      • ST6Gal-I Predicts Postoperative Clinical Outcome for Patients with Localized Clear-cell Renal Cell Carcinoma

        Hyperactivated ${\alpha}2$-6-sialylation on N-glycans due to overexpression of the Golgi enzyme ${\beta}$-galactoside: ${\alpha}2$-6-sialyltransferase (ST6Gal-I) often correlates with cancer progression, metastasis, and poor prognosis. This study was aimed to determine the association between ST6Gal-I expression and the risk of recurrence and survival of patients with localized clear-cell renal cell carcinoma (ccRCC) following surgery. We retrospectively enrolled 391 patients (265 in training cohort and 126 in validation cohort) with localized ccRCC underwent nephrectomy at a single center. Tissue microarrays were constructed for immunostaining of ST6Gal-I. Prognostic value and clinical outcomes were evaluated. High ST6Gal-I expression was associated with Fuhrman grade (p<0.001 and p=0.016, respectively) and the University of California Los-Angeles Integrated Staging System (UISS) score (p=0.004 and p=0.017, respectively) in both cohorts. Patients with high ST6Gal-I expression had significantly worse overall survival (OS) (p<0.001 and p<0.001, respectively) and recurrence free survival (RFS) (p<0.001 and p=0.002, respectively) than those with low expression in both cohorts. On multivariate analysis, ST6Gal-I expression remained associated with OS and RFS even after adjusting for the UISS score. Stratified analysis suggested that the association is more pronounced among patients with low and intermediate-risk disease defined by the UISS score. High ST6Gal-I expression is a potential independent adverse predictor of survival and recurrence in ccRCC patients, and the prognostic value is most prominent in those with low and intermediate-risk disease defined by the UISS score.

      • Association between Dietary Factors and Breast Cancer Risk among Chinese Females: Systematic Review and Meta-analysis

        Background: Evidence for associations between dietary factors and breast cancer risk is inconclusive among Chinese females. To evaluate this question, we conducted a systematic review of relevant case-control and cohort studies. Methods: Studies were systematically searched among 5 English databases (PudMed, ScienceDirect, Wiley, Clinicaltrials.gov, and Cochrane) and 3 Chinese databases (CNKI, WanFang, and VIP) until November 2012. Random effects models were used to estimate summary odds ratios (ORs) and the corresponding 95% confidence intervals (CIs). Results: Thirty one case-control studies and two cohort studies involving 9,299 cases and 11,413 controls were included. Consumption of both soy and fruit was significantly associated with decreased risk of breast cancer, with summary ORs of 0.65 (95% CIs: 0.43-0.99; I2=88.9%, P<0.001; N=13) and 0.66 (95% CIs: 0.47-0.91; $I^2$=76.7%, P<0.001; N=7), respectively. Consumption of fat was significantly associated with increased risk of breast cancer (OR=1.36; 95% CIs: 1.13-1.63; $I^2$=47.9%, P=0.088; N=6). There was nonsignificant association between consumption of vegetables and breast cancer risk (OR=0.72; 95% CIs: 0.51-1.02; $I^2$= 74.4%, P<0.001; N=9). However, sensitivity analysis based on adjusted ORs showed decreased risk of breast cancer was also associated with consumption of vegetables (OR=0.49; 95% CIs: 0.30-0.67). Conclusion: Both soy food and fruit are significantly associated with decreased risk of breast cancer among Chinese females, and vegetables also seems to be protective while dietary fatexerts a promoting influence.

        Experimental and Numerical Investigation on the Dynamic Responses of the Remaining Structure under Impact Loading with Column Being Removed

        The dynamic response and stability of building structures under accidental loads such as earthquake, explosion, and impact have been focused by experts. The significant influence will be induced once the damaged structure begins to collapse. In this study, a reinforced concrete frame structure model was designed and tested to study the dynamic response law of the structure under the condition of the failure of the middle column of the model structure, the remaining structure successively failed the load-bearing column and continued to be subjected to impact load and uniform load. The analytical method was used to calculate the ultimate uniform load subjecting to the structure under the failure of four load-bearing columns, because its value was less than the load in this test, the structure collapsed. The finite element calculation model was established, and compared the simulation results to the test results to verify its rationality. Then, the influence of the impact energy and the structural stores on the dynamic response of the structure were analyzed. The results showed that the peak value of the dynamic response of the damaged structure did not increase strictly with the increase of the number of failure columns, which may be related to the sequence of failure columns.

      • Strength of connection fixed by TOBs considering out-of-plane tube wall deformation-Part 1: Tests and numerical studies

        This paper presents a study on the behavior of a bolted T-stub to square tube connection using Thread-fixed Oneside Bolts (TOBs) through tests and numerical simulations. It outlines a research work of four connections with focus on the failure modes and strengths of the connection under tensile load. It was observed that the thread anchor failure caused by shear failure of hole threads controlled the final failure of the connection in the tests. Meanwhile, the out-of-plane deformation of tube wall resulted in the contact separation between hole threads and bolt threads, which in turn reduced the shear strength of hole threads. Finite element models (FEMs) allowing for the configuration details of the TOBs fixed connection are then developed and compared with the test results. Subsequently, the failure mechanism of hole threads and stress distribution of each component are analyzed based on FEM results. It was concluded that the ultimate strength of connection was not only concerned with the shear strength of hole threads, but also was influenced by the plastic out-of-plane deformation of tube wall. These studies lay a foundation for the establishment of suitable design methods of this type of connection.

      • Possibility of Using DNA Chip Technology for Diagnosis of Human Papillomavirus

        To explore the application of DNA chip technology for the detection and typing of Human Papillomavirus (HPV), the HPV6, 11, 16 and 18 gene fragments were isolated and printed onto aminosilane-coated glass slides by a PixSys 5500 microarrayer as probes to prepare the HPV gene chips. HPV samples, after being labeled with fluorescent dye by restriction display PCR (RD-PCR) technology, were hybridized with the microarray, which was followed by scanning and analysis. The experimental condition for preparing the HPV gene chips was investigated, and the possibility of HPV genotyping using gene chips was discussed. The technique that was established in this study for preparing HPV gene chips is practical. The results of the present study demonstrated the versatility and inspiring prospect of using this technology to detect and genotype HPV.

        Effects of Dietary Energy Density on Growth, Carcass Quality and mRNA Expression of Fatty Acid Synthase and Hormone-sensitive Lipase in Finishing Pigs

        A single factorial experiment was conducted to test the effects of three dietary levels of energy on mRNA expression of fatty acid synthase (FAS-mRNA) and hormone-sensitive lipase (HSL-mRNA) and their association with intramuscular fat in finishing pigs. 72 crossbred (Large $White{\times}Rongchang$) barrows with an average initial body weight of 20.71 (s.e. 0.1) kg, were randomly allotted to three dietary treatments (11.75, 13.05 and 14.36 MJ DE/kg) and fed until slaughtered at 100 or 101 kg. The diets were iso-nitrogenous and iso-essential amino acids. The growth performances including the duration of finishing were changed linearly (p<0.05) or quadratically (p<0.05) with increased dietary energy levels. The effects of dietary energy content on the percentage of external fat, intramuscular backfat and the fat thickness were linear (p<0.05). The content of dietary energy increased FAS-mRNA linearly or quadratically, while HSL-mRNA decreased linearly or quadratically in backfat and Longissmus dorsi muscle. Meanwhile, significant positive correlations (p<0.05) were found between energy level and intramuscular fat, FAS-mRNA or the ratio of FAS-mRNA to HSL-mRNA, between the ratio of FAS-mRNA to HSL-mRNA and intramuscular fat. However, the correlations between HSL mRNA and dietary energy or intramuscular fat were negative (p<0.05). The results indicated that dietary energy level regulates lipid accumulation, especially intramuscular fat, possibly by modulating the mRNA of FAS and HSL together rather than individually.

        Design of a Fresnel Lens for a Solar End-pumped Solid-state Laser

        A novel design for a Fresnel lens for a solar end-pumped laser is demonstrated in this paper. The new hybrid Fresnel lens includes two parts, inner and outer. The inner part is a twice-total-internal-reflection and vertical-transmission lens. The outer part is a once-total-reflection and vertical-transmission lens. The radius of the Fresnel lens is 600 mm, and its focal length is 750 mm. The concentrating performance of the Fresnel lens is examined using TracePro. The results show that the concentration efficiency has been greatly improved. The total concentration efficiency of the hybrid Fresnel lens reaches 73.2% when the radius of the laser rod is 3 mm. This design can simplify the concentration system of a solar endpumped solid-state laser.

      • Regeneration of Diesel Particulate Filter (DPF) using Ozone

        Recent years, automotive diesel engine emission standards become very strict, and it is hard to fulfill them just by using the current techniques of improving combustion. Therefore, the ceramic diesel particulate filter (DPF) is widely used for collecting soot from the exhaust gas of diesel engine. However, the DPF need being regenerated after the soot accumulation. This research deals with regeneration of the DPF using ozone, which comes from surface discharge occurred in a dielectric barrier discharge reactor. Pure oxygen was introduced into the reactor, and was discharged to form much activated species, including ozone and monatomic oxygen. When pure oxygen of 4 L/min at a temperature of 443 K is passed to the DPF through plasma reactor which is energized by 100 W for two hours, it has found a decrease in weight of soot by 2 grams. Comparing with the regenerating method via NO₂, the method using ozone is much friendlier to the environment, because ozone is very unstable.

      • A Basic Study on Regeneration System using Plasma for Diesel Particulate Filter

        Diesel Particulate Filter (DPF) is the most widely used method to control particulate emissions of diesel engine these days. However, DPF needs periodical regeneration because of soot deposition. Electric discharge plasma, with its strong oxidation characteristics can be utilized in the regeneration process even at lower exhaust temperatures, and the present study also confirmed that the regeneration of DPF was realized when the temperature was elevated from 200℃. Plasma oxidizes NO in the exhaust to NO<SUB>2</SUB> and NO<SUB>2</SUB> in turn oxidizes soot in presence of oxygen to release CO and CO<SUB>2</SUB>. The advantage with plasma regeneration is it proceeds at a lower temperature at which no damage to DPF can happen. Initial experiments were conducted on optimizing NO oxidation to NO<SUB>2</SUB> using plasma reactor. In the next step, regeneration of DPF was carried out. Different parameters such as applied high voltage and gas temperature are studied.
