymerase chain reaction. RESULTS: Pancreatic tumors were successfully induced. The tumor phenotype was a sarcomatoid carcinoma, which was verified through immunohistochemistry. Some cysts or duct-like structures suggested to be metaplastic acinar cells were visible in the induced tumor. CONCLUSIONS: The SB transposon enhanced with electroporation can readily generate pancreatic tumors in the mice, and thus, this model serves as a valuable resource for the mouse models of pancreatic cancer.

        Intravascular ultrasound-based machine learning for predicting fractional flow reserve in intermediate coronary artery lesions

        <P><B>Abstract</B></P> <P><B>Background and aims</B></P> <P>Intravascular ultrasound (IVUS)-derived morphological criteria are poor predictors of the functional significance of intermediate coronary stenosis. IVUS-based supervised machine learning (ML) algorithms were developed to identify lesions with a fractional flow reserve (FFR) ≤0.80 (<I>vs.</I> >0.80).</P> <P><B>Methods</B></P> <P>A total of 1328 patients with 1328 non-left main coronary lesions were randomized into training and test sets in a 4:1 ratio. Masked IVUS images were generated by an automatic segmentation model, and 99 computed IVUS features and six clinical variables (age, gender, body surface area, vessel type, involved segment, and involvement of the proximal left anterior descending artery) were used for ML training with 5-fold cross-validation. Diagnostic performances of the binary classifiers (L2 penalized logistic regression, artificial neural network, random forest, AdaBoost, CatBoost, and support vector machine) for detecting ischemia-producing lesions were evaluated using the non-overlapping test samples.</P> <P><B>Results</B></P> <P>In the classification of test set lesions into those with an FFR ≤0.80 <I>vs.</I> >0.80, the overall diagnostic accuracies for predicting an FFR ≤0.80 were 82% with L2 penalized logistic regression, 80% with artificial neural network, 83% with random forest, 83% with AdaBoost, 81% with CatBoost, and 81% with support vector machine (AUCs: 0.84–0.87). With exclusion of the 28 lesions with borderline FFR of 0.75–0.80, the overall accuracies for the test set were 86% with L2 penalized logistic regression, 85% with an artificial neural network, 87% with random forest, 87% with AdaBoost, 85% with CatBoost, and 85% with support vector machine.</P> <P><B>Conclusions</B></P> <P>The IVUS-based ML algorithms showed good diagnostic performance for identifying ischemia-producing lesions, and may reduce the need for pressure wires.</P> <P><B>Highlights</B></P> <P> <UL> <LI> ML provides the ability to automatically learn without being explicitly programmed, which improves diagnostic accuracies. </LI> <LI> The models using computed IVUS features predict the intermediate stenosis with an FFR ≤0.80 with an overall accuracy of 80%. </LI> <LI> The data-driven approach may help clinicians identify ischemia-producing coronary lesions. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
