Profiling of remote skeletal muscle gene changes resulting from stimulation of atopic dermatitis disease in NC/Nga mouse model

        Donghee Lee,Yelim Seo,Young-Won Kim,Seongtae Kim,Jeongyoon Choi,Sung-Hee Moon,Hyemi Bae,Hui-sok Kim,Hangyeol Kim,Jae-Hyun Kim,Tae-Young Kim,Eunho Kim,Suemin Yim,Inja Lim,Hyoweon Bang,Jung-Ha Kim,Jae-H 대한약리학회 2019 The Korean Journal of Physiology & Pharmacology Vol.23 No.5

Although atopic dermatitis (AD) is known to be a representative skin disorder, it also affects the systemic immune response. In a recent study, myoblasts were shown to be involved in the immune regulation, but the roles of muscle cells in AD are poorly understood. We aimed to identify the relationship between mitochondria and atopy by genome-wide analysis of skeletal muscles in mice. We induced AD-like symptoms using house dust mite (HDM) extract in NC/Nga mice. The transcriptional profiles of the untreated group and HDM-induced AD-like group were analyzed and compared using microarray, differentially expressed gene and functional pathway analyses, and protein interaction network construction. Our microarray analysis demonstrated that immune response-, calcium handling-, and mitochondrial metabolism-related genes were differentially expressed. In the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology pathway analyses, immune response pathways involved in cytokine interaction, nuclear factor-kappa B, and T-cell receptor signaling, calcium handling pathways, and mitochondria metabolism pathways involved in the citrate cycle were significantly upregulated. In protein interaction network analysis, chemokine family-, muscle contraction process-, and immune response-related genes were identified as hub genes with many interactions. In addition, mitochondrial pathways involved in calcium signaling, cardiac muscle contraction, tricarboxylic acid cycle, oxidation-reduction process, and calcium-mediated signaling were significantly stimulated in KEGG and Gene Ontology analyses. Our results provide a comprehensive understanding of the genome-wide transcriptional changes of HDM-induced AD-like symptoms and the indicated genes that could be used as AD clinical biomarkers.

Downlink Transmit Power Allocation in Soft Fractional Frequency Reuse Systems

        Donghee Kim,Jae Young Ahn,Hojoon Kim 한국전자통신연구원 2011 ETRI Journal Vol.33 No.1

Downlink transmit power allocation schemes are proposed for soft fractional frequency reuse (FFR) in loose and tightly coordinated systems. The transmit powers are allocated so that the loss of spectral efficiency from the soft FFR is minimized, and the required cell edge user throughput is guaranteed. The effect of the soft FFR on spectral efficiency is evaluated depending on the power allocation schemes and the number of subbands. Results show that the loss of spectral efficiency from the soft FFR can be reduced by configuring an appropriate number of subbands in the loosely coordinated systems. In tightly coordinated systems, results show that the loss of spectral efficiency can be minimized regardless of the number of subbands due to its fast coordination.

      • Solvent-Free Nanocomposite Colloidal Fluids with Highly Integrated and Tailored Functionalities: Rheological, Ionic Conduction, and Magneto-Optical Properties

Kim, Donghee,Kim, Younghoon,Cho, Jinhan American Chemical Society 2013 Chemistry of materials Vol.25 No.19

<P>We introduce a unique and facile strategy for the preparation of solvent-free nanocomposite colloidal fluids that allows accurate control over the integration of functionalities as well as the composition and dimensions of the nanocomposite structure. For the preparation of colloidal fluids with highly integrated functionalities, oleic acid (OA)-stabilized magnetic nanoparticles (i.e., OA-Fe3O4 NPs) and CdSe@ZnS quantum dots (QDs) were first synthesized in nonpolar solvent. In this case, OA-QDs dispersed in toluene were successively phase transferred to thiol-functionalized imidazolium-type ionic liquid (IL-SH) media with rheological and ionic conduction properties. After the functional NPs were synthesized, amine-functionalized dendrimers and OA-Fe3O4 NPs were alternately deposited onto silica colloids (i.e., SiO2/(dendrimer/OA-Fe3O4)<I>n</I>) using a ligand-exchange-induced LbL-assembly in organic media. Electrostatic LbL-assembled (anionic polyelectrolyte (PE)/cationic IL-SH-QD)<I>n</I> multilayers were then sequentially adsorbed onto the outermost dendrimer layer of the magnetic colloids. The resulting functional colloidal fluids were devoid of colloidal aggregation and exhibited strong superparamagnetic, fluorescent, rheological, and ionic conduction properties at room temperature. Furthermore, mixtures of photoluminescent colloidal fluids with and without OA-Fe3O4 NPs behaved effectively as magneto-optically separable colloidal fluids. Because a variety of inorganic NPs ranging from metal to transition-metal oxides can be easily incorporated into colloidal substrates via LbL-assembly, our approach provides a basis for exploiting and designing functional colloidal fluids with liquidlike behavior at room temperature.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/cmatex/2013/cmatex.2013.25.issue-19/cm401560r/production/images/medium/cm-2013-01560r_0009.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/cm401560r'>ACS Electronic Supporting Info</A></P>

      • Reactive Oxygen Species Enhance TLR10 Expression in the Human Monocytic Cell Line THP-1

Kim, Donghee,Kim, Yeon Ju,Koh, Hyun Sook,Jang, Tae Yang,Park, Hyo Eun,Kim, Jae Young Molecular Diversity Preservation International (MD 2010 INTERNATIONAL JOURNAL OF MOLECULAR SCIENCES Vol.11 No.10

<P>We investigated TLR10 expression in human monocytes, THP-1 cells, cultured in hypoxia (3% O2). Levels of both TLR10 mRNA and protein in THP-1 cells cultured in hypoxia were significantly higher than those cultured in normoxia (20% O2). We examined intracellular reactive oxygen species (ROS) content in hypoxic cells, and TLR10 expression in cells treated with hydrogen peroxide (H2O2), to determine whether the increase in TLR10 expression observed with hypoxia was due to an increase in intracellular ROS levels. We found that the level of intracellular ROS in cells subject to hypoxia was significantly higher than in normoxia. Experiments with ROS synthesis inhibitors revealed that hypoxia induced ROS production is mainly due to NADPH oxidase activity. TLR10 mRNA expression was increased by treatment with H2O2 at concentrations ranging from 50 to 250 μM. We screened the TLR10 promoter and found putative binding sites for transcription factors (TFs), such as NF-κB, NF-AT and AP-1. Next, we examined TF activities using a luciferase reporter assay. Activities of NF-κB, NF-AT and AP-1 in the cells treated with H2O2 were significantly higher than in untreated cells. The experiment with TF inhibitors revealed that ROS-induced upregulation of TLR10 expression is mainly due to NF-κB activation. Overall, our results suggest that hypoxia or ROS increase TLR10 expression in human monocytes and the transcriptional activities of NF-κB are involved in this process. Therefore, it is suggested that ROS produced by various exogenous stimuli may play a crucial role in the regulation of expression and function of TLR10 as second messengers.</P>

Scalable message translation mechanism for the environment of heterogeneous middleware

        Donghee Kim,Chang-Eun Lee,Jun Hee Park,KyeongDeok Moon,Kyungshik Lim IEEE 2007 IEEE transactions on consumer electronics Vol.53 No.1

<P>Various home network middleware such as HAVi, LnCP, LonWorks, and UPnP have been designed. They coexist because one middleware cannot support diverse requests of services and devices. This circumstance creates an interoperability problem among devices based on various types of home network middleware. The Universal Middleware Bridge system is designed and implemented to solve this interoperability problem using a device conversion mechanism and a message translation mechanism among the middleware. These conversion mechanisms make it possible for devices to distinguish and control other devices connected to different middleware networks in the Universal Middleware Bridge system. These mechanisms also easily support new device and middleware by adding some XML files.</P>

Automotive telematics market segmentation based on quality expectations

Kim, Dayoung,Kim, Donghee,Oh, Jungsuk Korean Marketing Association 2014 ASIA MARKETING JOURNAL Vol.16 No.3

This paper explores the possibility of segmentation of consumers based on their quality expectations toward the automotive telematics service. Hypotheses on utilizing consumers' expectations toward the automotive telematics service for the market segmentation and linking the segments with optimal product designs are formulated. Samples are segmented based on their perceived service quality to the service attributes from various configurations of automotive telematics service. Then, a regression analysis on the segmented groups of users is performed to check whether they have qualitatively differing evaluation on the service quality. The result indicates that the proposed segmentation is operational and differing product attributes configuration is desirable according to the characteristics of the consumer segment. Hence, according to the characteristics of each consumer segment formed based on their expectation toward the telematics service, a product differentiation strategy of the automotive telematics system can be designed and be proposed to the product line designer.

Effect of Nozzle Tip Size on the Fabrication of Nano-Sized Nickel Oxide Powder by Spray Pyrolysis Process

Kim, Donghee,Yu, Jaekeun Materials Research Society of Korea 2013 한국재료학회지 Vol.23 No.9

In this study, by using nickel chloride solution as a raw material, a nano-sized nickel oxide powder with an average particle size below 50 nm was produced by spray pyrolysis reaction. A spray pyrolysis system was specially designed and built for this study. The influence of nozzle tip size on the properties of the produced powder was examined. When the nozzle tip size was 1 mm, the particle size distribution was more uniform than when other nozzle tip sizes were used and the average particle size of the powder was about 15 nm. When the nozzle tip size increases to 2 mm, the average particle size increases to roughly 20 nm, and the particle size distribution becomes more uneven. When the tip size increases to 3 mm, particles with an average size of 25 nm and equal to or less than 10 nm coexist and the particle size distribution becomes much more uneven. When the tip size increases to 5 mm, large particles with average size of 50 nm partially exist, mostly consisting of minute particles with average sizes in the range of 15~25 nm. When the tip size increases from 1 mm to 2 mm, the XRD peak intensities greatly increase while the specific surface area decreases. When the tip size increases to 3 mm, the XRD peak intensities decrease while the specific surface area increases. When the tip size increases to 5 mm, the XRD peak intensities increase again while the specific surface area decreases.
