      • AFB₁ 대사에서 phloretion의 이중 활성 효과

        Aflatoxm B₁(AFB₁) is a potent hepatocarcinogen in experimental animals and a hazard to human health in several parts of the world. AFB₁ is activated to its ultimate carcinogenic intermediate, AFB₁-8,9-epoxide, by cytochrome P450(CYP) 1A2 and CYP3A4 in human liver and the intermediate is decomposed by several glutathione S-transferase(GST) including GSTA2, GSTM1 and GSTP1. In this study, we investigated the effects of phloretin on the enzyme systems which are involved in the activation and detoxification of AFB₁. The metabolic intermediate of AFB₁ was measured with HPLC. We found that phloretin could strongly inhibit the activities of CYP 3A4 and CYP1A2 in a dose dependent manner. Phloretin induced the antioxidant-response element(ARE)-mediated gene expression, including GSTs. The expressions of GSTA2, T1, M1, and GSTP1 were induced by 10μM phloretin. The decomposition of AFB₁-8,9-epoxide was measured with GSH conjugating activity of the epoxide. The rate was increased to 1.5 fold when HepG2 cells were treated by 10μM phloretin for 12h. In the mean while, the total GST activitives toward CDNB in HepG2 cells were not changed by the treatment with phloretin. The results demonstrate that phloretin has strong chemopreventive effects against AFB₁ toxicity through the inhibition of AFB₁ activation and induction of GSTs.

        Antimicrobial glycoalkaloids from the tubers of Stephania succifera

        Three new glycoalkaloids, N-formyl-asimilobine-2-O-b-D-glucoside (1), (-)-1-O-b-D-glucoside-8-oxotetrahydropalmatine(2), and 1-N-monomethylcarbamateargentinine-3-O-b-D-glucoside (3) were isolated fromtubers of Stephania succifera. The structures were establishedbased on spectroscopic analysis, and the antimicrobialactivities of the three glycoalkaloids are reported.

        Determinants of HMGB Proteins Required To Promote RAG1/2-Recombination Signal Sequence Complex Assembly and Catalysis during V(D)J Recombination

        <B>ABSTRACT</B><P>Efficient assembly of RAG1/2-recombination signal sequence (RSS) DNA complexes that are competent for V(D)J cleavage requires the presence of the nonspecific DNA binding and bending protein HMGB1 or HMGB2. We find that either of the two minimal DNA binding domains of HMGB1 is effective in assembling RAG1/2-RSS complexes on naked DNA and stimulating V(D)J cleavage but that both domains are required for efficient activity when the RSS is incorporated into a nucleosome. The single-domain HMGB protein from <I>Saccharomyces cerevisiae</I>, Nhp6A, efficiently assembles RAG1/2 complexes on naked DNA; however, these complexes are minimally competent for V(D)J cleavage. Nhp6A forms much more stable DNA complexes than HMGB1, and a variety of mutations that destabilize Nhp6A binding to bent microcircular DNA promote increased V(D)J cleavage. One of the two DNA bending wedges on Nhp6A and the analogous phenylalanine wedge at the DNA exit site of HMGB1 domain A were found to be essential for promoting RAG1/2-RSS complex formation. Because the phenylalanine wedge is required for specific recognition of DNA kinks, we propose that HMGB proteins facilitate RAG1/2-RSS interactions by recognizing a distorted DNA structure induced by RAG1/2 binding. The resulting complex must be sufficiently dynamic to enable the series of RAG1/2-mediated chemical reactions on the DNA.</P>

        The Contemporary Value of Xunzi`s Political Ethics Thought for the Social Administration

        Xunzi`s political ethical thought, including gentleman personality, people-oriented, prospering etiquettes and respecting law and so on, still has a relational meaning for the modern social political moral construction. Gentleman personality comes with an obligation to improve the leading officials` morality; the idea of people-oriented has a vital significance for establishing“ put people first“ thought throughout society and insisting the mass line; This statecraft that focuses both on ritual and law has a very important revelation for improving the foundation for anti-corruption and a clean government. Xunzi`s political ethical thought provides the value for our country`s conducting a harmonious and efficient social administration through the three dimensions mentioned above.

      • Implementation of online model updating with ANN method in substructure pseudo-dynamic hybrid simulation

        Substructure pseudo-dynamic hybrid simulation (SPDHS) is an advanced structural seismic testing method which combines physical experiment and numerical simulation. Generally, the key components which display nonlinearity first are taken as experimental substructures for actual test, and the remaining parts are modeled in simulation. Model updating techniques can be effectively applied to enhance the model precision of nonlinear numerical elements. Specifically, the constitutive model of the experimental substructure is identified online by the instantaneously-measured data, and the corresponding numerical elements with similar hysteretic behaviors are updated synchronously. Artificial neural network (ANN) can recognize the system which cannot be represented by definite numerical model, and thus avoids the structural response distortion caused by the inherent numerical model defects. In this study, a framework for online model updating in SPDHS with ANN method is expanded to implement actual test validation. Moreover, the effectiveness of ANN method is demonstrated by practical tests of a two-story frame model with bending dampers. Additionally, the unscented Kalman filter technique and offline ANN identification approach are both examined in the test validation. The experimental results show that, under the identical loading history, the online ANN method can significantly reduce the model errors and improve the accuracy of SPDHS.

        There are many key parts in the field of aerospace with aluminum alloy thin-walled parts, the problem of breaking the low efficiency in production that’s also followed. There will be a lot of deformation in processing because of its poor rigidity, poor process, which will lead to low processing efficiency. It is of great significance to improve the machining efficiency of the aluminum alloy thin-walled parts, and a series of analysis of the surface quality of aluminum alloy thin-walled parts there are carried out clamping, cutting tools, take the knife route so on, and the problems of low machining efficiency and poor surface quality are solved.

        Cadmium induces apoptosis in primary rat osteoblasts through caspase and mitogen-activated protein kinase pathways

        Exposure to cadmium (Cd) induces apoptosis in osteoblasts (OBs); however, little information is available regarding the specific mechanismsof Cd-induced primary rat OB apoptosis. In this study, Cd reduced cell viability, damaged cell membranes and induced apoptosis in OBs. We observed decreased mitochondrial transmembrane potentials, ultrastructure collapse, enhanced caspase-3 activity, and increasedconcentrations of cleaved PARP, cleaved caspase-9 and cleaved caspase-3 following Cd treatment. Cd also increased the phosphorylationof p38-mitogen-activated protein kinase (MAPK), extracellular signal-regulated kinases (ERK)1/2 and c-jun N-terminal kinase (JNK) in OBs. Pretreatment with the caspase inhibitor, N-benzyloxycarbonyl-Val-Ala-Asp-fluoromethylketone, ERK1/2 inhibitor (U0126), p38 inhibitor(SB203580) and JNK inhibitor (SP600125) abrogated Cd-induced cell apoptosis. Furthermore, Cd-treated OBs exhibited signs of oxidativestress protection, including increased antioxidant enzymes superoxide dismutase and glutathione reductase levels and decreased formationof reactive oxygen species. Taken together, the results of our study clarified that Cd has direct cytotoxic effects on OBs, which are mediatedby caspase- and MAPK pathways in Cd-induced apoptosis of OBs.

        Multimode participation of traditional Chinese medicine in the treatment of COVID-19

        Background The outbreak of COVID-19 has swiftly spread across China and all over the world, resulting in severe contagious pneumonia. However, no specific anti-COVID-19 drugs or methods are available for the treatment of this acute and fatal disease. In recent years, as the efficacy and safety of traditional Chinese medicine (TCM) have been universally acknowledged, it has been brought to a crucial status domestically and overseas for the treatment of COVID-19. Methods We searched relevant literature, electronic databases, and official statements, diagnoses and protocols to retrieve studies and applications related to traditional Chinese medicine for COVID-19 in terms of regulations and policies, clinical evidence, preclinical rationale and big data analysis and then summarized the discovery and development of potential drugs and their targets. Results Clinicians, researchers, governments, the public, colleges, institutes and companies collected and classified associated policies, regulations and actual contributions, searched clinical trials and preclinical experimental outcomes from databases, studied potential TCM drugs with possible mechanisms, retrieved numerous big data analysis method and gathered pooled results of compounds along with their effective targets to make traditional Chinese medicine vital to cover all stages of patients in the treatment and control of COVID-19. Conclusion Traditional Chinese medicine provides new evidence to support the clinical value of TCM for COVID-19.         The purpose of this study was to explore the relations LOHAS image, customer"s perceived quality, perceived values and attitude. The study used sampling and selected certain consumers who visited restaurant in China. The study used data collected from 461 restaurant consumers; Of 450 questionnaires, 11unsuitable data were excluded for data analysis. These data were processed using SPSS 20.0. The results of this study are summarized as follows. First, image of LOHAS exhibited a positive impact on perceived quality. Furthermore, the restaurant image of eco-friendly was the best predictors for perceived quality. Second, the factor of perceived quality had the positive effects on perceived value. Third, the factor of perceived quality had the positive effects on the attitude. Finally, the factor of perceived value had the positive effects on the attitude. On the basis of the findings, academic and practical implications were discussed. Results are expected to be valuable as a basic data for establishing various marketing strategies and improving management with consideration of LOHAS image, perceived quality and perceived value in China.

        The purpose of this study was to understand the influence of the physical environment of Small coffee shops on customer satisfaction and revisit intention. The study used sampling and selected certain consumers who visited Small coffee shops in Changchun. The study used data collected from 326 coffee shop consumers; Of 300 questionnaires, 26unsuitable data were excluded for data analysis. These data were processed using SPSS 20.0. The results of this study are summarized as follows. First, it was found that attractive and cleanliness affected customer satisfaction. While attractive was shown to have the largest influence on customer satisfaction, but practical space and convenience had no significant affect on customer satisfaction. Second, attractive and practical space affected the revisit intention. While attractive was shown to have the largest influence on revisit intention, but convenience and cleanliness did not affect visitation to Small coffee shops. Finally, customer satisfaction had significant influences on revisit intention for Small coffee shops. Results are expected to be valuable as a basic data for establishing various marketing strategies for improving management results with consideration of consumers" perceived experience in the Small coffee shop.
