      • Calpain-10 SNP43 and SNP19 Polymorphisms and Colorectal Cancer: a Matched Case-control Study

        Objective: Insulin resistance (IR) is an established risk factor for colorectal cancer (CRC). Given that CRC and IR physiologically overlap and the calpain-10 gene (CAPN10) is a candidate for IR, we explored the association between CAPN10 and CRC risk. Methods: Blood samples of 400 case-control pairs were genotyped, and the lifestyle and dietary habits of these pairs were recorded and collected. Unconditional logistic regression (LR) was used to assess the effects of CAPN10 SNP43 and SNP19, and environmental factors. Both generalized multifactor dimensionality reduction (GMDR) and the classification and regression tree (CART) were used to test gene-environment interactions for CRC risk. Results: The GA+AA genotype of SNP43 and the Del/Ins+Ins/Ins genotype of SNP19 were marginally related to CRC risk (GA+AA: OR = 1.35, 95% CI = 0.92-1.99; Del/Ins+Ins/Ins: OR = 1.31, 95% CI = 0.84-2.04). Notably, a high-order interaction was consistently identified by GMDR and CART analyses. In GMDR, the four-factor interaction model of SNP43, SNP19, red meat consumption, and smoked meat consumption was the best model, with a maximum cross-validation consistency of 10/10 and testing balance accuracy of 0.61 (P < 0.01). In LR, subjects with high red and smoked meat consumption and two risk genotypes had a 6.17-fold CRC risk (95% CI = 2.44-15.6) relative to that of subjects with low red and smoked meat consumption and null risk genotypes. In CART, individuals with high smoked and red meat consumption, SNP19 Del/Ins+Ins/Ins, and SNP43 GA+AA had higher CRC risk (OR = 4.56, 95%CI = 1.94-10.75) than those with low smoked and red meat consumption. Conclusions: Though the single loci of CAPN10 SNP43 and SNP19 are not enough to significantly increase the CRC susceptibility, the combination of SNP43, SNP19, red meat consumption, and smoked meat consumption is associated with elevated risk.

      • Suppression of Ku80 Correlates with Radiosensitivity and Telomere Shortening in the U2OS Telomerase-negative Osteosarcoma Cell Line

        Ku70/80 heterodimer is a central element in the nonhomologous end joining (NHEJ) DNA repair pathway, Ku80 playing a key role in regulating the multiple functions of Ku proteins. It has been found that the Ku80 protein located at telomeres is a major contributor to radiosensitivity in some telomerase positive human cancer cells. However, in ALT human osteosarcoma cells, the precise function in radiosensitivity and telomere maintenance is still unknown. The aim of this study was to investigate the effects of Ku80 depletion in the U2OS ALT cell line cell line. Suppression of Ku80 expression was performed using a vector-based shRNA and stable Ku80 knockdown in cells was verified by Western blotting. U2OS cells treated with shRNA-Ku80 showed lower radiobiological parameters (D0, Dq and SF2) in clonogenic assays. Furthermore, shRNA-Ku80 vector transfected cells displayed shortening of the telomere length and showed less expression of TRF2 protein. These results demonstrated that down-regulation of Ku80 can sensitize ALT cells U2OS to radiation, and this radiosensitization is related to telomere length shortening.

        Ni-NTA-COATED NANOWIRE MATERIALS FOR PROTEIN ENRICHMENT AND THE APPLICATION IN A MEDICAL DEVICE USED FOR BLOOD GLUCOSE DEGRADATION

        A general and reliable method has been developed to functionalize either the iron oxide or the silicon nanowires (NWs) with nickel–nitriloacetic acid (Ni–NTA) complex, which was manufactured to manipulate His-tagged proteins and enzymes. The Ni–NTA-functionalized sea-urchin-shaped α-Fe2O3 NWs exhibit the superior protein purification efficiency and excellent stability in the form of dry powder. Application of this new nanotechnology in biomedical research field has been explored. A glucose degradation bio-matrix was made via the Ni–NTA-modified silicon NW-chips, which were conjugated with an enzyme essential to glycolysis. The glucose level in a simulated blood solution was found to be reduced from 14.4 mM to 9 mM after incubating the hexokinase I-functionalized silicon NW-chips for 12 h. These results suggest a possible way to build up a medical device using enzymes functionalized NW-chips for the removal of excess blood glucose.

        Bayesian ballast damage detection utilizing a modified evolutionary algorithm

        This paper reports the development of a theoretically rigorous method for permanent way engineers to assess the condition of railway ballast under a concrete sleeper with the potential to be extended to a smart system for long-term health monitoring of railway ballast. Owing to the uncertainties induced by the problems of modeling error and measurement noise, the Bayesian approach was followed in the development. After the selection of the most plausible model class for describing the damage status of the rail-sleeper-ballast system, Bayesian model updating is adopted to calculate the posterior PDF of the ballast stiffness at various regions under the sleeper. An obvious drop in ballast stiffness at a region under the sleeper is an evidence of ballast damage. In model updating, the model that can minimize the discrepancy between the measured and model-predicted modal parameters can be considered as the most probable model for calculating the posterior PDF under the Bayesian framework. To address the problems of non-uniqueness and local minima in the model updating process, a two-stage hybrid optimization method was developed. The modified evolutionary algorithm was developed in the first stage to identify the important regions in the parameter space and resulting in a set of initial trials for deterministic optimization to locate all most probable models in the second stage. The proposed methodology was numerically and experimentally verified. Using the identified model, a series of comprehensive numerical case studies was carried out to investigate the effects of data quantity and quality on the results of ballast damage detection. Difficulties to be overcome before the proposed method can be extended to a long-term ballast monitoring system are discussed in the conclusion.

        Colonic Hypersensitivity and Sensitization of Voltage-gated Sodium Channels in Primary Sensory Neurons in Rats with Diabetes

        Background/Aims Patients with long-standing diabetes often demonstrate intestinal dysfunction and abdominal pain. However, the pathophysiology of abdominal pain in diabetic patients remains elusive. The purpose of study was to determine roles of voltage-gated sodium channels in dorsal root ganglion (DRG) in colonic hypersensitivity of rats with diabetes. Methods Diabetic models were induced by a single intraperitoneal injection of streptozotocin (STZ; 65 mg/kg) in adult female rats, while the control rats received citrate buffer only. Behavioral responses to colorectal distention were used to determine colonic sensitivity in rats. Colon projection DRG neurons labeled with DiI were acutely dissociated for measuring excitability and sodium channel currents by whole-cell patch clamp recordings. Western blot analysis was employed to measure the expression of NaV1.7 and NaV1.8 of colon DRGs. Results STZ injection produced a significantly lower distention threshold than control rats in responding to colorectal distention. STZ injection also depolarized the resting membrane potentials, hyperpolarized action potential threshold, decreased rheobase and increased frequency of action potentials evoked by 2 and 3 times rheobase and ramp current stimulation. Furthermore, STZ injection enhanced neuronal sodium current densities of DRG neurons innervating the colon. STZ injection also led to a significant upregulation of NaV1.7 and NaV1.8 expression in colon DRGs compared with age and sex-matched control rats. Conclusions Our results suggest that enhanced neuronal excitability following STZ injection, which may be mediated by upregulation of NaV1.7 and NaV1.8 expression in DRGs, may play an important role in colonic hypersensitivity in rats with diabetes.         Preparation of PAN/GO composite nanofiber membrane for oil-containing wastewater treatment

        The discharge of industrial oily wastewater and oil spill accidents has resulted in significant environmental pollution, creating an urgent need for the treatment of oily wastewater. Nanofiber membranes, known for their high stability, flux, and efficiency, are widely employed in oil–water separation. This study systematically compared the performance of polyacrylonitrile/graphene oxide (PAN/GO) composite nanofiber membranes prepared through immersion and electrospinning methods for oily wastewater treatment. Experimental findings demonstrated that immersion-prepared membranes achieved high flux (772.9 ± 2.9 L·m−2·h−1, n-hexane/water mixture) and efficiency (98.8%) at a GO concentration of 1.0 g/mL. On the other hand, electrospinning-prepared membranes exhibited superior flux (1500.3 ± 4.4 L·m−2·h−1, n-hexane/water mixture) and efficiency (99.6%) at a higher GO concentration (1.5 g/mL). This membrane displayed excellent stability, maintaining their exceptional performance even after ten cycles of separation. This study compared the advantages and disadvantages of the two methods for preparation of PAN/GO composite nanofiber membrane, offering valuable guidance for practical applications.

        A pairing-free key-insulated certificate-based signature scheme with provable security

        Certificate-based signature (CBS) combines the advantages of both public key-based signature and identity-based signature, while saving from the disadvantages of drawbacks in both PKS and IBS. The insecure deployment of CBS under the hostile circumstances usually causes the exposure of signing key to be inescapable. To resist the threat of key leakage, we present a pairing-free key insulated CBS scheme by incorporating the idea of key insulated mechanism and CBS. Our scheme eliminates the costly pairing operations and as a matter of fact outperforms the existing key insulated CBS schemes. It is more suitable for low-power devices. Furthermore, the unforgeability of our scheme has been formally proven to rest on the discrete logarithm assumption in the random oracle model.

        Joint Uplink and Downlink Resource Allocation in Data and Energy Integrated Communication Networks

        In this paper, we propose a joint power control strategy for both the uplink and downlink transmission by considering the energy requirements of the user equipments` uplink data transmissions in data and energy integrated communication networks (DEINs). In DEINs, the base station (BS) adopts the power splitting (PS) aided simultaneous wireless information and power transfer (SWIPT) technique in the downlink (DL) transmissions, while the user equipments (UEs) carry out their own uplink (UL) transmissions by exploiting the energy harvested during the BS`s DL transmissions. In our DEIN model, there are UEs served by the BS in order to fulfil both of their DL and UL transmissions. The orthogonal frequency division multiple access (OFDMA) technique is adopted for supporting the simultaneous transmissions of multiple UEs. Furthermore, a transmission frame is divided into time slots in the medium access control (MAC) layer. The mathematical model is established for maximizing the sum-throughput of the UEs` DL transmissions and for ensuring their fairness during a single transmission frame , respectively. In order to achieve these goals, in each transmission frame T, we optimally allocate the BS`s power for each subcarrier and the PS factor for each UE during a specific time slot. The original optimisation problems are transformed into convex forms, which can be perfectly solved by convex optimisation theories. Our numerical results compare the optimal results by conceiving the objective of maximising the sum-throughput and those by conceiving the objective of maximising the fair-throughput. Furthermore, our numerical results also reveal the inherent tradeoff between the DL and the UL transmissions.

        Expression of the Arabidopsis vacuolar $H^+$-pyrophosphatase gene AVP1 in peanut to improve drought and salt tolerance

        The Arabidopsis gene AVP1 encodes an $H^+$-pyrophosphatase that functions as a proton pump at the vacuolar membranes, generating a proton gradient across vacuolar membranes, which serves as the driving force for many secondary transporters on vacuolar membranes such as $Na^+/H^+$-antiporters. Overexpression of AVP1 could improve drought tolerance and salt tolerance in transgenic plants, suggesting a possible way in improving drought and salt tolerance in crops. The AVP1 was therefore introduced into peanut by Agrobacterium-mediated transformation. Analysis of AVP1-expressing peanut indicated that AVP1-overexpression in peanut could improve both drought and salt tolerance in greenhouse and growth chamber conditions, as AVP1-overexpressing peanuts produced more biomass and maintained higher photosynthetic rates under both drought and salt conditions. In the field, AVP1-overexpressing peanuts also outperformed wild-type plants by having higher photosynthetic rates and producing higher yields under low irrigation conditions.
