        Correlation Between Fiber Orientation and Geometrical Shrinkage of Injected Parts Under the Influence of Flow-Fiber Coupling Effect

        Fiber reinforced thermoplastics (FRP) have been widely used in automotive industry. However, how does the flow-fiber coupling effect influence the micro fiber orientation and further affect the geometrical shrinkage of the final part that is not fully understood yet. In this study, a complex center-gated plate has been applied to study the influence of the flow-fiber coupling effect on the fiber orientation variation and the geometrical change through numerical simulation. Then the practical verification through the micro-computed tomography (micro-CT) and image processing technology was carried out. Results show that in the presence of the flow-fiber coupling the required spruce pressure will be higher compared to no coupling case. In addition, the melt flow front pattern will be changed from “convex-flat” to “convex-concave” under the influence of this coupling. Moreover, in the presence of the flow-fiber coupling effect, the wider core width for fiber orientation tensor in the flow direction (A11) can be obtained from upstream to downstream regions for the same model. However, in the downstream region (i.e. in the FR), the flow-fiber coupling effect is more significantly due to the action of less shear rate in that region. Finally, through the measurement of the left–right asymmetrical shape of the FR for Model I (or Model II), the reason is that the flow-fiber coupling effect will switch the fiber orientation from the flow direction (A11) dominate to the cross-flow direction (A22) dominate. This asymmetrical fiber orientation distribution will further create that asymmetrical shrinkage shape of final part. The correlation between fiber orientation and geometrical shrinkage can be achieved.

        Original Article : A Disintegrin and Metalloprotease with Thrombospondin Motif 2 May Contribute to Cirrhosis in Humans through the Transforming Growth Factor-β/SMAD Pathway

        Background/Aims: We aimed to investigate the correlation between a disintegrin and metalloprotease with thrombospondin motif 2 (ADAMTS-2) and transforming growth factor-β1 (TGF-β1) in clinical human cirrhotic tissues. Methods: The liver tissues of 24 patients (16 cases with cirrhotic portal hypertension as the cirrhosis group and eight cases with healthy livers as the normal group) were collected. Immunohistochemistry and Western blots were performed to evaluate the protein expression levels of ADAMTS-2 and TGF-β1. Western blots for other key mediators of cirrhotic progression, including SMAD2, SMAD3, TGF-β receptor II (TGFβRII), matrix metalloproteinases 2 (MMP2), and tissue inhibitor of matrix metalloproteinases 2 (TIMP2), were also performed. Results: Cirrhotic tissues showed higher percentages of collagen. The protein expression levels of ADAMTS-2 and TGF-β1 were significantly higher in the cirrhotic group as compared to the matched normal group (p<0.05), and there was a positive correlation between these two proteins (r=0.862, p<0.01). The protein expressions of MMP2, TIMP2, and TGFβRII, as well as the phosphorylated forms of SMAD2 and SMAD3, were significant higher in the cirrhotic group (p<0.01 or p<0.05). Conclusions: These findings suggested that ADAMTS-2 and TGF-β1 may play important roles in the pathogenesis of human cirrhosis; specifically, TGF-β1 may induce the expression of ADAMTS-2 through the TGFβ/SMAD pathway.         Comparative Study of Two Common In Vitro Models for the Pancreatic Islet with MIN6

        BACKGROUND: Islet transplantation is currently considered the most promising method for treating insulin-dependent diabetes. The two most-studied artificial islets are alginate-encapsulated b cells or b cell spheroids. As three-dimensional (3D) models, both artificial islets have better insulin secretory functions and transplantation efficiencies than cells in twodimensional (2D) monolayer culture. However, the effects of these two methods have not been compared yet. Therefore, in this study, cells from the mouse islet b cell line Min6 were constructed as scaffold-free spheroids or alginate-encapsulated dispersed cells. METHODS: MIN6 cell spheroids were prepared by using Agarose-base microwell arrays. The insulin secretion level was determined by mouse insulin ELISA kit, and the gene and protein expression status of the MIN6 were performed by Quantitative polymerase chain reaction and immunoblot, respectively. RESULTS: Both 3D cultures effectively promoted the proliferation and glucose-stimulated insulin release (GSIS) of MIN6 cells compared to 2D adherent cells. Furthermore, 1% alginate-encapsulated MIN6 cells demonstrated more significant effects than the spheroids. In general, three pancreatic genes were expressed at higher levels in response to the 3D culture than to the 2D culture, and pancreatic/duodenal homeobox-1 (PDX1) expression was higher in the cells encapsulated in 1% alginate than that in the spheroids. A western blot analysis showed that 1% alginate-encapsulated MIN6 cells activated the phosphoinositide 3-kinase (PI3K)/serine/threonine protein kinase (AKT)/forkhead transcription factor FKHR (FoxO1) pathway more than the spheroids, 0.5% alginate-, or 2% alginate-encapsulated cells did. The 3D MIN6 culture, therefore, showed improved effects compared to the 2D culture, and the 1% alginate-encapsulated MIN6 cells exhibited better effects than the spheroids. The upregulation of PDX1 expression through the activation of the PI3K/AKT/FoxO1 pathway may mediate the improved cell proliferation and GSIS in 1% alginate-encapsulated MIN6 cells. CONCLUSION: This study may contribute to the construction of in vitro culture systems for pancreatic islets to meet clinical requirements.

      • The Structural Design of Tianjin Goldin Finance 117 Tower

        Tianjin Goldin Finance 117 tower has an architectural height of 597 m, total of 117 stories, and the coronation of having the highest structural roof of all the buildings under construction in China. Structural height-width ratio is approximately 9.5, exceeding the existing regulation code significantly. In order to satisfy earthquake and wind-resisting requirements, a structure consisting of a perimeter frame composed of mega composite columns, mega braces and transfer trusses and reinforced concrete core containing composite steel plate wall is adopted. Complemented by some of the new requirements from the latest Chinese building seismic design codes, design of the super high-rise building in high-intensity seismic area exhibits a number of new features and solutions to professional requirements in response spectrum selection, overall stiffness control, material and component type selection, seismic performance based design, mega-column design, anti-collapse and stability analysis as well as elastic-plastic time-history analysis. Furthermore, under the prerequisite of economic viability and a series of technical requirements prescribed by the expert review panel for high-rise buildings exceeding code limits, the design manages to overcome various structural challenges and realizes the intentions of the architect and the client.

        Deformation Characteristics of Soft Soil Induced by Deep Excavation and Its Impact on Adjacent Tunnels: A Case Study in Shanghai

        This research aims to comprehensively study the deformation characteristics of an excavation pit of 7,600 m2 (117 m long, 65 m wide and 20.8 m deep) in soft clay in Shanghai, China. The deformation characteristics of the mixing pile in both the vertical and lateral directions, ground surface movement, and surface displacement in the adjacent tunnels were comprehensively investigated. The results indicated that the maximum lateral deflection was approximately 85 mmat the end of the excavation and that the maximum lateral displacement, hm, was in the range between 0.12%He and 0.41%He. The factor of safety of the excavation was approximately 1.6, which was larger than that of most reported sites in Shanghai. The relationship between heaving and excavation depth was strong between 0.0047%He and 0.056%He, and a relatively large settlement was observed at a distance of approximately 0.2He – 0.3He. The tunnel surface of the eastbound line deformed upward and inward with a maximum horizontal convergence of 4 mm. A multilayer regression model was also validated, showing a maximum discrepancy of 26.5 mm. This research provides a valuable historical case for the documentation of the deformation characteristics of excavation pits close to existing foundation pits and tunnels.

        Depression and APOEε4 Status in Individuals with Subjective Cognitive Decline: A Meta-Analysis

        Objective To evaluate the associative role of depression and apolipoprotein E epsilon 4 allele (APOEε4) in subjective cognitive decline(SCD) and its progression to objective cognitive decline. Methods After literature search in electronic databases, studies were selected by following precise eligibility criteria. Meta-analyses were performed to examine the role of APOEε4 and depression in SCD or its progression to mild cognitive impairment (MCI) or dementia. Results APOEε4 positivity was not different between SCD and normal individuals but was significantly higher in individuals with SCD plus than in normal individuals [odds ratio: 2.39 (95% CI: 1.87, 3.05); p<0.00001] and in SCD converters than in non-converters [odds ratio: 5.19 (95% CI: 2.36, 11.42); p<0.00001]. Depression was significantly higher in individuals with SCD [standardized mean difference: 0.63 (0.45, 0.82); p<0.00001] and SCD plus [standardized mean difference: 0.83 (0.43, 1.22); p<0.0001] than in normal individuals. However, depression was not different between SCD and MCI or between SCD converters and non-converters. Age of SCD converters was higher than non-converters [mean difference: 2.95 years (0.58, 5.31)]. Conclusion Whereas APOEε4 positivity was higher in SCD plus and SCD converters, depression was higher in SCD and SCD plus but was not different between SCD and MCI.

        Laser Patterning Technology Based on Nanosecond Pulsed Laser for Manufacturing Bifacial Perovskite Solar Modules

        Bifacial semi-transparent perovskite (PVSK) solar cell is a promising candidate to achieve high photo-electrical conversion efficiency (PCE) in a tandem structure with Si solar cells. The gap between lab-scale cells and large area modules needs to be closed using innovative patterning technology. In this paper we demonstrate that a single nanosecond pulsed laser (wavelength 532 nm, pulse duration 7 ns) can be used to perform all scribing processes, i.e. P1, P2 and P3, to manufacture PVSK solar modules. Compared to picosecond or femtosecond lasers reported in the literature, our approach has the advantages of high stability and low cost, and is thus applicable to large scale manufacturing of PVSK solar modules. Detailed laser processing parameters such as laser power and overlap ratio etc. have been studied to achieve optimal results for each scribing process. A mini module with two cells was fabricated on a 2 × 2 cm2 substrate, showing an active area efficiency of 12.5%, FF of 72.4%, and high GFF of 94%.

      • Efficacy and Safety of 12 Weeks of Daclatasvir, Asunaprevir Plus Ribavirin for the Treatment of HCV Genotype 1b Infection without Baseline NS5A Resistance-Associated Variants (DARING)-Interim Report

        Aims: The current study aims to elucidate the treatment efficacy (defined as undetectable HCV RNA throughout 12 weeks of post-treatment follow-up, SVR12) and safety DCV/ASV plus ribavirin for 12 weeks in HCV-1b patients without NS5A RAS. Methods: This is a single-arm, open-label phase 2 study. Seventy directly acting antivirals (DAA)-naïve HCV-1b patients without L31/Y93 RAS are planned to receive daclatasvir (60 mg/ day) and asunaprevir (100 mg twice daily) plus weight-based ribavirin (1000-1200 mg/day) for 12 weeks. After treatment they were followed up for 12 weeks. Results: As of 31 Oct 2017, 58 eligible patients are allocated to treatment, with a mean age of 59.3 years and female predominance (67.2%, 39/58). The mean HCV RNA was 5.87+0.77 log10 IU/mL; 23 patients (39.7 %) had significant hepatic fibrosis (>F2). In the modified intention-to-treat analysis, the rate of undetectable HCV at week 1, week 2, week 4, week 8 and endof- treatment was 25 % (14/56), 84.8 % (39/46), 100 % (46/46), 100 % (38/38) and 100 % (27/27), respectively. Undetectable HCV RNA were observed in all of the patients with HCV RNA assessable 4 weeks (SVR4, 18/18) and 12 weeks (SVR12, 12/12) post treatment. None of the 18 patients who completed the 12-week treatment experienced relapse during post-treatment follow-up. The most common adverse event was fatigue (78.3 %), followed by pruritus (65.2 %) and dizziness (52.2 %), of which were considered as ribavirin related. None of the participating subjects withdrew treatment or follow-up throughout the trial peroid. Three serious adverse events were reported which included urosepsis, appendicitis and left ureteral stone. All were unrelated to the investigating drugs. Conclusions: 12 weeks of DCV/ASV plus ribavirin was highly effective and safe in HCV-1b patients without NS5A RAS in the interim analysis. The satisfactory results would be anticipated in the full patient set.

      • Enhanced Performance of Pseudo-Bilayer Organic Photovoltaic Devices via Small Molecule Doping

        <P>Controlling both the film crystallinity of the active layer for better charge transport and the interdiffusion between donor and acceptor materials for optimal bicontinuous networks is essential in producing pseudo-bilayer polymer solar cells. In this work, we investigated the influence of a doping solution-processable small molecule with high carrier mobility, 5,11-bis(triethylsilylethynyl) anthradithiophene (TES-ADT), on the performance of pseudo-bilayer polymer solar cells made of an underlayer of poly(3-hexylthiophene) (P3HT) and an upper layer of [6,6]-phenyl-C61-butyric acid methyl ester (PCBM). By analysis of the X-ray diffraction and UV–vis absorbance spectra of P3HT:TES-ADT blend films it was demonstrated that the film crystallinity was enhanced by TES-ADT doping in the P3HT underlayer. The hole mobility extracted from the current density–voltage curves of hole-only devices based on P3HT:TES-ADT demonstrated an optimized value with proper TES-ADT doping and thermal annealing. An intermixed photoactive layer was observed for the annealed device, indicating the occurrence of interdiffusion with a large interfacial area. With improved film crystallinity and interdiffusion, the optimal device performance was obtained when 5% TES-ADT was blended with P3HT and a thermal annealing treatment at 150 °C for 1 min was conducted.       • A Clinical Study on Juheli (Recombinant Human Interleukin - 11) in the Second Prevention of Chemotherapy Induced Thrombocytopenia

        Objective: to investigate the effect and side effects of recombinant human interleukin - 11 (rhIL - 11, in Chinese Juheli, produced by Qi Lu Biotechnology CO., LTD) in the second prevention of chemotherapy induced thrombocytopenia (CIT). Methods: Cancer patients with CIT were recruited and were treated with rhIL - 11 (treatment phase, TP), and in the following cycle, all these patients administered with rhIL - 11 24 hours immediately after chemotherapy (preventive treatment phase, PTP). Duration and severity of thrombocytopenia between two phases were compared. Results: for patients in TP or PTP, nadir values of platelet were ($29.28{\pm}20.08){\times}10^9/L$ and ($45.24{\pm}19.66){\times}10^9/L$, duration of thrombocytopenia in TP and PTP was ($11.52{\pm}4.33$) and ($8.20{\pm}+2.77$)days, recovery time was ($19.40{\pm}3.89$)and ($13.44{\pm}3.02$)days, duration of rhIL - 11 administration was ($10.68{\pm}2.46$)and ($6.28{\pm}1.77$)days, number of patients needing platelet infusion was 16and4 respectively, all differences were statistically significant (p value were 0.007, 0.002, 0.000, 0.000, 0.034 respectively). For TP and PTP, number of patients with hemorrhage was 8 and 4, duration of bleeding was ($5.00{\pm}0.82$) and ($4.50{\pm}0.71$) days respectively, with no statistically significant difference. Adverse reactions mainly included fever, edema, arrhythmia, joint pain, fatigue, skin rash, headache, dizziness, etc., all were not statistically significant between TP and PTP. Conclusion: rhIL - 11 could be well tolerated and is effective that could reduce the duration, severity of CIT, platelet transfusion, and incidence of bleeding, as well as shorten the recovery time, duration of rhIL - 11 administration. Thus, rhIL - 11 could be commended in the second prevention of CIT for patients with cancer.
