Objectives: Accumulating evidence indicates a strong link between knee osteoarthritis (KOA) and sarcopenia. However, the mechanisms involved have not yet been elucidated. This study primarily aims to explore the molecular mechanisms that explain the connection between these 2 disorders. Methods: The gene expression profiles for KOA and sarcopenia were obtained from the Gene Expression Omnibus database, specifically from GSE55235, GSE169077, and GSE1408. Various bioinformatics techniques were employed to identify and analyze common differentially expressed genes (DEGs) across the 3 datasets. The techniques involved the analysis of Gene Ontology and pathways to enhance understanding, examining proteinprotein interaction (PPI) networks, and identifying hub genes. In addition, we constructed the network of interactions between transcription factors (TFs) and genes, the co-regulatory network of TFs and miRNAs for hub genes, and predicted potential drugs. Results: In total, 14 common DEGs were found between KOA and sarcopenia. Detailed information on biological processes and signaling pathways of common DEGs was obtained through enrichment analysis. After performing PPI network analysis, we discovered 4 hub genes (FOXO3, BCL6, CDKN1A, and CEBPB). Subsequently, we developed coregulatory networks for these hub genes involving TF-gene and TF-miRNA interactions. Finally, we identified 10 potential chemical compounds. Conclusions: By conducting bioinformatics analysis, our study has successfully identified common gene interaction networks between KOA and sarcopenia. The potential of these findings to offer revolutionary understanding into the common development of these 2 conditions could lead to the identification of valuable targets for therapy. 1. Introduction Knee osteoarthritis (KOA) is a prevalent degenerative condition of the musculoskeletal system that predominantly impacts individuals in the middle-aged and older age groups. This progressive ailment gradually impairs the patient’s mobility and overall well-being [1,2]. The primary pathological feature of knee osteoarthritis (KOA) involves the breakdown and loss of articular cartilage. Different joint tissues are impacted to different extents, including remodeling of the subchondral bone, degeneration of the meniscus, weakening and looseness of ligaments, inflammation of the infrapatellar fat pad, and inflammation of the synovial membrane [2–5]. Furthermore, the presence of periarticular muscle atrophy plays a significant role in the progression of KOA. Decreased muscle strength can alter mechanical stress, reduce joint stability, and accelerate cartilage degeneration and abnormal subchondral bone changes [6–9]. Sarcopenia, which frequently coexists with KOA [10–12], is primarily characterized by the decline in both muscle mass and strengt

Objective: The purpose of this study was to investigate the clinical effects of BU pharmacopuncture therapy consisting of bear's gall(fel ursi) and ox bezoar(bovis calculus) on acute lumbar sprain. Methods: 12 patients diagnosed as acute lumbar sprain in 6 designated local Korean medicine clinics from October 2017 to February 2018 were treated by BU pharmacopuncture. Several acupoints in abdomen and lumbar region were selected by clinicians at their own discretion. The effectiveness of the therapy was evaluated using VAS and ODI. After that we reviewed the medical records of all these patients to evaluate the effectiveness and safety of the therapy. Results: The VAS and ODI scales were significantly decreased after BU pharmacopuncture therapy. And no major complications and adverse effects were reported. Conclusion: BU pharmacopuncture showed rapid pain relief in patients with acute lumbar sprain. It is possible to shorten the treatment period of acute lumbar sprain and prevent progressing to chronic back pain in advance. To establish the effects of BU pharmacopuncture therapy, more succeeding clinical and laboratory studies are needed.

Orange carotenoid protein (OCP) of photosynthetic cyanobacteria binds to ketocarotenoids noncovalently and absorbs excess light to protect the host organism from light-induced oxidative damage. Herein, we found that mutating valine 40 in the α3 helix of Gloeocapsa sp. PCC 7513 (GlOCP1) resulted in blue- or red-shifts of 6-20 nm in the absorption maxima of the lit forms. We analyzed the origins of absorption maxima shifts by integrating X-ray crystallography, homology modeling, molecular dynamics simulations, and hybrid quantum mechanics/molecular mechanics calculations. Our analysis suggested that the single residue mutations alter the polar environment surrounding the bound canthaxanthin, thereby modulating the degree of charge transfer in the photoexcited state of the chromophore. Our integrated investigations reveal the mechanism of color adaptation specific to OCPs and suggest a design principle for color-specific photoswitches.

We present an integrated scheme of 3-D surface profile measurements made at multiple sites concurrently by employing only a single fiber femtosecond laser as the common light source of low coherence scanning interferometry. This versatile use of an ultrashort mode-locked laser is enabled by linear scanning control of the pulse repetition rate on the source site, while diverse forms of unequal-path, non-symmetric measurements are taken with nanometer precision for different targets simply by delivering fr-scanned pulses through a fiber network. This proposed scheme has no restriction on the number of interferometer sites being integrated concurrently, allowing more diverse industrial applications of ultrashort lasers despite increased system cost and complexity.
Silver nanoparticle-induced degranulation observed with quantitative phase microscopy.
Yang, Wenzhong,Lee, Seungrag,Lee, Jiyong,Bae, Yoonsung,Kim, Dugyoung SPIE--the International Society for Optical Engine 2010 JOURNAL OF BIOMEDICAL OPTICS Vol.15 No.4

Monitoring a degranulation process in a live mast cell is a quite important issue in immunology and pharmacology. Because the size of a granule is normally much smaller than the resolution limit of an optical microscope system, there is no direct real-time live cell imaging technique for observing degranulation processes except for fluorescence imaging techniques. In this research, we propose optical quantitative phase microscopy (QPM) as a new observation tool to study degranulation processes in a live mast cell without any fluorescence labeling. We measure the cell volumes and the cross sectional profiles (x-z plane) of an RBL-2H3 cell and a HeLa cell, before and after they are exposed to calcium ionophore A23187 and silver nanoparticles (AgNPs). We verify that the volume and the cross sectional line profile of the RBL-2H3 cell were changed significantly when it was exposed to A23187. When 50 microg/mL of AgNP is used instead of A23187, the measurements of cell volume and cross sectional profiles indicate that RBL-2H3 cells also follow degranulation processes. Degranulation processes for these cells are verified by monitoring the increase of intracellular calcium ([Ca(2+)](i)) and histamine with fluorescent methods.

It has been found that both the number of paralleled grid-connected converters and the fi lter inductance value will aff ect the system’s stability in weak grid condition. However, a quantitative study on this eff ect is missing in the current literature. The main contribution of this paper is to quantitatively evaluates the infl uence of the number of paralleled grid-connected converters as well as the inductance value of the L fi lters on the system’s stable region. It is fi rst revealed that, when the same controller design principles are applied for L -type grid-connected converters, the stable region of the paralleled grid-connected converter system is dependent on the ratio between the fi lter inductances and the number of paralleled converters. Specifi - cally, the stable region is proportional to the ratio. Compared to the qualitative analysis in previous work, this fi nding can provide quantitative guidelines on the fi lter inductors or control system design for grid-connected converter applications in weak grid. Finally, experimental tests are carried out using two paralleled grid-connected converters to confi rm the fi ndings.

A single layer retarder possessing negative dispersion (ND) of birefringence as well as wide field-of-view (FOV) was long-term objective in optical science. We synthesized new guest reactive monomers with x-shape and mixed them with the host smectic reactive mesogen. The host-guest molecules formed two dimensionally self-organized nanostructure and showed both the ND of birefringence and wide FOV properties. We simulated the antireflection property of a circular polarizer using the optical properties of the retarder. The average reflectance of the retarder was 0.52% which was much smaller than that of the commercial single layer ND retarder 1.83%. (C) 2016 Optical Society of America

Hybrid and electric vehicles adopt electric motor driven system using rechargeable battery compared to conventional vehicles. These vehicles are eco- friendly les because of no fuel consumption. These chargeable vehicles using alternative power train driven by electric motor need high efficient thermal management system in battery system in high temperature status on going charging and discharging operation . To maintain proper temperature status in battery system, BCS(Battery Cooling System) is adopted. BCS is composed of BLDC motor, inverter, sirocco type wheel and BLWR. Unit and the operating noise of BCS located under the trunk makes NVH problem in vehicle. In this paper, FEA(Finite Element Analysis) and experiment are performed for analyzing NVH characteristics of BCS , verify the noise source of BCS and propose the noise reduced design for BCS NVH problem.
