      • Differentiated Troy<sup>+</sup> Chief Cells Act as Reserve Stem Cells to Generate All Lineages of the Stomach Epithelium

        Proliferation of the self-renewing epithelium of the gastric corpus occurs almost exclusively in the isthmus of the glands, from where cells migrate bidirectionally toward pit and base. The isthmus is therefore generally viewed as the stem cell zone. We find that the stem cell marker Troy is expressed at the gland base by a small subpopulation of fully differentiated chief cells. By lineage tracing with a Troy-eGFP-ires-CreERT2 allele, single marked chief cells are shown to generate entirely labeled gastric units over periods of months. This phenomenon accelerates upon tissue damage. Troy<SUP>+</SUP> chief cells can be cultured to generate long-lived gastric organoids. Troy marks a specific subset of chief cells that display plasticity in that they are capable of replenishing entire gastric units, essentially serving as quiescent ''reserve'' stem cells. These observations challenge the notion that stem cell hierarchies represent a ''one-way street.''

        Cultural Influences on the Ratings of Behavioral Problems, Emotional Problems, and School Adjustment for Korean and Korean American Adolescents

        본 연구는 한국의 청소년과 미국에 거주하는 한국계 청소년에게 발생하는 사회적ㆍ정서적ㆍ행동적 문제와 학교적응문제를 집단 간 비교함으로써, 다양한 문화적 배경에 근거한 미국에 거주하는 한인학생의 이해를 돕는데 목적이 있다. 한국 청소년 136명, 미국에 거주하는 한국계 청소년 88명으로 구성된 총 224명의 학생이 본 연구에 참여하였으며, 이들은 본 연구에 필요한 자기평가보고서를 작성하였다. 자기평가는 ‘Behavior Assessment System for Children (BASC-2)’를 통해 수집되었다. 분석결과, 미국에 거주하는 한국계 학생들이 전통적인 한국문화와 미국문화가 요구하는 상황사이에서 보여주는 심리ㆍ행동적 특징들을 발견할 수 있었다. 또한 한국 청소년과 미국거주 한국계 청소년의 차이를 보여주는 정서 및 행동 특성은 무엇인지를 발견할 수 있었다. 특히 본 연구에서 사용한 BASC-2(SRP) 측정도구가 한국 청소년 및 미국거주의 한인 청소년들의 인식과 정서 그리고 행동을 연구하는데 적합하며 타당하다는 것을 증명한 것은 매우 가치 있는 연구결과라고 할 수 있다. 즉, 이 결과를 통해 BASC-2(SRP) 측정도구가 인간의 정신병리학적 측면보다는 문화적 측면의 특성을 이해하는데 도움을 주고 있다는 것을 발견하게 되었다. 한편, 가족을 중심으로 한 한국의 집단주의적 가치와 미국의 개인주의적 가치는 한국 청소년과 미국에 거주하는 한국계 청소년 간에 나타난 주요한 차이라는 것을 본 연구를 통해 알 수 있었으며, 이러한 문화적 가치 충돌로 나타나는 다양한 이슈들은 본 연구의 토론과 결론에서 심도있게 거론되었다. Results also suggested that elevated BASC-2 SRP scores may reflect cultural factors rather than psychopathology.

        Gastrointestinal cancer organoids—applications in basic and translational cancer research

        Cancer is a major health problem and a leading cause of death worldwide. Early cancer detection and continuous changes in treatment strategies have improved overall patient survival. The recent development of targeted drugs offers new opportunities for personalized cancer treatment. Nevertheless, individualized treatment is accompanied by the need for biomarkers predicting the response of a patient to a certain drug. One of the most promising breakthroughs in recent years that might help to overcome this problem is the organoid technology. Organoid cultures exhibit self-renewal capacity, self-organization, and long-term proliferation, while recapitulating many aspects of their primary tissue. Generated patient-derived organoid (PDO) libraries constitute “living” biobanks, allowing the in-depth analysis of tissue function, development, tumor initiation, and cancer pathobiology. Organoids can be derived from all gastrointestinal tissues, including esophageal, gastric, liver, pancreatic, small intestinal and colorectal tissues, and cancers of these tissues. PDOs are amenable to various techniques, including sequencing analyses, drug screening, targeted therapy testing, tumor microenvironment studies, and genetic engineering capabilities. In this review, we discuss the different applications of gastrointestinal organoids in basic cancer biology and clinical translation.

The photophysical properties of the resulting SCCs in the presence of metal ions revealed that these dipyridyl-containing SCCs hold potential as metal-ion-responsive materials.</P></▼2>

      • Tetra-, Hexa-, Dodeca-Nuclear Ir Supramolecules via Bridge-Driven Self-Assembly of Tetrazolyl Ligands

        <P>Herein, we report the formation of multinuclear Ir<SUB>4</SUB>, Ir<SUB>6</SUB>, and Ir<SUB>12</SUB> supramolecular complexes via the bridge-driven self-assembly of tetrazolyl ligands. The synthesis of dimeric half-sandwich Ir units was made by the reaction of half-sandwich Ir units and tetrazolyl ligands in a molar ratio of 1:2. The use of different ligands containing multiple tetrazolyl units resulted in the formation of different Ir supramolecular architectures. The reaction of [Cp*IrCl<SUB>2</SUB>]<SUB>2</SUB>, AgOTf, and 1,2- or 1,3-ditetrazolyl benzene in a molar ratio of 1:3:1 resulted in the formation of rectangular tetranuclear or truncated trigonal pyramidal hexanuclear Ir complexes, respectively. On the other hand, the reaction of [Cp*IrCl<SUB>2</SUB>]<SUB>2</SUB>, AgOTf, and 1,3,5-tritetrazolyl benzene in a molar ratio of 6:18:4 produced a supramolecular dodecanuclear iridium complex. The molecular structure of the complex resembled a truncated tetrahedral structure with a large inner cavity, as determined by X-ray crystallography.</P><P>Tetra-, hexa-, and dodeca-nuclear Ir supramolecules have been synthesized from the bridge-driven self-assemblies of [Cp*IrCl<SUB>2</SUB>]<SUB>2</SUB> and multitetrazolyl benzene in the presence of AgOTf.</P> [FIG OMISSION]</BR>

      • Ruthenium–Cobalt Bimetallic Supramolecular Cages via a Less Symmetric Tetrapyridyl Metalloligand and the Effect of Spacer Units

        <P>The self-assembly of C-s-symmetric tetrapyridyl cobalt metalloligand 2 with three half-sandwich diruthenium acceptors, 3-5, led to the formation of A(4)D(2) (A = acceptor, D = donor) metallacages 6-8, as shown by ESI mass spectrometry, NMR spectroscopy, and X-ray crystallography. The solid-state structures of 6-8 revealed that the length of the acceptor unit greatly influences the molecular packing of these metallacages. Hence, in the solid state, 6-8 can be considered to have waterwheel-shaped, tweezer-shaped, and butterfly-like architectures, respectively.</P>

      • Biomedical and Biochemical Applications of Self-Assembled Metallacycles and Metallacages

        <P>Metal ions and metal complexes with organic molecules are ubiquitous in nature. Bulk metal ions of Na, K, Mg, and Ca constitute as much as 1% of human body weight. The remaining trace ions, most commonly of Fe, Ni, Cu, Mn, Zn, Co, Mo, and V, make up ∼0.01% by weight, but their importance in biological processes cannot be overstated.</P><P>Although nature is limited to the use of bioavailable metal ions, many rarer transition metals can elicit novel biological responses when they interact with biomolecules. For this reason, metal–biomolecule complexes are of interest in medicinal applications. A well-known example is cisplatin, which contains Pt, rare in nature, but highly effective in this context as an anticancer drug in the form of cis-Pt(NH<SUB>3</SUB>)<SUB>2</SUB>Cl<SUB>2</SUB> and analogous Pt(II) complexes. This and other examples have led to strong interest in discovering new metalloanticancer drugs.</P><P>In this Account, we describe recent developments in this area, particularly, using coordination-driven self-assembly to form tunable supramolecular coordination complexes (SCCs) with biomedical applications. Coordination-driven self-assembly describes the spontaneous formation of metal–ligand bonds in solution, transforming molecular building blocks into single, 2D metallacycles, or 3D metallacages depending on the directionality of the precursors used. Such SCCs have well-defined internal cavities and simple pre- or post-self-assembly functionalizations. They are highly tunable both spatially and electronically.</P><P>Metal ions are necessary structural elements for the directional bonding approach, which can be exploited to provide biological activity to an SCC, particularly for Pt- and Ru-based structures. Since these two metals are not only among the most commonly used for coordination-driven self-assembly but are also the basis for a number of small molecule anticancer agents, researchers have evaluated a growing number of SCCs for their antitumor properties.</P><P>The biological application of SCCs is still an emergent field of study, but the examples discussed in this Account confirm that supramolecular scaffolds have relevance to a wide variety of biochemical and biomedical targets. SCCs can serve as anticancer agents, act as selective sensors for biologically important analytes, or interact with DNA and proteins. The myriad of possible SCCs and their almost limitless modularity and tunability without significant synthetic penalty suggests that the biological applications of such species will continue along this already promising path.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/achre4/2013/achre4.2013.46.issue-11/ar400010v/production/images/medium/ar-2013-00010v_0016.gif'></P>
