Inactivated Sendai Virus Strain Tianjin Induces Apoptosis in Human Breast Cancer MDA-MB-231 Cells

Sendai virus strain Tianjin is a novel genotype. Here, we investigate the antitumor and proapoptotic effects of ultraviolet-inactivated Sendai virus strain Tianjin (UV-Tianjin) on human breast cancer MDA-MB-231 cells in vitro, as well as the involvement of the apoptotic pathway in the mechanism of UV-Tianjin-induced antitumor effects. MTT assays showed that treatment with UV-Tianjin dose-dependently inhibited the proliferation of MDA-MB-231 cells but not normal MCF 10A breast epithelium cells. Hoechst staining and flow cytometric analysis revealed that UV-Tianjin induced apoptosis of MDA-MB-231 cells in a dose-dependent manner. Moreover, UV-Tianjin treatment resulted in reduction in the mitochondria membrane potential (MMP) and release of cytochrome complex (cyt c) via regulation of Bax and Bcl-2, as well as activation of caspase-9, caspase-3, Fas, FasL and caspase-8 in MDA-MB-231 cells. In summary, our study suggests that UV-Tianjin exhibits anticancer activity in human breast cancer MDA-MB-231 cells through inducing apoptosis, which may involve both the endogenous mitochondrial and exogenous death receptor pathways.

Severe inflammatory reactions caused by macrophage activation can trigger a systemic immune response. In the present study, we observed the anti-inflammatory properties of hispidin on LPS induced RAW264.7 macrophage cells. Our results showed that hispidin treatment significantly reduced the production of cellular NO, IL-6 and reactive oxygen species (ROS) while has not inhibitory effect on TNF-α productions. Excitingly, hispidin treatment retains the phagocytosis ability of macrophages which enabling them to perform the function of removing foreign invaders. Signaling studies showed, hispidin treatment dramatic suppressed the LPS induced mitogen activated protein kinases (MAPK) and JAK/STAT activations. In conclusion, our findings suggest that hispidin may be a new therapeutic target for clinical treatment of macrophages-mediated inflammatory responses.

AlGaN/GaN heterostructures have been attracting a great deal of interest because of their great potential applications as light-emitting-diodes, high-electron-mobility transistors (HEMTs), and detectors operating in the visible-to-ultraviolet range. The performances of these devices are governed by the electronic properties of the two-dimensional electron gas (2DEG) formed at the interface of AlGaN/GaN heterostructure. In this work, we report transport measurements for an AlGaN/GaN 2DEG as functions of the magnetic field B over a wide range of temperature (4.682 K T 80 K). At the highest measurement temperature of 80 K, the longitudinal resistance is nominally Bindependent, compelling experimental evidence for Drude-Boltzmann-like transport in a 2D system.

The deforming process is complicated and both the end concave and the central defect can be easily formed in multi-step shafts shaped by the cross-wedge rolling technology. To realize the accurate forming of multi-step shafts without stub bar, this article breaks the bondage of traditional flat-end billet, and introduces into convex-end billet. Based on established mechanical models and the damage computation principles, the distribution and change features of stress fields, strain fields and microstructures in different segments of the multistep shaft during the progressive forming process are analyzed, and the location of the central defect is predicted. It is found that the concave depth of shaft ends decreases as the length of the convex-end of billet increases, the microcosmic grains are affected by the section shrinkage of the shaft segments and the larger the section shrinkage is, the smaller the size of the microcosmic grain will be. It records the longest duration of maximum stress, the largest fluctuation of lateral stress and the most frequent cycle of transverse strain in the multistep shaft end and therefore the central defect is most likely to occur. The research findings settle a dependable theoretical basis for enhancing the molding quality and realizing the accurate forming for multi-step shafts in cross wedge rolling.

The intestinal barrier function disrupted in sepsis, while little is known about the variation in different phases of sepsis. In this study, mouse models of sepsis were established by caecal ligation and puncture (CLP). The H&E staining of sections and serum diamine oxidase concentration were evaluated at different timepoint af-ter CLP. TUNEL assay and EdU staining were performed to evaluate the apoptosis and proliferation of intestinal epithelium. Relative protein expression was assessed by Western blotting and serum concentrations of pro-inflammatory cytokines was mea-sured by ELISA. The disruption of intestinal barrier worsened in the first 24 h after the onset of sepsis and gradually recovered over the next 24 h. The percentage of apop-totic cell increased in the first 24 h and dropped at 48 h, accompanied with the prolif-erative rate of intestinal epithelium inhibited in the first 6 h and regained in the later period. Furthermore, the activity of nuclear factor kappa B (NF-κB) presented similar trend with the intestinal barrier function, shared positive correction with apoptosis of intestinal epithelium. These findings reveal the conversion process of intestinal barrier function in sepsis and this process is closely correlated with the activity of NF-κB signaling.
Research on Design of Through-Diaphragm Connections between CFRT Columns and HSS Beams

The through-diaphragm connection detail has been identified as a good choice for attaching HSS beams to concrete-filledrectangular tube columns in engineering applications. In this research program an analytical study was conducted tocomprehend the behavior of this detail and develop the accompanying design guidelines. The experimental results are presentedelsewhere. In this paper two analytical models were established to predict the shear stiffness and yield shear strength of theHSS beam and panel zone, respectively. Theoretical results based on the proposed model well agreed with the experimentaldata. Finally, design provisions were introduced to check the strength in the connections.

This study aims to investigate the possible mechanisms of electroacupuncture (EA) at PC6to improve myocardial ischemia (MI) by regulating the cardiac transient outward potassiumcurrent channel (Ito). According to the random number table, the mice were dividedinto six groups of six mice each: control group, MI group, PC6, LU7 (Lieque-point), ST36(Zusanli-point), and nonacupoint group. Mice in the control group were injected with saline(20 mg/kg, 24 hours interval), and the other ASIC3 / mice were injected subcutaneouslytwice with isoproterenol (ISO) (20 mg/kg, 24 hours interval). In thepreexperiment, 5 mg/kg, 10 mg/kg, 20 mg/kg, and 30 mg/kg of ISO were used, andthe results showed that 5 mg/kg and 10 mg/kg of ISO both could induce acute MI, butshorter duration of sustained MI. On the other hand, an injection of 30 mg/kg can makethe mice experience arrhythmia or die immediately, and EA was operated at PC6, LU7,ST36 acupoints, and nonacupoint in the mice of PC6, LU7, ST36, and nonacupoint groups,respectively, after injecting twice. Then Western blotting techniques (Western Blot) wereused to analyze the protein expressions of Kv1.4, Kv4.2, Kv4.3, and KchIP2. The results ofthis experiment showed that the protein expressions of Kv1.4, Kv4.2, Kv4.3, and KChIP2 inMI group were significantly lower than those in the control group (p < 0.01). Comparedwith MI group, the results of PC6, LU7, and ST36 groups obviously increased (p < 0.05). Furthermore, the expressions of PC6 group were higher than LU7 group and ST36 group(p < 0.05). And electrocardiogram’s T-waves showed obvious pathological changes inthe MI group compared to the control group (p < 0.01). After EA, the abnormal T-waves voltage of ECG in PC6, LU7, and ST36 groups was improved (p < 0.05). In addition, therate change of PC6 group was larger than that of both LU7 and ST36 groups (p < 0.05). But the T-waves voltage of the nonacupoint group was not significantly different than thatof the MI group (p > 0.05).
