      • 생쥐의 발정주기에 따른 자궁내막의 점액질에 관한 조직화학적 연구

        Histochemical studies to elucidate the changes of properties and composition in the endomeritum of uterine horn and body of the normal adult mice were done during proestures, estrus, metestrus and diestrus. The determination of estrus cycle was confirmed by means of the papanicolaou smear method, and the mucosubstances were stained with alcian blue pH 2.5 and 1.0, PAS, alcian blue pH 2.5-PAS, alcian blue pH 1.0-PAS, aldehyde fuchsin pH 1.7-alcian blue pH 2.5, and high iron diamine-alcian blue pH 2.5 methods. The results obtained from this study were as follows: 1. The mucosubstances in the free surface of epithelial and glandular epithelial cells in endometrium of uterine horn and body were the mixture of acid and neutral mucins during proestrus, metestrus and diestrus. The acid mucins were the mixture of the sulfated and nonsulfated mucins, and the amount of the sulfated mucin was more prominent than nonsulfated mucin. During estrus the amount of mucosubstances was more increased than other periods of the estrus cycle. In the endomeritum of uterine horn and body, the free surface of epithelial cells contained a lot of strongly sulfated and neutral mucins, and that of glandular epithelial cells contained sulfated, nonsulfated and neutral mucins, and increased especially the amount of strongly sulfated mucin. 2. In the endometrium of uterine horn and body, the mucosubstances in the luminal substances and stroma did not show any significant charges according to eastrus cycle. The luminal substances contained a lot of neutral mucin and a a little of sulfated and nonsulfated mucins, and the stroma contained a little of neutral, sulfated and nonsulfated mucins.

        한국산 5종 개구리 피부선의 점액질에 대한 조직화학적 연구

        한국산 개구리 5종을 재료로 하여 복부, 흉부 및 배부의 피부진피내에 산재하는 피부선의 점액물질을 검색하기 위하여 hematoxylin-eosin염색, Weigert iron hematoxylin염색, alcian blue pH 2.5염색, alcian blue pH 2.5-PAS염색, alcian blue pH 1.0-PAS염색, aldehyde fuchsin pH 1.7염색, aldehyde fuchsin pH 1.0염색 및 aldehyde fuchsin pH 1.7-alcian blue pH 2.5염색을 실시하였다. 두꺼비를 제외한 모든 종에서 점액선, 장점액선 및 과립선이 관찰되었으며 점액선의 선세포가 편평형내지 입방형인데 비해 장점액선의 선세포는 원주형으로 선관부로 갈수록 편평형내지 입방형의 세포로 변하였다. 두꺼비에서는 점액선과 과립선만이 관찰되었다. 점액선의 점액질양은 무당개구리, 옴개구리 및 두꺼비에서 풍부하였으며 참개구리 및 청개구리는 점액질을 가지지 않은 휴지기의 선세포가 많았다. 모든 종의 점액선은 중성점액질과 산성점액질의 혼합성이며 무당개구리를 제외한 모든 종에서 중성점액질의 양이 많았다. 산성점액질의 조성으로 보아 참개구리, 무당개구리 및 청개구리에서는 sulfomucin과 sialomucin의 혼합성이며, 옴개구리에서는 강 sulfomucin만이 관찰되었으며 두꺼비에서는 대부분의 선세포가 강 sulfomucin만을 함유하나 일부 약 sulfomucin과 sialomucing 함유한 선세포가 관찰되었다. 장점액선의 염색정도는 점액선에 비해 미약하며 옴개구리는 중성점액질만 가지며 그 외 종은 중성점액질과 산성점액질의 혼합성이나 중성점액질의 양이 더 많았다. 산성점액질의 조성으로 보아 약 sulfomucin과 sialomucin의 혼합성이며 참개구리에서 일부 소수의 선세포가 강 sulfomucin을 함유하였다. 과립선은 점액질을 분비하지 않았다. This study was performed to observe the histochemical properties of the mucosubstances in the skin glands which scattered in the dermis of the abdominal, thoracic and dorsal skin of five frog species(Rana nigromaculata, Bombina orientalis, Rana rugosa, Hyla arobrea and Bufo bufo gargarizans). The mucosubstances were stained with hematoxylin-eosin, Weigert iron hematoxylin, PAS reaction, alcian blue pH 2.5, alcian blue pH 2.5-PAS, alcian blue pH 1.0-PAS, aldehyde fuchsin pH 1.7, aldehyde fuchsin pH 1.0 and aldehyde fuchsin pH 1.7-alcian blue pH 2.5 staining method. Three distinct types of gland, mucous, seromucous and granular gland, could be identified in Rana nigromaculata, Bombina orientalis, Rana rugosa and Hyla arobrea. The body of mucous glands was lined with cuboidal or squamous cells. The body of seromucous glands was columnar cells and toward the ductal pole the cells became flatten. Bufo bufo gargarizans possessed only two types of gland, mucous and granular gland. Mucosubstances were more abundantly observed in Bombina orientalis, Rana rugosa and Bufo bufo gargarizans than the other species, and in Rana nigromaculata and HyIa arobrea, a large number of resting mcuous cells which not contained mucosubstances were detected. The mucosubstances of mucous glands were composed of neutral and acid mucins. More neutral mucins than acid ones in the mucous gland were observed in all species examined except Bombina orientatis. According to their histochemical composition of acid mucins in Rana nigromaculata, Bombina orientalis and Hyla arobrea, mucous cells were composed of weak sulfomucin and sialomucin. Rana rugosa showed mucous cells having only strong sulfomucin. In Bufo bufo gargarizan, most of mucous cells contained strong sulfomucin whereas weak sulfomucin and sialomucin were detected in a few cells. The mucosubstances of seromucous glands in Rana nigromaculata, Bombina orientalis, Hyla arobrea, and Rana rugosa stained weakly with all staining methods as compared with the those of mucous gland. Rana rugosa exhibited neutral mucins only and the remaining three species detected neutral and acid mucins. More neutral mucins than acid ones were observed. According to their histochemical composition of acid mucins, most of seromucous cells were composed of weak sulfomucin and sialomucin, however, in Rana nigromaculata, a few cells contained strong sulfomucins only. Granular glands did not secrete mucins.

        흰쥐 혀 침샘 점액질에 미치는 paraquat 독성에 대한 녹차의 완화효과

        흰쥐의 혀 미각샘 및 점액샘과 그 점액질에 미치는 paraquat 독성과 녹차의 paraquat 독성완화 효과를 대조군, paraquat 투여군, Paraquat-녹차 투여군, 녹차 3주 투여후 paraquat-녹차 투여군으로 나누어 prelectin 조직화학법으로 연구하였다. Paraquat 투여군 미각샘은 대조군에 비해 장액샘꽈리 위축, 장액세포의 과립의 감소 및 소실, 공포변성 등이, 점액샘은 점액샘꽈리의 위축, 확장 또는 융합, 점액세포의 과립의 감소, 공포변성 및 미성숙 샘꽈리 등이 대조군에 비해 많이 관찰되었다. 미각샘 및 점액샘 도관상피의 점액화도 관찰되었다. Paraquat-녹차 투여군과 녹차 3주 투여후 paraquat-녹차 투여군에서는 paraquat 투여군의 미각샘과 점액샘의 형태적 변화가 대조군으로 회복되는 경향을 나타내었으며 후자에서 더 현저하였다. Paraquat 투여군의 미각샘의 중성점액질과 점액샘의 산성 및 중성점액질 양이 대조군에 비해 현저히 감소하였다 Paraquat 투여군의 혀 점액샘에서 sulfomucin을 분비하는 점액세포는 대조군에 비해 현저히 감소하였으나 sialomucin을 분비하는 점액세포는 오히려 증가하는 경향을 나타내었다. Paraquat-녹차 투여군과 녹차 3주 투여후 paraquat-녹차 투여군에서 대조군으로 회복되는 경향을 보였으며 후자에서 더 현저하였다. 이상의 결과로 보아 paraquat는 혀 미각샘 및 점액샘과 그 분비물인 점액질에 강한 독성을 나타내며 이 독성에 대해 녹차가 완화효과를 나타냄을 알 수 있고 녹차 3주 투여후 paraquat-녹차 투여군에서 녹차-paraquat 투여군보다 완화효과가 더 크게 나타나는 것으로 보아 paraquat 독성에 대한 완화효과는 녹차를 투여하는 시기와 그 양에 따라 더 효과가 있고 녹차에 paraquat 독성 발현억제 및 예방물질이 있음이 사료되어진다. Paraquat toxicity and the alleviating effects of green tea on the toxicity to the rat lingual mucins were investigated through prelectin histochemical methods. Mucosubstances properties were stained with PAS, AB pH 2.5, AB pH 1.0, AB pH 2.5-PAS, AF pH 1.7-AB pH 2.5 and HID-AB pH 2.5 methods. Sprague-Dawley male adult rats(250∼300g) were divided into one control group and three experimental groups, DP group which was administrated by only 0.04% paraquat in the drinking water for one week, GP group which was fed by both 0.04% paraquat and 3% green tea for one week, and GGP group which was applied with both 0.04% paraquat and 3% green tea for one week after 3% green tea in the drinking water was dosed for three weeks. For the morphological changes, the lingual gustatory glands of DP group, compared with the control group, were noted atrophy of serous acini, decrease and disappearance of zymogen granules, and vacuolation in the serous cells. And the lingual mucous glands of DP group showed atrophy of mucous acini, and decrease of mucigen granules. Especially, many enlarged, fused mucous acini, and vacuolated mucous cells were detected. Mucification of duct cells increased very notably in DP group. The groups of feeding green tea showed recovering tendency. The recovery degree of GGP group was greater than that of GP group. And GGP group recovered almost up to the state of the control group. Neutral mucin of the gustatory gland, and acid and neutral mucins of mucous gland decreased very notably in DP group compared with the control group. The decrease of acid mucin was more notable than that of neutral mucin in the mucous gland. And DP group showed that the mucous acini which secrete strong sulfomucin decreased, but the mucous acini which secrete sialomucin increased remarkably. GP and GGP group appeared the recovering tendency to the control group. The recovery of GGP group, comparing the degree, was more remarkable than that of GP group. And GGP group recovered almost up to the state of the control group. Consequently, the above results suggest that paraquat exhibites the toxicity which affects the histological structure and mucosubstances properties in the lingual salivary glands, and also suggest that green tea alleviates this toxicity. The different results of recovering tendency of paraquat toxicity in GP and GGP group indicate that the alleviating effects of green tea on paraquat toxicity to the histological structure and mucosubstances in the lingual salivary glands depend on the dosing time and the amount of green tea. Furthermore, green tea is presumed to have effects to recover from paraquat toxicity, to suppress exhibition of paraquat toxicity and to prevent paraquat toxicity.

      • 양서류 및 어류의 소화기관점막에 관한 비교조직화학적 연구

        A comparative histochemical study was done on the mucosubstance and alkaline phosphatase and acid phosphatase of the mucosa of the tongue, esophagus, stomach, small and large intestine in the frog and eel. The mucosubstance was observed with alcian blue staining (pH 2.5), mucicarmine staining, PAS reaction and combined staining of alcian blue (pH 2.5) and PAS. The alkaline and acid phosphatase were observed with Gomori's modified methods. The results were as follows : 1. PAS reaction : ⅰ) The lingual and esophageal glands were stained moderately to strongly with PAS in the frog. ⅱ) The goblet cells of the digestive tract were stained moderately, and the surface epithelial cells of the gastric mucosa and mucous neck cells of the gastric gland were stained strongly with PAS in the frog and eel. 2. Alcian blue staining : ⅰ) The lingual and esophageal glands were not stained with alcian blue in the frog. ⅱ) The goblet cells of the digestive tract were stained moderately to strongly with alcian blue in the frog and eel, but more strongly in the eel than the frog. ⅲ) The surface epithelial cells of the gastric mucosa and the mucous neck cells of the gastric gland were not stained with alcian blue in the frog, but the surface epithelial cells of the gastric mucosa were stained moderately in the eel. 3. Mucicarmine staining : ⅰ) The goblet cells and the lingual glands of the lingual mucosa were not stained, but the esophageal glands were stained weakly with mucicarmine in the frog, and the goblet cells of lingual mucosa were stained moderately with mucicarmine in the eel. ⅱ) The goblet cells of the esophagus were stained moderately, but the goblet cells of the intestine, the surface epithelial cells of the gastric mucosa and the mucous neck cells of the gastric gland were stained weakly with mucicarmine in the frog and eel. 4. Combined staining of alcian blue and PAS ⅰ) The goblet cells of the lingual and esophageal mucosa, lingual and esophageal glands were stained strongly with combined staining, but more strongly with PAS than alcian blue in the frog. ⅱ) The goblet cells of the lingual and esophageal mucosa were stained strongly with combined staining, but more strongly with alcian blue than PAS in the eel. ⅲ) The goblet cells of the intestine were stained strongly with combined staining, but more strongly with alcian blue than PAS in the frog and eel. ⅳ) The surface epithelial cells of the gastric mucosa were stained strongly with combined staining, and more strongly with PAS than alcian blue, but the mucous neck cells of the gastric gland were stained with PAS in combined staining in the frog and eel. 5. Alkaline phosphatase reaction : ⅰ) The surface epithelial cells of lingual and esophageal mucosa were almost negative, but the basal cells were moderately to strongly positive in the frog and eel. ⅱ) The lingual glands were moderately positive, but esophageal glands were negative in the frog. ⅲ) The gastric mucosa were negative, but the surface epithelial cells of intestine were strongly positive, but crypt epithelial cells were almost negative in the frog and eel. 6. Acid phosphatase reaction : ⅰ) The surface epithelial cells of the tongue were negative, but lingual glands were weakly positive, and the basal cells of the esophageal mucosa were moderately positive, but the esophageal glands were negative in the frog. ⅱ) The epithelial cells of the lingual and esophageal mucosa were almost negative in the eel. ⅲ) The gastric mucosa were negative in the frog and eel. The surface epithelial cells of the intestine were moderately to strongly positive, but crypt epithelial cells were almost negative in the frog and eel.

        개조개,백합,피조개 및 꼬막 발의 복합당질에 대한 Lectin 결합양상, Alkaline 및 Acid Phosphatase 분포에 관한 조직화학적 연구

        Lectin binding patterns of glycoconjugates of the foot of Saxidomus purpuratus, Meretrix lusoria, Scapharca broughtonii and Tegillarca granosa were studied lectin methods, i.e., 9 kinds of biotinylated lectins (DBA, SBA, PNA, BSL-1, RCA-1, sWGA, UEA-1, LCA and ConA). The activities of alkaline and acid phosphatases of the foot in these species were studied with modified Gomori's methods. Lectin binding patterns of glycoconjugates was different according to shellfish species, and protions of the foots. Lectin binding patterns of the epithelial cilia of the foots showed 9 biotinylated lectins binding in Saxidomus purpuratus, 8 biotinylated lectins binding except SBA in Meretrix lusoria, DBA, PNA, RCA-1 and LCA binding in Scapharca broughtonii and lectins of UEA-1, LCA, and ConA binding in Tegillarca granosa. Especially, these cilia showed moderate or moderate to intense with SBA and BSL-1 binding in Saxidomus purpuratus, and DBA binding in Scapharca broughtonii and Meretrix lusoria, respectively, but moderate with LCA binding in Tegillarca granosa. The cytoplasm of the foot epithelial cells appeared 6 lectins binding except SBA, UEA-1 and RCA-1 in Tegillarca granosa, while 9 lectins binding in other species. Especially, these cytoplasm showed a moderate with ConA binding in Tegillarca granosa, sWGA binding in Meretrix lusoria, LCA, sWGA, RCA-1 and PNA binding in Scapharca broughtonii, but moderate to intense with LCA binding in Saxidomus purpuratus, and DBA and LCA binding in Meretrix lusoria. Lectin binding patterns of glycoconjugates in the duct and mucous acini of the foot glands occurred 5 lectins affinity except PNA, sWGA, UEA-1, LCA and ConA in Saxidomus purpuratus, 8 lectins affinity except RCA-1 in Tegillarca granosa, and 9 lectins affinity in other speices. These mucous acini occurred intense with sWGA binding, moderate with BSL-1 and SBA in Scapharca broughtonii, and moderate to intense with sWGA in Saxidomus purpuratus, with BLS-1, RCA and sWGA in Meretrix lusoria, with SBA and BSL-1 in Tegillarca granosa, but moderate with DBA, ConA and sWGA in Tegillarca granosa. In the intraepithelial duct of these glands, sWGA binding was weak to moderate in Saxidomus purpuratus, and moderate in other species. These ducts showed a moderate affinity for SBA, PNA and RCA-1 in Scapharca broughtonii, ConA in Tegillarca granosa, and UEA-1 in Meretrix lusoria. On the other hand, the ducts of these glands within connective tissue showed a moderate to intense affinity for RCA-1 in Meretrix lusoria, a moderate affinity for SBA and sWGA in Scapharca broughtonii, and DBA, SBA, sWGA and ConA in Tegillarca granosa. In the granules of connective tissue, a weak to moderate affinity for only sWGA was found in Scapharca broughtonii. But in Tegillarca granosa, DBA, SBA, BSL-1, LCA and ConA binding were detected and DBA, SBA and BSL-1 was moderate to intense. Activities of alkaline and acid phosphatases of the epithelial cilia occurred presented faint in Meretrix lusoria, weak in Scapharca broughtonii and Tegillarca granosa. In the cytoplasms of epithelil cells, these activities were trace to weak in Saxidomus purpuratus and Meretrix lusoria, weak to moderate in Scapharca broughtonii and Tegillarca granosa. In the epithelial ducts of these glands, the activities were faint in Saxidomus purpuratus and Meretrix lusoria. In mucous acini and granules of connective tissue, the activities of alkaline and acid phosphatases were weak to moderate in only Scapharca broughtonii and Tegillarca granosa. 개조개, 백합, 피조개 및 꼬막 발의 복합당질 성상은 DBA, SBA, PNA, BSL-1, RCA-1, sWGA, UEA-1, LCA 및 Con A등 9종의 biotinylated lectin을 이용한 lectin 조직화학법 그리고 alkaline 및 acid phosphatase의 활성은 Gomori변법으로 관찰하였다. 복합당질에 대한 lectin의 결합 양상은 조개 종 및 부위에 따라 차이가 있었다. 상피세포의 섬모에 대한 lectin의 결합 양상은 개조개는 9종 lectin 모두, 백합은 SBA를 제외한 8종 lectin에, 피조개는 DBA, PNA, RCA-1, 및 LCA 4종 lectin에, 꼬막은 UEA-1, LCA 및 Con A 3종 lectin에 반응하였으며 개조개는 SBA 및 BSL-1에, 백합과 피조개는 DBA에 중등도 내지 강한 반응 또는 중등도 반응을, 그리고 꼬막은 LCA에 중등도 반응을 나타내었다. 상피세포의 세포질은 꼬막은 SBA, UEA-1 및 RCA-1를 제외한 6종 lectin에 그외 종은 9종 lectin 모두 반응하였으며 꼬막은 Con A에, 백합은 sWGA에, 피조개는 LCA, sWGA, RCA-1 PNA 및 DBA에 중등도 반응을, 그리고 개조개는 LCA에, 백합은 DBA에 중등도 내지 강한 반응을 나타내었다. 발샘 도관 및 점액샘꽈리 복합당질에 대한 lectin의 결합 양상을 보면 개조개는 PNA, sWGA, UEA-1, LCA, Con A 5종 lectin에, 꼬막은 RCA-1을 제외한 8종 lectin에, 그 외 종에서는 9종 lectin 모두에 반응하였다. 발샘의 점액샘꽈리는 피조개에서 sWGA에 강한 반응을, BSL-1 및 SBA에 중등도 반응을, 개조개에서는 sWGA에, 백합은 BSL-1, RCA 및 sWGA에 중등도 및 강한 반응을 나타내었으며, 꼬막은 SBA 및 BSL-1에 중등도 내지 강한 반응을, DBA, Con A 및 sWGA에 중등도 반응을 나타내었다. 도관에서 sWGA는 개조개에서 약한 반응 내지 중등도 반응을, 그 외 종에서는 중등도 반응을, SBA는 피조개에서, Con A는 꼬막에서 중등도 반응을 나타내었으며 결합조직 발샘 도관에서 백합은 RCA-1에 중등도 내지 강한 반응을, 꼬막은 SBA와 Con A에 중등도 반응을 나타내었다. 결합조직 과립은 피조개에 sWGA에만 약한 반응 내지 중등도 반응을 나타내었으나 꼬막에서는 DBA, SBA, BSL-1, LCA 및 Con A 5종 lectin에 반응 하였으며 DBA, SBA 및 BSL-1에 중등도 내지 강한 반응을 나타내었다. Alkaline 및 acid phosphatase의 활성은 상피세포의 섬모에서 개조개와 백합은 미약한 반응을, 피조개와 꼬막은 약한 반응을 나타내었으나, 세포질에서 개조개와 백합은 미약한 반응 내지 약한 반응을, 피조개와 꼬막은 약한 반응 내지 중등도 반응을 나타내었다. 상피의 발샘 도관은 개조개와 백합에서 미약한 반응을, 발샘의 도관과 점액샘꽈리 및 결합조직 과립은 피조개와 꼬막만 약한 반응 내지 중등도 반응을 나타내었다.

      • 새 교육과정에 의한 중학교 과학교과서(1 및 2)중 생물영역에 대한 분석적 연구

        In the science textbooks 1 and 2 according to the new educational curriculum, the results of investigation and analysis of biology course are as follow: 1. Science textbooks 1 and 2 consist of four parts: Earth science, biology, chemistry and physics. The area of biology was emphasized. 2. In the contents of the book, the levels of science textbook 1 was emphasized on 'individual' (65.7%), science textbook 2, on 'tissue and organ' (68.3%), and 'diversity and unity' (64.4%) were emphasized in textbook 1, and 'structure and function' (76.2%) in textbook 2 as a themes. 3. In view of the experiment and observation, there was not any experiment in textbook 1, but there were many observation (14 times) and study (45 times). On the contrary many experiments (9 times) have been performed in science textbook 2, but few observation (1 times) and study (13 times) comparing with textbook 1. The temes of experiments and observation many dealt with 'formation' in science textbook 1, but textbook 2 dealt with 'structure and function'. 4. The kinds of organisms which are use in experiments and observation were total 19 kinds, 12 in textbook 1 and 7 in textbook 2. It was show that water grass (3 times) and sprouted bean (2 times). There were total 11 kinds of chemicals in using frequency. They are 4 in textbook 1 and 7 in textbook 2. Much used chemical was iod-iodine solution. In using 36 kinds of instruments in total, such as 16 in textbook 1 and 20 in textbook 2, among these instruments in total, such as 16 in textbook 1 and 20 in textbook 2, among these instruments three were used duplicately. The using frequency order of the instruments is as follow: microscope> test tube, spoid, magnifying glass> water tank, slide glass. 5. This new science textbook contains total 164 pieces of pictures and photos which are affluent in chromaticity, such as 92 in textbook 1, and 72 in thextbook 2. So these science textbook lie emphasis in causing learns to have interest in their study and investigate.

      • 고등학교 새 생물교과서에서 관찰과 실험에 사용되는 재료생물에 대한 분석적 연구

        This study was carried out to analyze the appropriate topic of observation and experiment, and investigate the kinds and applicated frequency of material organisms requied for observation and experiment in the new biology textbook of the high school in Korea, 1984. The results were summarized as follows : 1. Comparision of five current biology textbook used in the high school revealed a great deal of difference in number of observation and experiment ranging from 18 to 27 items. 2. The material organisms were required 39 kinds in the editions Ⅰ, 38 kinds in the editions Ⅱ, and total the material prganisms were required 66 kinds in the edition Ⅰand Ⅱ. 3. The kinds of material organisms with the highest frequency of the application were appeared with four kinds, frog, man, onion and drosophila in the edition Ⅰ, and with four kinds, frog, man, spinach and onion in the editions Ⅱ. 4. The material organisms application together in both the edition Ⅰand Ⅱ were appeared with 16.7 percents (11 kinds) of the total 66 kinds. The names for the 11 kinds could be written from the highest to the lowest frequency such as, man>frog>onion>drosophila, yeast>paramecium, potato, Tradescantia>hen(cock), Euglena>corn. On the basis of the above results, it was suggested that the high school were required with the basic facilities such as botanical garden, zoological garden, farm, orchard, green house, animal room and animal case, two kinds of aquariums with sea and fresh water, and pond of pool.

        연골 및 경골어류 설 점액질의 성상에 관한 조직화학적 연구

        The studies were performed in order to investigate the histochemical properties and compositions of mucosubstances in the mucous cells and the matrices of the cartilages in the tongues of Chondrichthyes and Osteichthyes. The specimens used in this study were obtained from the tongues of 2 species of pisces; Chondrichthyes, Scylliorhinus torazame and Isteichthyes, Anguilla japonica. The mucosubstances were stained with alcian blue pH 2.5, alcian blue pH 1.0, alcian blue pH 0.4, alcian blue pH 2.5-PAS, alcian blue pH 1.0-PAS and aldegyde fuchsin pH 1.7-alcian blue pH 2.5 staning methods. The results were as follow: 1. It were seen mixed with most mucous cells were contained large amount of nonsulfated acid mucins and neutral mucins, and some contained a very small amount of weak sulfated acid mucins and large amount of neutral mucins in Osteichthyes (eels), but were seen mixed with most mucous cells were contained only a large amount of neutral mucins, and some contained a very small amount of weak sulfated acid mucins and a large amount of neutral mucins in Chondrichthyes (catsharks). 2. In Osteichthyes (eels), the mocosubstances in the matrices of the lingual cartilages, were composed of acid and neutral mucins, and more of acid mucins, but were different in the regions of the matrices, i.e., the mucosubstances were contained only a large amount of strong sulfated acid mucins in lacunar walls and territorial matrices, and composed of large amount of sulfated acid mucins and a very small amount of neutral mucins in granular matrices of interterritorial matrices, but composed of a small amount of weak sulfated mucins and neutral mucins in agranular matriecs of interterritorial matrices and subperichondrial matrices. In Chondrichthyes(catsharks), the mucosubstances in the matrices of the lingual cartilages, were composed of acid and neutral mucins, but were different in the regions of the matrices, i.e., the mucosubstances were contained only a large amount of strong sulfated acid nucins in lacunar walls, but composed of large amount of strong sulfated acid mucins and neutral mucins in territorial matrices and granular matrices of interterritorial matrices, and composed of a very small amount of weak sulfated acid mucins and large amount of neutral mucins in agranular matrices and subperichondrial matrices.

      • Cyclophosphamide가 흰쥐 혀침샘의 점액질에 미치는 영향에 대한 조직화학

        항암 및 항종양 치료제인 CP가 흰쥐의 혀 미각샘과 점액샘의 형태 및 그 분비물인 점액질에 미치는 영향을 PAS, AB pH 2.5, AB pH 2.5-PAS, AB pH 1.0, AF pH 1.7-AB pH 2.5 및 HID-AB pH 2.5 염색법으로 연구하였다. 1. CP군의 혀 미각샘의 형태적인 변화는 장액샘꽈리의 위축 및 파괴, 장액세포의 세포질과립의 감소 및 소실, 세포질의 공포변성 그리고 일부 도관상피세포의 점액화였으며, 혀 점액샘의 형태적 변화는 점액샘꽈리의 확장, 융합, 파괴, 점액세포의 세포질과립 감소 및 소실, 세포질의 공포변성 그리고 도관상칙의 현저한 점액화였다. 2. 혀 미각샘의 중성점액질이 대조군에 비해 현저히 감소하였으며 극소수의 미각샘세포에서 산성 점액질이 나타났다. 혀 점액샘의 중성점액질과 산성점액질은 대조군에 비해 다같이 현저히 감소하였고 산성점액질 중 sulfomucin은 현저히 감소하였으나 sialomucin은 다소 증가하는 경향을 나타내었다. The effect of cyclophophamide(CP) on the mucins of rat lingual salivary gland was examined by prelectin histochemical methods. Sprague-Dawley rats weighing about 250-300 gm were divided into control and CP groups. Control group was administrated by only drinking water for one week, CP group which was admirdstrated by 0.04% CP in the drinking water for one week. Mucins properties were stained with PAS, AB pH 2.5, AB pH 2.5-PAS, AB pH 1.0, AF pH 1.7-AB pH 2.5 and HID-AB pH 2.5 methods. 1. For the morphologic changes, the lingual gustatory glands of CP group, compared with the control group, were noted atrophy of serous adrd, decrease and disapperance of zymogen granules and vacuolation tn the serous cell. The mucification of duct cells was appeared in CP group. And the lingual mucous glands of CP group, compared with the control group, showed atrophy of mucous acini, and decrease of mudgen granules in mucous cells. Especially, many enlarged, fused mucous acini, and vacuolated mucous cells were detected. The mucification of duct cells increased very notably in CP group. 2. Neutral mucin of the lingual gustatory gland, and add and neutral mucins of lingual mucous gland decreased very remarkably in CP group compared with the control group. Especially, the decrease of acid mucin was more remarkable than that of neutral mucin in the mucous gland. But, in CP group, the mucous cells which secrete sulfomudn decreased, and the mucous cells which secrete sialomucin increased remarkably.

        피조개 발의 점액질에 대한 조직화학적 연구

        피조개 발의 조직학적 구조는 hematoxylin-eosin염색 및 Weigert iron hematoxylin염색을, 점액질 성상을 검색하기 위하여 PAS반응, diastase 침지 후 PAS반응, alcian blue pH 2.5염색, alcian blue pH 1.0염색, alcian blue pH 2.5-PAS염색, alcian blue pH 1.0-PAS염색, aldehyde fuchsin pH 1.7-alcian blue pH 2.5염색 및 0.25% toluidine blue염색을 실시하였다. 피조개의 발은 상피, 상피하결합조직 및 근층으로 이루어져 있었으며 상피는 단층섬모원주세포로 되어 있었다. 상피하결합조직 및 근층에 다세포성 점액선인 발선이 발달되어 있었고 도관은 결합조직과 상피세포를 거쳐 개구되어 있었다. 근층은 종주근, 수직근 및 횡근섬유로 구성되어 있었다. 점액질의 조성을 보면 상피세포섬모에는 sialomucin만을, 상피세포는 중성점액질만 함유하고 있었다. 상피세포 및 결합조직내 점액도관에는 산성과 중성점액질의 혼합성인 것과 산성점액질만 함유한 것이 혼재하나 전자가 더 많았다. 산성점액질의 성상으로 보아 대부분이 강 sulfomucin을 가지며 일부는 약 sulfomucin과 sialomucin의 혼합성이었다. 대부분 발선의 점액선포에 대한 점액질 조성은 점액도관과 유사하였으나 일부 상부발선의 점액선포는 중성점액질만을, 하부발선의 점액선포는 sialomucin만을 함유하였다. This study was performed to investigate the histological structure and the histochemical properties of the mucosubstances in the foot of Anadara broughonii. For the histological structure, hematoxylin-eosin and Weigert iron hematoxylin staining were used. The mucosubstances stained with PAS, PAS-diastase, 0.25% toluidine blue, alcian blue pH 2.5, alcian blue pH 1.0, alcian blue pH 2.5-PAS, alcian blue pH 1.0-PAS, aldehyde fuchsin pH 1.7-alcian blue pH 2.5 methods. The foot was composed basically of three layers: epithelium, subepithelial connective tissue and muscular layers. The foot was lined by simple ciliated columnar epithelial cell. Foot glands consisted of multicelluear mucous acini were present in the subepithelial connective tissue and muscular layer. The mucous duct of foot glands was distributed from connective tissue to epithelial cells, and open onto the surface of the foot. The foot muscle traveled in three planes as longitudinal, transverse and vertical muscle fiber. About the properties of mucosubstances on the foot, cilia of epithelial cell contained sialomucin only and epithelial cell neutral mucosubstances only. Most of mucous duct within epithelial cells and connective tissue showed a mixture of acidic and neutral mucosubstances, while a few mucous ducts were contained acidic ones only. Among acidic mucosubstances, notable strong sulfomucin were found in most mucous cell of duct, and a mixture of weak sulfomucin and sialomucin appeared in a few cells. The properties of mucosubstances in the food glands were similiar to those of mucous duct. But a few mucous acini contained neutral mucosubstances and sialomucin only in upper and lower portion of foot gland, respectively.
