Background: The leaves of Staphylea bumalda (S. bumalda), a deciduous tree distributed in Korea, China, and Japan, are used to treat respiratory diseases or inflammation. However, the immune-enhancing activity of S. bumalda leaves has yet to be examined. Thus, we investigated the effect of water extracts of S. bumalda leaves (SBL) on macrophage activity using RAW264.7 mouse macrophage cells. Methods and Results: SBL increased the production of immunomodulators such as NO, iNOS, IL-1β, IL-6, TNF-α, and MCP-1 and activated the phagocytic activity of RAW264.7 cells. SBL-mediated production of immunomodulators in RAW264.7 cells was blocked by inhibition of TLR2 and TLR4 and was attenuated by JNK inhibition. Moreover, SBL increased JNK phosphorylation, while inhibition of TLR2 and TLR4 blocked SBL-mediated JNK phosphorylation. These results suggest that SBL activates JNK through stimulating TLR2 and TLR4 in macrophages to induce immunomodulator production. In lipopolysaccharide-stimulated RAW264.7 cells, SBL inhibited the overproduction of immunomodulators. Conclusions: Given these results, SBL showed immunostimulatory activity under normal conditions and immunosuppressive activity during LPS-induced excessive immune response.

The use of natural plants for dyeing is a new green approach in the textile industry using eco-friendly, renewable,and non-toxic reductants for dyeing fabric. Recently, various templates have been reported to enhance the polymerization ofnatural phenols. In this paper, ferulic acid (FA) was synthesized using a new approach and arranged on poly(ethylene glycol)(PEG) templates using horseradish peroxide (HRP)-catalyzed oxidative polymerization. The polymerization mechanism wasstudied in detail using high-performance liquid chromatography-electrospray ionization mass spectrometry (HPLC-ESI-MS)analysis. In addition, the template effect of PEG on 13 natural phenols was also investigated. Upon analyzing the color depth,the K/S value of the silk fabric dyed using PEG as a template was significantly higher than that in the absence of PEG. Thehigher color depth of the poly(ferulic acid)/PEG-treated silk fabrics further validated the important role of PEG as a template. This study presents an environmentally friendly method for dyeing silk fabric under mild conditions and the polymerizationmechanism. Compared to chemical polymerization reactions, this enzyme (HRP)-based synthesis is more environmentallyfriendly due to its milder reaction conditions.

<P>This study assessed the expression of the p53 protein, beta-catenin, and HER2 and their prognostic implications in patients with EBV-associated gastric cancer (EBVaGC). After reviewing 1318 consecutive cases of surgically resected or endoscopic submucosal dissected gastric cancers, 117 patients were identified as EBV-positive using EBV-encoded RNA in-situ hybridization. The immunohistochemistry results were interpreted as follows: strong p53 nuclear expression in at least 50% of tumor nuclei was interpreted as a positive result, strong beta-catenin expression in at least 10% of cytoplasmic nuclei was interpreted as a positive result, and moderate or strong complete or basolateral membrane staining in 10% of tumor cells was interpreted as a positive result for HER2. Immunohistochemical staining for p53 was performed on tumor tissue from 105 patients, among whom 25 (23.8%) tested positive. Meanwhile, beta-catenin expression was positive in 10 patients (17.5%) and HER2 expression was positive in 8 patients (6.8%). The positive expression of p53 was significantly associated with a high T stage (p=0.006). More patients with lymph node metastasis were p53-positive (p=0.013). In the univariate analysis, the p53-positive patients showed significantly decreased disease-free survival (DFS) when compared with the p53-negative patients (p=0.022), although the p53 status was only marginally associated with overall survival (OS) (p=0.080). However, p53 expression showed no prognostic significance on DFS in the multivariate analysis. Moreover, beta-catenin and HER2 showed no association with DFS and OS in the survival analysis. The current study found a significant correlation between p53 expression and tumor progression and lymph node metastases in patients with EBVaGC.</P>

<P>BACKGROUND: An increase in the level of the vascular endothelial growth factor (VEGF) production has been reported in the peritoneal fluid (PF) of endometriosis patients. This suggests that changes in the vascular permeability and angiogenesis play an important role in the pathophysiology of this disease. This study examined the effects of the PF obtained from endometriosis patients on the release of VEGF by neutrophils and monocytes. METHODS: Neutrophils and monocytes were obtained from young healthy volunteers and cultured with the PF obtained from either endometriosis patients (EPF) (<I>n</I>=18) or a control group (CPF) (<I>n</I>=4). A human monocyte/macrophage cell line, THP-1, was cultured with either 10% EPF or 10% CPF. The PF and culture supernatants were assayed for VEGF using ELISA. Real-time PCR and Western blotting were used to measure the VEGF mRNA and protein expression level, respectively. RESULTS: The VEGF levels were higher in the EPF than in the CPF (591 ± 75 versus 185 ± 31 pg/ml, <I>P</I> < 0.05). However, the level of VEGF released by THP-1 cells in CPF and EPF was similar. The EPF induced the release of VEGF by neutrophils, but no VEGF was released by monocytes. The VEGF mRNA expression levels in the neutrophils were higher in the EPF, which was abrogated by cycloheximide, suggesting that the EPF induces the production of VEGF in neutrophils. Neutralizing antibodies against IL-8 and TNF-α did not completely prevent the EPF-induced release of VEGF by the neutrophils, even though these growth factors stimulated the release of VEGF by neutrophils. There was a positive correlation between the VEGF and IL-10 concentrations in the EPF (correlation coefficient=0.549, <I>P</I>=0.012, <I>n</I>=18), but the neutralizing antibody of IL-10 did not affect the release of VEGF by the EPF-treated neutrophils. CONCLUSION: The EPF induced the production and release of VEGF by neutrophils, suggesting that neutrophils may be a source of peritoneal VEGF. In addition, neutrophil-derived VEGF might be a marker for diagnosing endometriosis.</P>

Corrosion of reinforcing steel bars is a major concern for ocean engineers when reinforced concrete structures are exposed to marine environments. Evaluating the degree of corrosion and corrosion-induced defects is extremely necessary to pursue a proper retrofit or rehabilitation plan for reinforced concrete structures. A promising inspection should be carried out for the evaluation, otherwise the retrofit or rehabilitation process would be useless. Nowadays, ultrasonic guided wave-based inspection techniques become quite promising for the inspection, mainly because of their long-range propagation capability and their sensitivity to different types of defects or conditions. Evaluating how the guided waves response to the different types of defects or conditions is quite challenging and important. This study shows how surface conditions of reinforcing bars and a corrosion-induced defect, separation, affect guided wave propagation in reinforced concrete. Experiments and associated signal analysis show the sensitivity of guided waves to the surface conditions, as well as the amounts of separation at the interface between concrete and steel bar.

Bok choy is one of Brassica vegetables widely consumed worldwide. Brassica vegetables have been reported to exert various pharmacological activities such as antioxidant, anti-cancer and cardioprotective activity. However, studies on immunostimulatory activity of bok choy sprout have not been conducted properly. Thus, in this study, we investigated in vitro immunostimulatory activity of bok choy sprout extract (BCS) using mouse macrophage RAW264.7 cells. Our results showed that BCS increased the production of immunomodulators such as NO, iNOS, IL-1β, IL-6, IL-12, TNF-α and MCP-1, and phagocytic activity in RAW264.7 cells. BCS activated MAPK, NF-κB and PI3K/AKT signaling pathways. However, BCS-mediated production of immunomodulators was dependent on JNK, NF-κB and PI3K/AKT signaling pathways. the mRNA expression of TLR2 were significantly increased by BCS, TLR2 inhibition by anti-TLR2 dramatically suppressed the production of immunomodulators by BCS. In addition, TLR2 inhibition by anti-TLR2 significantly reduced BCS-mediated phosphorylation level of AKT, JNK and NF-κB. From these results, BCS may have immunostimulatory activity via TLR2-MAPK, NF-κB and PI3K/AKT signaling pathways. Therefore, BCS expected to be used as a potential immune-enhancing agent.
