A new eremophilane sesquiterpenoid, namely, 3β-angeloyloxy-6β,8β-dihydroxy-9β-senecioyloxyeremophil-7(11)-en-12,8α-lactone, along with eight known sesquiterpenoids, was isolated from the rhizome of Farfugium japonicum. The structures of all isolates were identified based on analyses of spectroscopic data (HRESIMS, IR, 1D, and 2D NMR) and comparison with literature data. The inhibitory effects of compounds 1-4 on nitric oxide production in lipopolysaccaride-activated mouse macrophages were also evaluated.

Load forecasting has always been essential to the operation and planning of power systems in deregulated electricity markets. Various methods have been proposed for load forecasting, and the neural network is one of the most widely accepted and used techniques. However, to obtain more accurate results, more information is needed as input variables, resulting in huge computational costs in the learning process. In this paper, to reduce training time in multi-layer perceptron-based short-term load forecasting, a graphics processing unit (GPU)-based computing method is introduced. The proposed approach is tested using the Korea electricity market historical demand data set. Results show that GPU-based computing greatly reduces computational costs.

In the last few decades, transgenic animal technology has witnessed an increasingly wide application in animal breeding. Reproductive traits are economically important to the pig industry. It has been shown that the bone morphogenetic protein receptor type IB (BMPR1B) A746G polymorphism is responsible for the fertility in sheep. However, this causal mutation exits exclusively in sheep and goat. In this study, we attempted to create transgenic pigs by introducing this mutation with the aim to improve reproductive traits in pigs. We successfully constructed a vector containing porcine BMPR1B coding sequence (CDS) with the mutant G allele of A746G mutation. In total, we obtained 24 cloned male piglets using handmade cloning (HMC) technique, and 12 individuals survived till maturation. A set of polymerase chain reactions indicated that 11 of 12 matured boars were transgene-positive individuals, and that the transgenic vector was most likely disrupted during cloning. Of 11 positive pigs, one (No. 11) lost a part of the terminator region but had the intact promoter and the CDS regions. cDNA sequencing showed that the introduced allele (746G) was expressed in multiple tissues of transgene-positive offspring of No.11. Western blot analysis revealed that BMPR1B protein expression in multiple tissues of transgene-positive $F_1$ piglets was 0.5 to 2-fold higher than that in the transgene-negative siblings. The No. 11 boar showed normal litter size performance as normal pigs from the same breed. Transgene-positive $F_1$ boars produced by No. 11 had higher semen volume, sperm concentration and total sperm per ejaculate than the negative siblings, although the differences did not reached statistical significance. Transgene-positive $F_1$ sows had similar litter size performance to the negative siblings, and more data are needed to adequately assess the litter size performance. In conclusion, we obtained 24 cloned transgenic pigs with the modified porcine BMPR1B CDS using HMC. cDNA sequencing and western blot indicated that the exogenous BMPR1B CDS was successfully expressed in host pigs. The transgenic pigs showed normal litter size performance. However, no significant differences in litter size were found between transgene-positive and negative sows. Our study provides new insight into producing cloned transgenic livestock related to reproductive traits.

This paper proposes a constrained optimization-based torque control allocation method aimed to improve energy efficiency, and thus, driving range for electric vehicles. In the proposed method, the cost function is defined not only to achieve desired yaw moment for vehicle handling and stability, but also to minimize power losses for energy efficiency. The particular attention is paid to the power losses due to tire slips both longitudinally and laterally. The constraints are also set based on thorough investigation on various causes of power disppation such that the torque is allocated with restraint to use regenerative braking in its maximum capacity. The proposed control allocation method has been tested and verified to be effective on energy efficiency improvement through both simulation and experiment under various driving maneuvers.

<P>UV-induced cell pigmentation represents an important mechanism against skin cancers. Sun-exposed skin secretes alpha-MSH, which induces the lineage-specific transcriptional factor MITF and activates melanogenesis in melanocytes. Here, we show that the autophagic tumor suppressor UVRAG plays an integral role in melanogenesis by interaction with the biogenesis of lysosome-related organelles complex 1 (BLOC-1). This interaction is required for BLOC-1 stability and for BLOC-1-mediated cargo sorting and delivery to melanosomes. Absence of UVRAG dispersed BLOC-1 distribution and activity, resulting in impaired melanogenesis in vitro and defective melanocyte development in zebrafish in vivo. Furthermore, our results establish UVRAG as an important effector for melanocytes' response to alpha-MSH signaling as a direct target of MITF and reveal the molecular basis underlying the association between oncogenic BRAF and compromised UV protection in melanoma.</P>

Objective: Substituting starch with digestible fiber (dF) can improve digestive health of rabbits and reduce costs. Therefore, it is necessary to develop a criterion for dF and starch supply. Effects of the dietary dF-to-starch ratio on pellet quality, growth and cecal microbiota of Angora rabbits were evaluated. Methods: Five isoenergetic and isoproteic diets with increasing dF/starch ratios (0.59, 0.66, 0.71, 1.05, and 1.44) were formulated. A total of 120 Angora rabbits with an average live weight of 2.19 kg were randomly divided into five groups with four replicates. At the end of 40 day feeding trial, cecal digesta were collected to analyse microbiota. Results: The results showed that the dF/starch ratio had linear effects on pellet variables (p<0.01). When the dF/starch ratio was 1.44, the pellets had the lowest powder and highest durability. The dF/starch ratio had unfavorable linear effects on growth variables (p<0.001). When analyzed by quadratic regression, the optimal dF/starch ratios for average weight gain and feed/gain were 0.59 and 0.74, respectively. There were differences in wool yield, fiber length and fiber diameter caused by the dF/starch ratio (p<0.05), and the dF/starch ratios that ranged from 0.66 to 1.06 were appropriate for good results. The cecal microbiota operational taxonomic unit (OTU) number index in the 1.05 dF/starch treatment was higher than that in the 0.66 and 0.71 dF/starch treatments. The higher dF/starch ratio resulted in a higher cecal microbiota OTU number index (p<0.05). The proportion of Ruminococcus in the 0.71 dF/starch treatment was higher than that in the 0.59 dF/starch treatment (p<0.05) Conclusion: The most suitable dF/starch ratio for feed pellet quality is 1.44, and for rabbit growth the optimal range of ratios is from 0.59 to 0.74. With combination of the wool growth, output cost, and cecal microbiota, we suggest that a dietary dF/starch ratio ranging from 0.74 to 1.06 is optimal.
Compact tri‐wideband bandpass filter with multiple transmission zeros

This paper presents a tri‐wideband bandpass filter (TWB‐BPF) with compact size, high band‐to‐band isolation, and multiple transmission zeros (TZs). The proposed TWB‐BPF is based on a multiple‐mode resonator (MMR), which is interpreted by the method of the even‐ and odd‐mode analysis technique. The MMR can excite 11 resonant modes, where the first two modes comprise the first passband, the next four modes form the second passband, and the last five modes are used to generate the third passband. In addition, 10 TZs are yielded to obtain high band‐to‐band isolation and wide stopband suppression characteristics up to 14.95fc1 (fc1 is the center frequency of the first passband). To verify the proposed filter, a TWB‐BPF with 3‐dB fractional bandwidths (FBWs) of 37.4%, 43.5%, and 40.4% is designed, fabricated, and measured.
PIK3CA and AKT Gene Polymorphisms in Susceptibility to Osteosarcoma in a Chinese Population

Purpose: To explore the association between PIK3CA and AKT single nucleotide polymorphisms(SNP) and osteosarcoma susceptibility. Methods: TaqMan polymerase chain reaction(PCR) was used to detect the genotypes of SNPs (rs7646409, rs6973569 and rs9866361) in peripheral blood samples from 59 patients with osteosarcoma and from 63 healthy controls. Unconditional logistic regression was used to analyze the correlation between SNPs and osteosarcoma risk. Results: No statistically significant difference was found between osteosarcoma patients and healthy controls in the genotype of AKT rs6973569 (P=0.7). However, after stratified analysis, the genotype AA of AKT rs6973569 carried a higher risk of osteosarcoma metastasis (OR:2.94, 95%CL:1.00-8.59); the difference of rs7646409 genotype distributions between the case and control groups was statistically significant (P=0.032). Taking genotype TT as a reference, the risk of osteosarcoma increased three fold in patients with genotype CC (OR:3.47, 95%CL:1.26-9.56). A statistically significant difference was found between the alleles C and T (P=0.005). Further analysis showed that the risk factor was more pronounced in male patients with Enneking's stage IIB and osteoblastic osteosarcoma. PIK3CA rs9866361 did not fit Hardy-Weinberg equilibrium (P<0.05). Conclusions: Genotype CC in locus PIK3CA rs7646409 may increase the risk of osteosarcoma in the Chinese population.

The incorporation of microwave irradiation with the prevalent "click chemistry" is currently of considerable synthetic interest. We describe here the introduction of such laboratorial shortcut into carbohydrate-based drug discovery, resulting in the rapid formation of a series of triazole-linked salicylic $\beta$-D-O-glycosides with biological activities. All "clicked" products were achieved in excellent yields ($\approx$ 90%) within only a quarter. In addition, based on the structural characteristics of the afforded glycomimetics, their inhibitory activities were evaluated toward protein tyrosine phosphatases 1B (PTP1B) and a panel of homologous protein tyrosine phosphatases (PTPs). Docking simulation was also conducted to plausibly propose binding modes of this glycosyl salicylate series with the enzymatic target.
