        Enhancing the Enzymatic Activity of the Multifunctional β-Glycosyl Hydrolase (Cel44C-Man26A<sub>P558</sub>) from Paenibacillus polymyxa GS01 Using DNA Shuffling

        본 연구자들은 이전에 cellulase, xyalnase 및 lichenase의 다기능 효소활성을 지니는 절단된 Cel44C-$Man26A_{P558}$의 ${\beta}$-glycosyl hydrolase를 보고하였다. 본 연구에서는 절단된 Cel44C-$Man26A_{P558}$ 효소의 다기능성 ${\beta}$-glycosyl hydrolase 활성을 증가시키기 위해 DNA shuffling을 시도하였다. DNA shuffling에 의해 단일변이(P438A)를 가진 M2Cel44C-$Man26A_{P558}$와 이중변이(A273T 및 P438A)를 가진 M21Cel44C-$Man26A_{P558}$를 얻었다. 이중변이를 가진 M21Cel44C-$Man26A_{P558}$은 단일변이를 가진 M2Cel44C-$Man26A_{P558}$ 보다 효소활성이 낮게 나타났으나, M2Cel44C-$Man26A_{P558}$와 M21Cel44C-$Man26A_{P558}$은 대조구인 Cel44C-$Man26A_{P558}$ 보다 약 1.3에서 2.2배 정도 높은 효소활성을 나타내었다. 특히, 단일변이를 가진 M2Cel44C-$Man26A_{P558}$는 대조구인 Cel44C-$Man26A_{P558}$보다 cellulase, xylanase 및 lichenase 효소활성이 약 1.5에서 2.2배 정도 높게 나타났다. ${\beta}$-Glycosyl hydrolase의 cellulase, linchenase 및 xylanase 최적 효소활성은 각각 pH 7.0, 7.0 및 6.0에서 이었다. 이러한 결과는, 아미노산 잔기인 Ala438이 다기능성 ${\beta}$-glycosyl hydrolase 활성을 증가시키는 중요한 역할을 한다고 추정할 수 있다. We previously reported that the truncated Cel44C-$Man26A_{P558}$ ${\beta}$-glycosyl hydrolase protein exhibits multifunctional activities, including cellulase, xylanase, and lichenase. DNA shuffling of the truncated Cel44C-$Man26A_{P558}$ enzyme was performed to enhance the enzymatic activity of the multifunctional ${\beta}$-glycosyl hydrolase. Two mutant enzymes, M2Cel44C-$Man26A_{P558}$ that carries one mutation (P438A) and M21Cel44C-$Man26A_{P558}$ that carries two mutations (A273T and P438A) were obtained. The enzymatic activity of the M21Cel44C-$Man26A_{P558}$ double mutant was lower than enzymatic activity of the single mutant (M2Cel44C-$Man26A_{P558}$). However, both mutants displayed the enhancements in their enzyme activities that were ${\approx}1.3$- to 2.2-fold higher than the original enzymatic activity in Cel44C-$Man26A_{P558}$. In particular, the mutant M2Cel44C-$Man26A_{P558}$ exhibited an approximate 1.5- to 2.2-fold increase in the cellulase, xylanase, and lichenase activities in comparison with the control (Cel44C-$Man26A_{P558}$). The optimum cellulase, linchenase, and xylanase activities of ${\beta}$-glycosyl hydrolase were observed at pH 7.0, pH 7.0 and pH 6.0, respectively. These results, therefore, suggest that the amino acid residue Ala438 plays important roles in the enhancement of the activity of multifunctional ${\beta}$-glycosyl hydrolase.

        The robustness of a seam tracking for the automatic welding system is studied. Ac servo motor is used to control the position and velocity of the weld torch. The CCD camera locates ahead of weld torch to preview the weld line, and brings about the inaccuracy on the weld speed. The advanced optimal preview control was used to enhance the tracking performance. The experiment results showed that the tracking performance was improved significantly.

        It has been very important to detect crack initiation during elasto-plastic fracture toughness[J_IC] test. In this study, Unloading compliance method, R-curve method are used to detect the onset of crack extension of SCM440. As the result of the test, the following results were obtained. Elasto-plastic fracture toughness [J_IC] estimation by R-curve method was overestimated than Unloading compliance method.       • Carotenoids 식이와 알코올이 혈중 지질 성분에 미치는 영향에 관한 연구

        Increasingly, humans are subjected to exposure to various foreign chemicals including drugs, food additives, pollutants. Some of them may be harmful and especially give rise to free radicals during metabolism in the tissues. These radicals such as superoxide, hydroxyl radical and hydrogen peroxide, induce oxidation of the macromolecules, resulting in a variety of damages in the body. However, humans have anti-oxidative mechanisms well developed against these damage. Anti-oxidants such as β-carotene and vitamin A, C, and E, are believed to play a major role in these mechanisms by acting as cofactors. On the basis that alcohol, among human's favorite foods, is one of the most common oxidative chemical, this study was carried out to elucidate biochemical changes in the lipid components of the blood after alcohol administration on Sprague-Dawley (SD) rats. Also, we investigated the effects of carotenoid and β-carotene diets on the alcohol-injected rats. At first, the concentrations of serum lipids were highly increased at alcohol-injected rats for one week (AL-1W), but significantly decreased at those dieted with β-carotene for one week (BC-1W). Second, after 2 weeks, the concentrations of serum lipids were exclusive decreased at alcohol-administrated rats. Third, the lipid concentration was increasing in β-carotene-dieted rats throughout the experiments for 3 weeks. These results may suggest that alcohol can cause damages in the liver, resulting in inflammation and increase of the concentration of serum lipids. Also, β-carotene and carotenoid diets are believed to protect the liver against the oxidative damages by alcohol administration.

      • 알코올 및 베타 카로틴이 혈액 성분에 미치는 영향에 관한 연구

        Human take in many kinds of foods, and also are exposed to various chemicals which may be harmful. Especially, some food and chemicals may give rise to free radicals during metabolism in the tissues. These free radicals, such as superoxide. hydroxy radical and hydrogen peroxide, induce oxidations of the macromolecules, which can cause a variety of damages in the body. But human have anti-oxidative mechanisms well developed against these damages. Anti-oxidants such as vitamines A, C, E, and β-carotene are believed to have a major role in these mechanisms acting as a cofactor of anti-oxidative system. On the basis that alcohol is one of the most common chemicals that induce oxidative damages in liver, this study was carried out to elucidate biochemical changes of blood after alcohol administration as well as to investigate the effects of β-carotene on the alcohol-injected Sprague-Dawley rats. The following results were obtained from this study. At first, the number of WBC after one week were lowest in the alcohol-treated group. But, β-carotene treatment by oral injection was shown to diminish the increasing rate of WBC in the alcohol-treated group. Second, RBC was increased all the time in all groups tested with alcohol and/or β-carotene. It is of interest that there was a sudden increase of RBC in the alcohol-treated group. Third, Hb was increased in the alcohol-treated groups on the second week. On the other hand, the groups of β-carotene treatment showed more higher rate of increase in Ht and MCV. It is of note worthy that these effects were maximized in the group of β-carotene treatment followed by diet with cheese for a week. Finally, alcohol had little or no effect on MCH and MCHC, although β-carotene showed atypical effects. These results made it clear that alcohol can cause damages in liver, resulting in inflammation and increase of RBC. Also β-carotene is believed to protect liver from oxidative effects by alcohol. In according to these results, it is suggested that diet of anti-oxidants such as Vt. A, B, C, E, and β-carotene is strongly recommended to prevent certain diseases caused by oxidative damages.

      • 원심펌프 성능에 관한 실험적 연구 (II)

        This work is to investigate effect of operating characteristics of pump according to the rotational speed of pump and the change of flow rate when centrifugal pump operates with the condition that regulated flow, head, revolution number, and specific speed are respectively 0.7 ㎥/min, 8 m, 1750 rpm, 180 (m, ㎥/min, rpm). The pump used in the experiment doesn't have a guide vane and directly connects with outside rim so that the rotational speed of volute pump in spiral or volute casing increase by 100 rpm from 1450 until it reaches 1750. The results of the relationship of Q-H, Q-L, Q-η and the dimensionless performance features such as head coefficient, power coefficient and efficiency are as follows; (1) According to the increase of evolution number, the head and power increase up to the square or cube of the evolution number. At the same evolution number, the power is increased according to the flow. In time of standard operation, the head has a stable feature in the early stage of flow increase from the segment point. (2) The pump's highest efficiency shows 52% with flow rate 0.17㏄/min and head 4.7 m in time of 1450 rpm and 50% with flow 0.18㎥/min and head 6.7 m in the pump's highest rotational speed of 1750 rpm. (3) In each rotational speed, curve of efficiency, head and power is different. Meanwhile, the dimensionless performance valuables show a line irrespective of rotational speed. As flow coefficient increases, power coefficient increases and head coefficient decreases. When the flow coefficient is 0.08, it reaches the maximum efficiency 52%.

      • 원발성 위암조직에서 fragile histidine triad 유전자의 이상 발현에 대한 연구

        Background : The fragile histidine triad(FHIT) gene, located at human chromosome 3p14.2, has been shown to produce aberrant transcripts in a number of human cancers. Also, FHIT protein is supposed to have tumor suppressor function. Methods : To determine the role of the FHIT gene in gastric carcinoma, FHIT mRNA transcripts were examined in 16 primary adenocarcinomas by reverse transcription-polymerase chain reaction(RT-PCR). Expression of FHIT protein was measured by immunohistochemistry. Results : In this study, we found normal and abnormal RT-PCR products in 7 cases, and no transcript in one case. Positive immunostaining was observed in normal gastic gland, whereas FHIT immunoreactivity was significantly decreased or absent in most cases tested. Gastric cancer is thought to be develop from carcinogenic exposure, possibly accompanying with the high frequency of the FHIT gene abnormalities. Conclusion : The above results suggest that the FHIT gene can be a preferential target of environmental carcinogens, and the FHIT inactivation may play a role in the gastric carcinogenicity.

        The purpose of this study is to apply and test lifestyle concept in the field of sports behavior research. The AIO(Activities, Interests, and Opinions) approach is used to operationalize the lifestyle concept. Questionnaires containing 63 AIO statements are admistered to a total 600 members of a private sport club in Busan. 452 usable questionnaires were returned, resulting in a response rate of 75.3%. The sampling was chosen by the stratified random sampling Method. Analysis Method was used factor analysis to identify. The overall structure of club member's lifestyle the main result of the study : There exist eight factors representing dimensions of club member's lifestyle, there are named as sensitive of fashion, preference of Brend, Achievement, stability directed, self-confidence, Nihilism, Health-Conscious, self-affirmative.

      • 음향방출법을 이용한 구조용강의 탄소성 파괴인성 Jic평가에 관한연구

        The estimation of elasto-plastic fracture toughness is necessary for design and manufacturing of machine and structure, economy stability in these use. In this study, J-integral values obtained by R-curve method and Acoustic Emission(AE) method are investigated. Elasto-plastic Fracture toughness Jic estimation by R-curve method are over-estimated than that by AE method, and then proved to be useful. According to the starting point determimed by acoustic emission technique, the clatulation J_IC value is stable, and even more near the demands of J-integral theory in comparison with R-curve method.

      • 스포츠 가치의식에 영향을 미치는 스포츠의식에 관한 연구

        The emergence of a new phenomenon called sports value consciousness and its diversification make itself a meaningful research task, in that it demands a change of healthy human consciousness according to various social structures. The present study, therefore, aims to establish a reasonable standard of sports value consciousness as well as a new social system of value paradigms regarding all sports phenomena by revealing the consciousness patterns and related factors affecting the sports value consciousness sports activities. For this purpose, 533 subjects were chosen by the systematic stratified cluster random sampling method from among the population of athletes and leaders in high schools and universities in Pusan as of 1997. The questionnaire for this research was adopted from the studies of Niwa (1985), Uesugi (1987, 1990), Kusaka (1988), and Asanuma (1990). The original inquiry items underwent two preparatory tests for the judgment of validity and reliability before actual administration. The entire data went through χ²test, discriminant analysis to minimize Wilk's λ. The following conclusions were made from this study: 1. As to sports consciousness, the more educated and the older one was, the more one was likely to develop Recreation (or Secular) orientation. 2. Concerning sports value consciousness, the tendency toward Ascetic and Agon orientation was stronger when one was older, more schooled or one's parents had specialized occupation. As a differentiation of sports value consciousness, 60.5% belonged to Ascetic orientation, 39.5% Agon orientation, 90.6% Leisure (or Play) orientation, and 9.4% Recreation orientation. 3. In the relationship between sports consciousness and sports sense of value, such factors as the principles of discipline, training, continutiy, spiritualism, development, and concentration affected Ascetic of Agon orientation. In addition, the factors or principles of development, spiritualism, and focus on sports had an influence on Leisure or Recreation orientation.
