Cyclophilins are originally identified as cytosolic binding protein of the immunosuppressive drug cyclosporine A. They have an activity of peptidyl prolyl cis/trans-isomerases (PPIase), which may play important roles in protein folding, trafficking, assembly and cell signaling. In this study, we report the cloning and characterization of a Bombyx mori cyclophilin A (bCypA) cDNA. The full-length cDNA of bCypA consist of 947 nucleotides with a polyadenylation signal sequence AATAAA and contain an open reading frame of 498 nucleotides encoding a polypeptide of 166 amino acids. The deduced amino acid sequence of bCypA shares a central peptidyl prolyl cis/trans-isomerase and a cyclosporin-A-binding domain with other cyclophilin sequences. Relative quantification real-time (RT) PCR analysis shows that mRNA transcripts of bCypA are detected in all the investigated tissues and highest expression level in the skin of 3-day-old 5 instar larva. Also, bCypA had PPIase activity on the proline-containing peptides. Accordingly, we suggest that bCypA is a new member of the cyclophilin A (CyPA) family and will be useful for quality control of bioactivity recombinant proteins with proline-containing peptides.

<P>This paper presents a comparative analysis of the characteristics of high-speed permanent magnet synchronous machines (PMSMs) with diametrically magnetized rotors under different construction conditions of the rotor magnetic circuit. An analytical method is adopted to predict the magnetic field distribution and to calculate the electrical parameters by using the transfer relations based on a two-dimensional model in a polar coordinate system. In addition, three types of PMSMs with diametrically magnetized permanent magnets are proposed in this work. The electrical parameters are estimated and the magnetic field distribution is analyzed for those models, and these results are compared with those obtained using the finite element method; the two sets of results are found to be in good agreement. On the basis of the analysis results, a real model manufactured and the validity of our results is demonstrated by performing suitable experiments.</P>

BACKGROUND AND AIM: The endoscopic techniques for treating a small rectal neuroendocrine tumor (NET) are performed in most large centers; however, the endoscopic management of this condition is not well established. This study was designed to determine the long-term prognosis of endoscopically resected NET. METHODS: We prospectively studied patients with endoscopically treated rectal NET in Seoul St Mary’s hospital, between January 2000 and June 2010. The long-term outcomes were analyzed in association with the pathological complete resection and resection procedures. RESULTS: Seventy-seven patients (48 men and 29 women; mean age, 52.3 years; range, 23–77 years) were included. The average NET size was 7.0±2.8 mm (range, 3–16 mm). There was no procedure-related complication. En-bloc removal was achieved for all lesions, and the rate of histological complete resection was 75.3% (58/77). Histological complete resection rates were 71.4% (10/14) by conventional endoscopic mucosal resection (EMR), 74.1% (43/58) by a two-channel EMR, and 100.0% (5/5) by endoscopic submucosal dissection (ESD). Among six patients with incomplete histological resection, two underwent additional EMR, two underwent transanal endoscopic microsurgery, and two underwent low anterior section with lymph node dissection. The remaining 13 patients with ‘possible’ remnant NET underwent regular endoscopic surveillance without additional resection. In the latter group, only one patient had local recurrence, detected on regular colonoscopic surveillance, after 56 months and was treated with additional EMR. All of the patients are alive and 98.7% (76/77) of the patients are free from disease during the follow-up periods. CONCLUSION: Endoscopic resection is a safe and effective modality and may potentially be used for the treatment of NETs smaller than 15 mm in diameter, those confined to the submucosal layer, and those without metastasis. Local treatment was believed to be curative in cases with complete histological resection. In addition, this treatment may have an excellent prognosis in patients with ‘possible’ remnant NET.
Detection of Differentially Expressed Genes in Glioblastoma by Suppression Subtractive Hybridization

Objective: A variety of genetic alterations in human glioblastoma comprises signal transduction and cell cycle arrest control of cellular processes. Subtractive hybridization is potentially a faster method for identifying differentially expressed genes associated with a particular disease state. Using the technique of subtraction, we isolated novel genes that are overexpressed in glioblastoma tissue as compared to normal brain tissue. Methods: We evaluated the differential expression of genes in each of hybridizing tester and driver cDNAs to digested 130 clones. After sequencing of 130 clones and homology search, this study performed to determine mRNA expression of the unknown gene, "clone 47", in brain tissue, glioblasoma, and several cancer cell lines by reverse transcription-polymerase chain reaction (RT-PCR). To test the time course for Go-phase arrest, serum stimulation and expression at various times for RT-PCR performed. Results: We identified 23 novel genes by BLAST of the digested 130 clones. The expressions of "clone 47" mRNA of glioblastoma and several cancer lines were significantly higher than normal brain tissues and several normal cell lines. We confirmed the mRNA expression of "clone 47" was up-regulation for $0.5{\sim}1hr$ of WI-38 cell differentiation. Conclusion: The novel gene, "Clone 47" is upregulated in glioblastoma tissue and several cancer cell lines. This gene is time dependent activation during time course of serum stimulation. This result suggests that "clone 47" playa role in brain tumorigenesis and the activation of this "clone 47" may be necessary for the development of cancer.

Purpose: Although metastasis occurs in 1 or 2 sentinel lymph nodes (SLNs), axillary lymph node dissection (ALND) has been widely not performed. For axillary staging and management, the necessity of intraoperative frozen section analysis of SLN has been controversial. The aim of this study is to evaluate the validity and benefit of SLN analysis by permanent section alone in clinically negative lymph node breast cancer patients. Methods: We conducted a retrospective study of 283 cases with negative node clinical findings between July 2018 and August 2019 in Samsung Medical Center. Clinical nodal stage was evaluated by physical examination, breast ultrasonography, breast magnetic resonance imaging, and chest computerized tomography. The cases were divided into 2 groups; the permanent group had 151 cases (53.4%) and the frozen group had 132 cases (46.6%). We retrospectively analyzed the differences in the number of metastatic lymph nodes and rates of performed ALND between the 2 groups. Results: Baseline and clinicopathologic characteristics between the 2 groups were well balanced. Three cases in the permanent group and 6 cases in the frozen group underwent additional or immediate ALND. The rates of ALND between the 2 groups were not significantly different (P = 0.312). The cased of 78.9% and 89.5% with metastatic lymph nodes in permanent and frozen groups were in the pathologic N1 stage, respectively. Conclusion: SLNs analysis by permanent section alone may be performed in clinically negative axillary node breast cancer patients. Our findings can help to avoid unnecessary intraoperative frozen section analysis.

Plasma cell vulvitis is a chronic inflammatory disease of vulva characterized by plasma cell infiltration during the lifetime of a woman. It can be seen especially during the climacterium. The most common symptoms associated with plasma cell vulvitis are pruritus, pain, burning sensation and dyspareunia. The specific cause of this condition is unknown and suggested predisposing factors include warmth, friction, poor hygiene, herpes simplex infection and other chronic infections. A 53-year-old woman presented with erythematous to whitish patch and plaque with severe itching on vulva for several years. Histopathological examination revealed perivascular infiltration of inflammatory cells mainly with plasma cells in superficial dermis.
Peripheral T cell lymphoma presented with extensive ulcerative cutaneous lesions

Peripheral T cell lymphoma is a heterogeneous category of mature T cell lymphoma and it does not correspond to any of the specifically defined types of mature T cell lymphoma. Although the signs and symptoms of peripheral T cell lymphoma vary according to the subtype, patients usually present with solitary, localized or generalized nodules or tumors without predilection for specific locations. Only a few cases of peripheral T cell lymphoma with extensive skin lesion were reported in the dermatologic literature. A 68-year-old man presented with erythematous to brownish ulcerative patch and plaque for 7 months. Previously, erythematous patches without ulcerative lesion were seen 1 year ago. On physical examination, large nodulo-ulcerative annular lesions with yellowish discharge were seen on the trunk and neck. Histopathology revealed diffuse cellular infiltration in dermis and subcutaneous layer. The cells were medium to large sized and pleomorphic. Immunohistochemistry were positive for CD3, CD4, CD56 and negative for CD8, TCR-gamma rearrangement. Lymphoma cells also had aberrant weak CD79a expression. Herein, we present a case of peripheral T cell lymphoma clinically showing widespread ulcerative lesion, which rarely been reported in the dermatologic literatures.
