Long Noncoding RNA HEIH Promotes Colorectal Cancer Tumorigenesis via Counteracting miR-939Mediated Transcriptional Repression of Bcl-xL

        Chunhui Cui,Duanyang Zhai,Lianxu Cai,Qiaobin Duan,Lang Xie,Jinlong Yu 대한암학회 2018 Cancer Research and Treatment Vol.50 No.3

Purpose Studies have found that long noncoding RNA HEIH (lncRNA-HEIH) is upregulated and facilitates hepatocellular carcinoma tumor growth. However, its clinical significances, roles, and action mechanism in colorectal cancer (CRC) remains unidentified. Materials and Methods lncRNA-HEIH expression in CRC tissues and cell lines was measured by quantitative realtime polymerase chain reaction. Cell Counting Kit-8, ethynyl deoxyuridine incorporation assay, terminal deoxynucleotidyl transferase dUTP nick end labeling staining, and nude mice xenografts assays were performed to investigate the roles of lncRNA-HEIH. RNA pull-down, RNA immunoprecipitation, chromatin immunoprecipitation, and luciferase reporter assays were performed to investigate the action mechanisms of lncRNA-HEIH. Results In this study, we found that lncRNA-HEIH is significantly increased in CRC tissues and cell lines. lncRNA-HEIH expression is positively associated with tumor size, invasion depth, and poor prognosis of CRC patients. Enhanced expression of lncRNA-HEIH promotes CRC cell proliferation and decreases apoptosis in vitro, and promotes CRC tumor growth in vivo. Whereas knockdown of lncRNA-HEIH inhibits CRC cell proliferation and induces apoptosis in vitro, and suppresses CRC tumor growth in vivo. Mechanistically, lncRNA-HEIH physically binds to miR-939. The interaction between lncRNA-HEIH and miR-939 damages the binding between miR-939 and nuclear factor B (NF-B), increases the binding of NF-B to Bcl-xL promoter, and promotes the transcription and expression of Bcl-xL. Moreover, Bcl-xL expression is positively associated with lncRNA-HEIH in CRC tissues. Blocking the interaction between lncRNA-HEIH and miR-939 abolishes the effects of lncRNA-HEIH on CRC tumorigenesis. Conclusion This study demonstrated that lncRNA-HEIH promotes CRC tumorigenesis through counteracting miR-939mediated transcriptional repression of Bcl-xL, and suggested that lncRNAHEIH may serve as a prognostic biomarker and therapeutic target for CRC.

Wei Sun,Chunhui Huo,Yang Yu 보안공학연구지원센터 2016 International Journal of Future Generation Communi Vol.9 No.11

Basic scientific research achievements management involves multiple knowledge resources. We propose a knowledge supernetwork model to integrate multiple knowledge resources. First, person network, material carrier network, and knowledge element network are established. Subsequently, the knowledge supernetwork is constructed to integrate the above three networks. Second, the characteristics of knowledge supernetwork are investigated from the perspective of the quantitative analysis. Based on the knowledge supernetwork, some methods for solving real problems are developed. Finally, we construct a knowledge supernetwork model using real basic scientific research achievement files. The methods of knowledge representation, basic scientific research proposal grouping, and knowledge classification are proposed to illustrate how to manage basic scientific research achievement from the perspective of knowledge.

Simultaneous Optimization of Passive Transit Priority Signals and Lane Allocation

        Weiguo Shi,Chunhui Yu,Wanjing Ma,Ling Wang,Lei Nie 대한토목학회 2020 KSCE JOURNAL OF CIVIL ENGINEERING Vol.24 No.2

This paper presents an integrated optimization model for the concurrent design of passive bus priority signal settings and lane allocation including lane markings for general traffic and exclusive bus lanes at isolated intersections. Traditional passive transit signal priority (TSP) strategies considered both lane markings for general traffic and exclusive bus lane settings as exogenous inputs. However, in our method, two modes of transportation (passenger cars and buses) have been considered in a centralized framework while exclusive bus lane settings, lane markings, and the optimal passive transit priority signal settings can be optimized concurrently. Minimization of the weighted delay of buses and cars is adopted as the aim of the integrated optimization model. Based on traditional National Electrical Manufacturers Association (NEMA) phase constructure, a precedence graph was employed, by which the mathematical optimization model is formulated. A set of restrictions have been set up to verify efficiency and safety for the resulting optimal lane markings and signal settings. A simulated annealing (SA) algorithm is designed to resolve the mix-integer-nonlinear-programming (MINLP) problem. In order to demonstrate the efficacy of the method proposed, numerical examples have been provided. The results from sensitivity analysis show that the proposed method can generate sub-optimal solutions for the intersection of different combinations of car demand, bus demand, and left turn ratios.

Properties of Bi2O3 thin films prepared via a modified Pechini route

        Timonah N. Soitah,Yang Chunhui,Yu Yong,Niu Yinghua,Sun Liang 한국물리학회 2010 Current Applied Physics Vol.10 No.6

Thin bismuth oxide films have been prepared by a modified Pechini route on glass substrate and annealed at temperatures ranging between 400 ℃ and 700 ℃ using bismuth nitrate as raw material. The thin films were then characterized for structural, surface morphological, optical and electrical properties by means of X-ray diffraction (XRD), Atomic force microscopy (AFM), scanning electron microscopy (SEM), optical absorption and d.c. two-probe, respectively. Structural investigations indicated that asprepared bismuth oxide films were polycrystalline and multiphase, and annealing temperatures played a key role in the composition and optical properties of these films. AFM and SEM images revealed well defined particles which are highly influenced by annealing temperatures. The optical studies showed a direct band gap which varied with annealing temperatures between 3.63 eV and 3.74 eV. The electrical measurement showed that the electrical resistivity increased with annealing temperatures and the films were typical semiconductors. As catalyst, bismuth oxide films annealed at 550 ℃ had the best photocatalytic performance for photodegradation of methyl orange.

Effects of exposure to trace pyriproxyfen on the intestinal bacterial diversity and immune signal pathways of silkworm (Bombyx mori) larvae

        Lu Qingyu,Li Guoli,Lan Huangli,Yu Dongliang,Yin Xingcan,Yang Wenqi,Yang Zehao,He Chunhui,Mo Danmei,Xu Kaizun 한국응용곤충학회 2022 Journal of Asia-Pacific Entomology Vol.25 No.2

Silkworm (Bombyx mori) is a widely used lepidopteran model insect. Pyriproxyfen is an insect growth regulator (IGR) that can be used to control pests such as lepidoptera and diptera, and there is no research about its in fluence on the intestinal bacterial diversity and immune signal pathway of non-target lepidopteran insect larvae. In this study, we treated fifth-instar silkworms with trace pyriproxyfen (10 − 4 mg/L). The results showed that the abundance and diversity of silkworm larvae intestinal flora at the level of phylum, class, order, family and genus were all changed. The function prediction result showed that the functional changes mainly focused on tran scription, amino acid transport and metabolism, carbohydrate transport and metabolism, signal transduction mechanisms, energy production and conversion et al. after exposure to trace pyriproxyfen. Furthermore, qRTPCR results showed that trace pyriproxyfen exposure activated the transcriptions of antimicrobial peptides (AMPs) genes regulated by Toll and IMD pathways. This research can lay the foundation for the study on the resistance of lepidopteran insects to pyriproxyfen and other juvenile hormone pesticides.

        Wang, Bo,Xie, Ying,Liu, Tong,Luo, Hao,Wang, Bin,Wang, Chunhui,Wang, Lei,Wang, Dianlong,Dou, Shixue,Zhou, Yu Elsevier 2017 Nano energy Vol.42 No.-

<P><B>Abstract</B></P> <P>Due to the relatively slow, diffusion-controlled faradaic reaction mechanisms of conventional LiFePO4 (LFP) materials, which is hard to deliver satisfied capacity for high rate applications. In this work, ultrafine LFP quantum dots (LFP-QDs) co-modified by two types of carbonaceous materials - amorphous carbon and graphitized conductive carbon (graphene) have been successfully synthesized through a novel microreactor strategy. Because of the very limited area constructed by the dual-carbon microreactor for the growth of LFP crystal, it's demension was furthest suppressed to a very small level (~ 6.5nm). Such a designed nano-composite possesses a large specific surface area for charge adsorption and abundant active sites for faradaic reactions, as well as ideal kinetic features for both electron and ion transport, and thus exhibits ultra-fast, surface-reaction-controlled lithium storage behavior, mimicking the pseudocapacitive mechanisms for supercapacitor materials, in terms of extraordinary rate capability (78mAhg−1 at 200C) and remarkable cycling stability (~ 99% over 1000 cycles at 20C). On the other side, due to the quasi-2D structure of the synthesized LFP-QDs composite, which can be used as the basic unit to further fabricate free-standing film, aerogel and fiber electrode without the addition of binder and conductive agent for different practical applications. In addition, to deeper understand its electrochemical behavior, a combined experimental and density functional theoretical (DFT) calculation study is also introduced.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A general microreactor strategy has been developed for structure-optimized Li-contained electrode materials. </LI> <LI> Ultrafine LiFePO4 quantum dots are first reported through the designed microreactor strategy. </LI> <LI> The synthesized G/LFP-QDs@C exhibits ultra-fast, surface-reaction-controlled Li storage behavior. </LI> <LI> A combined experimental and DFT calculation study is introduced to reveal the energy storage mechanism of G/LFP-QDs@C. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>Ultrafine LiFePO4 quantum dots (~ 6.5nm) co-modified by two types of carbonaceous materials - amorphous carbon and graphitized conductive carbon (graphene) have been successfully synthesized through a novel microreactor strategy, which exhibit ultra-fast, surface-reaction-controlled energy storage behavior, mimicking the pseudocapacitive mechanisms for supercapacitor materials, in terms of excellent rate capability and outstanding cycling stability.</P> <P>[DISPLAY OMISSION]</P>

Decline in non-smoking workers’ urine cotinine levels after increased smoking regulation in Korea

        Ju-Hyoung Park,Chae-Kwan Lee,Se-Yeong Kim,Chunhui Suh,Kun-Hyung Kim,Jeong-Ho Kim,Byung-Chul Son,Jong-Tae Lee,Seung-Do Yu,Wookhee Choi,Hosub Im 대한직업환경의학회 2015 대한직업환경의학회지 Vol.27 No.-

Objectives: To identify any association between implementing smoking regulation policies and workers’ urine cotinine concentration levels in Korea. Methods: From the first stage of the Korean National Environmental Health Survey conducted by the National Institute of Environmental Research from 2009 to 2011, 2,475 non-smoking workers selected. We analyzed the trend in the changes of cotinine concentration in urine using the general linear model and linear regression, in various jobs as categorized by the National Center for Health Statistics (NCHS) and Korea Standard Classification of Occupations (KSCO). Results: The urine cotinine concentration tended to decrease every year (2.91 ng/ml in 2009, 2.12 ng/ml in 2010, and 1.31 ng/ml in 2011), showing a decreasing trend (P < 0.001). The total subjects’ decreased cotinine concentration in urine between 2009 and 2011 was 2.72 ng/ml (54.1 % relative decrease). The changes in each subgroup’s urine cotinine concentration ranged from 1.59 to 6.03 ng/ml (33.2 to 77.5 %). All groups except for the managerial group (n = 49), which had a small sample size, had statistically significant negative regression coefficients (p < 0.05). The ranges of the decrease in urine cotinine were 2.75 ng/ml (53.6 %) for males and 2.72 ng/ml (54.9 %) for females. The negative slope in urine cotinine level was statistically significantly greater in men than women. The changes in urine cotinine by occupation as classified by the NCHS occupational categories ranged from 2.43 to 3.36 ng/ml (46.6 to 61.5 % relative decrease). The negative slopes in urine cotinine levels of the white-collar and farm workers were statistically significantly greater than those of the service workers and blue-collar workers. The change by occupation as classified by the KSCO ranged from 1.59 to 6.03 ng/ml (a 33.2 to 77.5 % relative decrease). The negative slopes in urine cotinine levels of the professionals and related workers and clerks were statistically significantly greater than those of the service workers and plant and machine operators and assemblers. Conclusions: The cotinine concentration in urine among non-smoking worker groups tended to decline from 2009 to 2011. Such a result may be an indirect indicator of the effectiveness of smoking regulation policies including the revision of the National Health Promotion Act.
