Enhancement of Aggregation-Induced Emission in Dye-Encapsulating Polymeric Micelles for Bioimaging

<P>Three amphiphilic block copolymers are employed to form polymeric micelles and function as nanocarriers to disperse hydrophobic aggregation-induced emission (AIE) dyes, 1,1,2,3,4,5-hexaphenylsilole (HPS) and/or bis(4-(N-(1-naphthyl) phenylamino)-phenyl)fumaronitrile (NPAFN), into aqueous solution for biological studies. Compared to their virtually non-emissive properties in organic solutions, the fluorescence intensity of these AIE dyes has increased significantly due to the spatial confinement that restricts intramolecular rotation of these dyes and their better compatibility in the hydrophobic core of polymeric micelles. The effect of the chemical structure of micelle cores on the photophysical properties of AIE dyes are investigated, and the fluorescence resonance energy transfer (FRET) from the green-emitting donor (HPS) to the red-emitting acceptor (NPAFN) is explored by co-encapsulating this FRET pair in the same micelle core. The highest fluorescence quantum yield (∼62%) could be achieved by encapsulating HPS aggregates in the micelles. Efficient energy transfer (>99%) and high amplification of emission (as high as 8 times) from the NPAFN acceptor could also be achieved by spatially confining the HPS/NPAFN FRET pair in the hydrophobic core of polymeric micelles. These micelles could be successfully internalized into the RAW 264.7 cells to demonstrate high-quality fluorescent images and cell viability due to improved quantum yield and reduced cytotoxicity.</P> <B>Graphic Abstract</B> <P>Highly efficient fluorescence probes are achieved through the encapsulation of aggregation-induced emission molecules, 1,1,2,3,4,5-hexaphenylsilole (HPS) and/or bis(4-(N-(1-naphthyl) phenylamino)-phenyl)fumaronitrile (NPAFN) in the core of polymeric micelles. Design and Synthesis of Carbohydrate-based HIV Vaccine

The HIV-1 envelope is a basic lipid bilayer that contains gp120 and gp41 glycoproteins. Several animal models studies have demonstrated that antibodies can provide considerable benefit against HIV or SIV (Simian Immunodeficiency Virus) challenge and prompted an interest in inducing broadly neutralizing antibodies, an ultimate goal in HIV vaccine research. However, the HIV vaccine development has been deterred by the low antigenicity and immunogenicity of the HIV envelope glycoprotein gp120 and the efficient hiding of highly immunogenic epitopes by their surface glycans. Recently, a new class of bNABs (PG9, PG16, PGT121-137, and PGT141-145) isolated from HIV positive donors was shown to be potent in neutralizing primary HIV-1 strains across clades. Here we describe our efforts towards chemical and enzymatic synthesis of various gp-120 related high-mannose, hybrid, and complex type N-Glycans. We studied specificities of various bNABs towards synthetic N-glycans printed on glass slides using carbohydrate microarray. Based on recognition patterns of bNABs to quaternary structure at variable domain (V1/V2 and V3 regions) of gp-120, we created synthetic oligomannose constructs that more effectively mimic the glycan display found on gp120 and further optimized strategies to conjugate these constructs to carrier protein molecule.
Synthesis of mesoporous SiO2 xerogel/chitosan mixed-matrix membranes for butanol dehydration

Novel mesoporous SiO2 xerogel/chitosan (CS) mixed-matrix membranes (MMMs) were successfully prepared by incorporating sol–gel synthesized SiO2 xerogels into glutaraldehyde (GA) cross-linked CS membranes. The effects of the SiO2 doping amount and the feed temperature on the performance of the separation of butanol and water was also investigated in this study. The 0.25 wt% SiO2 xerogel/CS MMMs possessed the best pervaporation performance and the largest PSI value of 1102 kg/m2h with a permeate flux of 736 g/m2h (Permeability of 3.25 × 10−5 g m m−2 h −1 Pa−1) and a separation factor of 1498 at a feed temperature of 50 °C. This PSI value is not only comparable but also exceeds that of other membranes reported in the literature. The separation factor and the flux of the SiO2 xerogel/CS MMMs in this study clearly exceed the upper limit of the previously reported MMMs. As a result, the as-prepared novel SiO2 xerogel/CS MMMs showed an improved performance during the butanol and water separation process, which raises the possibility of future pervaporation applications using this novel SiO2 xerogel/CS MMMs.

The traditional sulfonylureas with long half-lives have sustained stimulatory effects on insulin secretion compared to the short-acting insulin secretagogue. In this study, we used the frequently sampled intravenous glucose tolerance test (FSIGT) to evaluate the insulin sensitivity (IS), glucose sensitivity (SG), and acute insulin response after glucose load (AIRg) after 4months treatment with either gliclazide or repaglinide. The design of study was randomizedcrossover. We enrolled 20 patients with new-onset type 2 diabetes (mean age, 49.3 years). Totally three FSIGTs were performed, one before and one after each of the two treatment periods as aforementioned. No significant differences in fasting plasma glucose, insulin, body mass index, blood pressure, glycated hemoglobin, or lipids were noted between the two treatments. After the repaglinide treatment, higher AIRg, lower IS, and lower SG were noted, but they did not reach statistical significance. The disposal index (DI) was also not significantly different between the two treatments. In conclusion, since non-significantly higher DI, AIRg, lower IS and SG were noted after repaglinide treatment, it might be a better treatment for diabetes, relative to gliclazide.

Purpose: Metastatic castration-resistant prostate cancer (mCRPC) has a poor prognosis. Abiraterone acetate (AA), enzalutamide, and chemotherapy are first-line treatments for patients with mCRPC. This study examined prognostic factors for AA response in the form of prostate-specific antigen (PSA) kinetics throughout androgen-deprivation therapy (ADT) in chemonaïve patients with mCRPC. Materials and Methods: We retrospectively included data from 34 chemonaïve patients with mCRPC who had received AA at some point between January 2017 and December 2018. We separated patients into two study arms according to the decrease in PSA percentages after use of AA for 3 months. We correlated PSA kinetics parameters with response and compared the two study groups with respect to PSA kinetics. Results: The patients’ median age was 77 years. In the total group of patients, 64% had a response to AA, whereas 35% did not. The ratio of the PSA level at nadir to the level during ADT was significantly higher in the AA-sensitive group (19.78 vs. 1.03, p=0.019). Conclusions: Patients who experienced a dramatic change in PSA level during ADT were more likely to be resistant to AA after progression to mCRPC. Chemotherapy rather than AA might be more suitable as a first-line treatment for these patients.
한국인 및 중국 한족 정신분열병 환자의 5-HT2A 수용체 유전자 -1438A/G 다형성

Objectives : The purpose of the present study was to investigate the association between -1438A/G polymorphism of 5-HT2A receptor gene and schizophrenia in Korean and Han Chinese population. Methods : A sample of 184 Korean patients with schizophrenia and 96 Korean healthy normal controls and 96 Han Chinese patients with schizophrenia and 96 Han-Chinese healthy normal controls were genotyped for a single nucleotide polymorphism with in 5-HT2A receptor gene (promoter region, A-1438G) by Msp I Resthction Fragment Length Polymorphism (RFLP). Results : There was no difference in allelic frequencies and genotype frequencies of -1438A/G polymorphism between Korean schizophrenics and controls (p=0.13) and Han Chinese schizophrenics and controls (p=0.40). Also, -1438A/G Poly-morphism did not show ethnical difference between Korean and Han Chinese controls. The Scale for the Assessment of Negative Symptoms (SANS) scores showed no significant differences between genotypes of -1438A/G polymorphism in both of Korean and Han Chinese schizophrenics. Conclusion : These results suggest that -1438A/G polymorphism of the 5-HT2A receptor gene is not causally related to the development of schizophrenia in Korean and Han Chinese population, and there no ethnic difference between Korean and Han Chinese population.
A Fair-Exchange E-Payment Protocol For Digital Products With Customer Unlinkability

Along with the development of Information Technology, online transactions through Internet have become more popular for the reasons of convenience and efficiency. In order to provide secure and reliable online transactions, an effective electronic payment protocol is crucial. In this paper, we propose a novel electronic payment protocol for digital product transactions with an offline arbiter to achieve fair exchange, automated dispute resolution, customer anonymity, and customer unlinkability. In our protocol a product token is adopted to eliminate the need of key management for digital product decryption in the offline arbiter. In addition, Elliptic Curve Cryptography (ECC)-based self-certified public key is utilized to further reduce computing overheads. According to our analysis, the efficiency of our protocol can be greatly increased in comparison with previous literatures.
