The DcuS-R two-component system of Escherichia coli senses C4-dicarboxylates of the medium and regulates expression of the genes related to utilization of them. It is known that phospho-DcuR induces expression of genes such as the dcuB-fumB operon, the frdABCD operon, and the dctA gene. We analyzed promoters of the dcuS-R operon to elucidate the transcriptional regulation system. We found a novel internal promoter within the dcuS gene that is regulated by the transcriptional regulator, CRP-cAMP, in both aerobic and anaerobic conditions.

The pulp yield was improved by about 4.5-5% when polysulfide (PS) and anthraquinone (AQ) were added to the kraft cooking liquor (white liquor). The exchange of the black liquor with fresh white liquor further increased the yield. The highest pulp yield was obtained when the PS cooking liquor containing 70% of total active alkali (AA) and 100% of AQ was used from the beginning of the reaction and the black liquor was exchanged with fresh white liquor containing the residual 30% of AA just after temperature reached 135℃. There was a good correlation between kraft pulp yields of a hardwood species and the ratios of the amount of xylose to glucose (X/G ratio), liberated by an acid hydrolysis of the pulps. However, the correlation was dependent on raw material wood species. Therefore, it is required in advance to establish a correlation between the yields and X/G ratios for raw material wood species of a target pulp in order to estimate pulp yield using X/G ratio. The X/G ratios of relatively high yield pulps showed higher values than those expected from the correlation. In a mill trial, the superiority of the PS-AQ isothermal cooking (ITC) process over the kraft ITC process was confirmed by examining X/G ratio of pulps obtained. The pulp yield in the PS-AQ ITC process was estimated at about 57.0%. This yield is very high, which indicates that reaction conditions of the PS-AQ ITC process are optimal.
Identification of PhoB Binding Sites of the yibD and ytfK Promoter Regions in Escherichia coli

By using a lacZ operon fusion genomic library of the Escherichia coli O157:H7 Sakai, we identified phosphatestarvation-inducible (psi) promoters located upstream of the yibD and ytfK genes. They have been previously proposed to belong to the phosphate regulon (pho regulon) by Beak and Lee (2006), based on the DNA array and in vivo transcriptional experiments. However, the direct interaction of these promoters with the activator protein of the pho regulon, PhoB, has not been determined. We determined the binding regions of PhoB in these promoter regions by DNase I footprinting. Both regions contained two pho boxes similar to the consensus sequence for PhoB binding.
General Tendency of Lignin Chemical Structure and its Relation to Reactivity

Hardwood lignins are different from softwood lignins by the presence of syringyl type aromatic nuclei. In addition, there are wide varieties in the structure and amount of hardwood lignins depending on wood species, environment of growing site, portion in the wood, portion in the cell wall, and so on. We have shown that the wide variety of lignin structure and amount can be sorted out by taking the syringyl/guaiacyl ratio as an index. Furthermore, we have also shown that lignin structure can be quantitatively related to the chemical reactivity such as delignification during chemical pulping by taking the syringyl/guaiacyl ratio as an index. In this report, we review our recent achievements about the quantitative relationships between lignin structure and reactivity.
