        Dynorphin B analogues [H-Tyr-DAla-Gly-Phe-Leu-Ala-DArg-Gln-Phe-Lys-DArg- DAla-Thr] was synthesized. All amino groups of amino acids were protected with t-Boc group and 2.6-dichlorobenzyl and nitro groups were used to protect the side chains of Tyr and Arg, respectively. stepwise synthetic method was applied for synthesis of the products. Dicyclohexylcarbodiimide (DCC) and 1-hydroxy benzotriazole (HOBT) were used as the coupling reagents. The crude peptides were purified by sephadex LH-20 column and then characterized with HPLC and amino acid analyzer.

        경도인지장애 환자에서 복합-일상활동의 제한

        Objectives : Mild cognitive impairment (MCI) is distinguished from mild dementia by an absence of global intellectual deterioration and the preservation of activities of daily living (ADL). Recently, however, it became apparent that impairment of instrumental activities of daily living (IADL) is present before the threshold of dementia is reached. Thus, we want to examine whether IADL are impaired in patients with MCI, and which items of IADL are particularly involved. We divided the MCI group into amnestic (aMCI) and non-amnestic MCI (naMCI), and compared to the cognitively nonnal controls. In this study, we focused on the instrumental activities of daily living (IADL). Methods : The sample consisted of 69 community-dwelling older adults from a welfare center for the aged in Korea. The subjects were divided into three diagnostic groups; aMCI [N=19, memory domains below -1.5 standard deviation (SD)], naMCI (N= 19, other cognitive domains below -1.5 SD, except memory domains) and cognitive normal controls (N=31). Subjects were assessed both on IADL and the cognitive function. In order to assess the IADL, we used the Seoul -Instrumental Activities of Daily Living (S-IADL). Included measures of cognitive tests are as follows ; Seoul Verbal Learning Test (SVLT), Rey Complex Figure Test (RCFT), Korean-Boston Naming Test (K-BNT), Stroop test, and Korean-Mini Mental Status Examination (K-MMSE). Groups were compared on the S-IADL and the cognitive tests. Results : The three groups did not differ in the mean age, gender distribution and years of education. S-IADL were shown to be different between the groups in this study. Suects with aMCI were significantly more impaired in S-IADL in comparison to the controls [F (2,50) = 4.251, P=0.020]. And on four items ofS-IADL (shopping, transportation, medication and talking about recent events), subjects with aMCI showed higher impairment compared to the controls. However, the S-IADL did not differ between the subjects with naMCI and controls. Conclusion : In this study, impairment of S-IADL was shown in subjects with aMCI. And S-IADL ofnaMCI was not Significantly differed from aMCI and controls. The results suggest that naMCI would be distinguished from aMCI in characteristics and prognosis.

        고상법에 의한 Dermorphin 유사체의 합성과 생물학적 활성에 관한 연구

        N/A Dermorphin is a hepta peptide(H-Tyr-DAla-Phe-Gly-Tyr-Pro-Ser-NH_2) with exceptionally potent and long-lasting peripheral and central activity. Dermorphin analogues, dermorphionyl(DMP)-Lys-NH_2 and DMP-Lys-Lys-NII_2 have been prepared in order to examine the effect of opiod activity. Dermorphin analogues were synthesized by the solid phase method. The crude peptide was purified by gel filter on a Sephadex LH-20, characterized with HPLC and amino acid analyzer. Analgesic potency was estimated by writhing syndrome method and Randall-Selitto method. As a result, dennorphin analogues have lower potency than that of morphine.

        벤조페논 유도체의 합성과 전기화학 및 분광학특성에 관한 연구

        The benzophenone derivatives(4-CH₃O-4 -NO₂ and 3,4 -di-NO₂) are synthesized by the Fridel-Craft acylation and the nitration method. Electrochemical redox potentials of the benzophenone derivatives (4-CH₃O, H, 3-Cl, 3-NO₂, 4-NO₂, 4-CH₃O-4 -NO₂, 3,4 -di-NO₂) are measured by using cyclic voltammometry. In the relationship of summing Hammett value and redox potential, we find a proportional constant(ρ) that shows a good relation with an electrochemical property and a reactivity of the benzophenone derivatives. The benzophenone substituted with the electron donating groups(4-OCH₃ and 4-OCH₃-4 -NO₂) are higher the energy in the LUMO level, then increasing a band-gap energy(Eg), their Egs are obtained as a 3.94 eV and 3.59 eV, respectively.

        주조에 의한 Core와 CAD-CAM에 의한 Core의 적합도 비교평가

        Dental CAD (computer-aided design)/CAM (computer-aided manufacturing) systems facilitate the use of zirconia core for all-ceramic crown. The purpose of this study was to evaluate the marginal and internal fit of zirconia core fabricated using a dental CAD/CAM system and to compare the fit of metal cores by a conventional method. Ten identical cases of single coping study models (abutment of teeth 11) were manufactured and scanned. Ten zirconia cores were fabricated using dental CAD/CAM system. An experienced dental technician fabricated 10 samples of metal cores for the control group using the lost wax technique. Marginal and internal fit was measured by the silicone replica technique. Fit was measured with magnification of 160 using a digital Microscope. Margin, rounded chamfer, axial wall and incisal fits were measured for comparison. T-test of independent sample for statistical analysis was executed with SPSS 12.0 for Windows (SPSS Inc., Chicago, IL, USA) (a=0.05). The mean (SD) for marginal, rounded chamfer, axial wall and incisal were: 97.0 (25.3) mm, 104.0 (22.0) mm, 59.6 (21.4) mm and 124.8 (33.3) mm for the zirconia core group, and 78.2 (18.4) mm, 83.8 (15.1) mm, 42.7 (9.6) mm and 83.4 (14.4) mm for the metal core group. T-test showed significant differences between groups for margin (p<.001), rounded chamfer (p<.001), axial wall (p<.001) and incisal (p<.001). But zirconia core group observed that the marginal and internal fit values in the present study were within clinically acceptable range.

        Canine Renal Failure Caused by Ochratoxin A and Citrinin in the Commercial Dog Food

        Five dogs with renal failure were refered to the Veterinary Medical Teaching Hospital at Kangwon Nationalfor over 1 month. The dogs showed anorexia, emaciation, vomiting, and polydipsia/polyuria. And in one severelyaffected dog, bloody diarrhea and hypothermia were seen. The remarkable clinicopathological signs were high valueof BUN and creatinine. In some dogs, GGT, phosphorus and lipase were increased. However, no significant changesof complete blood count were found. In urinalysis, hematuria, low specific gravity urine, proteinuria, and calciumoxalate-like crystals were observed. Two severely affected dogs were died. The remained dogs were recovered graduallyafter change of dog food and supportive therapy. Pathological findings were seen typically in kidneys. Renal atrophy,tion, necrosis, dystrophic calcification and regeneration inthe tubular epithelium were seen. Yellowish brown fluorolucent laminated materials or particles were quite often foundin the lumina of the necrotizing renal tubules of cortex and medulla. Proliferation of fibrous tissue in the interstitiumwas also seen. By the mycotoxin analysis of the Pedigree dry dog fod, ochratoxin A (OTA) and citrinin were detectedas much as the concentration of 372.8 ppb and 8.3 ppb, respectively. The final diagnosis of renal failure caused byOTA and citrinin toxicosis was made on the basis of history takings, clinical signs, clinicopathological and pathologicalfindings, and analysis of mycotoxins.

        Preparation and Characterization of Human Adipose Tissue-Derived Extracellular Matrix, Growth Factors, and Stem Cells: A Concise Review

        BACKGROUND: Human adipose tissue is routinely discarded as medical waste. However, this tissue may have valuable clinical applications since methods have been devised to effectively isolate adipose-derived extracellular matrix (ECM), growth factors (GFs), and stem cells. In this review, we analyze the literature that devised these methods and then suggest an optimal method based on their characterization results. METHODS: Methods that we analyze in this article include: extraction of adipose tissue, decellularization, confirmation of decellularization, identification of residual active ingredients (ECM, GFs, and cells), removal of immunogens, and comparing structural/physiological/biochemical characteristics of active ingredients. RESULTS: Human adipose ECMs are composed of collagen type I–VII, laminin, fibronectin, elastin, and glycosaminoglycan (GAG). GFs immobilized in GAG include basic fibroblast growth factor (bFGF), transforming growth factor beta 1(TGF-b1), insulin like growth factor 1 (IGF-1), vascular endothelial growth factor (VEGF), platelet-derived growth factor (PDGF), BMP4 (bone morphogenetic protein 4), nerve growth factor (NGF), hepatocyte growth factor (HGF), and epithermal growth factor (EGF). Stem cells in the stromal-vascular fraction display mesenchymal markers, self-renewal gene expression, and multi-differentiation potential. CONCLUSION: Depending on the preparation method, the volume, biological activity, and physical properties of ECM, GFs, and adipose tissue-derived cells can vary. Thus, the optimal preparation method is dependent on the intended application of the adipose tissue-derived products.
