      • UBE2Q1 in a Human Breast Carcinoma Cell Line: Overexpression and Interaction with p53

        The p53 tumor suppressor protein is a principal mediator of growth arrest, senescence, and apoptosis in response to a broad array of cellular damage. p53 is a substrate for the ubiquitin-proteasome system, however, the ubiquitin-conjugating enzymes (E2s) involved in p53 ubiquitination have not been well studied. UBE2Q1 is a novel E2 ubiquitin conjugating enzyme gene. Here, we investigated the effect of UBE2Q1 overexpression on the level of p53 in the MDA-MB-468 breast cancer cell line as well as the interaction between UBE2Q1 and p53. By using a lipofection method, the p53 mutated breast cancer cell line, MDA-MB-468, was transfected with the vector pCMV6-AN-GFP, containing UBE2Q1 ORF. Western blot analysis was employed to verify the overexpression of UBE2Q1 in MDA-MB-468 cells and to evaluate the expression level of p53 before and after cell transfection. Immunoprecipitation and GST pull-down protocols were used to investigate the binding of UBE2Q1 to p53. We established MDA-MB-468 cells that transiently expressed a GFP fusion proteins containing UBE2Q1 (GFP-UBE2Q1). Western blot analysis revealed that levels of p53 were markedly lower in UBE2Q1 transfected MDA-MB-468 cells as compared with control MDA-MB-468 cells. Both in vivo and in vitro data showed that UBE2Q1 co-precipitated with p53 protein. Our data for the first time showed that overexpression of UBE2Q1can lead to the repression of p53 in MDA-MB-468 cells. This repression of p53 may be due to its UBE2Q1 mediated ubiquitination and subsequent proteasome degradation, a process that may involve direct interaction of UBE2Q1with p53.

        Dynamic behavior of a functionally graded plate resting on Winkler elastic foundation and in contact with fluid

        A semi-analytical method is developed to consider free vibrations of a functionally graded elastic plate resting on Winkler elastic foundation and in contact with a quiescent fluid. Material properties are assumed to be graded distribution along the thickness direction according to a power-law in terms of the volume fractions of the constituents. The fluid is considered to be incompressible and inviscid. In the analysis, the effect of an in-plane force in the plate due to the weight of the fluid is taken into account. By satisfying the compatibility conditions along the interface of fluid and plate, the fluid-structure interaction is taken into account and natural frequencies and mode shapes of the coupled system are acquired by employing energy methods. The results obtained from the present approach are verified by those from a finite element analysis. Besides, the effects of volume fractions of functionally graded materials, Winkler foundation stiffness and in-plane forces on the dynamic of plate are elucidated.

        Development and Microstructural Characterization of a New Wrought High Entropy Superalloy

        In this study, a newly developed γ′ precipitation hardened high entropy superalloy (designated as HES-A1 hereafter) wasproduced and its microstructural features were characterized. The new alloy composition is based on the major elementsof conventional IN718 alloy, however other elements were added or removed to minimize possible presence of δ (or γʺ)phase and increase the volume fraction of very fine γ′ precipitates instead. Broadening the hot working window, lowerdensity, and avoiding the use of expensive elements were other considerations taken into account in the design of the newalloy. CALPHAD (CALculation of PHAse Diagrams) and PhaComp methods were used for the prediction of phases andtheir evolutions. The microstructure of the HES-A1 alloy in as-cast, homogenized, hot rolled, annealed and aged conditionswere characterized using optical, scanning, and transmission electron microscopes as well as X-ray diffraction technique. The as-cast microstructure of HES-A1 contained 5.5% of Laves phase, which was reduced to less than 0.3% through thedevelopment of a homogenization treatment. Hot rolling with reductions up to 36% at 1015 °C did not produce any cracking,indicating a good forming potential for the new alloy. The application of double aging treatment, similar to the one forIN718 alloy, showed no presence of γʺ or formation of δ phase in the microstructure. HES-A1 has been designed with ahigher Al/(Ti + Nb) ratio and higher proportion of Al + Ti + Nb so that it could be mainly strengthened by γ′ precipitation. The average size of monomodal γ′ precipitates was smaller than that observed in conventional alloys after similar treatments.

        Effect of PCaRD DGB Flipped Learning on EFL Learners’ Grammar Skill

        This research examines the employing of a Play Curricular Activity Reflection Discussion (PCaRD) and Digital Game-based (DGB) through flipped learning on the EFL learners’ grammar skills to improve their achievement and attitudes. The participants were 66 EFL female intermediate students in Iran; the research was conducted over 10 weeks. A PCaRD DGB flipped learning was used for the experimental group (N = 33), and a standard flipped classroom was used for the control group (N = 33). Both groups constructed using the same teacher, time, coursebook, and videos. The PCaRD DGB flipped learning provided mobile-based game apps and a game-based feedback mechanism in the form of collaborative response and created a setting in which learners could engage in more grammar activity, pay more attention to their answers, and improving their grammar skills. The results indicated that the PCaRD DGB flipped learning outperformed the standard flipped learning in grammar achievement and in terms of their positive attitudes. The researcher speculates that the PCaRD DGB flipped learning strengthens EFL learners’ grammar skills and a positive attitude.

        The Art of Captivating Writing: Exploring Digital Storytelling from a New Dimension

        While previous research has primarily focused on evaluating the effectiveness of digital storytelling (DST) itself, there is a dearth of knowledge regarding the impacts of flow, group engagement, and perceived instructional support on learners' DST experiences. Addressing this gap, the present study examines data collected from a sample of 225 students enrolled in DST-based writing courses. The results of this study indicate that learners' perceived instructional support significantly and positively influences their writing skills. Furthermore, the findings demonstrate that the relationship between instructional support and learning engagement is mediated by flow and group engagement. These findings underscore the significance of offering appropriate support to foster group engagement and facilitate students' flow experiences within the DST context. By shedding light on these less-explored aspects of the DST strategy, this study contributes to the existing literature in the field. It suggests that educators and instructional designers should consider the interplay of flow, group engagement, and instructional support when incorporating DST into their pedagogical practices. Understanding and effectively addressing these factors can enhance the learning experiences and outcomes of students engaged in DST-based writing activities. Further research is warranted to explore additional dimensions of DST and its impact on various educational contexts.

        An assessment of dietary intake, food avoidance and food beliefs in patients with ulcerative colitis of different disease status

        Background/Aims: Ulcerative colitis (UC) is a chronic, relapsing and remitting inflammation of the gastrointestinal tract. Little is known about the link between dietary intake, food avoidance, and beliefs among UC patients of different disease severity. Therefore, this study aimed to assess the dietary intake, food avoidance, and beliefs among active and inactive UC patients. Methods: A cross-sectional study was conducted among UC patients from a tertiary medical center in Kuala Lumpur, Malaysia. Demographic, anthropometric, dietary intake, food avoidance and beliefs were assessed. Disease activity of UC patients was evaluated using the Powell Tuck Index. Results: UC patients were recruited (64.1% inactive UC and 35.9% active UC). As compared to inactive UC patients, active UC patients were likely to lose weight (75.0% vs. 0%), possess certain food beliefs (95.7% vs. 39.0%), and frequently practiced dietary avoidance (95.7% vs. 43.9%). The dietary intake among inactive UC patients was higher than active UC patients. However, neither of them met the standard nutrients recommendation for protein, calcium, iron, folate, zinc, vitamin D, vitamin B<sub>12</sub>, and vitamin E. Conclusions: Active UC patients had poorer dietary intake, were more prone to practicing food avoidance and exhibited certain food beliefs as compared to inactive UC patients. Both macro- and micronutrients intakes were inadequate regardless of patient’s disease status. These findings emphasized the importance for patients to be provided with the nutrition-related knowledge as part of strategies to avoid nutritional inadequacies. (Intest Res 2020;18:447-458)

        Enlarging Bulblet by Magnetic and Chelating Structures of Nano-chitosan as Supplementary Fertilizer in Lilium

        The nutrition is the effective component on the increasing size of the bulb in growth period. In this work,carboxymethyl chitosan and magnetic nano-chitosan in a size ≤ 20 nm were synthesized that can be taken up byplant roots. The purpose of this work was to evaluate carboxymethyl chitosan and magnetic nano-chitosan withchelating structures as supplementary fertilizers to increase yearling bulblet growth in Lilium. The application ofmagnetic nano-chitosan at a concentration of 15 mg・L-1 as compared to its lower concentrations, carboxymethylchitosan and iron sulphate had better effect on the promoting the growth, development and protecting the shoot, andgrowth of bulblet. Higher growth and development of shoots and roots by high concentration of magneticnano-chitosan could affect the enlarging bulblet. There was a positive magnetic effect on the enzyme structures andsupplying the essential iron for plant. Furthermore, magnetic nano-chitosan can act as a chelating component tofacilitate the uptake of elements.

        Anti-inflammatory diet and inflammatory bowel disease: what clinicians and patients should know?

        Current treatment for inflammatory bowel disease (IBD) includes the application of anti-inflammatory agents for the induction and remission of IBD. However, prolonged use of anti-inflammatory agents can exert adverse effects on patients. Recently, formulated dietary approach in treating IBD patients is utilized to improve clinical activity scores. An alteration of gastrointestinal microbiota through dietary therapy was found to reduce IBD and is recognized as a promising therapeutic strategy for IBD. One of the recommended formulated diets is an anti-inflammatory diet (AID) that restricts the intake of carbohydrates with modified fatty acids. This diet also contains probiotics and prebiotics that can promote balanced intestinal microbiota composition. However, scientific evidences are limited to support this specific dietary regime in maintaining the remission and prevention relapse of IBD. Therefore, this review aimed to summarize available data from various studies to evaluate the AID diet effectiveness which will be useful for clinicians to manage their IBD patients by application of improved dietary therapy.
