Multiple sclerosis is a chronic disorder commonly occurred in the central nervous system and is an autoimmune disease. Corticosteroids are used as a short-term treatment for multiple sclerosis, but they cause side effects. Therefore, interferon beta having antiviral and anti-inflammatory effects, is used for long-term treatment. Interferon beta-1a, produced in Chinese hamster ovary cells, is glycosylated upon biosynthesis and forms glycoform, which is similar to naturally occurring protein. However, its process has disadvantages such as slow cell growth, low yield and high contamination possibility. Also, recombinant Escherichia coli is widely used for the production of interferon beta-1b due to its rapid growth and easy process scale up, but it lacks post-translational modification, leading to low activity. In addition, the produced Interferon beta-1b might cause protein aggregation due to misfolding. Alternatively, yeast can be used as a production host possessing N-glycosylation activity. Therefore, the choice of expression system in the production of interferon beta should be carefully selected in relation to the quality and yield.
Predictor for deteriorating ward patient with repetitive Medical Emergency Team activation

Purpose: Recurrent clinical deterioration and repetitive activation of Medical Emergency Team (MET) may be associated with poor outcome. The purpose of this study is to analyze the clinical characteristics of patients with repetitive MET activation and to predict the repetitive MET activation at first MET activation. Methods: We retrospectively reviewed the data of deteriorating patients who were contacted to the MET of Asan Medical Center from Jan 2012 to Nov 2016. The repetitive MET activation means the case of activation within 72 hours after the first MET activation. Results: A total of 7,470 patients were included during the study period. Of these patients, 892 (11.9%) experienced repetitive MET activation. ICU admission rate (49.0% vs. 27.8%, p<0.001) and in-hospital mortality rate (51.7% vs. 36.6%, p<0.001) were higher in the repetitive MET activation group than in the group who were once MET activation. The repetitive MET activation were independently associated with hematologic malignancies (OR: 1.45, 95% CI: 1.13-1.90), activation by respiratory distress(OR: 1.51, 95% CI: 1.20-1.89), airway suctioning (OR: 1.57, 95% CI: 1.24-1.98), apply of Bipap (OR: 1.97, 95% CI: 1.20-3.22), high SOFA (OR: 1.11, 95% CI: 1.07-1.15), and high SpO2/FiO2 score (OR: 1.14, 95% CI: 1.05-1.24) at first MET activation. Conclusion: These risk factors may be useful to identify patients at risk of repetitive MET activation at first MET activation, and then we could classify the high risk group and apply the aggressive intervention for better outcome.

Two new (1 and 3) two known diarylhep-tanoids (2 and 4), along with two tetralones (5 and 6), one naphthoquinone (7), four phenylpropanoids (8-11), and phenol (12) were isolated from the leaves of Juglans mandshurica. Their structures were elucidated on the basis of spectral and chemical data. Compounds 2 and 10 are firstly isolated from this plant and 8 and 12 were isolated from the Juglans genus for the first time. Among these compounds, only 7 cxhibitcd moderate cytotoxicities against cultured Mgc-803, A549, K562, and Hela tumor cell lines with IC50 values of 25.90, 28.60, 39.06, 44.90 LLM, respectively.

<P><B>ABSTRACT</B></P><P>Tissue microenvironment adjusts biological properties of different cells by modulating signaling pathways and cell to cell interactions. This study showed that epithelial–mesenchymal transition (EMT)/ mesenchymal–epithelial transition (MET) can be modulated by altering culture conditions. HPV E6/E7‐transfected immortalized oral keratinocytes (IHOK) cultured in different media displayed reversible EMT/MET accompanied by changes in cell phenotype, proliferation, gene expression at transcriptional, and translational level, and migratory and invasive activities. Cholera toxin, a major supplement to culture medium, was responsible for inducing the morphological and biological changes of IHOK. Cholera toxin per se induced EMT by triggering the secretion of interleukin 6 (IL‐6) from IHOK. We found IL‐6 to be a central molecule that modulates the reversibility of EMT based not only on the mRNA level but also on the level of secretion. Taken together, our results demonstrate that IL‐6, a cytokine whose transcription is activated by alterations in culture conditions, is a key molecule for regulating reversible EMT/MET. This study will contribute to understand one way of cellular adjustment for surviving in unfamiliar conditions. J. Cell. <P><B>Abstract</B></P> <P>We study thermal annealing-driven surface sealing of polymer hollow particles with a different surface property. Use of phase separation between polystyrene (PS) solid particles incorporated with sulfonate (SO<SUB>3</SUB> <SUP>-</SUP>) group and decane allows their transformation into hollow particles with an opening on the surface. Control of the phase separation enables us to tune the structure (outer and inner diameters, and size of the opening) of the hollow particles. While thermally annealed at a temperature above <I>T</I> <SUB> <I>g</I> </SUB> of the polymer, structural change of the hollow particles is experimentally observed and theoretically analyzed. The inhibitory activity of ginsenoside Rp4 in adenosine diphosphate-induced platelet aggregation

Background: Korean ginseng, Panax ginseng Meyer, has been used as a traditional oriental medicine to treat illness and promote health for several thousand years. Ginsenosides are the main constituents for the pharmacological effects of P. ginseng. Since several ginsenosides, including ginsenoside (G)-Rg3 and G-Rp1, have reported antiplatelet activity, here we investigate the ability of G-Rp4 to modulate adenosine diphosphate (ADP)-induced platelet aggregation. The ginsenoside Rp4, a similar chemical structure of G-Rp1, was prepared from G-Rg1 by chemical modification. Methods: To examine the effects of G-Rp4 on platelet activation, we performed several experiments, including antiplatelet ability, the modulation of intracellular calcium concentration, and P-selectin expression. In addition, we examined the activation of integrin ${\alpha}IIb{\beta}_3$ and the phosphorylation of signaling molecules using fibrinogen binding assay and immunoblotting in rat washed platelets. Results: G-Rp4 inhibited ADP-induced platelet aggregation in a dose-dependent manner. We found that G-Rp4 decreased calcium mobilization and P-selectin expression in ADP-activated platelets. Moreover, fibrinogen binding to integrin ${\alpha}IIb{\beta}_3$ by ADP was attenuated in G-Rp4-treated platelets. G-Rp4 significantly attenuated phosphorylation of extracellular signal-regulated protein kinases 1 and 2, p38, and c-Jun N-terminal kinase, as well as protein kinase B, phosphatidylinositol 3-kinase, and phospholipase C-${\gamma}$ phosphorylations. Conclusion: G-Rp4 significantly inhibited ADP-induced platelet aggregation and this is mediated via modulating the intracellular signaling molecules. These results indicate that G-Rp4 could be a potential candidate as a therapeutic agent against platelet-related cardiovascular diseases.
