        Protein profiling predicts the response to anthracycline and taxanes based neo-adjuvant chemotherapy in breast cancer

        Neo-adjuvant chemotherapy for breast cancer substantially benefits patients who achieve pathological response. However, clinical or pathological response information can only be obtained a period of time after chemotherapy. The identification of novel bio-markers or the application of new technique that can be used to predict treatment response before che-motherapy would allow therapy to be tailored on an individual patient basis. The purpose of this study is to identify the chemo-sensitivity and chemo-resistance related proteins using antibody microarray profiling, and to develop a multi-protein predictive model for breast cancer. Total protein was extracted from core needle biopsy samples obtained from 15 patients before treatment with neo-adjuvant TA(combination of taxanes and anthracycline) chemotherapy. Protein profiling was analyzed by antibody microarray. 10 pati-ents were used as training set to develop the predictive model using the software PAM(prediction analysis of microarray). Another 5 patients were used as a validation set to test the model. In cross-validation, the mole-cular predictive model showed an accuracy of 90%, in independent validation, the model classified the cases with an accuracy of 80%. In conclusion, the proteomic predictive model has the potential to predict pathological response to neo-adjuvant TA chemotherapy.

        Multi-RIS Aided 3D Secure Precise Wireless Transmission

        In this paper, multiple reconfigurable intelligent surfaces (RIS) aided secure precise wireless transmission (SPWT) schemes are proposed in the three-dimensional (3D) wire- less communication scenario. Unavailable direct path channels from transmitter to receivers are considered when the di- rect paths are obstructed by obstacles. Then, multiple RISs are utilized to achieve SPWT through the reflection path among transmitter, RISs and receivers in order to enhance the communication performance and energy efficiency simulta- neously. First, a maximum-signal-to-interference-and-noise ratio (MSINR) scheme is proposed in a single user scenario. Then, the multi-user scenario is considered where the illegitimate users are regarded as eavesdroppers. A maximum-secrecy-rate (MSR) scheme and a maximum-signal-to-leakage-and-noise ratio (MSLNR) are proposed. The former achieves a better secrecy rate (SR) performance but incurs a higher complexity. The latter has a lower complexity than the MSR scheme with an SR performance loss. Simulation results show that both single- user scheme and multi-user scheme can achieve SPWT which transmits confidential message precisely to location of desired users. Moreover, MSLNR scheme has a lower complexity than the MSR scheme, while the SR performance is close to that of the MSR scheme.

        Association of Measures of Glucose Metabolism with Colorectal Cancer Risk in Older Chinese: A 13-Year Follow-up of the Guangzhou Biobank Cohort Study-Cardiovascular Disease Substudy and Meta-Analysis

        Background: Abnormal glucose metabolism is a risk factor for colorectal cancer (CRC). However, association of glycosylated hemoglobin (HbA1c) with CRC risk remains under-reported. We examined the association between glycemic indicators (HbA1c, fasting plasma glucose, fasting insulin, 2-hour glucose, 2-hour insulin, and homeostasis model of risk assessment-insulin resistance index) and CRC risk using prospective analysis and meta-analysis.Methods: Participants (<i>n</i>=1,915) from the Guangzhou Biobank Cohort Study-Cardiovascular Disease Substudy were included. CRC events were identified through record linkage. Cox regression was used to assess the associations of glycemic indicators with CRC risk. A meta-analysis was performed to investigate the association between HbA1c and CRC risk.Results: During an average of 12.9 years follow-up (standard deviation, 2.8), 42 incident CRC cases occurred. After adjusting for potential confounders, the hazard ratio (95% confidence interval [CI]) of CRC for per % increment in HbA1c was 1.28 (95% CI, 1.01 to 1.63) in overall population, 1.51 (95% CI, 1.13 to 2.02) in women and 1.06 (95% CI, 0.68 to 1.68) in men. No significant association of other measures of glycemic indicators and baseline diabetes with CRC risk was found. Meta-analyses of 523,857 participants including our results showed that per % increment of HbA1c was associated with 13% higher risk of CRC, with the pooled risk ratio being 1.13 (95% CI, 1.01 to 1.27). Subgroupanalyses found stronger associations in women, colon cancer, Asians, and case-control studies.Conclusion: Higher HbA1c was a significant predictor of CRC in the general population. Our findings shed light on the pathology of glucose metabolism and CRC, which warrants more in-depth investigation.

      • MicroRNA-155 Expression has Prognostic Value in Patients with Non-small Cell Lung Cancer and Digestive System Carcinomas

        Objective: Published data have shown that microRNAs (miRNAs) could play a potential role as diagnostic and prognostic indicators in cancers. Data for the predictive value of microRNA-155 are inconclusive. The aim of the present analysis was therefore to evaluate the role of miR-155 in prognosis for patients with a variety of carcinomas. Methods: Relevant studies were identified by searching PubMed and EMBASE. Data were extracted from studies comparing overall survival (OS), recurrence-free survival (RFS) or cancer-specific survival (CSS) in patients with carcinoma with higher miR-155 expression and those with lower levels. The pooled hazard ratios (HRs) and 95% confidence intervals (CIs) of miR-155 for clinical outcome were calculated. Results: A total of 15 studies were included. The pooled hazard ratio (HR) for OS of higher miR-155 expression in cancerous tissue was 1.89 (95% CI: 1.20-2.99, P=0.006), which could markedly predict poorer survival in general cancer. For RFS/CSS, elevated miR-155 was also associated with poor prognosis of cancer (HR=1.50, 95% CI: 1.10-2.05, P=0.01). On subgroup analysis, the pooled HR for OS in non-small cell lung cancer (NSCLC) was 2.09 (95% CI: 0.68-6.41, P > 0.05), but for RFS/CSS was 1.28 (95% CI: 1.05-1.55, P=0.015), with statistical significance; the pooled HRs for OS and RFS/CSS in digestive system neoplasms were 3.04 (95% CI: 1.48-6.24, P=0.003) and 2.61 (95% CI: 1.98-3.42, P<0.05), respectively. Conclusions: The results indicated that the miR-155 expression level plays a prognostic role in patients with cancer, especially NSCLCs and digestive system carcinomas.

        Darapladib, a Lipoprotein-Associated Phospholipase A2 Inhibitor, Reduces Rho Kinase Activity in Atherosclerosis

        Purpose: Increased lipoprotein-associated phospholipase A2 (Lp-PLA2) activity and Rho kinase activity may be associated with atherosclerosis. The principal aim of this study was to examine whether darapladib (a selective Lp-PLA2 inhibitor) could reduce the elevated Lp-PLA2 and Rho kinase activity in atherosclerosis. Materials and Methods: Studies were performed in male Sprague-Dawley rats. The atherosclerosis rats were prepared by feeding them with a high-cholesterol diet for 10 weeks. Low-dose darapladib (25 mg·kg-1·d-1) and high-dose darapladib (50 mg·kg-1·d-1) interventions were then administered over the course of 2 weeks. Results: The serum levels of triglycerides, total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoproteincholesterol (HDL-C), high-sensitivity C-reactive protein (hs-CRP), and Lp-PLA2, significantly increased in atherosclerosis model groups, as did Rho kinase activity and cardiomyocyte apoptosis (p<0.05 vs. sham group), whereas nitric oxide (NO) productionwas reduced. Levels of TC, LDL-C, CRP, Lp-PLA2, and Rho kinase activity were respectively reduced in darapladib groups, whereas NO production was enhanced. When compared to the low-dose darapladib group, the reduction of the levels of TC, LDL-C, CRP, and Lp-PLA2 was more prominent in the high-dose darapladib group (p<0.05), and the increase of NO productionwas more prominent (p<0.05). Cardiomyocyte apoptosis of the high-dose darapladib group was also significantly reduced compared to the low-dose darapladib group (p<0.05). However, there was no significant difference in Rho kinase activity between the low-dose darapladib group and the high-dose darapladib group (p>0.05). Conclusion: Darapladib, a Lp-PLA2 inhibitor, leads to cardiovascular protection that might be mediated by its inhibition of both Rho kinase and Lp-PLA2 in atherosclerosis.

        An integrated blade optimization approach based on parallel ANN and GA with hierarchical fair competition dynamic-niche

        This work presented an available multi-point blade optimization procedure for better aerodynamic performances. Based on the proposed Parallel ANN and GA with hierarchical fair competition dynamic-niche (GA-HFCDN), an integrated approach for the blade optimization design was put forward by combining Bezier parameterization with FINE/TURBO solver. In the optimization design, parallel ANN was employed to build a more proper approximate model. GA-HFCDN was proposed to obtain the global optimization solution more efficiently for the blade design. Two research cases, including plane cascades blade optimization and the optimization and experimental study of a low specific speed centrifugal blower blade, were performed by using the above approach. The conclusions showed the rationality and validity of the optimization approach.

        Design of Coordination-Crosslinked Nitrile Rubber with Self-Healing and Reprocessing Ability

        In this work, we describe a simple approach to design of a coordination crosslinked elastomer with extraordinary mechanical properties: self-healing and recycling abilities by introducing Co-cyano coordination into nitrile rubber(NBR) matrix. The formation of reversible coordination crosslink network results in good mechanical properties superior to conventional sulfur-cured NBR, such as high stretchability and toughness. The reversibility of metal-ligand bond endows the polymer with self-healing capability and recycling or reprocessing abilities by rearrangement at elevated temperature. The fully cut samples after self-healing or reprocessing can restore their original tensile strength. It is also worth noting that the reclaimed NBR exhibited higher modulus than its original sample due to the compensation of new metal-ligand bonds formed during heating. This study provides possibilities to extend lifetime, reshaping and recycling capabilities of commercial rubber and reduce toxicity and environmental issues caused by waste rubber.

      • Paris polyphylla Smith Extract Induces Apoptosis and Activates Cancer Suppressor Gene Connexin26 Expression

        Background: The inhibition of tumor cell growth without toxicity to normal cells is an important target in cancer therapy. One possible way to increase the efficacy of anticancer drugs and to decrease toxicity or side effects is to develop traditional natural products, especially from medicinal plants. Paris polyphylla Smith has shown anti-tumour effects by inhibition of tumor promotion and inducement of tumor cell apoptosis, but mechanisms are still not well understood. The present study was to explore the effect of Paris polyphylla Smith extract (PPSE) on connexin26 and growth control in human esophageal cancer ECA109 cells. Methods: The effects of PPSE on Connexin26 were examined by RT-PCR, western blot and immunofluorescence; cell growth and proliferation were examined by the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl-tetrazolium bromide (MTT) assay. Results: PPSE inhibited the growth and proliferation on esophageal cancer ECA109 cells, while increasing the expression of connexin26 mRNA and protein; conversely, PPSE decreased Bcl-2 and increased Bad. Conclusion: This study firstly shows that PPSE can increase connexin26 expression at mRNA and protein level, exerting anti-tumour effects on esophageal cacner ECA109 cells via inhibiting cell proliferation and inducing cell apoptosis.

      • Hyperin Extracted from Manchurian Rhododendron Leaf Induces Apoptosis in Human Endometrial Cancer Cells Through a Mitochondrial Pathway

        Background: A number of effective prevention measures have been introduced in attempts to substantially reduce both the incidence and mortality due to many kinds of cancer. The search for new anti-cancer compounds in foods or in plant medicines is one realistic and promising approach to prevention. Chinese medicines provide a rich pool of novel and efficacious agents for cancer prevention and treatment. Previously it was demonstratrated that hyperin extracted from the Manchurian rhododendron leaf reduces the proliferation of many cancer cells. The present study was carried out to evaluate its effects on human endometrial cancer cell viability and apoptosis and to investigate its mechanisms of action in RL952 cells. Methods: Cell viability was measured using the MTT assay. Intracellular calcium ions were detected using laser-scanning confocal microscopy. The effects of hyperin on apoptosis related proteins in RL952 cells were examined using Western blot analysis. Results: The growth of RL952 cells was inhibited by treatment with hyperin. OD values of caspase-3 and caspase-9 were increased and expression of bcl-2 was increased and bax was decreased in protein levels in RL952 cells after 24 h of hyperin treatment, Moreover, intracellular calcium accumulation occurred in hyperin-treated cells. Conclusion: These results suggest that hyperin may play an important role in tumor growth suppression by inducing apoptosis in human endometrial cells via a $Ca^{2+}$-related mitochondrion apoptotic pathway in RL952 cells.
