        MiR-182-5p Knockdown Targeting PTEN Inhibits Cell Proliferation and Invasion of Breast Cancer Cells

        Purpose: Breast cancer (BC) is one of the most common malignant tumors, affecting a significant number of women worldwide. MicroRNAs (miRNAs) have been reported to play important roles in tumorigenesis. The aim of this study was to determine theroles of miR-182-5p in BC progression. Materials and Methods: The expressions of miR-182-5p and phosphatase and tensin homolog deleted on chromosome 10(PTEN) were measured in BC tissues and cells by quantitative real-time polymerase chain reaction or Western blot. Cell proliferationand invasion were detected by cell counting kit-8 assay and trans-well assay, respectively. The interaction between miR-182-5p and PTEN was probed by bioinformatics analysis, luciferase activity, and RNA immunoprecipitation. A murine xenograftmodel was established to investigate the role of miR-182-5p in BC progression in vivo. Results: An abundance of miR-182-5p was noted in BC tissues and cells. High expression of miR-182-5p was associated with poorsurvival. Abrogation of miR-182-5p inhibited cell proliferation and invasion in BC cells. Interestingly, PTEN was indicated as atarget of miR-182-5p, and its restoration reversed miR-182-5p-mediated promotion of proliferation and invasion of BC cells. Moreover, depletion of miR-182-5p suppressed tumor growth via up-regulating PTEN expression in the murine xenograft model. Conclusion: MiR-182-5p exhaustion blocked cell proliferation and invasion by regulating PTEN expression, providing a noveltherapeutic avenue for treatment of BC.

        Design and Implementation of a Universal System Control Strategy Applicable to VSC-HVDC Systems

        This paper proposes a universal system control strategy for voltage source converter (VSC) based high voltage direct current (HVDC) systems. The framework of the designed control strategy consists of five layer structures considering the topology and control characteristics of the VSC-HVDC system. The control commands sent from the topmost layer can be transmitted to the next layer based on the existing communication system. When the commands are sent to each substation, the following transmission of commands between the four lower layers are realized using the internal communication system while ignoring the communication delay. This hierarchical control strategy can be easily applied to any VSC-HVDC system with any topology. Furthermore, an integrated controller for each converter is designed and implemented considering all of the possible operating states. The modular-designed integrated controller makes it quite easy to extend its operating states if necessary, and it is available for any kind of VSC. A detailed model of a VSC-HVDC system containing a DC hub is built in the PSCAD/EMTDC environment. Simulation results based on three operating conditions (the start-up process, the voltage margin control method and the master-slave control method) demonstrate the flexibility and validity of the proposed control strategy.

        Anatomical Study of the Accessory Tendon of the Extensor Hallucis Longus Muscle and Its Clinical Application

        Background: The accessory tendon of the extensor hallucis longus (ATEHL) muscle is a common abnormal structure, and its clinical significance remains debatable. In this study, we provide the incidence of the ATEHL and characterize its morphological types in Asian cadavers and investigate its clinical applications. Methods: The tendons from 50 adult cadaveric feet, fixed in 10% formalin, were analyzed. We measured the length and width of both the ATEHL and the extensor hallucis brevis (EHB). Results: All dissected specimens had an ATEHL. The first metatarsophalangeal joint was surrounded by an accessory tendon that inserted onto the joint capsule and the dorsal base of the proximal phalanx. We classified the ATEHL into 3 types based on their directions. Differences in ATEHL type based on sex were not statistically significant. Conclusions: We found an ATEHL in all cadaveric specimens in this study. We surmise that the ATEHL acts as an antagonist with the EHB when the toe is extending, which might help prevent the occurrence of hallux valgus deformity.

        Property Modification of Caseinate Responsible to Transglutaminase-induced Glycosylation and Crosslinking in the Presence of a Degraded Chitosan

        Transglutaminase at a concentration of 10 kU/kg of protein and degraded chitosan were used for glycosylation and crosslinking of caseinate at a fixed molar ratio of the acyl acceptor to the acyl donor of 3:1, a protein concentration of 50 g/L, a pH 7.5 at 37oC, and a reaction time of 4 h. Electrophoretic and chemical analyses showed glycosylation and crosslinking of caseinate. Glycosylated and crosslinked caseinate (GC-caseinate) contained glucosamine at 12.77 g/kg of protein, and the protein fraction had fewer reactable amino groups than original caseinate (0.58 vs. 0.64 mol/kg of protein). GC-caseinate exhibited an enhanced surface hydrophobicity, in vitro digestibility, water-binding capacity, and rheological properties, with poor protein dispensability and emulsification activity, but a similar oilbinding capacity and emulsion stability, compared with original caseinate. GC-caseinate also exhibited better properties than transglutaminase-crosslinked caseinate. Glycosylation and crosslinking was effective for better water-binding and rheological properties of caseinate.

        PACKING DIMENSIONS OF GENERALIZED RANDOM MORAN SETS

        We consider random fractal sets with random recursive constructions in which the contracting vectors have different distributions at different stages. We prove that the random fractal associated with such construction has a constant packing dimension almost surely and give an explicit formula to determine it.

      • Taxol Produced from Endophytic Fungi Induces Apoptosis in Human Breast, Cervical and Ovarian Cancer Cells

        Currently, taxol is mainly extracted from the bark of yews; however, this method can not meet its increasing demand on the market because yews grow very slowly and are a rare and endangered species belonging to first-level conservation plants. Recently, increasing efforts have been made to develop alternative means of taxol production; microbe fermentation would be a very promising method to increase the production scale of taxol. To determine the activities of the taxol extracted from endophytic fungus N. sylviforme HDFS4-26 in inhibiting the growth and causing the apoptosis of cancer cells, on comparison with the taxol extracted from the bark of yew, we used cellular morphology, cell counting kit (CCK-8) assay, staining (HO33258/PI and Giemsa), DNA agarose gel electrophoresis and flow cytometry (FCM) analyses to determine the apoptosis status of breast cancer MCF-7 cells, cervical cancer HeLa cells and ovarian cancer HO8910 cells. Our results showed that the fungal taxol inhibited the growth of MCF-7, HeLa and HO8910 cells in a dose-and time-dependent manner. IC50 values of fungal taxol for HeLa, MCF-7 and HO8910 cells were $0.1-1.0{\mu}g/ml$, $0.001-0.01{\mu}g/ml$ and $0.01-0.1{\mu}g/ml$, respectively. The fungal taxol induced these tumor cells to undergo apoptosis with typical apoptotic characteristics, including morphological changes for chromatin condensation, chromatin crescent formation, nucleus fragmentation, apoptotic body formation and G2/M cell cycle arrest. The fungal taxol at the $0.01-1.0{\mu}g/ml$ had significant effects of inducing apoptosis between 24-48 h, which was the same as that of taxol extracted from yews. This study offers important information and a new resource for the production of an important anticancer drug by endofungus fermentation.

        EVALUATION OF THE ANTITUMOR ACTIVITY BY CdTe QDs WITH VERBASCOSIDE

        Cadmium telluride quantum dots (QDs) have received significant attention in biomedical research because of their potential in drug delivery. In this study, the chemotherapeutic agent Verbascoside (VB) was immobilized successfully onto CdTe QDs by covalent bonding between the –OH group of VB and the –COOH group of CdTe QDs. The amount of VB that could be immobilized depended on the concentration of –COOH groups (succinic acid) on the surface of the CdTe QDs. Apoptotic staining, DNA fragmentation and flow cytometry analysis further demonstrated that compared with CdTe QDs or VB treatment alone, the apoptosis rate increased after the treatment of CdTe QDs together with VB (VB–QDs) in HepG2/ADM cells. We observed that VB–QDs treatment could clearly activate apoptosis-related Caspase 3 expression in HepG2/ADM cells. Moreover, our in vivo study indicated that the treatment of VB–QDs effectively inhibited the human hepatoma HepG2/ADM nude mice tumor growth. The increased cell apoptosis rate was closely correlated with the enhanced inhibition of tumor growth in the studied animals. Thus, VB–QDs may serve as a possible alternative for therapeutic approaches for some cancer treatments. In summary, studies have shown that cancer cells are treated by the presence of VB–QDs nanoparticles based on analysis of cell apoptosis.

        Effects of monochromatic lights on the growth performance, carcass characteristics, eyeball development, oxidation resistance, and cecal bacteria of Pekin ducks

        Objective: Light is a significant component of housing environment in commercial poultry industry. This study was conducted to investigate whether Pekin ducks perform better under monochromatic lights than under white light with respect to their growth performance, carcass quality, eyeball development, oxidation resistance, and cecal bacterial communities. Methods: A total of 320 one-day-old male Pekin ducklings were randomly distributed into five rooms with different light treatments, white, red, yellow, green, and blue light. Each room consisted of 4 replicated pens with 16 ducklings per pen. Results: Blue light significantly decreased fat deposition by decreasing abdominal fat. Long wavelength light, such as red, green, and yellow light, considerably increased the back-to-front eyeball diameter and the red light potentially enlarged the side-to-side eyeball diameter. Besides, the blue light had adverse effects on the oxidation resistance status in terms of increasing the product malonaldehyde of lipid oxidation and decreasing the plasma concentration of total superoxide dismutase. The phyla of Firmicutes had the greatest abundance in the green and blue treatments, while Bacteroidetes in blue treatment was the least. The genus of Faecalibacterium was significantly lower under the red light. Conclusion: The high risk of cecal health status and decreased anti-oxidation activity were observed under blue light. Red, yellow, and green light might increase the risk of oversized eyeball and cecal illness. Therefore, monochromatic lights compared to white light did not show advantages on the performance of housing ducks, it turns out that the white light is the best light condition for grow-out ducks.

      • Research on Combination of Controllable Reactor and Magnetic Saturation Transformer

        With intermittent energy grid connected, the requirements of transformer and reactive power compensation are developed. A novel combination technique of controllable reactor and magnetic saturation transformer is proposed in this paper. The optimal control target is implemented by combining the advantages of controllable reactor and magnetic saturation type transformer to suppress flicker of voltage effectively. The corresponding mathematical representation and the equivalent circuit are presented. The design and realization approach of this kind of transformer is described. The traditional transformer capacity can be promoted effectively.

        "Printing" is an ancient technique for recording and publishing. Recently, this technique has extended its activities down to a nanometer scale. In this paper, we review the progress of the printing technique focusing on the most recent developments of one technique called "transfer printing" and its applications in nanophotonics.
