        Bile Acid Transporters Are Expressed and Heterogeneously Distributed in Rat Bile Ducts

        Background/Aims: Cholangiocytes are capable of reabsorbing bile salts from bile, but the pathophysiological significance of this process is unclear. To this end, we detected the expression and distribution of bile acid transport proteins in cholangiocytes from normal rat liver and analyzed the possible pathophysiological significance. Methods: Bile duct tissues of Sprague-Dawley rats were isolated by enzymatic digestion and mechanical isolation, and then divided into large and small bile duct tissues. Immunohistochemistry, real-time polymerase chain reaction and Western blotting were used to determine the expression of the apical sodium-dependent bile acid transporter (ASBT), ileal bile acid binding protein (IBABP), and basolateral organic solute transporter α (Ostα) in the biliary tract system of rats. Differences in the expression and distribution of these proteins were analyzed. Results: In cholangiocytes, ASBT and IBABP were mainly expressed in cholangiocytes of the large bile ducts, in which the expression of both was significantly higher than that in the small ducts (p<0.05). Ostα was simultaneously expressed in cholangiocytes of both the large and small bile ducts, showing no significant difference in expression between the two groups of bile ducts (p>0.05). Conclusions: Bile acid transporters are expressed and heterogeneously distributed in rat bile ducts, indicating that bile acid reabsorption by cholangiocytes might mainly occur in the large bile ducts. These findings may help explore the physiology of bile ducts and the pathogenesis of various cholangiopathies.

        Ischemic postconditioning protects cardiomyocytes against ischemia/reperfusion injury by inducing MIP2

        Cardiomyocytes can resist ischemia/reperfusion(I/R) injury through ischemic postconditioning (IPoC) which is repetitive ischemia induced during the onset of reperfusion. Myocardial ischemic preconditioning up-regulated protein 2 (MIP2) is a member of the WD-40 family proteins, we previously showed that MIP2 was up-regulated during ischemic preconditioning (IPC). As IPC and IPoC engaged similar molecular mechanisms in cardioprotection, this study aimed to elucidate whether MIP2 was up-regulated during IPoC and contributed to IPoC-mediated protection against I/R injury. The experiment was conducted on two models, an $in$ $vivo$ open chest rat coronary artery occlusion model and an $in$ $vitro$ model with H9c2 myogenic cells. In both models, 3 groups were constituted and randomly designated as the sham, I/R and IPoC/hypoxia postconditioning (HPoC) groups. In the IPoC group, after 45 min of ischemia, hearts were allowed three cycles of reperfusion/ischemia phases (each of 30 s duration) followed by reperfusion. In the HPoC group, after 6 h of hypoxia, H9c2 cells were subjected to three cycles of 10 minute reoxygenation and 10 minute hypoxia followed by reoxygenation. IPoC significantly reduced the infarct size, plasma level of Lactate dehydrogenase and creatine kinase MB in rats. 12 h after the reperfusion, MIP2 mRNA levels in the IPoC group were 10 folds that of the sham group and 1.4 folds that of the I/R group. Increased expression of MIP2 mRNA and attenuation of apoptosis were similarly observed in the HPoC group in the $in$ $vitro$ model. These effects were blunted by transfection with MIP2 siRNA in the H9c2 cells. This study demonstrated that IPoC induced protection was associated with increased expression of MIP2. Both MIP2 overexpression and MIP2 suppression can influence the IPoC induced protection.

This paper presents a developed graphical modelling method for super-lift converters and gives a thorough analysis with a consideration of the effects caused by parasitic parameters and diodes' forward voltage drop. The general guidelines for constructing and deriving graphical models are provided for system analysis. By applying it to examples, the proposed method shows the advantages of high convenience and feasibility. Both the circuit simulation and experimental results are given to support the theoretical analysis.

        MicroRNAs: their involvement in fibrosis pathogenesis and use as diagnostic biomarkers in scleroderma

        miRNAs are important post-transcriptional regulators. The aberrant expression of miRNAs is strongly associated with the initiation and progression of pathophysiologic processes in a wide range of human diseases. Scleroderma (systemic sclerosis;SSc) is a highly heterogeneous autoimmune disease that includes the progressive fibrotic replacement of normal tissue architecture in multiple organs. Our previous studies have suggested that SSc skin tissues display a different miRNA expression signature than that found in normal controls. miRNAs with pro- or antifibrotic properties are found to be dysregulated in SSc skin fibrosis. Serum miRNA levels are associated with SSc activity and severity. miRNAs have the potential to be therapeutic targets and serve as biomarkers for SSc diagnosis and assessment of disease state and severity. This review summarizes the SSc miRNA expression signature and the roles of dysregulation of miRNAs in SSc tissues and serum and examines the future therapeutic potential of targeting miRNAs in the management of SSc patients.

      • Influences of Doping Methods on Microstructure and Fracture Toughness of Mo-La Alloys

        Lanthanum oxide was introduced to molybdenum powder by liquid-liquid doping and liquid-solid doping respectively. Mo alloys were prepared by powder metallurgy technology. The size distribution and feature of dopant particles and the fractographs of Mo alloys were investigated by TEM and SEM respectively. The results indicated that liquid-liquid doping method is favorable for refining and dispersing particles uniformly in matrix. Fracture toughness of Mo alloys prepared by liquid-liquid doping showed better results than that of liquid-solid doping. Furthermore, the influences of the size distribution of on properties of Mo alloys was discussed by dislocation pile-up theory.

      • XPC 939A>C and 499C>T Polymorphisms and Skin Cancer Risk: a Meta-analysis

        The xeroderma pigmentosum complementation group C gene (XPC) has been identified as important for repairing UV-related DNA damage. Some subtle changes in this gene may impair repair efficiency and influence susceptibility to human cancers, including skin cancer. Two polymorphisms in XPC, 939A>C (rs2228001) and 499C>T (rs2228000), are considered to have possible associations with the risk of skin cancer, but the reported results have been inconsistent. Here we performed a meta-analysis of the available evidence regarding the relationship between these two polymorphisms and the risk of skin cancer. All relevant studies were searched using PubMed, Embase and Web of Science before February 2012. A total of 8 case-control studies were included in this analysis, and no convincing associations between the two polymorphisms and risk of skin cancer were observed in any of the genetic models. Stratified analyses by skin cancer type also did not detect significant associations in any subgroup. This meta-analysis suggested that the XPC 939A>C and 499C>T polymorphisms may have little involvement in susceptibility to skin cancer.

      • Improving Bricks (kaolin) Performance by adding Incinerator Bottom Ash and Adjusting Kiln Calcination Temperatures

        The bottom ash (BA) is the residuals of the municipal trashes after incinerated in an incinerator plant. The BA is usually collected and landfilled as the final disposal treatment. However, it becomes harder finding locations for Taiwan as an island with a very scarce land for the landfill treatment. Moreover, the amount of the BA recycled was far behind the amount produced in which leading to a hard problem for central and local governments. How to properly recycle or reuse of the BA becomes an urgent need. From the results obtained by many different researchers, the use of the BA as a ceramic material may help improve the strength of related products. In this study, the BA was applied as part of the replacement for kaolin in the manufacture of the brick specimens. To investigate the effects of the kiln temperature on the brick specimens and evaluate the related properties of the specimens, the kiln temperatures were set at 850, 900, and 950℃. The municipal solid waste incineration BA was applied as part of kaolin replacement in the manufacture of bricks. Test results showed that the shrinkage increment of bricks tended to be smooth when the BA replacement was larger than 10%. Moreover, the results of loss on ignition indicated that the organic and inorganic matters and heavy metal were mostly burned off at the kiln temperature of 900℃ or above. The compressive strengths of bricks increased with the increase amount of BA replacement. The study of the micro structure observations showed that the silicification of the brick specimen bodies reduced with the increase amount of the BA replacement. Moreover, the results obtained from XRD indicated that the peak intensities of the SiO<sub>2</sub> in the brick specimens decreased with increase amount of the BA replacement. This study suggested that the optimal amount of BA replacement and the kiln temperature were 20% and 950℃, respectively.
