Poly-γ-glutamic acid (γ-PGA) is a novel polyamino acid formed through microorganism fermentation and biosynthesis. In the present test, membrane (PGA-C) formation by γ-PGA and collodion was performed by using 0.1%glutaraldehyde as a cross-linking agent. A study was conducted on the PGA-C adsorption of Cu2+, specifically the related adsorption equilibrium and kinetics, desorption and regeneration. The results show that with an initial solution pH=5.5and at 318 K, the static adsorption isotherm behavior of PGA-C is in compliance with the Langmuir model and is beneficial to the adsorption of the metal. Meanwhile, with the reaction lasting for 30min, adsorption equilibrium was reached with a maximum adsorption capacity up to 7.431 mg/g. The entire reaction process follows the pseudo-second-order kinetics. By using PGA-C, good regeneration results were obtained after adsorption-generation-adsorption cycling with an HCl solution (0.1 mol/L) as regeneration liquid.

PEG-Dextran and PEG-salt aqueous two-phase systems (ATPS) have been applied to separate glutathione (GSH) from crude yeast extracts. Single-factor experiments were carried out to determine the important factors influencing the partition coefficient and extraction yield. The effect of PEG molecular weight, phase-forming components,PEG and Dextran concentration, pH value, and temperature on the GSH partitioning behavior in ATPS was investigated. Three factors, Dextran concentration, pH value, and temperature, were confirmed to have significant influence on the partition coefficient and extraction yield. These factors were further analyzed with the aid of central composite rotatable design and response surface methodology. The optimal conditions for GSH extraction in the PEGDextran system were determined, including PEG molecular weight 6,000, 10% PEG concentration, 14% Dextran concentration,pH 5.2, and temperature 32 oC. A high extraction yield (83.55%) of GSH from crude yeast extracts was achieved under these optimized conditions. This work is very helpful for developing one efficient and cost-effective process for the separation and purification of GSH from yeast broths.

Polysaccharides from Ganoderma lucidumshowed multiple biological activities, such as immunomodulating,antitumor, antioxidant, and hepatoprotectiveactivity, etc. Adlay oil was added into the media to enhancepolysaccharide production by submerged culture of G. lucidumin this work. The results revealed the optimal concentrationof adlay oil was 1.5 % for polysaccharideproduction of G. lucidum. Analysis of the polysaccharidecomponents confirmed that no novel components werebiosynthesized by the addition of adlay oil. The mainfraction of extracellular polysaccharide, GLEP-2, wasisolated from the submerged culture broth of G. lucidum byethanol precipitation, filtration, DEAE cellulose-52 andSepharose CL-6B chromatography. GLEP-2, which wascomposed of glucose, galactose, mannose, arabinose, andrhamnose in a ratio of 332:55:32:13:3 respectively, had anaverage molecular weight of *2.08 9 104 Da. The bioactivitytests demonstrated that GLEP-2 enhanced the Tlymphocyte proliferation significantly at a concentration of200 lg/mL and B lymphocyte proliferation at lower concentrationsof 50 lg/mL. The results suggest polysaccharidesfrom the submerged culture of G. lucidum arepotential candidates for further development and possiblecommercial applications, especially in the pharmaceuticaland functional foods industries.
