        Sesquiterpenoids from Farfugium japonicum and Their Inhibitory Activity on NO Production in RAW264.7 Cells

        A new eremophilane sesquiterpenoid, namely, 3β-angeloyloxy-6β,8β-dihydroxy-9β-senecioyloxyeremophil-7(11)-en-12,8α-lactone, along with eight known sesquiterpenoids, was isolated from the rhizome of Farfugium japonicum. The structures of all isolates were identified based on analyses of spectroscopic data (HRESIMS, IR, 1D, and 2D NMR) and comparison with literature data. The inhibitory effects of compounds 1-4 on nitric oxide production in lipopolysaccaride-activated mouse macrophages were also evaluated.

        Production of Transgenic Pigs with an Introduced Missense Mutation of the Bone Morphogenetic Protein Receptor Type IB Gene Related to Prolificacy

        In the last few decades, transgenic animal technology has witnessed an increasingly wide application in animal breeding. Reproductive traits are economically important to the pig industry. It has been shown that the bone morphogenetic protein receptor type IB (BMPR1B) A746G polymorphism is responsible for the fertility in sheep. However, this causal mutation exits exclusively in sheep and goat. In this study, we attempted to create transgenic pigs by introducing this mutation with the aim to improve reproductive traits in pigs. We successfully constructed a vector containing porcine BMPR1B coding sequence (CDS) with the mutant G allele of A746G mutation. In total, we obtained 24 cloned male piglets using handmade cloning (HMC) technique, and 12 individuals survived till maturation. A set of polymerase chain reactions indicated that 11 of 12 matured boars were transgene-positive individuals, and that the transgenic vector was most likely disrupted during cloning. Of 11 positive pigs, one (No. 11) lost a part of the terminator region but had the intact promoter and the CDS regions. cDNA sequencing showed that the introduced allele (746G) was expressed in multiple tissues of transgene-positive offspring of No.11. Western blot analysis revealed that BMPR1B protein expression in multiple tissues of transgene-positive $F_1$ piglets was 0.5 to 2-fold higher than that in the transgene-negative siblings. The No. 11 boar showed normal litter size performance as normal pigs from the same breed. Transgene-positive $F_1$ boars produced by No. 11 had higher semen volume, sperm concentration and total sperm per ejaculate than the negative siblings, although the differences did not reached statistical significance. Transgene-positive $F_1$ sows had similar litter size performance to the negative siblings, and more data are needed to adequately assess the litter size performance. In conclusion, we obtained 24 cloned transgenic pigs with the modified porcine BMPR1B CDS using HMC. cDNA sequencing and western blot indicated that the exogenous BMPR1B CDS was successfully expressed in host pigs. The transgenic pigs showed normal litter size performance. However, no significant differences in litter size were found between transgene-positive and negative sows. Our study provides new insight into producing cloned transgenic livestock related to reproductive traits.

      • Prognostic Value of CD44 Variant exon 6 Expression in Non-Small Cell Lung Cancer: a Meta-analysis

        Background: CD44v6 (CD44 variant exon 6) is the chief CD44 variant isoform regulating tumor invasion, progression, and metastasis. The prognostic value of CD44v6 expression in non small cell lung cancer (NSCLC) has been evaluated in many studies, but the results have remained controversial. Thus, we performed a meta-analysis of currently available studies to investigate the prognostic value of CD44v6 expression in NSCLC patients and the relationship between the expression of CD44v6 and clinicopathological features. Materials and Methods: Two independent reviewers searched the relevant literature in Pubmed, Medline and Embase from 1946 to January 2014. Overall survival (OS) and various clinicopathological features were collected from included studies. This meta-analysis was accomplished using STATA 12.0 and Revman 5.2 software. Pooled hazard ratios (HRs) with 95% confidence intervals (95%CIs) were calculated to estimate the effects. Results: A total of 921 NSCLC patients from ten studies met the inclusion criteria. The results showed that CD44v6 high expression was a prognostic factor for poor survival (HR=1.91, 95%CI=1.12-3.26, p<0.05). With respect to clinicopathological features, CD44v6 high expression was related to histopathologic type (squamous cell carcinoma versus adenocarcinoma: OR=2.72, 95%CI=1.38-5.38, p=0.004), and lymph node metastasis (OR=3.02, 95%CI=1.93-4.72, p<0.00001). Conclusions: Our results suggested CD44v6 high expression as a poor prognostic factor for NSCLC, and CD44v6 expression is associated with lymph node metastasis and histopathologic type. Therefore, CD44v6 expression can be used as a novel prognostic marker in NSCLC cases.

        The role of discoid domain receptor 1 on renal tubular epithelial pyroptosis in diabetic nephropathy

        Pyroptosis, a form of cell death associated with inflammation, is known to be involved in diabetic nephropathy (DN), and discoid domain receptor 1 (DDR1), an inflammatory regulatory protein, is reported to be associated with diabetes. However, the mechanism underlying DDR1 regulation and pyroptosis in DN remains unknown. We aimed to investigate the effect of DDR1 on renal tubular epithelial cell pyroptosis and the mechanism underlying DN. In this study, we used high glucose (HG)-treated HK-2 cells and rats with a single intraperitoneal injection of streptozotocin as DN models. Subsequently, the expression of pyroptosis-related proteins (cleaved caspase-1, GSDMD-N, Interleukin-1β [IL-1β], and interleukin-18 [IL-18]), DDR1, phosphorylated NF-κB (p-NF-κB), and NLR family pyrin domain-containing 3 (NLRP3) inflammasomes were determined through Western blotting. IL-1β and IL-18 levels were determined using ELISA. The rate of pyroptosis was assessed by propidium iodide (PI) staining. The results revealed upregulated expression of pyroptosisrelated proteins and increased concentration of IL-1β and IL-18, accompanied by DDR1, p-NF-κB, and NLRP3 upregulation in DN rat kidney tissues and HG-treated HK-2 cells. Moreover, DDR1 knockdown in the background of HG treatment resulted in inhibited expression of pyroptosis-related proteins and attenuation of IL-1β and IL-18 production and PI-positive cell frequency via the NF-κB/NLRP3 pathway in HK-2 cells. However, NLRP3 overexpression reversed the effect of DDR1 knockdown on pyroptosis. In conclusion, we demonstrated that DDR1 may be associated with pyroptosis, and DDR1 knockdown inhibited HG-induced renal tubular epithelial cell pyroptosis. The NF-κB/NLRP3 pathway is probably involved in the underlying mechanism of these findings.

        Influence of ultrasonic impact treatment on microstructure and mechanical properties of nickel-based alloy overlayer on austenitic stainless steel pipe butt girth joint

        Ultrasonic impact treatment (UIT) is carried out on the Ni-based alloy stainless steel pipe gas tungsten arc welding (GTAW) girth weld, the differences of microstructure, microhardness and shear strength distribution of the joint before and after ultrasonic shock are studied by microhardness test and shear punch test. The results show that after UIT, the plastic deformation layer is formed on the outside surface of the Ni-based alloy overlayer, single-phase austenite and g type precipitates are formed in the overlayer, and a large number of columnar crystals are formed on the bottom side of the overlayer. The average microhardness of the overlayer increased from 221 H V to 254 H V by 14.9%, the shear strength increased from 696 MPa to 882 MPa with an increase of 26.7% and the transverse average residual stress decreased from 102.71 MPa (tensile stress) to 18.33 MPa (compressive stress), the longitudinal average residual stress decreased from 114.87 MPa (tensile stress) to 84.64 MPa (compressive stress). The fracture surface has been appeared obvious shear lip marks and a few dimples. The element migrates at the fusion boundary between the Ni-based alloy overlayer and the austenitic stainless steel joint, which is leaded to form a local martensite zone and appear hot cracks. The welded joint is cooled by FA solidification mode, which is forming a large number of late and skeleton ferrite phase with an average microhardness of 190 H V and no obvious change in shear strength. The base metal is all austenitic phase with an average microhardness of 206 H V and shear strength of 696 MPa

        Prediction Model of Austenite Growth and the Role of MnS Inclusions in Non- Quenched and Tempered Steel

        The austenite growth behavior of non-quenched and tempered steels (casted by continuous casting and moldingcasting processes) was studied. The austenite grain size of steel B casted by continuous casting process is smallerthan that of steel A casted by molding casting process at the same heating parameters. The abnormal austenitegrowth temperature of the steels A and B are 950 °C and 1000 °C, respectively. Based on the results, the models forthe austenite grain growth below and above the abnormal austenite growth temperature of the investigated steelswere established. The dispersedly distributed fine particles MnS in steel B is the key factor refining the austenitegrain by pinning the migration of austenite grain boundary. The elongated inclusions MnS are ineffective in preventingthe austenite grain growth at high heating temperature. For the non-quenched and tempered steel, the continuouscasting process should be adopted and the inclusion MnS should be elliptical, smaller in size anddistributed uniformly in order to refine the final microstructure and also improve the mechanical properties.

        Investigation on microstructure and mechanical properties of ni-based alloy overlayer by ultrasonic impact treatment

        Ultrasonic impact treatment (UIT) was performed on the Ni-based alloy overlayer of austenitic stainless steel pipe girth weld. The difference of microstructure, microhardness and maximum shear strength distribution of the joint before and after UIT was studied by autobiographic test and shear punch test. The electrode potential data were tested by a threeelectrode electrochemical workstation and tafel curve was drawn. The results show that a grain refinement layer of about 30 um and a transition layer of about 60 um are formed in the overlayer of nickel-based alloy after UIT. Meanwhile, the transition layer has high corrosion sensitivity to oxalic acid solution, and the main precipitates in the overlayer are austenite and M23C6. In 3.5 % NaCl solution, the self-corrosion potential is -432 mV. The average microhardness of the overlayer of nickel-based alloy is 270 HV, which is 18.9 % higher than that of the sample without UIT. In the meantime, the maximum shear strength of nickel-based alloy is 955 MPa, which is 9.6 % higher than the sample without UIT. It is found that the ferrite is mainly lath and skeleton with no obvious change under UIT or not. Meanwhile, an average microhardness is 191 HV, and shear strength is 678 MPa. The microstructure of the welded joint is mainly consisted of austenite phase and ferrite phase, and FA solidification mode has been a major influence in the welding process. At the boundary between overlayer and circumferential welded joint of 304 austenitic stainless steel, reheat cracks in this area are occurred easily. Due to the migration of carbon, local martensite bands will be formed which will lead to large heterogeneity of microhardness.

        Efficient Production of Diltiazem Chiral Intermediate using Immobilized Lipase from Serratia marcescens

        The lipase from Serratia marcescence ECU1010(Sml) was capable of enantioselectively catalyzing the synthesis of many chiral drug precursors. This paper investigated the immobilization of Sml on appropriate supporting materials and its performance in bioreactor. Chitosan, Celite 545, and DEAE-cellulose were found to be the ideal supports among 8 carriers tested with respect to enzyme load and activity recovery of lipase. When Sml was immobilized, significant improvements of stability against pH, thermal, and operational deactivation were observed with all the 3 better supports, and the best stability was observed when the lipase was immobilized on glutaraldehyde activated chitosan. As for the effect of organic solvent in the biphasic reaction system, the hydrolytic activity of the immobilized lipase on trans-3-(4'-methoxyphenyl)glycidic acid methyl ester ((±)-MPGM)observed in isopropyl ether was 6 and 3 times higher than those in toluene and methyl tert-butyl ether. The lipasecatalyzed production of (−)-MPGM by enzymatic resolution of (±)-MPGM with chitosan-Sml in isopropyl etherwater biphasic system was carried out in a 2 L stirred-tank reactor. The batch operation was more efficient operation mode for the enantioselective hydrolysis of (±)-MPGM,affording enantiopure (−)-MPGM in 44.3% overall yield,in contrast to 29.3% in a continuous reactor.
