        Feasibility, accuracy and acceptability of self-sampled human papillomavirus testing using careHPV in Cambodia: a cross-sectional study

        Objective: Self-sampled human papillomavirus (HPV) testing is a potential option forcer vical cancer screening, but research is scarce in Cambodia. We evaluated the feasibility,accuracy, and acceptability of self-sampled HPV testing using careHPV. Methods: A cross-sectional study including women aged 20–49 years attending 2 nationalhospitals in the capital city was conducted. Women under went both self-sampling andclinician-sampling of specimens, and were then asked to complete an acceptabilityquestionnaire. The paired samples were analyzed for high-risk HPV by careHPV andgenotyped by polymerase chain reaction (PCR). Results: A total of 375 women were eligible for inclusion. Based on PCR, 78.9% were negativefor HPV in both self and clinician-samples, 9.9% had a complete HPV type match, and 6.1%had all HPV types in clinician-samples also detected in self-samples. In 5.1%, one or more HPVtypes identified in the clinician-samples were missed in self-samples. When using careHPV,the overall agreement between the 2 sampling methods was 95.7% (95% confidence inter val[CI]=95.8–95.6) with good concordance (κ=0.66, 95% CI=0.56–0.76). Nearly 90% of the womenpreferred clinician-sampling over self-sampling, citing greater comfort, ease, and speed. Conclusion: Self-sampled HPV testing using careHPV could be an option for cer vical cancerscreening in Cambodia; however, it requires periodic quality control of handling procedures. In addition, women’s health education regarding the accuracy of self-sampled HPV testingand the importance of follow-up in cases of positive results is needed.

        A phase I dose-finding trial of hyperthermic intraperitoneal docetaxel combined with cisplatin in patients with advanced-stage ovarian cancer

        Objective: To identif y the maximum tolerated dose (MTD) of docetaxel combined with afixed dose of cisplatin (75 mg/m2) delivered as hyperthermic intraperitoneal chemotherapy(HIPEC) in patients with ovarian cancer. Methods: In this phase I trial, a time-to-event Bayesian optimal inter val design was used. Docetaxel was given at a starting dose of 60 mg/m2 and was increased in 5 mg/m2 incrementsuntil the MTD was determined or the maximum dose level of 75 mg/m2 was reached. The dose-limiting toxicity (DLT) rate was set at 25%, with a total sample size of 30 patients. HIPEC wasdelivered immediately following debulking surger y at a target temperature of 43°C for 90 minutes. Results: From August 2022 to November 2022, 30 patients were enrolled. Among thepatients who received a dose of docetaxel ≤65 mg/m2, no DLT was reported. DLTs wereobser ved in one patient who received 70 mg/m2 docetaxel (grade 3 anaemia) and in threepatients who received 75 mg/m2 docetaxel (one case of grade 3 anaemia, one case of grade3 hepatic impairment and one case of grade 4 thrombocytopenia). Patients treated withdocetaxel 75 mg/m2 in combination with cisplatin 75 mg/m2 had an estimated DLT rate of25%, which was the closest to the target DLT rate and was therefore chosen as the MTD. Conclusion: Docetaxel, in combination with a fixed dose of cisplatin (75 mg/m2), can be usedsafely at intraperitoneal doses of 75 mg/m2 in ovarian cancer patients who received HIPEC(43°C, 90 minutes) following debulking surger y.         Expression profiles of circular RNAs in sheep skeletal muscle

        Objective: Circular RNAs (circRNAs) are a newfound class of non-coding RNA in animals and plants. Recent studies have revealed that circRNAs play important roles in cell proliferation, differentiation, autophagy and apoptosis during development. However, there are few reports about muscle development-related circRNAs in livestock. Methods: RNA sequencing analysis was employed to identify and annotate circRNAs from longissimus dorsi of sheep. Reverse transcription followed by real-time quantitative (q) polymerase chain reaction (PCR) analysis verified the presence of these circRNAs. Targetscan7.0 and miRanda were used to analyse the interaction of circRNA-microRNA (miRNA). To investigate the function of circRNAs, an experiment was conducted to perform enrichment analysis hosting genes of circRNAs using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) pathways. Results: About 75.5 million sequences were obtained from RNA libraries of sheep skeletal muscle. These sequences were mapped to 729 genes in the sheep reference genome. We identified 886 circRNAs, including numerous circular intronic RNAs and exonic circRNAs. Reverse transcription PCR (RT-PCR) and DNA sequencing analysis confirmed the presence of several circRNAs. Real-Time RT-PCR analysis exhibited resistance of sheep circRNAs to RNase R digestion. We found that many circRNAs interacted with muscle-specific miRNAs involved in growth and development of muscle, especially circ776. The GO and KEGG enrichment analysis showed that hosting genes of circRNAs was involved in muscle cell development and signaling pathway. Conclusion: The study provides comprehensive expression profiles of circRNAs in sheep skeletal muscle. Our study offers a large number of circRNAs to facilitate a better understanding of their roles in muscle growth. Meanwhile, we suggested that circ776 could be analyzed in future study.

        Curcumin Inhibits the Proliferation, Migration, Invasion, and Apoptosis of Diffuse Large B-Cell Lymphoma Cell Line by Regulating MiR-21/VHL Axis

        Purpose: Curcumin exerts its anti-cancer effects, partly by targeting special microRNAs, in human cancers. MiR-21 is a key oncomirin carcinogenesis of multiple human cancers. Here, we aimed to further explore the mechanistic insight into the link betweencurcumin and miR-21 on diffuse large B-cell lymphoma (DLBCL). Materials and Methods: Quantitative real-time PCR assays were performed to assess the levels of miR-21 and Von Hippel-Lindau(VHL) mRNA. In situ hybridization assay was used for miR-21 expression visualization in lymphoma tissues. Western blot was usedfor determination of VHL protein, Ki-67, caspase-3, and cleaved caspase-3 levels. Dual-luciferase reporter assay and RNA immunoprecipitationassay were employed to confirm the direct target of miR-21. MTT assay, flow cytometric analysis, and transwell assaywere used to evaluate cell proliferation, apoptosis, and migration and invasion capacities, respectively. Results: Curcumin repressed the proliferation, migration, and invasion abilities and promoted apoptosis in SU-DHL-8 cells. Curcumininhibited miR-21 expression and curcumin exerted its anti-proliferation, anti-migration, anti-invasion, and pro-apoptosiseffects by miR-21 in SU-DHL-8 cells. VHL was a direct target of miR-21. Moreover, curcumin exerted its regulatory effects on SUDHL-8 cells by VHL. Conclusion: Curcumin exerted its anti-proliferation, anti-migration, anti-invasion, and pro-apoptosis functions, at least partly, byrepressing miR-21 and regulating VHL expression in DLBCL cell line. Our findings provided a possible molecular mechanism ofcurcumin-mediated anti-cancer effect.
