      • 들깻잎에서 동정한 Phytol의 항암 및 면역활성증강 효과

        Several studies have shown that extracts from yellow-green vegetables reveal antitumor activities. In the present study we investigated the effect of phytol in order to elucidate the immunological mechanism of antitumor activity of this substance. The results obtained from the experiment as follows : 1) Phytol showed cytotoxic effect on sarcoma 180 cells in vitro. 2) When phytol was injected into the peritoneal cavity of mice transplanted with sarcoma 180 cells, the average survival time(24.0 days) tended to increase as compared with the nontreated control(19.2 days). 3) When sarcoma 180 cells were injected subcutaneously into the right groin of mice and then phytol was injected into the peritoneal cavity the tumor inhibition ratio was 33%. 4) The natural killer(NK) cell activity was significantly augmented by phytol in vitro and in vivo. Similar augmentations of NK cell activity were obtained with culture supernatants of phytol exposed spleen cells and peripheral blood mononuclear cells. 5) Phytol on the macrophage from peritoneal cavity showed a higher effectiveness in vivo than in vitro. These results indicate that phytol shows the inhibitory effect for growth of sarcoma 180 cells in vitro, also it can augment macrophage and NK cell activities in vivo.

      • Synechococcus PCC7002의 세포 및 틸라코이드 막의 Polyvinylalcohol 고정화에 의한 광합성 특성

        산소발생능에 있어 오랜 기간 안정성을 유지하는 세포와 분리 틸라코이드 막을 단세포성 남조류인 Synechococcus PCC7002로보터 PVA로 고정화하여 구할 수 있었다. 분리 틸라코이드 막 및 고정화한 세포나 틸라코이드 막의 흡수스펙트럼의 굽대가 청색 쪽이로 3㎚ 정도 전이하였다. PVA에 고정화한 세포나 틸라코이드 막은 4°C 에 보관할 경우 광합성에 의한 전자전달 활성, 특히 광계 2의 활성을 오랜기간 보존할 수 있었다. 세포를 고온헤 처리하였을 때 Fo와 Fv가 증가하였으나 고정화한 후에는 Fo의 변화가 거의 없었다. 이 결과는 고정화가 열처리에 의한 광계 2 복합체의 손상 특히 물분해기구의 손상에 대해 보호작용을 할 수 있음을 시사한다. Highly stable O₂-evolving cells and thylakoid membranes have been obtained from the cyanobacterium, Synechococcus PCC7002, by immobilization with polyvinylalcohol(PVA). The absorption peak showed the blue-shift of about 3 nm after immobilization of intact cells and thylakoid membranes as well as isolation of thylakoid membranes. Photosynthetic electron transport activities, especially PS Ⅱ activity showed greater stability in the PVA-immobilized cells and thylakoid membranes when stored at 4°C than in those at 25°C. When the cells were threated at higher temperature, the level of Fo and Fv increased. After immobilization, however, Fo showed no change. This suggests that the immobilization can protect against the damages of PS Ⅱ complex, especially a water-spiliting system, by heat treatment.       • Linoleic acid와 Ursolic acid가 마우스 Natural Killer 세포활성에 미치는 효과

        Several studies have reported that linoleic acid(LA) and ursolic acid(UA) extracted from yellow-green vegetables reveal antitumor activities. In the present study we examined the natural killer(NK) cell activity in order to elucidate the mechanism of antitumor activity of these substances. The findings in this study are summerized as follows. The NK cell activity was significantly augmented by LA and UA both in vitro and in vivo. The effect of LA appeared to be proportional to its concentration, however the effect of UA was inhibitory at high concentration(5.00㎍/㎖). Similar augmentations of NK cell activity were obtained with culture supernatants of LA or UA-exposed spleen cells and peripheral blood mononuclear cells. With these results, we suggest that the LA or UA-exposed cells produce cytokines related to enhancement of NK cell activity.

      • DNA 합성관련 효소 억제제 및 인삼 Saponin이 Chinese Hamster Ovary(CHO) 세포손상에 미치는 영향

        The physiological activity of adriamycin(ADR) is related to its ability to bind specifically with DNA by intercalation between adjacent base pairs of the double helical structure and ADR induce DNA damage, chromosome aberrations and sister chromatid exchange in mammalian cell as the inhibitors of DNA synthesis related enzyme such as the aphidicolin(APC), novobiocin(Novo) and ginseng saponin(GS) on the frequencies of chromosome aberration, sister chromatid exchanges(SCEs), we treated ADR in CHO cells and post-treated APC, Novo and GS. 1. CHO cell viability by ADR was 60% in- 1ng/ml and 0% l g/ml concentration. 2. When we treated ADR, APC, Novo and GS separately, only ADR increased, chromosome aberrations and SCEs frequency. In proportion to the concentration of ADR increased, the aberration frequency was also increased. The more the concentration of ADR went up, the better the frequency of aberration rose. 3. In the group treated with ADR+APC or ADR+Novo, the frequency of the chromosome aberration and sister chromatid exchange were higher than the group treated with ADR alone, but decreased by post-treated ginseng saponin. The group treated with ADR, NOVO and GS inhibited SCEs frequency significantly among the groups. These results suggest that GS can inhibit chromosome aberration and sister chromatid exchange induced by ADR, APC or ADRNovo.

      • Linoleic acid, Ursolic acid, Phytol, 들미나리추출물이 마우스 Phagocyte에 미치는 효과

        We previously reported that linoleic acid (LA), ursolic acid (UA), phytol (Pt), and small water dropwort (SWD, Oenanthe javanica DC, will type) extract revealed strong antimutagenic and antitumor activities. In the present study we examined the phagocytic activity (engulfment) and nitroblue tetrazolium (NBT) reduction (cytotoxicity) in order to elucidate the mechanism of antitumor activity of these substances. The findings in this study are summerized as follows. The phagocytic activity of peritoneal phagocyte of mice was significantly augmented by LA, UA, Pt, and SWD in vitro. The NBT reduction of peritoneal phagocyte of mice was significantly increased by 1 time exposure of LA, Pt, SWD in vivo. From the above results, we suggest that the LA, UA, Pt, or SWD-exposed cells show the enhancement of phagocytic activity and the peritoneal phagocyte of the LA, Pt, or SWD-injected mice produce higher superoxide than in vitro exposure.

      • Photosynthetic Characteristics of Polyvinylalcohol-Immobilized Spinach Chloroplasts

        Photosynthetic properties of polyvinylalcohol (PVA)-immobilized chloroplast especially regarded to stability of photosynthetic electron transport and the fluorescence induction pattern were studied. When isolated spinach chloroplasts were immobilized with PVA, it showed good preservation of photosynthetic electron transport activity, especially PS Ⅱ activity, during storage at -15℃, 4℃, and 20℃. And immobilized chloroplasts revealed similar thermostability of whole chain electron transport to free chloroplasts. And the absorption peak or red band of chloroplasts showed the blue-shift of 2-4 nm after immobilization. F_v/F_m ratio of chlorophyll fluorescence slightly decreased after immobilization. White light pulse after continuous light do not induced the additional fluorescence rise. This means chlorophyll fluorescence at room temperature reached to F_max under continuous light in the immobilized chloroplasts. It seems that PVA may be a good candidate for immobilization matrix for the preservation of photosynthetic function of thylakoids and for the continuous use of chloroplast membranes of higher plants for solar energy storage and conversion.

        시금치에서 분리한 엽록체를 폴리비닐알콜에 고정화한후 광합성특성을 조사하였다. 고정화한 엽록체의 경우 고정화하지 않은 엽록체에 비하여 광합성전자전달활성의 안정성이 크게 증가하였는데 4℃에서 보관할 경우 1개월 후까지 전자전달활성이 보존되었으며, 열처리에 대한 내성은 유사하였다. 고정화한 경우 엽록체의 흡수스펙트럼의 적색피크가 청색쪽으로 2-4 ㎚ 전이하였다. 그러나 이와 같은 엽록체의 구조적인 변화가 고정화 후의 전자전달활성이 대조구에 비하여 큰 저하를 일으키지 않는 것으로 보아 전자전달계의 직접적인 활성변화를 유발하는 구조적변화는 아닌 것으로 생각된다. 고정화한 엽록체의 엽록소 형광패턴은 백색광에 의한 spike전이가 일어나지 않아 이미 actinic 광하에서 F?로의 전이가 일어난 것으로 생각된다. 또한 actinic 광하에서 F?로의 전이시간이 길어짐을 볼 수 있었다. 이와 같은 현상은 고정화한 엽록체의 광계에 서의 여기에너지의 열발산이나 광계 Ⅱ의 전자공여부위의 저해에 의하여 나타날 수 있다고 보여지나 이에 대한 보다 자세한 연구가 필요하다고 사료된다.         Anabaena 의 광합성 전자전달 활성에 미치는 Simazine 의 억제효과

        Effects of simazine [2-chloro-4,6-bis(methylamino)-s-triazine] on the photochemical reactions of isolated spinach chloroplasts and crude thylakoids of Anabaena inequalis UTEX B-381 were compared. Simazine inhibited photosynthetic O_2 evolution and increased the chlorophyll fluorescence in whole cells of Anabaena. The electron transfer from diphenylcarbazide to 2,6-dichlorophenolindophenol was inhibited by simazine treatment in spinach chloroplasts, but not in crude thylakoids of Anabaena. In spinach chloroplasts, the chlorophyll fluorescence was increased by simazine treatment in the presence of diphenylcarbazide and ferricyanide, but not in the presence of diphenylcarbazide and silicomolybdate. In crude thylakoids of Anabaena, simazine treatment did not increase the chlorophyll fluorescence in the presence of either diphenylcarbazide and silicomolybdate, or diphenylcarbazide and ferricyanide. These results suggest that the inhibitory site of simazine on photosynthetic electron transport chain of Anabaena is different from that of spinach chloroplasts. And there may be a possibility that the inhibition site of simazine in Anabaena lies on the donor side of photosystem Ⅱ, before the site of electron donation by diphenylcarbazide.
