Purpose: The molecular links between metabolism and inflammation that drive different inflammatory phenotypes in asthma are poorly understood. We aimed to identify the metabolic signatures and underlying molecular pathways of different inflammatory asthma phenotypes. Methods: In the discovery set (n = 119), untargeted ultra-high-performance liquid chromatography-mass spectrometry (UHPLC-MS) was applied to characterize the induced sputum metabolic profiles of asthmatic patients with different inflammatory phenotypes using orthogonal partial least-squares discriminant analysis (OPLS-DA), and pathway topology enrichment analysis. In the validation set (n = 114), differential metabolites were selected to perform targeted quantification. Correlations between targeted metabolites and clinical indices in asthmatic patients were analyzed. Logistic and negative binomial regression models were established to assess the association between metabolites and severe asthma exacerbations. Results: Seventy-seven differential metabolites were identified in the discovery set. Pathway topology analysis uncovered that histidine metabolism, glycerophospholipid metabolism, nicotinate and nicotinamide metabolism, linoleic acid metabolism as well as phenylalanine, tyrosine and tryptophan biosynthesis were involved in the pathogenesis of different asthma phenotypes. In the validation set, 24 targeted quantification metabolites were significantly expressed between asthma inflammatory phenotypes. Finally, adenosine 5′-monophosphate (adjusted relative risk [adj RR] = 1.000; 95% confidence interval [CI] = 1.000–1.000; P = 0.050), allantoin (adj RR = 1.000; 95% CI = 1.000–1.000; P = 0.043) and nicotinamide (adj RR = 1.001; 95% CI = 1.000–1.002; P = 0.021) were demonstrated to predict severe asthma exacerbation rates. Conclusions: Different inflammatory asthma phenotypes have specific metabolic profiles in induced sputum. The potential metabolic signatures may identify therapeutic targets in different inflammatory asthma phenotypes.
A Hybrid Method on Video Mixing for Multimedia Videoconference

In this paper, we propose a fast video mixing method for reducing the computational complexity in the MCU (Multipoint Control Unit) used in the video conferencing. The conventional mixing method is based on the pixeldomain transcoder, of which computational complexity is linearly increased as the number of participants is increased. Basically the method requires N decoders and one huge encoder to mix the bitstreams from the N participants. To reduce the computational complexity, we propose a hybrid mixing method based on the bitstreamdomain and pixel-domain transcoding methods. The proposed method reduces the computational complexity about 45% at the improved quality, compared with the conventional mixing method based on the pixel-domain transcoders.
Promoter demethylation mediates the expression of ZNF645, a novel cancer/testis gene

Cancer/testis (CT) antigens exhibit highly tissue-restricted expression and are considered promising targets for cancer vaccines. Here we identified a novel CT gene ZNF645 which restrictively expresses in normal human testes and lung cancer patients (68.3%). To investigate the promoter methylation status of ZNF645, we carried out bisulfite genomic sequencing and found that the CpG island in its promoter was heavily methylated in normal lung tissues without the expression of ZNF645, whereas there was high demethylation in normal human testes and lung carcinoma tissues with its expression. Also ZNF645 could be remarkably activated in A549 and HEK293T cells treated by DNA demethylation agent 5`-aza-2`-deoxycytidine. And the dual luciferase assay revealed that the promoter activity of the ZNF645 was inhibited by methylation of the CpG island region. Therefore, we proposed that ZNF645 is a CT gene and activated in human testis and lung cancers by demethylation of its promoter region. [BMB reports 2010; 43(6): 400-406]

Fuji’ apple fruit harvested 10 days before normal harvest (H1) and at normal harvest (H2) were untreated or treated with 1 ㎕·L-1 1-methylcyclopropene (1-MCP) and stored at 0 for up to 30 weeks. Fruits from H1 were firmer and had higher titratable acidity (TA) but lower soluble solids concentrations than those from H2. 1-MCP treatment delayed loss of firmness and TA in fruit from both harvests during storage. Superficial scald incidence was decreased by 1-MCP treatment, but flesh browning was unaffected. H2 fruit had higher total phenolics, flavonoid, and glutathione content as well as total antioxidant activity than H1 fruit at harvest and throughout storage. 1-MCP treated fruit tended to have higher levels of these constituents than untreated fruit in peel, but not in flesh tissues. These results suggest that fruit harvested at the mature stage have better integral quality with 1-MCP treatment.

MED19 is a member of the Mediator that plays a key role in the activation and repression of signal transduction or the regulation of transcription in carcinomas. To tested the functional role of MED19 in human prostate cancer, we downregulated MED19 expression in prostate cancer cells (PC-3 and DU145) by lentivirus- mediated short hairpin (shRNA), and analyzed the effect of inhibition of MED19 on prostate cancer cell proliferation and tumorigenesis. The in vitro prostate cancer cell proliferation, colony formation, and in vivo tumor growth in nude mice xenografts was significantly reduced after the downregulation of MED19. Knock- down of MED19 caused S-phase arrest and induced apoptosis via modulation of Bid and Caspase 7. It was suggested that MED19 serves as a novel proliferation regulator that promotes growth of prostate cancer cells. [BMB reports 2011; 44(8): 547-552]

Background: Evidence for associations between dietary factors and breast cancer risk is inconclusive among Chinese females. To evaluate this question, we conducted a systematic review of relevant case-control and cohort studies. Methods: Studies were systematically searched among 5 English databases (PudMed, ScienceDirect, Wiley, Clinicaltrials.gov, and Cochrane) and 3 Chinese databases (CNKI, WanFang, and VIP) until November 2012. Random effects models were used to estimate summary odds ratios (ORs) and the corresponding 95% confidence intervals (CIs). Results: Thirty one case-control studies and two cohort studies involving 9,299 cases and 11,413 controls were included. Consumption of both soy and fruit was significantly associated with decreased risk of breast cancer, with summary ORs of 0.65 (95% CIs: 0.43-0.99; I2=88.9%, P<0.001; N=13) and 0.66 (95% CIs: 0.47-0.91; $I^2$=76.7%, P<0.001; N=7), respectively. Consumption of fat was significantly associated with increased risk of breast cancer (OR=1.36; 95% CIs: 1.13-1.63; $I^2$=47.9%, P=0.088; N=6). There was nonsignificant association between consumption of vegetables and breast cancer risk (OR=0.72; 95% CIs: 0.51-1.02; $I^2$= 74.4%, P<0.001; N=9). However, sensitivity analysis based on adjusted ORs showed decreased risk of breast cancer was also associated with consumption of vegetables (OR=0.49; 95% CIs: 0.30-0.67). Conclusion: Both soy food and fruit are significantly associated with decreased risk of breast cancer among Chinese females, and vegetables also seems to be protective while dietary fatexerts a promoting influence.
Thalidomide Combined with Chemotherapy in Treating Patients with Advanced Colorectal Cancer

Objective: To assess the safety and effectiveness of thalidomide (produced by CHANGZHOU PHARMACEUTICAL FACTORY CO.LTD) combined with chemotherapy in treating patients with advanced colorectal cancer. Method: A consecutive cohort of pretreated patients with advanced colorectal cancer were treated with thalidomide combined with chemotherapy. And chemotherapy for patients with advanced colorectal cancer were administered according to the condition of patients. Thalidomide was orally administered at a dosage of 50mg/day to 150mg/day before sleeping for at least 14 days. After at least 14 days of treatment, safety and side effects were evaluated. Results: There were 12 female and 3 male patients with advanced cancer recruited into this study, including 9 patients with colon, 6 patients with rectal cancer. The median age of patients was 57(41-82) years. Partial response was observed in 2 patients (2/15), and stable disease in 3 patients(3/15). Incidences of Grade 1 to 2 myelosuppression was observed in 1/15 patients, and Grade 1 to 2 elevation of hepatic enzyme was recorded in 1/15 patients. Adverse effects on the gastrointestinal tract were documented in 1/15 patients, and were Grade 1. No Grade 3-4 toxicities were diagnosed. No treatment related death was found. Conclusions: Thalidomide combined with chemotherapy was safe and mildly effective in treating patients with advanced colorectal cancer. However, further study should be conducted to clarify the effectiveness of this combination.

According to the reinforcement requirements of load-bearing members of existing transmission tower, a new type of in-situ reinforcement scheme for the angle steel T-shaped composite section is proposed. Through the compression test of 10 groups of members, the infl uence of slenderness ratio, the number of fi xtures, the number of bolt rows and other parameters on the failure mode and bearing performance of members was studied. A simulation model was established using ABAQUS software, the load-displacement curve, failure models and other aspects verify the reliability and validity of simulation results. The results show that when the slenderness ratio of member reaches a certain value, the ultimate bearing capacity of member reinforced with the reinforcement angle steel of same specifi cation and strength can be multiplied and increased several times, the slenderness ratio has little eff ect on the force transmission effi ciency of reinforcement angle steel. The number of bolt rows has a small eff ect on the ultimate bearing capacity of member, the increase rate of ultimate bearing capacity, the failure mode and the force transmission effi ciency of reinforcement angle steel, the infl uence of strength of reinforcement angle steel, the specifi cation of reinforcement angle steel on the ultimate bearing capacity, and the increase rate of ultimate bearing capacity of member obviously, the higher strength and specifi cation of reinforcement, the greater the ultimate bearing capacity of reinforced member, and the higher the rate of improvement of ultimate bearing capacity, the smaller the thickness of fi ller plate, that is the smaller the eccentricity, the higher force transmission effi ciency of reinforcement. The research results can provide experimental and theoretical basis for the reinforcement of tower structure and engineering design.
