Background and Purpose This study investigated the contribution of white-matter hyperintensities (WMH) and lacunar infarcts (LI) to the risk of Alzheimer’s disease (AD) in an elderly cohort in China. Methods Older adults who were initially cognitively normal were examined with MRI at baseline, and followed for 5 years. WMH were classified as mild, moderate, or severe, and LI were classified into a few LI (1 to 3) or many LI (≥4). Cognitive function was assessed using the Mini Mental State Examination and the Activities of Daily Living scale. Results Among the 2,626 subjects, 357 developed AD by the end of the 5-year follow-up period. After adjusting for age and other potential confounders, having only WMH, having only LI, and having both WMH and LI were associated with an increased risk of developing AD compared with having neither WMH nor LI. Moderate and severe WMH were associated with an increased risk of developing AD compared with no WMH. Furthermore, patients with many LI had an increased risk of developing AD compared with no LI. Conclusions Having moderate or severe WMH and many LI were associated with an increased risk of developing AD, with this being particularly striking when both WMH and LI were present.

A new quinoline-based acylhydrazone (1) has been synthesized and applied as a fluorescent probe. Probe 1 exhibits high selectivity and sensitivity to Cu2+ with fluorescence “ON-OFF” behavior in HEPES buffered (1‰ DMSO, HEPES 20 mM, pH = 7.4) solution. The on-site generated 1-Cu2+ complex displays excellent selectivity to sulfide ions with fluorescence “OFF-ON” performance through copper displacement approach.

This paper reports the production and characterization of crude xylanase from the newly isolated Humicola sp. Ly01. The highest (41.8 U/ml) production of the crude xylanase was obtained under the optimized conditions (w/v): 0.5% wheat bran, 0.2% KH2PO4, and 0.5% peptone; initial pH 7.0; incubation time 72 h; 30℃; and 150 rpm. A considerable amount of the crude xylanase was induced using hulless barley bran or soybean meal as the carbon source, but a small amount of the enzyme was produced when supplementary urea was used as the nitrogen source to wheat bran. The crude xylanase showed apparent optimal cellulase-free xylanase activity at 60℃ and pH 6.0, more than 71.8% of the maximum xylanase activity in 3.0-30.0% (v/v) ethanol and more than 82.3% of the initial xylanase activity after incubation in 3.0-30.0% (v/v) ethanol at 30℃ for 2 h. The crude xylanase was moderately resistant to both acid and neutral protease digestion, and released 7.9 and 10.9 μmol/ml reducing sugar from xylan in the simulated gastric and intestinal fluids, respectively. The xylooligosaccharides were the main products of the hydrolysis of xylan by the crude xylanase. These properties suggested the potential of the crude enzyme for being applied in the animal feed industry, xylooligosaccharides production, and high-alcohol conditions such as ethanol production and brewing.

Five novel Nocardiopsis strains isolated from hypersaline soils in China were subjected to a polyphasic analysis to determine their taxonomic position. All of the novel isolates could grow on agar plates at NaCl concentrations of up to 18 % (w/v), with optimum growth at 5-8 %. The DNA G+C contents of the novel strains ranged from 67.9 to 73.2 mol%. The morphological and chemotaxonomic characteristics of the isolates matched those described for members of the genus Nocardiopsis. Based on their 16S rRNA gene sequence analysis, DNA-DNA hybridization values and phenotypic characteristics, including the composition of cell-wall amino acids and sugars, menaquinones, polar lipids and cellular fatty acids, the isolates are proposed as representing five novel species of the genus Nocardiopsis. The novel species are proposed as Nocardiopsis gilva sp. nov. [type strain YIM 90087T (=KCTC 19006T=CCTCC AA 2040012T=DSM 44841T)], Nocardiopsis rosea sp. nov. [type strain YIM 90094T (=KCTC 19007T=CCTCC AA 2040013T=DSM 44842T), Nocardiopsis rhodophaea sp. nov. [type strain YIM 90096T (=KCTC 19049T=CCTCC AA 2040014T=DSM 44843T), Nocardiopsis chromatogenes sp. nov. [type strain YIM 90109T (=KCTC 19008T=CCTCC AA 2040015T=DSM 44844T) and Nocardiopsis baichengensis sp. nov. [type strain YIM 90130T (=KCTC 19009T=CCTCC AA 2040016T=DSM 44845T). On the basis of the chemotaxonomic data, the description of the recently described species Nocardiopsis salina Li et al. 2004 is emended.

A 990 bp full-length gene (xynAHJ2) encoding a 329- residue polypeptide (XynAHJ2) with a calculated mass of 38.4 kDa was cloned from Bacillus sp. HJ2 harbored in a saline soil. XynAHJ2 showed no signal peptide, distinct amino acid stretches of glycoside hydrolase (GH) family 10 intracellular endoxylanases, and the highest amino acid sequence identity of 65.3% with the identified GH 10 intracellular mesophilic endoxylanase iM-KRICT PX1-Ps from Paenibacillus sp. HPL-001 (ACJ06666). The recombinant enzyme (rXynAHJ2) was expressed in Escherichia coli and displayed the typical characteristics of low-temperatureactive enzyme (exhibiting optimum activity at 35 o C, 62% at 20 o C, and 38% at 10 o C; thermolability at ≥45 o C). Compared with the reported GH 10 low-temperature-active endoxylanases, which are all extracellular, rXynAHJ2 showed low amino acid sequence identities (<45%), low homology (different phylogenetic cluster), and difference of structure (decreased amount of total accessible surface area and exposed nonpolar accessible surface area). Compared with the reported GH 10 intracellular endoxylanases, which are all mesophilic and thermophilic, rXynAHJ2 has decreased numbers of arginine residues and salt bridges, and showed resistance to Ni 2+ , Ca 2+ , or EDTA at 10 mM final concentration. The above mechanism of structural adaptation for low-temperature activity of intracellular endoxylanase rXynAHJ2 is different from that of GH 10 extracellular low-temperature-active endoxylanases. This is the first report of the molecular and biochemical characterizations of a novel intracellular low-temperatureactive xylanase.

Aim: Apoptosis has been considered as a fundamental component in cancer pathogenesis, and related genetic factors might play an important role in gastric cardiac adenocarcinoma (GCA) genesis. Methods: We conducted a hospital based case.control study to evaluate the genetic effects of functional single nucleotide polymorphisms (SNPs): BCL2 rs17757541 C>G, BCL2 rs12454712 T>C, FAS rs2234767 G>A, FASL/FASLG rs763110 C>T, ERBB2 rs1136201 A>G and VEGFR2/KDR rs11941492 C>T on the development of GCA. A total of 243 GCA cases and 476 controls were recruited for the study and genotypes were determined using a custom-by-design 48-Plex SNPscan$^{TM}$ Kit. Results: The BCL2 rs17757541 C>G polymorphism was associated with increased risk of GCA. However, there was no significant associations with the other five SNPs. Stratified analyses indicated a significantly increased risk of GCA associated with the BCL2 rs17757541 C>G polymorphism among males, older patients and those with a history of smoking or drinking. Conclusion: These findings indicated that the functional polymorphism BCL2 rs17757541 C>G might contribute to GCA susceptibility. However, our results were limited by small sample size. Future larger studies are required to confirm our current findings.
A New Chemosensing Ensemble for Colorimetric Detection of Oxalate in Water
Li-Jun Tang,Ming-Hui Liu 대한화학회 2010 Bulletin of the Korean Chemical Society Vol.31 No.11
To realize highly selective recognition of oxalate in water, a new chemosensing ensemble that behaves highly selective colorimetric recognition of oxalate in water at pH 7.4 has been developed. The ensemble was constructed by a pyrrole containing mononuclear copper complex and chromeazurol S. The ensemble shows a highly selective recognition of oxalate through an obvious color change from blue to yellow upon the addition of oxalate, whereas, other dicarboxylates such as malonate, succinate, fumarate, maleate, glutarate, adipate, phthalate, isophthalate and terephthalate do not induce any noticeable color changes. The oxalate recognition process is not significantly affected by other coexisting dicarboxylate.

In this work, flake-like poly(2,3-dimethylaniline) (P(2,3-DMA)) with enhanced thermal stability andanticorrosive ability was synthesized by in situ polymerization using H2O2/Fe2+ catalytic oxidation system, comparingwith traditional oxidant ammonium persulfate (APS) synthetic method. The structure and morphology of thesamples were characterized Fourier transform infrared (FTIR) spectra, X-ray diffraction (XRD) and field-emissionscanning electron microscope (FESEM). The experimental results demonstrated that using H2O2/Fe2+ catalytic oxidationsystem was more inclined to form the two-dimensional P(2,3-DMA) flakes. The enhancement in thermostabilityand corrosion resistance was attributed to the formation of phenazine-like structures in the polymer chains, which could serve as templates to form the flake-like morphology. In addition, using H2O2/Fe2+ catalytic oxidationsystem is more environmental friendly than the APS method that can avoid ammonium pollution on aquatic life aswell as waters.
