<P>The stimulation of H3K4 trimethylation (H3K4me3) by H2B monoubiquitination (H2Bub) has been widely studied, and multiple mechanisms have been proposed for this form of histone cross-talk. Thornton et al. combine biochemical, structural, and in vivo approaches to provide a novel mechanism for the role of H2B ubiquitination machinery in the regulation of histone H3K4 methylation by COMPASS. This study demonstrates that the H2Bub machinery and Cps35/Swd2 function to focus the H3K4me3 activity of COMPASS at promoter-proximal regions in a context-dependent manner.</P>

The effect of different feeding ratios of concentrate to roughage on ruminal lipid synthesis in vitro was examined. Three sheep fitted with a rumen fistula were fed three different ratios (8:2, 4:6 and 0:10) of concentrate and roughage, and their rumen liquor were used for incubation. $^{13}C$-labeled glucose or sodium acetate as substrate was added to cultures of rumen liquor, and they were incubated for 6 h. The total lipid in the culture of the rumen liquor was extracted, and the percentage of $^{13}C$ excess was analyzed. The percentage of $^{13}C$ excess recovered when incubated with glucose increased with increased ratio of concentrate in the diet. The values of cultures incubated with glucose were higher than those incubated with sodium acetate except the roughage-only feeding. In the roughage-only diet, the percentage of $^{13}C$ excess when incubated with sodium acetate was highest of all diets. The recovery percentage of $^{13}C$ from glucose increased with increased ratio of concentrate. The recovery percentage of $^{13}C$ from sodium acetate addition in only roughage feeding was highest among the three diets. The recovery percentage of $^{13}C$ from glucose was markedly higher than that of sodium acetate addition in all feedings. The results indicate that high concentrate feeding facilitates lipid synthesis by rumen microorganisms, and that glucose may be the precursor for lipid synthesis rather than acetic acid.

In a CHS experiment, various types of transport barrier have been studied. An internal transport barrier (ITB) for electrons in a stellarator was first found in CHS, and the ITB for improved ion confinement was also found. H-mode (edge transport barrier) study in CHS showed many common features in transition to tokamak experiments, although the magnetic surface quantities are very different. Discharges with a combination of ETB and ITB were also found. Two heavy ion beam diagnostics in CHS showed a new direct measurement of zonal flow structure in toroidal confinement. The relation between the fluctuation and the transport barrier was also studied.

Skeletal myogenesis is precisely regulated by multiple transcription factors. Previously, we demonstrated that enhancer of polycomb 1 (Epc1) induces skeletal muscle differentiation by potentiating serum response factor (SRF)-dependent muscle gene activation. Here, we report that an interacting partner of Epc1, ret finger protein (RFP), blocks skeletal muscle differentiation. Our findings show that RFP was highly expressed in skeletal muscles and was downregulated during myoblast differentiation. Forced expression of RFP delayed myoblast differentiation, whereas knockdown enhanced it. Epc1-induced enhancements of SRF-dependent multinucleation, transactivation of the skeletal α-actin promoter, binding of SRF to the serum response element, and muscle-specific gene induction were blocked by RFP. RFP interfered with the physical interaction between Epc1 and SRF. Muscles from rfp knockout mice (Rfp<SUP>−/−</SUP>) mice were bigger than those from wild-type mice, and the expression of SRF-dependent muscle-specific genes was upregulated. Myotube formation and myoblast differentiation were enhanced in Rfp<SUP>−/−</SUP> mice. Taken together, our findings highlight RFP as a novel regulator of muscle differentiation that acts by modulating the expression of SRF-dependent skeletal muscle-specific genes.
PATIAL DISTRIBUTION OF STAR FORMATION ACTIVITY ON NGC 253 BY FIR AND RADIO EMISSION LINES

The aim of this research is to reveal the spatial distribution of the star formation activity of nearby galaxies by comparing CO molecular emission lines with the large area observation in far-infrared (FIR) lines. We report the imaging observations of NGC 253 by FIR forbidden lines via FIS-FTS and CO molecular lines from low to high excitation levels with ASTE, which are good tracers of star forming regions or photo-dissociation regions, especially spiral galaxies, in order to derive the information of the physical conditions of the ambient interstellar radiation fields. The combination of spatially resolved FIR and sub-mm data leads to the star formation efficiency within galaxy. The ratio between the FIR luminosity and molecular gas mass, $L_{FIR}/M_{H_2}$, is expected to be proportional to the number of stars formed in the galaxy per unit molecular gas mass and time. Moreover the FIR line ux shows current star formation activity directly. Furthermore these can be systematic and statistical data for star formation history and evolution of spiral galaxies.

We present the results of far-infrared spectroscopic observations of the Large Magellanic Cloud (LMC) with FIS-FTS. We covered a large area across the LMC, including 30 Doradus (30 Dor) and N44 star-forming regions, by 191 pointings in total. As a result, we detect the [OIII] and [CII] line emission as well as far-infrared dust continuum emission throughout the LMC. We find that the [OIII] emission is widely distributed around 30 Dor. The observed size of the distribution is too large to be explained by massive stars in 30 Dor, which are assumed to be enshrouded by clouds with the constant gas density estimated from the [OIII] line intensities. Therefore the surrounding structure is likely to be highly clumpy. We also find a global correlation between the [OIII] and the far-infrared continuum emission, suggesting that the gas and dust are well mixed in the highly-ionized region where the dust survives in clumpy dense clouds shielded from energetic photons. Furthermore we find that the ratios of [CII]/CO are as high as 110,000 in 30 Dor, and 45,000 even on average, while they are typically 6,000 for star-forming regions in our Galaxy. The unusually high [CII]/CO is also consistent with the picture of clumpy small dense clouds.
