EVALUATION OF THE ANTITUMOR ACTIVITY BY CdTe QDs WITH VERBASCOSIDE

        XIAO-HUI ZHAO,HUI-LAN YUE,PING LI,XIN ZENG,GEN ZHANG 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2013 NANO Vol.8 No.3

Cadmium telluride quantum dots (QDs) have received significant attention in biomedical research because of their potential in drug delivery. In this study, the chemotherapeutic agent Verbascoside (VB) was immobilized successfully onto CdTe QDs by covalent bonding between the –OH group of VB and the –COOH group of CdTe QDs. The amount of VB that could be immobilized depended on the concentration of –COOH groups (succinic acid) on the surface of the CdTe QDs. Apoptotic staining, DNA fragmentation and flow cytometry analysis further demonstrated that compared with CdTe QDs or VB treatment alone, the apoptosis rate increased after the treatment of CdTe QDs together with VB (VB–QDs) in HepG2/ADM cells. We observed that VB–QDs treatment could clearly activate apoptosis-related Caspase 3 expression in HepG2/ADM cells. Moreover, our in vivo study indicated that the treatment of VB–QDs effectively inhibited the human hepatoma HepG2/ADM nude mice tumor growth. The increased cell apoptosis rate was closely correlated with the enhanced inhibition of tumor growth in the studied animals. Thus, VB–QDs may serve as a possible alternative for therapeutic approaches for some cancer treatments. In summary, studies have shown that cancer cells are treated by the presence of VB–QDs nanoparticles based on analysis of cell apoptosis.

      • A Multi-center Survey of HPV Knowledge and Attitudes Toward HPV Vaccination among Women, Government Officials, and Medical Personnel in China

        Zhao, Fang-Hui,Tiggelaar, Sarah M.,Hu, Shang-Ying,Zhao, Na,Hong, Ying,Niyazi, Mayinuer,Gao, Xiao-Hong,Ju, Li-Rong,Zhang, Li-Qin,Feng, Xiang-Xian,Duan, Xian-Zhi,Song, Xiu-Ling,Wang, Jing,Yang, Yun,Li, Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.5

Objectives: To assess knowledge of HPV and attitudes towards HPV vaccination among the general female population, government officials, and healthcare providers in China to assist the development of an effective national HPV vaccination program. Methods: A cross-sectional epidemiologic survey was conducted across 21 urban and rural sites in China using a short questionnaire. 763 government officials, 760 healthcare providers, and 11,681 women aged 15-59 years were included in the final analysis. Data were analyzed using standard descriptive statistics and logistic regression. Results: Knowledge of HPV among the general female population was low; only 24% had heard of HPV. Less than 20% of healthcare providers recognized sexually na$\ddot{i}$ve women as the most appropriate population for HPV vaccination. There was high acceptance of the HPV vaccine for all categories of respondents. Only 6% of women were willing to pay more than US $300 for the vaccine. Conclusions: Aggressive education is necessary to increase knowledge of HPV and its vaccine. Further proof of vaccine safety and efficacy and government subsidies combined with increased awareness could facilitate development and implementation of HPV vaccination in China.

Posterior Nutcracker Syndrome Associated with Interrupted Left Inferior Vena Cava with Azygos Continuation and Retroaortic Right Renal Vein

        Xiao-Li Luo,Gen-Nian Qian,Hui Xiao,Chun-Lei Zhao,Xiao-Dong Zhou 대한영상의학회 2012 Korean Journal of Radiology Vol.13 No.3

Various anatomic anomalies have been considered the causes of nutcracker syndrome (NCS). Posterior NCS refers to the condition, in which vascular narrowing was secondary to the compression of the retroaortic left renal vein while it is crossing between the aorta and the vertebral column. Here, we report an unusual case of posterior NCS associated with a complicated malformation of the interrupted left inferior vena cava with azygos continuation and retroaortic right renal vein, diagnosed by both color Doppler ultrasonography and CT angiography.

Comparative proteomic analysis of proteins expression changes in the mammary tissue of cows infected with Escherichia coli mastitis

        Xiao-wei Zhao,Yong-xin Yang,Dong-wei Huang,Guang-long Cheng,Hui-ling Zhao 대한수의학회 2015 Journal of Veterinary Science Vol.16 No.3

Cows infected with Escherichia (E.) coli usually experience severe clinical symptoms, including damage to mammary tissues, reduced milkyield, and altered milk composition. In order to investigate the host response to E. coli infection and discover novel markers for mastitistreatment, mammary tissue samples were collected from healthy cows and bovines with naturally occurring severe E. coli mastitis. Changesof mammary tissue proteins were examined using two-dimensional gel electrophoresis and label-free proteomic approaches. A total of 95differentially expressed proteins were identified. Of these, 56 proteins were categorized according to molecular function, cellular component, and biological processes. The most frequent biological processes influenced by the proteins were response to stress, transport, andestablishment of localization. Furthermore, a network analysis of the proteins with altered expression in mammary tissues demonstrated thatthese factors are predominantly involved with binding and structural molecule activities. Vimentin and -enolase were central “functionalhubs” in the network. Based on results from the present study, disease-induced alterations of protein expression in mammary glands andpotential markers for the effective treatment of E. coli mastitis were identified. These data have also helped elucidate defense mechanismsthat protect the mammary glands and promote the pathogenesis of E. coli mastitis.

      • Single Nucleotide Polymorphisms in the NER Pathway and Clinical Outcome of Patients with Bone Malignant Tumor

Sun, Xiao-Hui,Hou, Wen-Gen,Zhao, Hong-Xing,Zhao, Yi-Lei,Ma, Chao,Liu, Ying Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.3

The effects of polymorphisms in ERCC5, ERCC6, XPC, CCNH and MMS19L on osteosarcoma response to chemotherapy and the survival of the affected patients were assessed. Genotyping of ERCC5, ERCC6, XPC, CCNH and MMS19L was performed by PCR-RFLP assay. The median PFS was 12.8 months, and the median OS was 18.6 months. Individuals carrying homozygous genotypes of ERCC5 rs17655 and ERCC5 rs1047768 were more like to have good response to treatment, while those carrying homozygous genotypes of MMS19L rs29001322 showed poor response. Osteosarcoma patients carrying TT genotype of ERCC5 rs1047768 showed a significantly longer PFS (16.8 months) and OS (21.4 months) than CC genotype, with HRs(95% CI) of 0.31 (0.10-0.93) and 0.32 (0.06-0.97), respectively. Conversely, those with the TT genotype of MMS19L rs29001322 demonstrated shorter PFS and OS, the HRs (95% CI) being 2.23 (1.08-4.15) and 4.62 (1.45-16.08), respectively. Our findings showed polymorphisms in ERCC5 rs1047768 and MMS19L rs29001322 to be associated with clinical outcome of osteosarcoma patients undergoing chemotherapy.

      • Perioperative Comprehensive Supportive Care Interventions for Chinese Patients with Esophageal Carcinoma: a Prospective Study

        Zhang, Xiao-Dan,Zhao, Qing-Yu,Fang, Yi,Chen, Guan-Xuan,Zhang, Hui-Fang,Zhang, Wen-Xiao,Yang, Xiao-Ping Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.12

Objective: To assess the effects of perioperative comprehensive supportive care interventions on outcome of Chinese esophageal cancer patients in a prospective study. Methods: 60 patients with primary esophageal carcinoma were randomized into an intervention group (IG, n=31) and a control group (CG, n=29). The Chinese version of symptom checklist-90 (SCL-90) was adopted to assess their psychological status. The interventions, including health education, psychological support, stress management, coping strategies and behavior training, were carried out in 3 phases (preoperative, postoperative I and postoperative II), and psychological effects were thereafter evaluated accordingly before surgery, and 1 week, 4 weeks and 24 weeks post-surgery. Medical costs were estimated at discharge. Survival of patients was estimated each year post-surgery. General health status and satisfaction-with-hospital were surveyed by a follow-up questionnaire 4 years post-surgery. Results: All the subjects demonstrated higher scores in the preoperative phase than the normal range of Chinese population concerning 7 psychological domains including somatization, obsessive-compulsive, depression, anxiety, hostility, phobic anxiety and paranoid ideation. Although no significant difference was observed between the two groups at admission, the scores of IG, which tended to decrease at a faster rate, were generally lower than those of CG at weeks 1, 4 and 24 post-surgery. The length of hospital stay and medical costs of IG were significantly less than those of CG and satisfaction-with-hospital was better. However, there was no significant difference in 4-year survival or health status between two groups. Conclusions: Appropriate perioperative comprehensive supportive care interventions help to improve the psychological state of Chinese patients with esophageal carcinoma, to reduce health care costs and to promote satisfaction of patients and their families with hospital.

      • Endostar Combined with Cisplatin Inhibits Tumor Growth and Lymphatic Metastasis of Lewis Lung Carcinoma Xenografts in Mice

Dong, Xiao-Peng,Xiao, Tian-Hui,Dong, Hong,Jiang, Ning,Zhao, Xiao-Gang Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.5

Objective: To investigate the effects of endostar, a recombined humanized endostatin, plus cisplatin on the growth, lymphangiogenesis and lymphatic metastasis of the Lewis lung carcinoma (LLC) in mice. Methods: A tumor model were established in C57BL/6 mice by intravenious transplantation of LLC cells. Then the mice were randomized to receive administration with NS, endostar, cisplatin, or endostar plus cisplatin. After the mice were sacrificed, tumor multiplicity, tumor size and lymph node metastasis were assessed. Then the expression of vascular endothelial growth factor-c (VEGF-C) and podoplanin were determined by immunohistochemical staining. Results: Endostar plus cisplatin significantly suppressed tumor growth. lymphatic metastasis and prolonged survival time of the mice without obvious toxicity. The inhibition of lymphatic metastasis was associated with decreased microlymphatic vessel density (MLVD) and expression of VEGF-C. Conclusions: Endostar combined with cisplatin was more effective to suppress tumor growth and lymphatic metastasis than either agent alone. Thus this may provide a rational alternative for lung carcinoma treatment.

Mechanisms of Extracellular NO and Ca2+ Regulating the Growth of Wheat Seedling Roots

        Xiang Zhao,Xiao-wei Zhao,Hui He,Yan-xiao Wang,Xiao Zhang 한국식물학회 2010 Journal of Plant Biology Vol.52 No.4

Our previous studies suggested the cross talk of nitric oxide (NO) with Ca2+ in regulating stomatal movement. However, its mechanism of action is not well defined in plant roots. In this study, sodium nitroprusside (SNP, a NO donor) showed an inhibitory effect on the growth of wheat seedling roots in a dose-dependent manner, which was alleviated through reducing extracellular Ca2+ concentration. Analyzing the content of Ca2+ and K+ in wheat seedling roots showed that SNP significantly promoted Ca2+ accumulation and inhibited K+ accumulation at a higher concentration of extracellular Ca2+, but SNP promoted K+ accumulation in the absence of extracellular Ca2+. To gain further insights into Ca2+ function in the NOregulated growth of wheat seedling roots, we conducted the patch-clamped protoplasts of wheat seedling roots in a whole cell configuration. In the absence of extracellular Ca2+,NO activated inward-rectifying K+ channels, but had little effects on outward-rectifying K+ channels. In the presence of 2 mmol L−1 CaCl2 in the bath solution, NO significantly activated outward-rectifying K+ channels, which was partially alleviated by LaCl3 (a Ca2+ channel inhibitor). In contrast,2 mmol L−1 CaCl2 alone had little effect on inward or outward-rectifying K+ channels. Thus, NO inhibits the growth of wheat seedling roots likely by promoting extracellular Ca2+ influx excessively. The increase in cytosolic Ca2+ appears to inhibit K+ influx, promotes K+outflux across the plasma membrane, and finally reduces the content of K+ in root cells.

Follistatin N terminus differentially regulates muscle size and fat in vivo

        Hui Zheng,Chunping Qiao,Ruhang Tang,Jianbin Li,Karen Bulaklak,Zhenhua Huang,Chunxia Zhao,Yi Dai,Juan Li,Xiao Xiao 생화학분자생물학회 2017 Experimental and molecular medicine Vol.49 No.-

Delivery of follistatin (FST) represents a promising strategy for both muscular dystrophies and diabetes, as FST is a robust antagonist of myostatin and activin, which are critical regulators of skeletal muscle and adipose tissues. FST is a multi-domain protein, and deciphering the function of different domains will facilitate novel designs for FST-based therapy. Our study aims to investigate the role of the N-terminal domain (ND) of FST in regulating muscle and fat mass in vivo. Different FST constructs were created and packaged into the adeno-associated viral vector (AAV). Overexpression of wild-type FST in normal mice greatly increased muscle mass while decreasing fat accumulation, whereas overexpression of an N terminus mutant or N terminusdeleted FST had no effect on muscle mass but moderately decreased fat mass. In contrast, FST-I-I containing the complete N terminus and double domain I without domain II and III had no effect on fat but increased skeletal muscle mass. The effects of different constructs on differentiated C2C12 myotubes were consistent with the in vivo finding. We hypothesized that ND was critical for myostatin blockade, mediating the increase in muscle mass, and was less pivotal for activin binding, which accounts for the decrease in the fat tissue. An in vitro TGF-beta1-responsive reporter assay revealed that FST-I-I and N terminus-mutated or -deleted FST showed differential responses to blockade of activin and myostatin. Our study provided direct in vivo evidence for a role of the ND of FST, shedding light on future potential molecular designs for FST-based gene therapy.

Study on mass flow characteristics of fluidized bed with a circulative accelerating tube

Xiao Wang,Hui Si,Qi Cheng,Jinchao Kong,Dong Zhao 한국공업화학회 2015 Journal of Industrial and Engineering Chemistry Vol.25 No.-

The heat transfer between annular fluidized-bed and the furnace can be enhanced by adding circulative accelerating tubes in the reactor. Cold state experiments were conducted to test the effect of operation conditions on particles flow in fluidized bed with a circulative accelerating tube. Results showed that the spouting velocity, fluidizing velocity and static bed height all had a beneficial effect on particles circulating efficiency. Shorter and moderate outlet needle nozzles were suitable for circulative accelerating tube to maximize circulating rate. What’s more, smaller size particles which had better fluidity should be chosen for the bed material to improve circulating efficiency.
