Background: Previous studies accessing the association of CYP2C19 with outcomes of patients using tamoxifen for breast cancer have yielded conflicting results. The aim of this meta-analysis is to obtain a more precise estimate of effects of CYP2C19 polymorphisms and to clarify their effects on survival of the breast cancer patients using tamoxifen. Materials and Methods: A systematic search of PubMed and Embase was performed, comparing patients with or without $CYP2C19^*2$ and $CYP2C19^*17$, relevant articles searched for. The following outcomes were included from the eligible studies: disease-free survival (DFS) and overall survival (OS), expressed by hazard ratios (HR) with corresponding 95% confidence interval (CI). Subgroup analysis by genotypes was also performed. Pooled estimates were calculated using random-effect model in accordance to the heterogeneity. Results: Six studies met the inclusion criteria. The integrated OR on the association between CYP2C19 and DFS, calculated by the random-effect model, was 0.54 (95%CI=0.34-0.84, p=0.013). Subgroup analysis showed that both $CYP2C19^*2$ and $CYP2C19^*17$ were associated with increased survival. The pooled results of two studies for OS were OR=0.46 (95%CI=0.21-1.01, p=0.233). Conclusions: This meta-analysis suggests that the $CYP2C19^*2$ and $CYP2C19^*17$ genotypes are associated with increased survival in breast cancer patients using tamoxifen.

<P><B>Abstract</B></P><P><B>BACKGROUND</B></P><P><I>Pyropia yezoensis</I>, rich in porphyran, is a medicine‐edible red alga. In the present study, the physicochemical characteristics, conformational states and antitumor activities of a novel porphyran extracted from the high‐yield algal strain <I>Pyropia yezoensis</I> Chonsoo2 and its two degraded derivatives by gamma irradiation were investigated.</P><P><B>RESULTS</B></P><P><I>Pyropia yezoensis</I> porphyran is a water‐soluble, triple‐helical sulfated hetero‐galactopyranose, named PYP. PYP was degraded by gamma irradiation at 20 kGy and 50 kGy, giving two low molecular weight derivatives comprising PYP‐20 and PYP‐50, respectively. PYP with a higher molecular weight has a solution conformation different from PYP‐20 and PYP‐50. Three porphyrans had no toxicity in normal human liver cells (HL‐7702) and showed antitumor effects on Hep3B, HeLa and MDA‐MB‐231. They had better antitumor against HeLa cells, exhibiting a similar inhibition ratio compared to 5‐fluorouracil, with PYP especially exhibiting a higher inhibition ratio than 5‐fluorouracil. With respect to HeLa cells, the different antitumor activities might be related to porphyran molecular weight and solution conformation. Furthermore, the HeLa cell cycle was blocked in the G2/M phase after PYP treatment, leading to cell proliferation inhibition. The induction of cell cycle arrest was related to the changes in the expression of p21, p53, Cyclin B1 and cyclin‐dependent kinase 1.</P><P><B>CONCLUSION</B></P><P><I>Pyropia yezoensis</I> porphyran, as applied to medicine and functional food, could potentially be used as a non‐toxic natural adjuvant in cancer therapy. © 2019 Society of Chemical Industry</P>
Improved Model Predictive Control for H-bridge Cascaded STATCOM

H-bridge cascaded static synchronous compensator (STATCOM) is often used to compensate the harmonic and reactive current derived from non-linear load. In order to improve the performance of STATCOM, an improved model predictive control (MPC) method is put forward in this paper. In the proposed MPC, the difference equations which are derived from the transfer functions of controlling variables and controlled variables in STATCOM are used as the predictive control model, the error between the output of STATCOM and predictive model is used to design feedback correction controller and rolling optimization controller. Meanwhile, the introduced repetitive controller is used to eliminate the inherent steady error. The actual H-bridge cascaded STATCOM is constructed and a series of verification tests are executed. The experimental results prove that the output voltage and current of H-bridge cascaded STATCOM with the proposed MPC have smaller distortion and better sinusoidal shape than those of the traditional Proportion Integral (PI) control. Moreover, the performance of H-bridge cascaded STATCOM is great in high power experiment.

Background: The purpose of this study was to investigate Tim-3 expression on peripheral CD3-CD56+ natural killer (NK) cells and CD3+CD56+ natural killer T (NKT) cells in lung cancer patients. Materials and Methods: We analyzed Tim-3+CD3-CD56+ cells, Tim-3+CD3-$CD56^{dim}$ cells, Tim-3+CD3-$CD56^{bright}$ cells, and Tim-3+CD3+CD56+ cells in fresh peripheral blood from 79 lung cancer cases preoperatively and 53 healthy controls by flow cytometry. Postoperative blood samples were also analyzed from 21 members of the lung cancer patient cohort. Results: It was showed that expression of Tim-3 was significantly increased on CD3-CD56+ cells, CD3-$CD56^{dim}$ cells and CD3+CD56+ cells in lung cancer patients as compared to healthy controls (p=0.03, p=0.03 and p=0.04, respectively). When analyzing Tim-3 expression with cancer progression, results revealed more elevated Tim-3 expression in CD3-CD56+ cells, CD3-$CD56^{dim}$ cells and CD3+CD56+ cells in cases with advanced stages (III/IV) than those with stage I and II (p=0.02, p=0.04 and p=0.01, respectively). In addition, Tim-3 expression was significantly reduced on after surgical resection of the primary tumor (p<0.01). Conclusions: Tim-3 expression in natural killer cells from fresh peripheral blood may provide a useful indicator of disease progression of lung cancer. Furthermore, it was indicated that Tim-3 might be as a therapeutic target.

A valve-less electro-hydraulic servo steering device (short: VSSD) for ships was chosen as a study object, and its mathematic model of hydraulic shock was established on the basis of flow properties and force balance of each component. The influence of system structure parameters, changing rate of motor speed and external load on hydraulic shock strength was simulated by the method of numerical simulation. Experiment was designed to test the hydraulic shock mathematic model of VSSD. Experiment results verified the correctness of the model, and the model provided a correct theoretical method for the calculation and control of hydraulic shock of valve-less electro-hydraulic servo steering device.
Prognostic Significance of Overexpression of EZH2 and H3k27me3 Proteins in Gastric Cancer

The enhancer of zeste homolog 2 (EZH2) methyl transferase and histone 3 lysine 27 (H3K27me3) protein can repress gene transcription, and their aberrant expression has been observed in various human cancers. This study determined their expression levels in gastric cancer tissues with reference to clinicopathological features and patient survival. We collected 117 gastric cancer and corresponding normal tissues for immunohistochemistry analysis. In gastric cancers, 82/117 (70.1%) were positive for EZH2 and 66/117 (56.4%) for H3K27me3 proteins in contrast to only 5.41% and 7.25% of normal gastric mucosa specimens, respectively. Kaplan-Meier survival data showed the average overall and disease-free survival of EZH2 high expression patients was 25.2 and 20.2 months, respectively, shorter than that with EZH2 low expression (40.5 and 35.9 months). The average overall survival and disease-free survival of high H3K27me3 expression patients was 23.4 and 17.4 months, shorter than without H3K27me3 expression (37.6 and 34.5 months). The average overall survival and disease-free survival of patients with both EZH2 and H3K27me3 expression was 18.8 and 12.9 months, respectively, shorter than that with either alone (34.7 and 31.2 months) or with low levels of both (43.9 and 39.9 months). Multivariate Cox regression analysis showed that H3K27me3 and EZH2 expression, tumor size differentiation and clinical stage were all independent prognostic factors for predicting patient survival. This study demonstrated that detection of both EZH2 and H3K27me3 proteins can predict poor survival of gastric cancer patients, superior to single protein detection. In addition, H3K27me3 and EZH2 protein expression could predict lymph node metastasis.
Reconstituted Fibril from Heterogenic Collagens-A New Method to Regulate Properties of Collagen Gels

Heterotypic collagen fibril has long been found in the tissues of organisms, which plays an important role in the formation and function of complex structures of organisms. Inspiring by the phenomenon, scholars tried to incubate collagens from different sources into novel collagen materials in vitro, and the forming of heterogenic reconstituted collagen fibrils (RF) was often demonstrated by differential scanning calorimetry (DSC) and fluorescence quenching analysis. In this work, we used two type-I collagens from different species (bovine tendon and grasscarp fish skin) to coassemble in vitro, and verified the formation of RF from a new rheological perspective. In addition, we also investigated the assembly behavior, surface hydrophilicity and hydrophobicity, microscopic morphology and cell proliferation ability of the RF. The results showed that the assembly rate, surface properties, fibril size, viscoelastic properties of RF can be delicately regulated by the method of heterogenic collagen reconstitution. This study provides new experimental evidence for the reconstitution of heterogenic collagens, and also offers a new means for the regulation of collagen gel performance, which would help to expand the application range of collagen gel materials.

Three compounds, vanillic acid, p-hydroxylcinnamic acid, p-hydroxybenzaldehyde have been isolated from the ethylacetate extract of Sparganium stoloniferum Buch.-Ham roots using silica gel open column chromatography, preparative thin-layer chromatography (pTLC) and reverse phase high performance liquid chromatography. The structures of the compounds were established on the basis of IR, extensive 1D NMR, and MS analyses. The ethylacetate (EtOAc) extract, vanillic acid, and p-hydroxybenzaldehyde showed $\alpha$-glucosidase inhibition activity of 72.71%, 20.13%, and 30.42%, at the concentration of 10 ${\mu}g/mL$, respectively. The EtOAc extract exhibited strong antioxidant activity with an $IC_50$ value of 24.37 ${\mu}g/mL$ against DPPH radical scavenging activity, the vanillic acid, p-hydroxylcinnamic acid, and p-hydroxybenzaldehyde with an $IC_50$ value of 2.10 ${\mu}M$, 1.59 ${\mu}M$, and 2.72 ${\mu}M$ against DPPH, respectively.
