      • S-510 A case of peritoneal dialysis-associated peritonitis caused by Agromyces mediolanus

        Introduction:?Agromyces mediolanus (A.mediolanus) is an unusual pathogen in human infection. No case of peritoneal dialysis (PD)-associated peritonitis by A.mediolanus has been reported. This is the first case report of PD-associated peritonitis by A.mediolanus in the world. We describe a case of PD-associated peritonitis by A.mediolanus that was cured after catheter removal.?Case: A 59-year-old man who had been treated with CAPD was admitted to our hospital because of turbid peritoneal effluent accompanied by constant diffuse abdominal pain. The patient had an exit-site infection with purulent secretion. The laboratory findings showed PD peritonitis: the white blood cell (WBC) count of the peritoneal effluents was 1157/mm3 with a neutrophil predominance (91.3%). Cefazolin and ceftazidime were given intraperitoneally per day. The peritoneal WBC decreased, but the patient’s clinical condition didn’t improve on the 5th day after starting intraperitoneal antibiotics. Culture of the peritoneal dialysate revealed Agromyces species, which was susceptible to meropenem, imipenem, vancomycin, but resistant to ceftriaxone. Thus, we changed antibiotics to intraperitoneal vancomycin and meropenem on the 5th day. Despite the intraperitoneal antibiotics for 4 days, the patient’s abdominal pain persisted and the peritoneal WBC count became elevated. Therefore, we removed the PD catheter and the patient was switched to hemodialysis on the 9th day. Vancomycin and imipenem was continued for another 2 weeks and the patient’s clinical condition improved. The patient has been maintained on hemodialysis.?Conclusions: There is no previous report of peritonitis associated with A.mediolanus in a CAPD patient. In this case, we used intraperitoneal cefazolin and ceftazidime and changed to vancomycin and imipenem according to the result of peritoneal dialysate. However the patient improved after removal of catheter. Therefore, early removal of the PD catheter in A. mediolanus related peritonitis without prompt reaction to antibiotic therapy should be considered.

        청소년 우울증 측정 및 관리를 위한 스마트폰 어플리케이션 콘텐츠 개발

        본 연구는 청소년 정신건강의 핵심문제 중 하나인 우울증의 측정과 관리를 위한 스마트폰 어플리케이션 콘텐츠 개발을 목적으로 하며, 이는 충남 소재 A대학의 청소년상담 관련학과, 컴퓨터공학 관련학과의 교수와 학생들의 협업으로 이루어졌다. 사용대상은 우울증의 예방 및 관리를 위해 집단상담에 참여하는 청소년들이며, 개발된 어플리케이션은 집단상담을 돕기 위해 사용될 예정이다. 콘텐츠 구성을 위해 먼저, 청소년 우울증의 발생원인과 특성, 수준측정방안, 심리치료이론을 고찰하였다. 다음으로 기존에 개발, 발표된 국내·외 어플리케이션과 선행연구를 통해 콘텐츠 내용과 실용성 등을 비교분석하였다. 이를 통해 청소년 우울증수준 측정방법으로 BDI-Ⅱ(우울척도), 심리치료 기법으로 인지행동치료, 치료 촉진방략으로 집단상담을 선정했다. 따라서 본 연구는 이에 초점을 맞춰 콘텐츠를 구성 및 개발하였으며, 개발 목적 및 실용화의 적절성 파악을 위해 청소년 5인을 대상으로 예비조사를 실시하였다. 예비조사 결과에서 제시된 제안점은 전문가와 실제 사용대상의 의견을 수렴하여 수정 보완할 예정이다. 결론적으로 본 콘텐츠는 일상성을 바탕으로 한 능동적 정서관리를 통해 효율적으로 청소년의 정신건강 웰니스를 지원할 것이다. The present study aims to develop the digital content of a smart-phone app for the measurement and management of depression – a key issue in adolescents’ mental health. This app has been developed by a psychology laboratory and a computer software engineering laboratory for an interdisciplinary convergence study. The target users of the app are adolescents who participate in group counseling for prevention and management for depression. The app is planned to be used as a tool for the group counseling. To develop the app content, we conducted a literature review. We then comparatively analyzed the key aspects of existing domestic and foreign apps designed to help for depression. The results of the analysis suggested the Beck Depression Inventory-II for the assessment method, the cognitive behavioral approach for the psychotherapeutic approach, and the group counseling that allows mutual support. Therefore, in this study, we organized and developed the app content by focusing on the identified aspects and conducted a pilot test with five adolescents to determine if the app met the development objectives. Modifications and improvements will be made based on the suggestions obtained from the pilot test and expert reviews. In conclusion, this app content is expected to enhance the wellness of adolescents effectively by improving proactive emotional management on a daily basis.

        Effects of Pyrolysis Temperature on Structural, Raman, and Infrared Properties of Perovskite PbTiO3 Nanotubes

        In this work, we investigated the effects of the pyrolysis temperature on the structural, Raman, and infrared properties of PbTiO3 nanotubes (PTO-NTs). The PTO-NTs were synthesized by spin coating a sol-gel solution on porous anodic alumina membranes, followed by the pyrolysis step at 400 − 600 C. Then, PTO-NTs were finally crystallized at 600 − 700 C in an oxygen atmosphere to get the perovskite phase. The PTO-NTs had an outer diameter of about 420 nm and a wall thickness of about 10 nm. X-ray diffraction patterns showed that the tetragonality (c/a) increased from 1.019 to 1.028 as the pyrolysis temperature was increased from 400 to 600 C. Raman spectroscopy showed that the phonon modes of A1(nTO) and E(nTO) increased with a redshift of the A1(3TO) mode as the pyrolysis temperature was increased. In the Fourier-transform infrared spectra of PTO-NTs embedded in the porous anodic alumina membrane, the transmittance of the band at 499 cm−1 increased as the pyrolysis temperature was increased, which might be due to an increase in the tetragonality.

        두 가지 삼원교잡종 (랜드레이스 x 요크셔 x 두록 및 랜드레이스 x 요크셔 x 우리흑돈)의 육질특성 비교

        본 연구는 외화 낭비를 줄이기 위해 우리흑돈이 종료종모돈으로써 두록을 대체할 수 있는지 조사하고자 하였다. 본 논문에서는 랜드레이스 X 요크셔 X 두록(LYD)과 랜드레이스 X 요크셔 X 우리흑돈(LYW)의 두 삼원교잡종에서 등심의 육질 특성을 비교하기 위해 이화학적 분석을 실시하였다. 공시 동물은 180일령의 LYD 59두와 LYW 60두, 총 119두를 이용하였으며, 도축 후 24시간 동안 냉각시키고, 돈육에 대한 이화학적 분석, 육색, 지방산 조성 및 관능평가를 실시하였다. 일반 성분 결과 수분 및 보수력은 두 품종간의 유의한 차이를 나타내지 않았다. 지방 함량 및 가열감량은 LYW에서 LYD에 비해 유의적으로 높게 나타났고(p <0.05), 전단력 및 pH는 LYW가 LYD보다 유의적으로 낮게 나타났다(p <0.05). 육색지수를 확인한 결과 적색도(a*) 및 황색도(b*)가 LYW에서 유의적으로 높게 나타났다(p <0.05). 두 품종의 지방산 함량을 비교한 결과 Stearic acid(C18:0)가 LYD에 비해 LYW에서 유의적으로 높게 나타났다(p <0.05). 관능검사 결과 LYW가 LYD에 비해 육색과 풍미가 유의적으로 높게 나타났다(p <0.05). 이러한 결과는 LYD와 비교하여 LYW에서 특정 화학적 특징이 개선되었음을 나타낸다. 따라서 우리흑돈이 수입 종돈인 두록의 대체 종돈으로 사용 가능할 것으로 사료된다. This study was undertaken to investigate whether the local Woori black pigs could replace Duroc as the terminal sire in order to reduce foreign currency outgoings. This study compared the physicochemical properties of pork loins in two cultivars of three-way crossbred pigs: Landrace X Yorkshire X Duroc (LYD) and Landrace X Yorkshire X Woori black pig (LYW). A total of 119 pigs (59 LYD and 60 LYW) aged 180 days were used in the experiment. After 24 hours of cooling, pork loins were assessed for their physicochemical traits, meat color, fatty acid and sensory evaluation. The moisture content and water holding capacity showed no significant difference between the two cultivars. Fat content and cooking loss were significantly higher in LYW than in LYD (p<0.05), whereas shear force and pH were significantly lower in LYW than LYD (p<0.05). Redness (a*), yellowness (b*), and stearic acid (C18: 0) content were also significantly higher in LYW than in LYD (p<0.05). Sensory evaluation revealed significantly higher meat color and flavor in LYW than LYD (p<0.05). Taken together, these results indicate that certain physicochemical qualities are improved in the LYW cultivar, as compared to LYD. Therefore, Woori black pig (W) has the potential to be substituted for Duroc, a breeding pig used currently.

        Suppression of Lipopolysaccharide-Induced Microglial Activation by a Benzothiazole Derivative

        We previously reported that KHG21834, a benzothiazole derivative, attenuates the beta-amyloid (A${\beta}$)-induced degeneration of both cortical and mesencephalic neurons in vitro. Central nervous system inflammation mediated by activated microglia is a key event in the development of neurodegenerative disease. In this study, we show that KHG21834 suppresses inflammation-mediated cytokine upregulation. Specifically, KHG21834 induces significant reductions in the lipopolysaccharide-induced activation of microglia and production of proinflammatory mediators such as tumor necrosis factor-${\alpha}$, interlukin-1${\beta}$, nitric oxide, and inducible nitric oxide synthase. In addition, KHG21834 blocks the expression of mitogen-activated protein kinases, including ERK, p38 MAPK, JNK, and Akt. In vivo intracerebroventricular infusion of KHG21834 also leads to decreases the level of interleukin-1${\beta}$ and tumor necrosis factor-${\alpha}$ in brain. These results, in combination with our previous findings on A${\beta}$-induced degeneration, support the potential therapeutic efficacy of KHG21834 for the treatment of neurodegenerative disorders via the targeting of key glial activation pathways.

        능이버섯 첨가가 우육의 물리화학적 및 관능적 특성에 미치는 영향

        단백질 가수분해 효소가 함유되어 있는 능이버섯이 우육의 연화, 보수력 및 품질특성에 미치는 영향을 알아보기 위하여 능이버섯 첨가량을 달리하여 조직학적 관찰, 수분함량, texture, 색도의 기계적인 측정과 관능적인 특성을 키위, 배를 첨가한 것과 비교하여 품질평가를 하였다. 투과전자현미경에 의한 미세구조 관찰 결과, 능이버섯(1,000 unit)을 25℃에서 10분간 처리했을 때 근원섬유 단백질이 분해되기 시작하였으며, 60분간 처리시 근절, a-band, Z-line 등을 전혀 구분할 수 없을 정도로 근원섬유의 분해현상이 현저하였다. 연육효과가 있는 능이버섯, 키위, 배를 우육에 첨가하였을 때 각 처리군의 수분함량은 첨가량이 증가할수록 유의적으로 증가하는 것으로 나타났다. Texture는 첨가량이 증가할수록 shear force는 감소하였으며 배를 첨가하는 것보다는 키위나 능이버섯을 첨가하는 것이 더 부드러운 조직감을 나타내었다. 색도에 있어서 L값은 능이버섯의 첨가량이 많아질수록 감소하였으나, 키위와 배의 첨가량이 많아질수록 증가하였으며 a값과 b값은 능이버섯, 배, 키위의 첨가량이 많아질수록 감소하였다. 관능검사 결과에서는 항목별로 유의적인 차이를 나타냈는데, 맛과 향미는 대조군을, 색은 배 10% 첨가군을 가장 선호하는 것으로 나타났다. 다즙성은 키위 25% 첨가군이 가장 좋았고 능이버섯, 키위, 배의 첨가량이 많아질수록 증가하였다. 연화도는 능이버섯과 키위 첨가군이 배 첨가군에 비해 높았다. 기호도에서는 대조군, 능이버섯 0.05%, 0.1% 첨가군, 배 10% 첨가군이 차이를 보이지 않았으며, 보다 부드러운 조직감과 다즙성을 고려하면 능이버섯 0.05~0.1% 정도의 첨가가 바람직한 것으로 보인다. An instrumental analysis of cooked beef was carried out along with sensory evaluation to find out the effect of Sarcodon aspratus on the physicochemical and sensory characteristics in comparision with kiwi fruit and pear. Transmission electron microscopy showed the muscle fiber started to be degraded when treated with Sarcodon aspratus (1,000 unit) for 10 min at 25℃. No distinct sarcomere, A-band, and Z-line was observed when treated with Sarcodon aspratus for 60 min at same condition. The moisture content of cooked beef was increased in proportion to the increment of Sarcodon aspratus, kiwi fruit and pear. In the texture, shear force of cooked beef was decreased with the increment of Sarcodon aspratus, kiwi fruit and pear. In terms of color, L-value was decreased by addition of Sarcodon aspratus, whereas L-value was increased by addition of kiwi fruit and pear in dose-dependent manners. a-value and b-value was decreased with the increment of Sarcodon aspratus, kiwi fruit and pear. There were significant differences (p<0.05) in the sensory characteristics of the samples in which control was most preferred in taste and flavor. As the content of Sarcodon aspratus, kiwi fruit and pear was increased, the score of juiciness and tenderness was increased. In the overall acceptance, score of 0.05~0.1% Sarcodon aspratus and 10% pear was not different from that of control. Therefore, it can be concluded that 0.05~0.1% addition of Sarcodon aspratus might be desirable for the improvement of texture and juiciness of cooked beef.

        Inhibitory Effect of Clavicepitaceae on Serotonin Release out of Human Platelets and Human Platelet Aggregation

        We have investigated the effects of hypha-water extracts (HWE), fruit body-water extracts (FWE) and cordycepin from Cordyceps militaris on serotonin release out of human platelets and human platelet aggregation. HWE and FWE inhibited the release of [^(3)H]-serotonin from human platelet stimulated by thrombin (2 U/ml) or collagen (20 ㎍/ml) in a dose-dependent manner. Furthermore, cordycepin, a major component of Cordyceps militaris, inhibited the human platelet aggregation induced by collagen (10 ㎍/ml) in a dose-dependent manner. These results suggest that cordycepin containing in HWE and FWE may inhibit the serotonin release by suppressing the collagen-induced human platelet aggregation. Accordingly, our data demonstrate that HWE and FWE containing much cordycepin might have antithrombotic and antimigrainous functions.

      • Fabrication of A/R-TiO<sub>2</sub> composite for enhanced photoelectrochemical performance: Solar hydrogen generation and dye degradation

        <P><B>Abstract</B></P> <P>Anatase/rutile TiO<SUB>2</SUB> nanorods composites were prepared by a facile hydrothermal method followed by dip coating method using titanium isopropoxide in acetic acid and ethanol solvent. The effects of the titanium isopropoxide precursor concentration, on the formation of dip coated anatase/rutile TiO<SUB>2</SUB> nanorods composite were systematically explored. The growth of anatase on rutile TiO<SUB>2</SUB> nanorods can be controlled by varying the titanium isopropoxide concentration. The morphological study reveals that anatase TiO<SUB>2</SUB> nanograins formed on the surface of rutile TiO<SUB>2</SUB> nanorod arrays through dip coating method. Photoelectrochemical analyses showed that the enhancement of the photocatalytic activities of the samples is affected by the anatase nanograins present on the rutile TiO<SUB>2</SUB> nanorods, which can induce the separation of electrons and holes. To interpret the photoelectrochemical behaviors, the prepared photoelectrodes were applied in photoelectrochemical solar hydrogen generation and orange II dye degradation. The optimized photocurrent density of 1.8mAcm<SUP>−2</SUP> and the 625μmolhydrogen generation was observed for 10mM anatase/rutile TiO<SUB>2</SUB> NRs composites. Additionally, 96% removal of the orange II dye was achieved within 5h during oxidative degradation under solar light irradiation. One of the benefits of high specific surface area and the efficient photogenerated charge transport in the anatase/rutile TiO<SUB>2</SUB> nanorod composite improves the photoelectrochemical hydrogen generation and orange dye degradation compared to the rutile TiO<SUB>2</SUB>.         Spinach Saponin-Enriched Fraction Inhibits Platelet Aggregation in cAMP- and cGMP-Dependent Manner by Decreasing TXA2 Production and Blood Coagulation

        In this study, we investigated the effect of spinach saponin-enriched fraction (SSEF) on collagen (10 μg/ml)-stimulated platelet aggregation. SSEF inhibited collagen-induced platelet aggregation, and which was involved in the inhibition of thromboxane A2 (TXA2) production, an intracellular Ca2+ -agonist as an aggregation-inducing autacoidal molecule. In addition, SSEF significantly increased the formation of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), intracellular Ca2+ -antagonists as aggregation-inhibiting molecules, in collagen-stimulated platelets. These results suggest that SSEF might inhibit Ca2+-elevation and TXA2 formation by increasing the production of Ca2+-antagonistic molecules cAMP and cGMP. These mean that SSEF is a potent inhibitor of collagen-stimulated platelet aggregation. On the other hand, prothrombin time (PT) and activated partial thromboplastin time (APTT) were potently prolonged by SSEF. These findings suggest that SSEF prolongs the internal time between the conversion of fibrinogen to fibrin. Accordingly, our data demonstrate that SSEF may be a crucial tool for a negative regulator during platelet activation and blood coagulation on thrombotic diseases.

        Korean Red Ginseng water extract inhibits cadmium-induced lung injury via suppressing MAPK/ERK1/2/AP-1 pathway

        Background: Few studies reported the therapeutic effect of Korean Red Ginseng (KRG) in lung inflammatory diseases. However, the anti-inflammatory role and underlying molecular in cadmium-induced lung injury have been poorly understood, directly linked to chronic lung diseases (CLDs): chronic obstructive pulmonary disease (COPD), cancer etc. Therefore, in this study we aim to investigate the therapeutic activities of water extract of KRG (KRG-WE) in mouse cadmium-induced lung injury model. Method: The anti-inflammatory roles and underlying mechanisms of KRG-WE were evaluated in vitro under cadmium-stimulated lung epithelial cells (A549) and HEK293T cell line and in vivo in cadmium-induced lung injury mouse model using semi-quantitative polymerase chain reaction (RT-PCR), quantitative real-time PCR (qPCR), luciferase assay, immunoblotting, and FACS. Results: KRG-WE strongly ameliorated the symptoms of CdSO4-induced lung injury in mice according to total cell number in bronchoalveolar lavage fluid (BALF) and severity scores as well as cytokine levels. KRG-WE significantly suppressed the upregulation of inflammatory signaling comprising mitogen-activated protein kinases (MAPK) and their upstream enzymes. In in vitro study, KRG-WE suppressed expression of interleukin (IL)-6, matrix metalloproteinase (MMP)-2, and IL-8 while promoting recovery in CdSO<sub>4</sub>-treated A549 cells. Similarly, KRG-WE reduced phosphorylation of MAPK and c-Jun/c-Fos in cadmium-exposed A549 cells. Conclusion: KRG-WE was found to attenuate symptoms of cadmium-induced lung injury and reduce the expression of inflammatory genes by suppression of MAPK/AP-1-mediated pathway.
