        Study on the differential gene expression of elm leaves fed on by Tetraneura akinire Sasaki

        Background To study the essential molecular mechanism of gall formation is very important. Objective To investigate the differential gene expression in leaves fed on by Tetraneura akinire Sasaki and to provide a basis for the better understanding of the essential molecular mechanism of gall formation. Methods The infected leaves of the elm were divided into three periods: initial formation period (T2), growth and differentiation period (T3), and cracking period (T4). The untouched leaves were used as the control (T1). RNA-Seq was performed, and the high-quality sequences were mapped to the reference genome and the elm gene database to obtain the gene expression profiles. The expression level of each gene was calculated by the RPKM method. A combination of FDR ≤ 0.01 and the absolute value of |log2 ratio (T/CK)| ≥ 2 was used as the threshold to determine the significance of gene expression. Finally, GO and pathway enrichment analyses were used to identify the significantly enriched functional classification and metabolic pathways in DEGs. Results The results revealed that approximately 244 mRNAs were detected between T1 and T2, including 192 up-regulated and 52 down-regulated mRNAs; approximately 175 mRNAs were detected between T1 and T3, including 145 up-regulated and 30 down-regulated mRNAs; and approximately 372 mRNAs were detected between T1 and T4, including 360 up-regulated and 12 down-regulated mRNAs. Approximately 34 differentially expressed genes were identified by Venn analysis. Comparing the three infection periods to the control, there were 28 up-regulated and six down-regulated mRNAs. Additionally, 562 genes were used for cluster analysis, which revealed that the gene expression in T2 and T3 changed greatly. Genes related to cell proliferation and respiration, such as microtubulin and 6-phosphoric acid fructose kinase were mainly up-regulated during the T2 period. Genes encoding lipoxygenase, glutathione-S-transferase, superoxide dismutase and protease inhibitor were up-regulated during T2 and T3. Genes encoding lignocellulose synthase were up-regulated during T4, which suggests the reinforcement of the cell wall to improve the resistance to the damage of the Tetraneura akinire Sasaki. Conclusions The results showed that the feeding of Tetraneura akinire Sasaki caused the differential expression of elm genes and influenced cellular energy metabolism. These changes in physiological response and gene expression of the elm compose the physiological and molecular basis of the gall formation and may improve the resistance of elm to Tetraneura akinire Sasaki.

        The effi cacy of daily chlorhexidine bathing for preventing healthcare-associated infections in adult intensive care units

        Background/Aims: Healthcare-associated infections (HAIs) in critically ill patients with prolonged length of hospital stay and increased medical costs. The aim of this study is to assess whether daily chlorhexidine gluconate (CHG) bathing will significantly reduce the rates of HAIs in adult intensive care units (ICUs). Methods: PubMed, EMBASE, and the Cochrane Central Register of Controlled Trials were systematically searched until December 31, 2014 to identify relevant studies. Two authors independently reviewed and extracted data from included studies. All data was analyzed by Review Manager version 5.3. Results: Fifteen studies including three randomized controlled trials and 12 quasi- experimental studies were available in this study. The outcomes showed that daily CHG bathing were associated with significant reduction in the rates of primary outcomes: catheter-related bloodstream infection (risk ratio [RR], 0.44; 95% confidence interval [CI], 0.32 to 0.63; p < 0.00001), catheter-associated urinary tract infection (RR, 0.68; 95% CI, 0.52 to 0.88; p = 0.004), ventilator-associated pneumonia (RR, 0.73; 95% CI, 0.57 to 0.93; p = 0.01), acquisition of methicillin-resistant Staphylococcus aureus (RR, 0.78; 95% CI, 0.68 to 0.91; p = 0.001) and vancomycin-resistant Enterococcus (RR, 0.56; 95% CI, 0.31 to 0.99; p = 0.05). Conclusions: Our study suggests that the use of daily CHG bathing can signifi- cantly prevent HAIs in ICUs. However, more well-designed studies are needed to confirm these findings.

        The effects of the circulating water tunnel wall and support struts on hydrodynamic coefficients estimation for autonomous underwater vehicles

        This paper investigates the influence of the Circulating Water Channel (CWC) side wall and support struts on the hydrodynamic coefficient prediction for Autonomous Underwater Vehicles (AUVs) experiments. Computational Fluid Dynamics (CFD) method has been used to model the CWC tests. The hydrodynamic coefficients estimated by CFD are compared with the prediction of experiments to verify the accuracy of simulations. In order to study the effect of side wall on the hydrodynamic characteristics of the AUV in full scale captive model tests, this paper uses the CWC non-dimensional width parameters to quantify the correlation between the CWC width and hydrodynamic coefficients of the chosen model. The result shows that the hydrodynamic coefficients tend to be constant with the CWC width parameters increasing. Moreover, the side wall has a greater effect than the struts.

        <P>The effects of catalponol (1) on dopamine biosynthesis and l-DOPA-induced cytotoxicity in PC12 cells were investigated. Catalponol at concentration ranges of 1-5 muM increased the intracellular levels of dopamine at 12-48 h. Catalponol at concentrations of up to 10 muM did not alter cell viability. Tyrosine hydroxylase (TH) activity was enhanced by 1 at 3 muM in a time-dependent manner, but aromatic l-amino acid decarboxylase activity was not. Catalponol also increased the intracellular levels of cyclic AMP and TH phosphorylation. In addition, catalponol at 3 muM associated with l-DOPA (20-50 muM) further enhanced the increases in dopamine levels induced by l-DOPA (50-100 muM) at 24 h. Catalponol at 2-5 muM inhibited l-DOPA (100-200 muM)-induced cytotoxicity at 48 h. These results suggest that 1 enhanced dopamine biosynthesis by inducing TH activity and protected against l-DOPA-induced cytotoxicity in PC12 cells, which was mediated by the increased levels of cyclic AMP.</P>

        Anti-Diabetic Effect of Pectinase-Processed Ginseng Radix (GINST) in High Fat Diet-Fed ICR Mice

        In the present study, we investigate anti-diabetic effect of pectinase-processed ginseng radix (GINST) in high fat diet-fed ICR mice. The ICR mice were divided into three groups: regular diet group, high fat diet control group (HFD), and GINST-treated group. To induce hyperglycemia, mice were fed a high fat diet for 10 weeks, and mice were administered with 300 mg/kg of GINST once a day for 5 weeks. Oral glucose tolerance test revealed that GINST improved glucose tolerance after glucose challenge. Compared to the HFD control group, fasting blood glucose and insulin levels were decreased by 57.8% (p<0.05) and 30.9% (p<0.01) in GINST-treated group, respectively. With decreased plasma glucose and insulin levels, the insulin resistance index of the GINST-treated group was reduced by 68.1% (p<0.01) compared to the HFD control group. Pancreas of GINST-treated mice preserved a morphological integrity of islets and consequently having more insulin contents. In addition, GINST up-regulated the levels of phosphorylated AMP-activated protein kinase (AMPK) and its target molecule, glucose transporter 4 (GLUT4) protein expression in the skeletal muscle. Our results suggest that GINST ameliorates a hyperglycemia through activation of AMPK/GLUT4 signaling pathway, and has a therapeutic potential for type 2 diabetes.

        Ginseng Leaf Extract Prevents High Fat Diet-Induced Hyperglycemia and Hyperlipidemia through AMPK Activation

        This study evaluated the protective effects of ginseng leaf extract (GLE) against high fat-diet-induced hyperglycemia and hyperlipidemia, and explored the potential mechanism underlying these effects in C57BL/6J mice. The mice were randomly divided into four groups: normal control, high fat diet control (HFD), GLE-treated at 250 ㎎/㎏, and GLE-treated at 500 ㎎/㎏. To induce hyperglycemic and hyperlipidemic states, mice were fed a high fat diet for 6 weeks and then administered GLE once daily for 8 weeks. At the end of the treatment, we examined the effects of GLE on plasma glucose, lipid levels, and the expression of genes related to lipogenesis, lipolysis, and gluconeogenesis. Both GLE groups lowered levels of plasma glucose, insulin, triglycerides, total cholesterol, and non-esterified fatty acids when compared to those in HFD group. Histological analysis revealed significantly fewer lipid droplets in the livers of GLE-treated mice compared with HFD mice. To elucidate the mechanism, Western blots and RT-PCR were performed using liver tissue. Compared with HFD mice, GLE-treated mice showed higher levels of phosphorylation of AMP-activated protein kinase (AMPK) and its substrate, acetyl-CoA carboxylase, but no differences in the expression of lipogenic genes such as sterol regulatory element-binding protein 1a, fatty acid synthase, sterol-CoA desaturase 1 and glycerol-3-phosphate acyltransferase. However, the expression levels of lipolysis and fatty acid uptake genes such as peroxisome proliferator-activated receptor-α and CD36 were increased. In addition, phospho-α and CD36 were increased. In addition, phosphoenolpyruvate carboxykinase gene expression was decreased. These results suggest that GLE ameliorates hyperglycemia and hyperlipidemia by inhibiting gluconeogenesis and stimulating lipolysis, respectively, via AMPK activation.

        Cavitation Diagnosis Method for Centrifugal Pumps based on Agglomerative Hierarchical Clustering Algorithm

        Cavitation in the pump induces vibration and noise, which leads to the degradation of pump performance and damage to the impeller. Therefore, it is of great significance to accurately identify the cavitation state of the centrifugal pump. To determine the cavitation state using the vibration characteristic frequency of the centrifugal pump, it is necessary to accurately identify the characteristic frequency due to the noise's effect. Therefore, a cavitation fault diagnosis method of centrifugal pump based on a cohesive hierarchy algorithm was proposed. Firstly, Singular Value Decomposition (SVD) is used to de-noise the vibration signal. Then the root means square of the vibration signal after de-noising is obtained as the eigenvalue of the vibration signal. Secondly, the hierarchical clustering algorithm is used to classify the vibration eigenvalues and accurately identify the non-cavitation and cavitation states of centrifugal pumps. Finally, through experimental tests, the method can effectively and quickly identify the cavitation state of the centrifugal pump, with an accuracy of 95%. The study provides a new way for rapidly diagnosing centrifugal pump cavitation
