      • UHRF2 mRNA Expression is Low in Malignant Glioma but Silencing Inhibits the Growth of U251 Glioma Cells in vitro

        UHRF2 is a member of the ubiquitin plant homeo domain RING finger family, which has been proven to be frequently up-regulated in colorectal cancer cells and play a role as an oncogene in breast cancer cells. However, the role of UHRF2 in glioma cells remains unclear. In this study, we performed real-time quantitative PCR on 32 pathologically confirmed glioma samples (grade I, 4 cases; grade II, 11 cases; grade III, 10 cases; and grade IV, 7 cases; according to the 2007 WHO classification system) and four glioma cell lines (A172, U251, U373, and U87). The expression of UHRF2 mRNA was significantly lower in the grade III and grade IV groups compared with the noncancerous brain tissue group, whereas its expression was high in A172, U251, and U373 glioma cell lines. An in vitro assay was performed to investigate the functions of UHRF2. Using a lentivirus-based RNA interference (RNAi) approach, we down-regulated UHRF2 expression in the U251 glioma cell line. This down-regulation led to the inhibition of cell proliferation, an increase in cell apoptosis, and a change of cell cycle distribution, in which S stage cells decreased and G2/M stage cells increased. Our results suggest that UHRF2 may be closely related to tumorigenesis and the development of gliomas.

        Effect of meridional shape on performance of axial-flow fan

        In this study, the effect of impeller meridional shape on the performance of axial-flow fan is investigated by CFD method. Three axialflow fan impellers with different meridional shapes are designed. The blade angle, blade stacking condition and other structure factors of the impellers are all remained consistent. The performance curves of the three impellers are calculated and compared. In almost all the interested flowrate range, the impeller W3 with an inverted-isosceles-trapezoid meridional shape and the longer blade camber achieves both the higher pressure rise and the higher efficiency than the other two impellers. A two-stage axial-flow fan designed on basis of W3 is manufactured and tested. Test results show good agreement with the calculated performance curves. Further, analyses of the CFD results are conducted to reveal the reasons for the different performance. A newly-defined Local Euler head (LEH) is introduced to represent the distribution of the major Euler work in the axial-flow fan. And the LEH distributions in the three impellers are obtained. W3 achieves the highest LEH at blade Trailing edge (TE), because it could perform the most Euler work to the fluid with the longest blade camber. Then losses in the impellers are analyzed by means of the entropy generation. Among the losses in impeller, the tip leakage loss and endwall friction loss are dominated at design flowrate. The generation condition of the tip leakage loss shows significant differences among the three impellers. And the whole power loss in impeller of W3 is slightly higher than those of the other two models. However, the power loss difference among the three impellers is negligible. And due to the highest shaft power, the efficiency loss of W3 is the lowest of all.

      • The Study of Mobile Teaching System Based on Micro-Lecture: JAVA Flipped Classroom for Example

        With the development of wireless technology, education services through the mobile terminal are emerging. The research on mobile education is getting more and more attention in university. In view of the particular group of college students, this paper combines mobile education with a specific course- JAVA Programming as example, and takes advantage of the new "Flipped Classroom” teaching mode to study how to carry out mobile teaching activity with the use of Micro-Lecture resources on mobile platform. In this article, composition of the mobile teaching system, teaching process and mobile teaching resource construction were also discussed. Teaching practice shows that the study has a positive role in improving the effect of programming teaching and it also be used for reference to develop china’s mobile education.

      • Differential Protein Expression in EC304 Gastric Cancer Cells Induced by Alphastatin

        Objective: To explore the differential protein expression profile in EC304 gastric cancer cells induced by alphastatin. Methods: Cultured EC304 cells in the exponential phase of growth were randomly divided into alphastatin and control groups. Total proteins were extracted and the two dimensional electrophoresis (2-DE) technique was applied to analyze differences in expression with ImageMaster 2D Platinum 5.0 software. Proteins were identified using the MASCOT database and selected differently expressed proteins were characterised by western blotting and immunofluorescence. Results: $1350{\pm}90$ protein spots were detected by the ImageMaster software in the 2-DE gel images from the control and alphastatin groups. The match rate was about 72-80% for the spectrum profiles, with 29 significantly different protein spots being identified, 10 upregulated, 16 downregulated, two new and one lost. The MASCOT search scores were 64-666 and the peptide matching numbers were 3-27 with sequence coverage of 8-62%. Twenty-three proteins were checked by mass spectrometry, including decrease in Nm23 and profilin-2 isoform b associated with the regulation of actin multimerisation induced by extracellular signals. Conclusion: The proteome in EC304 cells is dramatically altered by alphastatin, which appears to play an important role in modulating cellular activity and anti-angiogenesis by regulating protein expression and signal transduction pathways through Nm23 and profilin-2 isoform b, providing new research directions for anti-angiogenic therapy of gastric cancer.

        Prevalence of Spina Bifida Occulta and Its Relationship With Overactive Bladder in Middle-Aged and Elderly Chinese People

        Purpose: To investigate the prevalence of spina bifida occulta (SBO) and its relationship with the presence of overactive bladder (OAB) in middle-aged and elderly people in China. Methods: A cross-sectional community-based survey was carried out at 7 communities in Zhengzhou City, China from December 15, 2013 to June 10, 2014, where residents aged over 40 years were randomly selected to participate. All of the participants underwent lumbosacral radiographic analysis and relevant laboratory tests. A questionnaire including basic information, past medical history and present illness, and the OAB symptom score was filled out by all participants. Chi-square tests and logistic regression were used for data analysis with a P-value of <0.05 denoting statistical significance. Results: A total of 1,061 subjects were qualified for the final statistical analysis (58.8±11.7 years; male, 471 [44.4%]; female, 590 [55.6%]). The overall prevalence of SBO was 15.1% (160 of 1,061): 18.3% (86 of 471) in men and 12.5% (74 of 590) in women. Among these subjects, 13.7% (145 of 1,061) had OAB: 13.2% (62 of 471) in men and 14.1% (83 of 590) in women. The results of logistic regression showed that age, SBO, history of cerebral infarction (HCI), and constipation were risk factors for OAB (P<0.05), while sex, history of childhood enuresis (HCE), body mass index (BMI), and diabetes mellitus (DM) were not (P>0.05). In men, age, SBO, and constipation were risk factors for OAB (P<0.05), while HCE, BMI, DM, HCI, and benign prostate hyperplasia were not (P>0.05). In women, age, SBO, and HCI were risk factors for OAB (P<0.05), while HCE, BMI, DM, vaginal delivery, and constipation were not (P>0.05). Conclusions: The prevalence of SBO is high and it is related to OAB in middle-aged and elderly people in China.

        First principles calculation of boron diffusion in fcc-Fe

        The diffusion mechanism of boron in fcc-Fe was studied by first-principles calculations. The sites where B atoms tend to occupy and the diffusion behavior were calculated. Results indicated that the main mechanism of boron diffusion in fcc-Fe was the B–monovacancy complex mechanism instead of the interstitial mechanism. The diffusion coefficient D1 of the B–monovacancy complex mechanism was calculated without considering the backward jump of the B atoms. The calculated D1=1.26×10−4×exp(–2.01eV/kBT) m2·s−1 is consistent with the reported results from experiments.

        Accurate delivery of pristimerin and paclitaxel by folic acid-linked nano-micelles for enhancing chemosensitivity in cancer therapy

        Chemoresistance remains a huge challenge for effective treatment of non-small cell lung cancer (NSCLC). Previous studies have shown Chinese herbal extracts possess great potential in ameliorating tumor chemoresistance, however, the efficacy is clinically limited mainly because of the poor tumor-targeting and in vivo stability. The construction of nano-delivery systems for herbal extracts has been shown to improve drug targeting, enhance therapeutic efficacy and reduce toxic and side effects. In this study, a folic acid (FA)-modified nano-herb micelle was developed for codelivery of pristimerin (PRI) and paclitaxel (PTX) to enhance chemosensitivity of NSCLC, in which PRI could synergistically enhance PTX-induced growth inhibition of A549 cancer cell. PTX was firstly grafted with the FA-linked polyethylene glycol (PEG) and then encapsulated with PRI to construct the PRI@FA-PEG-PTX (P@FPP) nano-micelles (NMs), which exhibited improved tumor-targeting and in vivo stability. This active-targeting P@FPP NMs displayed excellent tumor-targeting characteristics without obvious toxicity. Moreover, inhibition of tumor growth and metastasis induced by P@FPP NMs were significantly enhanced compared with the combined effects of the two drugs (PRI in combination of PTX), which associated with epithelial mesenchymal transition inhibition to some extent. Overall, this active-targeting NMs provides a versatile nano-herb strategy for improving tumor-targeting of Chinese herbal extracts, which may help in the promotion of enhancing chemosensitivity of NSCLC in clinical applications.

      • Identification of Homer1 as a Potential Prognostic Marker for Intrahepatic Cholangiocarcinoma

        Background: The aim of the present study was to analyze whether Homer1 is a potential prognostic marker for intrahepatic cholangiocarcinoma (ICC). Materials and Methods: The expression of Homer1 in ICC tissue was detected with immunohistochemistry and levels of protein in ICC and paratumor tissues were evaluated by Western blotting. Survival analysis by the Kaplan-Meier method was performed to assess prognostic significance. Results: Homer1 expression was high in 67.4% (58/86) of ICC samples, and there was significant difference between ICC and adjacent noncancerous tissues (p<0.001); high expression was associated with poor histologic differentiation (p=0.019), TNM stage (p=0.014), lymph node metastasis (p=0.040), and lymphatic invasion (p=0.025). On Kaplan-Meier analysis, a comparison of survival curves of low versus high expressors of Homer1 revealed a highly significant difference in OS (p=0.001) and DFS (p=0.006), indicating that high expression of Homer1 was linked with a worse prognosis. Multivariate analyses showed that Homer1 expression was an independent risk factor predicting overall survival[Hazard ratio(HR), 7.52; 95% confidence interval (CI), 2.63-21.47; p=0.002] and disease-free survival (HR, 11.56; 95%CI, 5.17-25.96; p<0.001) in ICC. Conclusions: Homer1 promotes lymphatic invasion and associates with lymph node metastasis and poor prognosis of ICC. The current study shows that Homer1 may be an independent prognostic factor for ICC patients after curative resection, and it provides an important basis for screening/treating high-risk patients.

      • Therapeutic Effects and Adverse Drug Reactions are Affected by Icotinib Exposure and CYP2C19 and EGFR Genotypes in Chinese Non-Small Cell Lung Cancer Patients

        Background: The aim of this study was to evaluate how CYP2C19 affects icotinib and metabolite' exposure, and to determine whether the exposure and EGFR genotype influences survival time, tumor metastasis and adverse drug reactions. Materials and Methods: 274 NSCLC patients who accepted 125mg icotinib/t.i.d. were chosen from a phase III study. Blood samples were obtained in $672^{nd}$ ($4^{th}$ week) and $1,680^{th}$ hours ($10^{th}$ week), and plasma was used to quantify the concentration of icotinib and blood cells were sampled to check the genotypes. Clinical data were also collected at the same time, including EGFR genotypes. Plasma concentrations were assessed by HPLC-MS/MS and genotype by sequencing. All data were analyzed through SPSS 17.0 and SAS 9.2. Results: CYP 2C19 genotypes affected bio-transformation from icotinib to M24 and M26, especially in poor-metabolisers. Higher icotinib concentrations (>1000 ng/mL) not only increased patient PFS and OS but also reduced tumor metastasis. Patients with mutant EGFR experienced a higher median PFS and OS (234 and 627 days), especially those with the 19del genotype demonstrating higher PR ratio. Patients who suffered grade II skin toxicity had a higher icotinib exposure than those with grade I skin toxicity or no adverse effects. Liver toxic reactions might occur in patients with greater M20 and M23 plasma concentrations. Conclusions: CYP2C19 polymorphisms significantly affect icotinib, M24 and M26 exposure. Patients with mutant EGFR genotype and higher icotinib concentration might have increased PFS and OS and lower tumor metastasis. Liver ADR events and serious skin effects might be respectively induced by greater M20, M23 and icotinib concentrations.

        Effects of Supplementary Chinese Milk Vetch Silage and Rapeseed Meal on the Performance and Rumen Fermentation of Lambs Given Ammoniated Rice Straw Based Diet

        This study was proposed to investigate effects of inclusion of Chinese milk vetch silage (MVS) and rapeseed meal (RSM) on the growth and rumen fermentation of Hu-sheep. Fifty weanling lambs were randomly divided into five equal groups and offered ammoniated rice straw (ABRS) ad libitum along with 100 g concentrate (Trial 1). The animals in $T_0$, $T_1$, $T_2$, $T_3$ and $T_4$ group were respectively supplemented with MVS at levels of 0, 0, 7, 14 or 21% and with RSM at levels of 0, 15, 10, 5 or 0%. Daily gain of lambs was significantly (p<0.05) higher in $T_1$, $T_2$ and $T_3$ group than that in $T_0$ and $T_4$ group. Feed conversion ratio was greatly reduced in supplemented groups as compared with $T_0$ group. In trial 2, five sheep with rumen cannulae were used in a $5{\times}5$ Latin square design. The experimental treatments were as described in Trial 1, but without concentrate. The intake of AVRS was significantly (p<0.05) lower in $T_4$ group than that in $T_0$ group, and also significantly (p<0.05) lower than those in $T_1$ and $T_2$ group. Little difference among all treatments was found in 48h DM degradability of ABRS, MVS and RSM, and in rumen pH value and microbial protein concentration. Rumen concentrations of individual and total VFA tended to be higher in supplemented groups than those in $T_0$ group.These rusults suggest that supplementation with RSM or RSM plus MVS can effectively improve the performance of lambs, and may fail to influence markedly the rumen digestion of ABRS and rumen environments.
