        Effect of Urolithin A on Bone Repair in Mice with Bone Defects

        BACKGROUND: Bone defect difficult to manage clinically and it is a big challenge to repair it. Secondary metabolites source from herb has shown potential for the treatment of bone defect. METHODS: Mesenchymal stem cells (MSCs) were isolated from mice and incubated with urolithin A (UA) (10, 25, and 50 lg/mL). 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay was performed to estimate apoptosis and mineralisation was evaluated by alkaline phosphatase assay and alizarin red S staining. A middle femoral defect was induced in mice and bone tissue was prepared for endochondral ossification by treating with UA. The effect of UA was estimated by determining markers of osteoblast proliferation in serum and micro-computed tomography to analyse bone defects. RESULTS: UA enhanced mineralisation of MSCs and osteogenic gene markers in MSCs in vitro. Also, the bone defect score and bone mineral density were improved by UA. Moreover, UA ameliorated the altered Wnt3a protein and histopathological changes in bone defect mice. CONCLUSION: Presented report conclude that UA enhances osteoblast proliferation in bone-defect mice by activating the Wnt pathway.

        Probabilistic Risk Assessment of Unsaturated Slope Failure Considering Spatial Variability of Hydraulic Parameters

        Probabilistic risk assessment of slope failure evaluates the slope safety in a quantitative manner, which considers the failure probability and failure consequence simultaneously. However, risk assessment of unsaturated slope accounting for spatially variable soil-water characteristic curve (SWCC) model parameter and saturated hydraulic conductivity has been rarely reported. A probabilistic risk assessment approach is proposed in current study for rationally quantifying the unsaturated slope failure risk with the aid of Monte Carlo (MC) simulation. The SEEP/W and SLOPE/W modules contained in Geostudio software are applied to carry out deterministic analysis, where factor of safety (FS) of the unsaturated slope is calculated by Morgenstern–Price method. The spatially variable hydraulic parameters are characterized by their respective random fields that are transferred from the random void ratio field in this study, rather than generating them separately. The proposed approach is subsequently employed to an unsaturated slope example for exploring the influences of spatially variable void ratio. Results show that the unsaturated slope failure risk is considerably affected by the spatially variable void ratio, and the single exponential autocorrelation function (ACF) popularized in geotechnical engineering tends to underestimate the failure risk in the unsaturated slope risk assessment.

        Exploring molecular mechanisms underlying the pathophysiological association between knee osteoarthritis and sarcopenia

        Objectives: Accumulating evidence indicates a strong link between knee osteoarthritis (KOA) and sarcopenia. However, the mechanisms involved have not yet been elucidated. This study primarily aims to explore the molecular mechanisms that explain the connection between these 2 disorders. Methods: The gene expression profiles for KOA and sarcopenia were obtained from the Gene Expression Omnibus database, specifically from GSE55235, GSE169077, and GSE1408. Various bioinformatics techniques were employed to identify and analyze common differentially expressed genes (DEGs) across the 3 datasets. The techniques involved the analysis of Gene Ontology and pathways to enhance understanding, examining proteinprotein interaction (PPI) networks, and identifying hub genes. In addition, we constructed the network of interactions between transcription factors (TFs) and genes, the co-regulatory network of TFs and miRNAs for hub genes, and predicted potential drugs. Results: In total, 14 common DEGs were found between KOA and sarcopenia. Detailed information on biological processes and signaling pathways of common DEGs was obtained through enrichment analysis. After performing PPI network analysis, we discovered 4 hub genes (FOXO3, BCL6, CDKN1A, and CEBPB). Subsequently, we developed coregulatory networks for these hub genes involving TF-gene and TF-miRNA interactions. Finally, we identified 10 potential chemical compounds. Conclusions: By conducting bioinformatics analysis, our study has successfully identified common gene interaction networks between KOA and sarcopenia. The potential of these findings to offer revolutionary understanding into the common development of these 2 conditions could lead to the identification of valuable targets for therapy. 1. Introduction Knee osteoarthritis (KOA) is a prevalent degenerative condition of the musculoskeletal system that predominantly impacts individuals in the middle-aged and older age groups. This progressive ailment gradually impairs the patient’s mobility and overall well-being [1,2]. The primary pathological feature of knee osteoarthritis (KOA) involves the breakdown and loss of articular cartilage. Different joint tissues are impacted to different extents, including remodeling of the subchondral bone, degeneration of the meniscus, weakening and looseness of ligaments, inflammation of the infrapatellar fat pad, and inflammation of the synovial membrane [2–5]. Furthermore, the presence of periarticular muscle atrophy plays a significant role in the progression of KOA. Decreased muscle strength can alter mechanical stress, reduce joint stability, and accelerate cartilage degeneration and abnormal subchondral bone changes [6–9].
