        A clinical and pathological update on hepatocellular carcinoma

        It is estimated that more than 1 million individuals will be affected annually by hepatocellular carcinoma (HCC) by 2025. HCC can be broadly grouped into two major molecular subgroups, each of which is characterized by specific morphological and phenotypic features that mirror the genetic background. The use of these tissue biomarkers in the daily practice of pathologists promises to better allocate patients with HCC with adequate treatments. In turn, this will likely boost the attitude of clinicians toward obtaining a pre-treatment biopsy. (J Liver Cancer 2022;22:14-22)

      • Impaired melanoma growth in VASP deficient mice

        Progression of tumors depends on interactions of cancer cells with the host environment. Expression of the cytoskeleton protein VASP is upregulated in various cancer entities. We analyzed the role of VASP for melanoma growth in murine allograft models. Growth of VASP expressing melanomas was retarded in VASP<SUP>-/-</SUP> versus wild-type animals. Over time tumor size was <50% in VASP<SUP>-/-</SUP> versus wild-type animals and independent of expression levels of Ena/VASP protein family members. Histological analyses showed smaller cells with impaired nutrition status and less vascularization in melanomas derived from VASP<SUP>-/-</SUP> versus counterparts from wild-type mice. Cumulatively, the data reveal a critical role of VASP in non-tumor cells in the tumor environment for melanoma growth in vivo.

        Effects of energy-protein supplementation frequency on performance of primiparous grazing beef cows during pre and postpartum

        Objective: Twenty-four pregnant Nellore primiparous grazing cows were used to evaluate the effects of energy-protein supplementation and supplementation frequency during pre (105 d before calving) and postpartum (105 d after calving) on performance and metabolic characteristics. Methods: Experimental treatments consisted of a control (no supplementation), daily supplementation (1.5 kg/d of concentrate/animal) and infrequent supplementation (4.5 kg of concentrate/ animal every three days). During the pre and postpartum periods, concentrations of blood metabolites and animal performance were evaluated. Ureagenesis and energy metabolism markers were evaluated at prepartum period. Results: Supplementation frequency did not alter (p>0.10) body weight (BW), average daily gain (ADG), and carcass traits during pre and postpartum. The BW (p = 0.079), adjusted BW at day of parturition (p = 0.078), and ADG (p = 0.074) were greater for supplemented cows during the prepartum. The body condition score (BCS; p = 0.251), and carcass traits (p>0.10) were not affected by supplementation during prepartum. On postpartum, supplementation did not affect animal performance and carcass traits (p>0.10). The dry mater intake was not affected (p>0.10) by supplementation and supplementation frequency throughout the experimental period. Daily supplemented animals had greater (p<0.001) glucose levels than animals supplemented every three days. Supplementation and supplementation frequency did not alter (p>0.10) the levels of blood metabolites, neither the abundance of ureagenesis nor energy metabolism markers. Conclusion: In summary, our data show that the reduction of supplementation frequency does not cause negative impacts on performance and metabolic characteristics of primiparous grazing cows during the prepartum.

        Provision of a protein-rich supplement for grazing suckling female beef calves to improve productive performance and metabolic response

        Objective: This study was conducted to evaluate the effects of the provision of a proteinrich supplement on productive performance, and metabolic profile on grazing suckling female beef calves in tropical conditions during 150 d of experimentation. Methods: Fifty-six Nellore suckling female calves, and their respective dams were distributed in a completely randomised design and made to undergo two treatments as follows: UNS (without supplementation), and SUP (supplementation with 5 g/kg body weight [BW] of a protein supplement). Throughout the experiment, animal performance and metabolic profile were evaluated. Also, ureagenesis and gluconeogenesis were assessed for gene expression. Results: SUP female calves showed a higher voluntary intake (p≤0.03) of the diet components evaluated, digestibility of organic matter (p≤0.02) and microbial nitrogen production (MICN; p≤0.02) compared to UNS female calves. In its turn, serum urea nitrogen (p≤0.01) and insulin-like growth factor-1 (p≤0.03) levels and ureagenesis (p≤0.04) increased in SUP female calves compared to UNS female calves. Blood glucose and triglyceride levels were not affected by supplementation. The average daily gain (ADG) from SUP female calves was higher (p≤0.02) compared with UNS female calves. However, supplementation did not affect the body measures of the animals. Conclusion: In summary, provision of a protein-rich supplement improves the intake and nutrients digestibility, ADG and final BW and increases metabolic indicators of the protein status in grazing suckling female beef calves in tropical conditions.         AMP-activated Protein Kinase Impairs Endothelial Actin Cytoskeleton Assembly by Phosphorylating Vasodilator-stimulated Phosphoprotein

        <P>Vasodilator-stimulated phosphoprotein (VASP) is an actin regulatory protein that links signaling pathways to remodeling of the cytoskeleton. VASP functions are modulated by protein kinases, which phosphorylate the sites Ser-157, Ser-239, and Thr-278. The kinase responsible for Thr-278 phosphorylation, biological functions of the phosphorylation, and association with disease states have remained enigmatic. Using VASP phosphorylation status-specific antibodies, we identified AMP-activated protein kinase (AMPK), a serine-threonine kinase and fundamental sensor of energy homeostasis, in a screen for kinases that phosphorylate the Thr-278 site of VASP in endothelial cells. Pharmacological AMPK inhibitors and activators and AMPK mutants revealed that the kinase specifically targets residue Thr-278 but not Ser-157 or Ser-239. Quantitative fluorescence-activated cell sorter analysis and serum response factor transcriptional reporter assays, which quantify the cellular F-/G-actin equilibrium, indicated that AMPK-mediated VASP phosphorylation impaired actin stress fiber formation and altered cell morphology. In the Zucker Diabetic Fatty (ZDF) rat model for type II diabetes, AMPK activity and Thr-278 phosphorylation were substantially reduced in arterial vessel walls. These findings suggest that VASP is a new AMPK substrate, that VASP Thr-278 phosphorylation translates metabolic signals into actin cytoskeleton rearrangements, and that this signaling system becomes down-regulated in diabetic vessels.</P>

        A light-induced photochromic nanoswitch capable of non-destructive readout via fluorescence emission: cluster vs. single-molecule excitation of dihydroindolizines

        We have synthesized a prototype of a photochromic styrylquinolyl-dihydroindolizine (DHI), which forms a highly coloured and fluorescent betaine upon irradiation with $\lambda$ < 400 nm. Embedding this photochromic DHI in a thin polymethyl methacrylate (PMMA) film permits the non-destructive readout via fluorescence at low temperature (77 K). Thus, either a non-destructive photoswitch or an information recording system becomes available. Both possibilities have been explored: image recording and read-out, as well as information storage (at 77 K) have been demonstrated. Cluster- and single molecule-fluorescence upon laser excitation ($\lambda$ = 355 nm) of the styrylquinolyl-dihydroindolizine in a PMMA matrix, and the effect of fluorescence blinking has been observed.
