        Sevoflurane induces neuronal apoptosis via enhancing DNMT3L expression and promoting methylation of PSD95 promoter in postoperative cognitive dysfunction

        Background Postoperative cognitive dysfunction (POCD) is a central nervous system (CNS) complication and occurs quite frequently after anesthesia, especially in elderly patients. DNA (cytosine-5)-methyltransferase 3-like (DNMT3L) is an enzymatically inactive regulatory factor that can modulate gene expression via methylation. Few studies specifically focused on the role of DNMT3L in sevoflurane-induced POCD. Objective The cognitive dysfunction was determined by Morris water maze assay. Hippocampal neuron apoptosis was measured by TdT-mediated dUTP nick-end labeling (TUNEL) assay. Neuron proliferation and apoptosis were measured by cell counting kit-8 (CCK-8) and flow cytometry assay. The expression levels of DNMT3L and postsynaptic density protein-95 (PSD95) were measured by qRT-PCR and western blot. Results The sevoflurane-induced POCD aging rat model was established for in vivo study. Suppression of DNA methylation by 5-Aza-2′-deoxycytidine (5-AZA; a widely used methylation inhibitor) reversed sevoflurane-induced cognitive dysfunction and hippocampal neuron apoptosis in rats. Moreover, sevoflurane enhanced DNMT3L expression and induced methylation of PSD95 promoter in vivo. In vitro, sevoflurane induced neuronal apoptosis and DNMT3L expression to promote PSD95 methylation. Finally, rescue experiments indicated that sevoflurane induced neuron apoptosis by increasing DNMT3L expression and promoting methylation of PDS95 in POCD. Conclusion Sevoflurane induced neuronal apoptosis by increasing DNMT3L expression and promoting methylation of PDS95 in vivo and in vitro, providing a novel therapeutic strategy for POCD.

        Sweet Olive Extract Attenuates Dextran Sulfate Sodium-Induced Colitis in Mice

        Sweet olive (Osmanthus fragrans flowers) is used to treat dysentery and reduce phlegm and stasis in traditional Chinese medicine. Recently, we found that verbascoside, the major component in the sweet olive ethanolic extract (OFE), inhibited IL-8 secretion in human colorectal adenocarcinoma WiDr cells. However, evidence-based treatment of inflammatory bowel disease (IBD) with the extract is yet to be performed. To evaluate the therapeutic effect of OFE, we measured IL-8 suppression by OFE and verbascoside in a WiDr cell culture assay. In the IL-8 secretion assay, both OFE (100 μg/mL) and verbascoside (10 μM) significantly inhibited IL-8 production in WiDr cells. Furthermore, we designed cotreated (dextran sulfate sodium [DSS]+OFE-treated) and post-treated (DSS–OFE-treated) protocols to access the therapeutic effects of OFE in vivo. Mice treated with 500 mg/kg per day OFE exhibited significant improvement in IBD symptoms, including disease activity index score, body weight, and colon length maintenance. The suppressive effects on myeloperoxidase expression and lower histopathology scores (including neutrophil infiltration) for the colon were also found. These findings suggest that OFE exerts anti-inflammatory effect on DSS-induced colitis.

      • Proteomic Analysis of the Aging-related Proteins in Human Normal Colon Epithelial Tissue

        In order to screen the aging related proteins in human normal colon epithelia, the comparative proteomics analysis was applied to get the two-dimensional electrophoresis (2-DE) profiles with high resolution and reproducibility from normal colon epithelial tissues of young and aged people. Differential proteins between the colon epithelia of two age groups were found with PDQuest software. The thirty five differential protein-spots were identified by peptide mass fingerprint (PMF) based on matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) and database searching. Among them there are sixteen proteins which are significantly up-regulated in the colonic mucosal epithelia of young people group, which include ATP synthase beta chain, electron transfer flavoprotein alpha-subunit, catalase, glutathione peroxidase 1, annexin A2 and heat shock cognate 71 kDa protein, etc.; There are nineteen proteins which are significantly up-regulated in the colonic mucosal epithelia of aged people group, which include far upstream element-binding protein 1, nucleoside diphosphate kinase B, protein disulfide-isomerase precursor and VDAC-2, etc.. The identified differential proteins appear to be involved in metabolism, energy generation, chaperone, antioxidation, signal transduction, protein folding and apoptosis. The data will help to understand the molecular mechanisms of human colon epithelial aging.

        Synthesis and Electrochemical Properties of LiV3O8/PAn Composite as a Cathode Material for Lithium Secondary Batteries

        A LiV3O8/polyaniline (PAn) composite was prepared by the in-situ polymerization method assisted by sodium dodecyl sulfate and ammonium persulfate. The as-prepared powders were investigated by XRD, SEM, and galvanostatic discharge/charge analysis. It was found that the introduction of PAn to LiV3O8 can effectively buffer the mechanical stress and restrain the number of phase changes of composite material during the electrochemical cycling. Compared with pristine LiV3O8, LiV3O8/PAn composite maintains a reversible capacity of 212.1 mAh g−1 at the current density of 30 mA g−1 after 50 cycles, approximately 22.6%, much higher than the former.

      • Association between Pax8-PPARγ1 Rearrangement and Follicular Thyroid Cancer: a Meta-Analysis

        Background: Pax8 and peroxisome proliferator-activated receptor gamma 1 gene (Pax8-$PPAR{\gamma}1$) are important factors in tumors. Several studies have suggested that follicular thyroid cancer may arise from Pax8- $PPAR{\gamma}1$ rearrangement. In order to have a better understanding of the association between Pax8-$PPAR{\gamma}1$ rearrangement and follicular thyroid cancer, we conducted the presenmt meta-analysis. Materials and Methods: The information was extracted from PubMed, EMBASE and Web of Science. Statistic analysis was performed with Stata12.0 software. Odds ratios (ORs) were calculated using a fixed-effects model. We also performed heterogeneity and publication bias analyses. Results: Nine studies including 198 follicular thyroid cancer patients and 268 controls were considered eligible. The frequency of Pax8-$PPAR{\gamma}1$ rearrangement was significantly higher in the follicular thyroid cancer group than in the control group, with a pooled OR of 6.63 (95%CI=3.50-12.7). In addition, through subgroup analysis, the OR between Pax8-$PPAR{\gamma}1$ rearrangement and follicular thyroid cancer was 6.04 (95%CI = 3.18-11.5) when using benign tumor tissues as controls. The OR for the method subgroup was 9.99 (95% CI =4.86-20.5) in the RT-PCR. Conclusions: The final results demonstrated that Pax8-$PPAR{\gamma}1$ rearrangement has significant association with follicular thyroid cancer.

        Co3(PO4)2-Coated LiV3O8 as Positive Materials for Rechargeable Lithium Batteries

        Co3(PO4)2-coated LiV3O8 has been successfully synthesized and used as positive material for rechargeable lithium batteries by a facile liquid phase method. The as-prepared powders were characterized by x-ray diffraction (XRD), scanning electron microscope (SEM), transmission electron microscope (TEM), and the galvanostatic discharge/charge experiments. As-prepared Co3(PO4)2-coated LiV3O8 forms a good layered structure with a poor cyrstallinity. SEM reveals that Co3(PO4)2-coated LiV3O8 has uniform particle distribution and reduced particle size when compared with bare one. The Co3(PO4)2 coating layer is about 33 - 59 nm forming a continuous lumps attached to LiV3O8 particle surface. Co3(PO4)2-coated LiV3O8 electrode shows increased capacity and more stable cycling. The first and 35th discharge capacities of the Co3(PO4)2-coated LiV3O8electrode are 322.8 mAh g−1 and 235.7 mAh g−1 in the range of 4.0 - 1.8 V at a current rate of 30 mA g−1, respectively. The improved electrochemical performance is assigned to the greatly reduced LiV3O8 particle with uniform morphology. Co3(PO4)2-coating further benefits the phase transitions of LiV3O8 during discharge/charge while preventing parasite reactions between electrode surface and electrolyte.

        Spontaneous Sm<sup>3+</sup>→Sm<sup>2+</sup> reduction ability of MAl<sub>2</sub>Si<sub>2</sub>O<sub>8</sub> (M = Sr, Ba): Sm

        <P><B>Abstract</B></P> <P>In air atmosphere, the spontaneous reduction abilities of Sm<SUP>3+</SUP>→Sm<SUP>2+</SUP> in Sm ions doped monoclinic SrAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (SASO) and BaAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (M-BASO) as well as hexagonal BaAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (H-BASO) are investigated. Sm ions doped MAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (M = Sr, Ba) samples were synthesized using Pechini-type sol-gel technology and sintered in air atmosphere and reducing condition, respectively. By comparing the photoluminescence properties of Sm doped MAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (M = Sr, Ba) calcined in air with that in reducing condition, the spontaneous reduction of Sm<SUP>3+</SUP>→Sm<SUP>2+</SUP> can be detected only in the host lattice H-BASO. The essential that makes the reduction of Sm<SUP>3+</SUP> to Sm<SUP>2+</SUP> was investigated by calculating their chemical bonds parameter quantitatively. The important key environmental factor of Ba<SUP>2+</SUP> in H-BASO is the largest among the three samples MAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB>. It indicates that the hexagonal H-BASO sample is the easiest one to realize the spontaneous reduction of Sm<SUP>3+</SUP>→Sm<SUP>2+</SUP> among Sm doped MAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (M = Sr, Ba) samples. The needed energy transferring one electron of the ligand to the central ions Sm<SUP>3+</SUP> in H-BASO:Sm is the lowest. The theoretical analysis agrees well with the experimental result.       • Expression of Endogenous Hypoxia Markers in Vulvar Squamous Cell Carcinoma

        Objective: To investigate the expression of endogenous hypoxia-related markers identified as being involved in vulvar squamous cell carcinoma (VSCC). Methods: We performed immunohistochemical staining of hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$), glucose transporter-1 (GLUT-1), carbonic anhydrase 9 (CA-9) and vascular endothelial growth factor (VEGF), on tissue sections of 25 VSCC patients, 10 vulvar intraepithelial neoplasia (VIN) patients and 12 healthy controls. Results: HIF-$1{\alpha}$ expression was found in all sections, with no significant difference between controls, VIN and VSCC sections (all P<0.05). Glut-1 expression was found in 25% of control, 90% of VIN and 100% of VSCC sections. A significant difference between control and VIN or VSCC was observed (all P<0.05), while no difference was found between VIN and VSCC sections (P>0.05). CA-9 expression was negative in control sections, but it was found in 30% of VIN sections and 52% of VSCC sections with strong staining. Similarly, CA-9 expression also showed obvious differences between controls and VIN or VSCC sections (all P<0.05). However, there was no significant difference between VIN and VSCC (P>0.05). There were only 25% of control sections with weak VEGF expression, while strong staining was found in about 60% of VIN sections and 25% of VSCC sections (all P<0.05). In addition, a difference was also found between VIN and VSCC sections (P<0.05). Conclusion: Expression of endogenous hypoxia markers (HIF-$1{\alpha}$, GLUT-1, CA-9 and VEGF) might be involved in the malignant progression of VSCC.

        Combined Assessment of Serum Alpha-Synuclein and Rab35 is a Better Biomarker for Parkinson’s Disease

        Background and Purpose It is essential to develop a reliable predictive serum biomarker for Parkinson’s disease (PD). Te accumulation of alpha-synuclein (αSyn) and up-regulated expression of Rab35 participate in the etiology of PD. Te purpose of this investigation was to determine whether the combined assessment of serum αSyn and Rab35 is a useful predictive biomarker for PD. Methods Serum levels of αSyn or Rab35 were determined in serum samples from 59 sporadic PD patients, 19 progressive supranuclear palsy (PSP) patients, 20 multiple system atrophy (MSA) patients, and 60 normal controls (NC). Receiver operating characteristics (ROC) curves were calculated to determine the diagnostic accuracy of αSyn or/and Rab35 in discriminating PD patients from NC or atypical parkinsonian patients. Results The levels of αSyn and Rab35 were increased in PD patients. The serum level of Rab35 was positively correlated with that of αSyn in PD patients. Compared to analyzing αSyn or Rab35 alone, the combined analysis of αSyn and Rab35 produced a larger area under the ROC curve and performed better in discriminating PD patients from NC, MSA patients, or PSP patients. When age was dichotomized at 55, 60, 65, or 70 years, the combined assessment of αSyn and Rab35 for classifying PD was better in the group below the cutof age than in the group above the cutof age. Conclusions Combined assessment of serum αSyn and Rab35 is a better biomarker for discriminating PD patients from NC or atypical parkinsonian patients, and is a useful predictive biomarker for younger sporadic PD patients.
