The marine red alga Pyropia tenera (Rhodophyta) is one of the most valuable and widely cultivated Pyropia species. Temperature is a major factor that affects the growth and life cycle of Pyropia. Small heat-shock proteins (sHSPs) play a crucial role in heat stress response. Among sHSPs isolated from P. tenera, PtsHSP19.6 showed the strongest response to heat stress but not to other abiotic stresses such as desiccation and freezing. Although an α-crystallin domain was found in PtsHSP19.6, its amino acid sequence showed low homology with known sHSPs, except those from red algae. This led us to investigate whether PtsHSP19.6 from red algae has chaperone activity and is involved in high-temperature tolerance. PtsHSP19.6 significantly reduced the thermal aggregation of alcohol dehydrogenase used as a substrate. When PtsHSP19.6 was introduced into Escherichia coli, the transformed cells grew better than the control cells under high-temperature conditions. Fluorescence of the PtsHSP19.6-GFP fusion protein was detected from granules in the cytoplasm. The in vitro analysis data showed that PtsHSP19.6 forms oligomers with a molecular weight greater than 240 kDa in solution. This study showed that PtsHSP19.6 from P. tenera forms oligomers in solution and plays a role in heat tolerance, with a chaperone function similar to that observed in green plants.

In this study, a hydrogen/deuterium (H/D) exchange method using gas chromatography–electrospray ionization/mass spectrometry (GC–ESI/MS) was first investigated as a novel tool for online H/D exchange of multitarget analytes. The GC and ESI source were combined with a homemade heated column transfer line. GC–ESI/MS-based H/D exchange occurs in an atmospheric pressure ion source as a result of reacting the gas-phase analyte eluted from GC with charged droplets of deuterium oxide infused as the ESI spray solvent. The consumption of the deuterated solvent at a flow rate of 2 μL min–1 was more economical than that in online H/D exchange methods reported to date. In-ESI-source H/D exchange by GC–ESI/MS was applied to 11 stimulants with secondary amino or hydroxyl groups. After H/D exchange, the spectra of the stimulants showed unexchanged, partially exchanged, and fully exchanged ions showing various degrees of exchange. The relative abundances corrected for naturally occurring isotopes of the fully exchanged ions of stimulants, except for etamivan, were in the range 24.3–85.5%. Methylephedrine and cyclazodone showed low H/D exchange efficiency under acidic, neutral, and basic spray solvent conditions and nonexchange for etamivan with an acidic phenolic OH group. The in-ESI-source H/D exchange efficiency by GC–ESI/MS was sufficient to determine the number of hydrogen by elucidation of fragmentation from the spectrum. Therefore, this online H/D exchange technique using GC–ESI/MS has potential as an alternative method for simultaneous H/D exchange of multitarget analytes.

Graphic Abstract

Aims: Treatment responses of unresectable hepatocellular carcinoma (HCC) remain unacceptably low and treatment modalities are limited. We compared the efficacy and safety of concurrent chemoradiotherapy (CCRT). Methods: In retrospective cohort study, data on 74 patients with unresectable HCC, with Child-Turcotte-Pugh (CTP) scores of 5-8, were collected from a university hospital between January 2009 and October 2018. All patients were treated with CCRT (5-fluorouracil 500mg/㎡ via intraarterial chemoport at Day 1 to 5 and Day 20 to 25, plus radiotherapy 6,250 cGy/25 times at Day 1 to 25). Results: From 74 patients with unresectable HCC, 71.6% were classified as Child-Pugh (CP)-A, 86.4% as Barcelona Clinic Liver Cancer (BCLC)-C. The median overall survival (OS) and time to progression (TTP) were 13 months, and 8 months in the CCRT group. In univariate analysis, operation, ECOG, Child-Pugh score, diffuse type, main portal vein invasion, bile duct invasion, albumin, PT, HBV DNA were significant prognostic factors of OS (P=0.014, 0.005, 0.024, 0.029, 0.012, 0.008, 0.017, 0.000, 0.023), whereas operation, antiviral agent, ECOG, Child-Pugh score, age, creatinine, albumin, HBV DNA, PIVKA-II were significant prognostic factors of TTP (P=0.000, 0.008, 0.012, 0.031, 0.023, 0.023, 0.024, 0.024, 0.006). In multivariate analysis, operation, diffuse type, ECOG were significant prognostic factors of OS (P=0.028, 0.037, 0.043), whereas operation, antiviral agent, Child-Pugh score were significant prognostic factors of TTP (P=0.008, 0.044, 0.037). Major complications included hyperbilirubinemia (44.8%), ALT elevation (34.5%), ascites (13.8%), gastric ulcer (14.5%), catheter-related complications (3.4%) and radiation pneumonitis (2.3%). Conclusions: For managing unresectable HCC, CCRT may be a valuable and safe treatment modality.
Anomalous extinction in index-matched terahertz nanogaps
Jeong, Jeeyoon,Kim, Dasom,Park, Hyeong-Ryeol,Kang, Taehee,Lee, Dukhyung,Kim, Sunghwan,Bahk, Young-Mi,Kim, Dai-Sik De Gruyter 2018 Nanophotonics Vol.7 No.1

Slot-type nanogaps have been widely utilized in transmission geometry because of their advantages of exclusive light funneling and exact quantification of near-field enhancement at the gap. For further application of the nanogaps in electromagnetic interactions with various target materials, complementary studies on both transmission and reflection properties of the nanogaps are necessary. Here, we observe an anomalous extinction of terahertz waves interacting with rectangular ring-shaped sub-30 nm wide gaps. Substrate works as an index matching layer for the nanogaps, leading to a stronger field enhancement and increased nonlinearity at the gap under substrate-side illumination. This effect is expressed in reflection as a larger dip at the resonance, caused by destructive interference of the diffracted field from the gap with the reflected beam from the metal. The resulting extinction at the resonance is larger than 60% of the incident power, even without any absorbing material in the whole nanogap structure. The extinction even decreases in the presence of an absorbing medium on top of the nanogaps, suggesting that transmission and reflection from nanogaps might not necessarily represent the absorption of the whole structure.

Microchannel plates (MCPs) have been developed by introducing new materials and process technologies. Main body was made of alumina by programmable punching, laminating, and firing. The channel walls of pore arrays of an MCP were deposited with thin films by electro1ess copper plating and sol-gel process. Our MCP has advantages such as easy fabrication, durability, high temperature endurance, and applicability to the large size comparing with the conventional MCPs. Experiments on the brightness of an MCP incorporated FED revealed that the FED with a MCP is three to four times brighter than a conventional FED. Moreover, the focusing in a FED is improved. Incorporating an MCP into a FED is one of promising methods to enhance the characteristics of the FED. In addition, amplification yield of the MCP is measured for varying the aspect ratio and the input current.
