        Controlled synthesis and luminescence properties of doped NaLa(WO4)2 microstructures

        In this work, well-defined NaLa(WO4)2microstructures were prepared by a facile hydrothermal processin the presence of oleylamine and oleic acid as a mixed surfactant. The phase, morphology, size andluminescence properties were well characterized by means of X-ray diffraction (XRD), scanning electronmicroscopy (SEM), transmission electron microscopy (TEM), photoluminescence (PL) spectra, and decaytime, respectively. The results reveal that the morphology and size of the as-synthesized samples can betuned by altering the reaction time and the amounts of oleic acid. A possible formation mechanism oftetragonal NaLa(WO4)2 is proposed on the basis of oleic acid amount-dependent experiments. Uponultraviolet (UV) excitation, NaLa(WO4)2:Eu3+ and NaLa(WO4)2:Tb3+ exhibit the characteristic downconversionemissions of Eu3+ and Tb3+. Yb3+/Er3+ and Yb3+/Tm3+ co-doped NaLa(WO4)2 samples showstrong green (Er3+, 4S3/2, 2H11/2!4I15/2) and blue (Tm3+, 1G4!3H6) up-conversion emission under980 nm laser diode excitation. Upconversion emission mechanisms of NaLa(WO4)2:Ln (Ln = Yb/Er, Yb/Tm) are also presented based on the emission spectra and the plot of luminescence intensity to pumppower. It should be noted that the intensity of the upconversion emissions can be dramatically enhancedcompared with sodium free tungstates, which can be attributed to the charge compensation and meansthe introduction of Na+ ions in WO42 causes less distortion in the crystal structure.

      • Alternative Messenger RNA Splicing of Autophagic Gene Beclin 1 in Human B-cell Acute Lymphoblastic Leukemia Cells

        Beclin 1 is a key factor for initiation and regulation of autophagy, which is a cellular catabolic process involved in tumorigenesis. To investigate the role of alternative splicing of Beclin1 in the regulation of autophagy in leukemia cells, Beclin1 mRNA from 6 different types of cell lines and peripheral blood mononuclear cells from 2 healthy volunteers was reversely transcribed, subcloned, and screened for alternative splicing. New transcript variants were analyzed by DNA sequencing. A transcript variant of Beclin 1 gene carrying a deletion of exon 11, which encoded a C-terminal truncation of Beclin 1 isoform, was found. The alternative isoform was assessed by bioinformatics, immunoblotting and subcellular localization. The results showed that this variable transcript is generated by alternative 3' splicing, and its translational product displayed a reduced activity in induction of autophagy by starvation, indicating that the spliced isoform might function as a dominant negative modulator of autophagy. Our findings suggest that the alternative splicing of Beclin 1 might play important roles in leukemogenesis regulated by autophagy.

        In Vitro Screening for Compounds Derived from Traditional Chinese Medicines with Antiviral Activities Against Porcine Reproductive and Respiratory Syndrome Virus

        Seventeen compounds derived from traditional Chinese medicines (TCMs) were tested for their antiviral activity against porcine reproductive and respiratory syndrome virus (PRRSV) in vitro. Visualization with the cytopathologic effect (CPE) assay and the 3-(4, 5-dimethyithiazol- 2-yl)-2,5-diphenyltetrazolium bromide test were used to determine the 50% cytotoxic concentration (CC50) and 50% effective concentration (EC50) in cultured Marc-145 cells. Among the tested compounds, chlorogenic acid and scutellarin showed potential anti-PRRSV activity. The EC50 values were 270.8 ± 14.6 μg/ml and 28.21 ± 26.0 μg/ml and the selectivity indexes were >5.54 and 35.5, respectively. The time-of-addition and virucidal assay indicated that the anti-PRRSV activity of the two compounds could be due to their inhibiting the early stage of virus replication and/or inactivating the virus directly. The inhibition of the virus attachment was not observed in the adsorption inhibition assay. The inhibition ratios of chlorogenic acid and scutellarin were, respectively, 90.8% and 61.1% at the maximum non-cytotoxic concentrations. The results have provided a basis for further exploration of their antiviral properties and mechanisms in vivo. We believe that the chlorogenic acid and scutellarin have a great potential to be developed as new anti-PRRSV drugs for clinical application.

        Analysis of the Ecological Efficiency of Chinese Provincial Based on the Three-stage DEA Model

        에코 효율성은 경제 및 자원 환경의 지속가능한 발전의 중요한 요인이다. 에코 효율성은 중국의 지역 경제 및 자원 환경의 평가 척도가 된다. 본 연구는 3단계 DEA 모델을 이용하여 2014년도의 중국에 있는 31개성의 에코 효율성을 분석 하였다. 분석결과, 외부환경변수인 환경 거버넌스의 총 투자액, 2차 산업 및 1인당 자동차 소유량이 중국 지역의 에코 효율성에 많은 영향을 미치고 있는 것으로 분석되었다. 외부환경 및 임의오류를 제외한 후 중국 지역의 에코 효율성은 0.526에서 0.639로 증가하였고, 순수 기술 효율성은 0.650에서 0.858로 증가하였으며, 규모 효율성은 0.833에서 0.740로 감소하였다. 또한 순수 기술 효율성 및 규모 효율성의 측정을 위해서 중국에 있는 31개성을 4가지 유형으로 구분하였다. 분석결과, 중국의 각 성은 에코 효율성을 개선하기 위해 각성의 특성에 적합한 에코 효율성 유형에 따라 관리수준을 제고하거나 생산규모를 확대해야 하는 것으로 분석되었다. Ecological efficiency is an important index measuring and reflecting the sustainable development level of economy, resources and environment in a region. This paper makes an empirical study on the ecological efficiency of 31 provinces in China in 2014 with the three-stage DEA model. The results show that the three indexes, the total investment in environmental governance (Unit: hundred million Yuan), the second industry proportion(%), and per capita automobile ownership (car/ten thousand people) functioning as the external environmental variables have significantly impacted the regional ecological efficiency. Excluding the impact of the external environment and statistical noise, the technical efficiency of regional ecological efficiency has increased from 0.526 to 0.639, and the pure technical efficiency has increased from 0.650 to 0.858, with the scale efficiency decreased from 0.833 to 0.740, accurately reflecting the regional ecological efficiency level. 31 Chinese provinces are classified into four different types according to the pure technical efficiency and scale efficiency. Every region shall, according to the characteristics of its efficiency, emphasize differently on improving the management level or expanding the scale of production so as to improve the ecological efficiency.

        An integrated strategy for production of four anthocyanin compounds from Ribes nigrum L. by deep eutectic solvents and flash chromatography

        This study explored an integrated strategy for extraction and preparative separation of four anthocyaninsfrom Ribes nigrum L. using DES-based microwave assisted extraction andflash chromatography. Theextraction yield of anthocyanins reached 2.03 mg/g after optimization. An efficient preparativeseparation process of four main anthocyanins by macroporous resin andflash chromatography wasestablished afterwards. The yields of D3G, D3R, C3G and C3R reached 0.32 mg/g, 0.47 mg/g, 0.18 mg/g and0.43 mg/g with the purities of more than 95%, respectively. The developed procedure represents anefficient and convenient alternative for the systematic utilization of natural products from plantmaterials.

        Effect of Dietary Structural to Nonstructural Carbohydrate Ratio on Rumen Degradability and Digestibility of Fiber Fractions of Wheat Straw in Sheep

        The effect of different dietary structural carbohydrate (SC) to nonstructural carbohydrate (NSC) ratios on fiber degradation, digestion, flow, apparent digestibility and rumen fluid characteristics was studied with a design using 18 wethers fitted with permanent rumen and duodenum cannulae. All sheep were divided into six groups randomly, receiving six diets with varying SC to NSC ratios. All diets contained the same proportion of wheat straw and concentrate. The dietary SC to NSC ratios were adjusted by adding cornstarch to the concentrate supplements. The duodenal and fecal flows of dry matter (DM), neutral detergent fiber (NDF), acid detergent fiber (ADF), hemicellulose (HC) and cellulose (CEL) were estimated using chromium-mordanted wheat straw as a flow marker. The degradation parameters of wheat straw DM, NDF, ADF, HC and CEL were determined by incubating the ground wheat straw in nylon bags in the rumen for different periods of time. There was no effect (p>0.05) of the different dietary SC to NSC ratios on rumen pH or $NH_3$-N, but acetate, propionate and butyrate concentrations were significantly affected (p<0.05 or p<0.01) by dietary SC to NSC ratios in the rumen fluid. When the dietary SC to NSC ratio was 2.86, the highest rumen degradability of wheat straw DM, NDF, ADF and CEL was found, but the highest apparent rumen digestibilities of DM, NDF, ADF, HC and CEL occurred at a 2.64 SC to NSC ratio. However, because of compensatory digestion in the hindgut, the apparent digestibilities of DM, NDF, ADF, HC and CEL were highest when the dietary SC to NSC ratio was 2.40. In conclusion, there is a optimal range of dietary SC to NSC ratios (between 2.86 and 2.40) that is beneficial to maximize wheat straw fiber degradation and apparent digestibility.

        Effects of Dietary Nitrogen Sources on Fiber Digestion and Ruminal Fluid Characteristics in Sheep Fed Wheat Straw

        Fifteen Inner Mongolian wethers with permanent ruminal and duodenal cannulas were used to study the effects of dietary rumen-undegradable protein (RUP) to rumen-degradable protein (RDP) ratios or protein sources on fiber digestion in the gastrointestinal tract and ruminal fluid characteristics. Fiber digestion and ruminal fermentation were not affected (p>0.05) by dietary RUP to RDP ratios (from 1.54 to 0.72). Soybean meal supplementation improved ruminal digestion. Fish meal supplementation increased (p<0.05) the ruminal degradability of fiber. The different RUP to RDP ratios (from 1.54 to 0.72) did not influence (p>0.05) ruminal fluid pH, but there were differences (p<0.05) in ruminal fluid $NH_3-N$ concentration because of urea replacement. Soybean meal as a dietary protein source decreased (p<0.05) ruminal fluid pH and increased (p<0.05 or p<0.01) $NH_3-N$, acetate, propionate and butyrate concentrations in the rumen. Fish meal as a dietary protein source decreased (p<0.05 or p<0.01) ruminal $NH_3-N$ and acetate concentrations and increased (p<0.05) ruminal propionate concentration. It can be concluded that dietary protein sources have more significant effect on fiber digestion and ruminal fermentation than different dietary RUP to RDP ratios, when the dietary crude protein requirements of growing sheep are satisfied.

        Beneficial effects of natural Jeju groundwaters on lipid metabolism in high-fat diet-induced hyperlipidemic rats

        BACKGROUND: Groundwater is believed to possess many beneficial effects due to its natural source of various minerals. In this study, we examined the effects of natural Jeju groundwater S1 (SamdasooTM), S2 and S3 pumped up from different locations of Jeju Island, Korea, along with local tap water, on body weight gain, serum lipids and lipoproteins, and liver histopathology in high-fat diet-induced hyperlipidemic rats. MATERIALS/METHODS: Rats were randomly and equally divided into 6 groups. Different water samples were supplied to the hyperlipidemic rats as their daily drinking water and the widely-used anti-hyperlipidemic drug simvastatin was used as a positive control. Body weight, serum lipids and lipoproteins were measured weekly. Liver weight, liver index and liver histopathology were examined after the execution of the rats. RESULTS: After drinking Jeju groundwaters for two months, S2 but not S3 significantly reduced weight growth and serum triglycerides levels and increased high density lipoprotein-C (HDL-C) without affecting total cholesterol or LDL-C. S1 and particularly S2 significantly reduced the severity of liver hypertrophy and steatosis. All Groundwaters had much higher contents of vanadium (S3>S2>S1>>tap water) whereas S1 and S2 but not S3 markedly blocked autoxidation of ferrous ions. CONCLUSION: Jeju Groundwater S1 and particularly S2 exhibit protective effects against hyperlipidemia and fatty liver and hypothesize that the beneficial effect of Jeju Groundwaters may be contributed from blockade of autoxidation of ferrous ions rather than their high contents of vanadium.

        Carboxymethyl Flavonoids and A Monoterpene Glucoside from Selaginella moellendorffii

        A new dihydroflavone, 5-carboxymethyl-7,4'-dihydroxyflavonone (1), and its glucoside 5-carboxymethyl-7,4'-dihydroxyflavonone-7-O-β-D-glucopyranoside (2), and one new monoterpene glucoside, (4Z,6E)-2,7-dimethyl-8-hydroxyocta-4,6-dienoic acid 8-O-β-D-glucopyranoside (3), were isolated from the whole plants of Selaginella moellendorffii. Their structures were determined by spectroscopic methods and chemical transformation. Compound 2 was evaluated for the ability to enhance glucose consumption in normal and insulin-resistant L6 muscle cells induced by high concentrations of insulin and glucose. Glucose consumption in insulin-resistant cells (but not in normal cells) was increased 15.2 ± 3.3% (p < 0.01) by compound 2 at a concentration of 0.1 μM in the presence of insulin (1 nM).
