Identification of Arcanobacterium pyogenes isolated by post mortem examinations of a bearded dragon and a gecko by phenotypic and genotypic properties

        H. Ülbegi-Mohyla,M. Hijazin,J. Alber,A. A. Hassan,A. Abdulmawjood,E. Prenger-Berninghoff,R. Weiß,M. Zschöck,C. Lämmler 대한수의학회 2010 Journal of Veterinary Science Vol.11 No.3

The present study was designed to identify phenotypically and genotypically two Arcanobacterium (A.) pyogenes strains isolated by post mortem examinations of a bearded dragon and a gecko. The A. pyogenes strains showed the typical biochemical properties and displayed CAMP-like synergistic hemolytic activities with various indicator strains. The species identity could be confirmed genotypically by amplification and sequencing of the 16S rDNA gene and, as novel target gene, by sequencing of the beta subunit of RNA polymerase encoding gene rpoB, of both strains and of reference strains representing nine species of the genus Arcanobacterium. The species identity of the two A. pyogenes strains could additionally be confirmed by PCR mediated amplification of species specific parts of the 16S-23S rDNA intergenic spacer region, the pyolysin encoding gene plo and by amplification of the collagen-binding protein encoding gene cbpA. All these molecular targets might help to improve the future identification and further characterization of A. pyogenes which, as demonstrated in the present study, could also be isolated from reptile specimens.

Effect of Acibenzolar-S-methyl and Rahnella aquatilis (Ra39) on Chitinase and β-1, 3-glucanase Activities and Disease Resistance of Apple Plants

        Kamal A. M. Abo-Elyousr,M. A. A. Sallam,M. H. A. Hassan,W. Zeller 한국식물병리학회 2010 Plant Pathology Journal Vol.26 No.1

The effect of Acibenzolar-S-methyl (ASM) and Rahnella aquatilis Ra39 against apple fire blight disease caused by Erwinia amylovora were tested as a possible alternative to streptomycin. In vitro studies, no inhibition effect against the pathogen was found when ASM was tested. Under greenhouse conditions, application of R. aquatilis Ra39 with the highly susceptible M26 rootstock resulted in a marked disease suppression. Application of ASM and strain Ra39 caused a high decrease of the disease, 82% and 58% respectively; this was correlated with a reduction of the growth of the pathogen within host plants up to 64% and 49.5% respectively. Further studies in the field under artificial infection condition during full bloom revealed that application of ASM and R. aquatilis Ra39 with Gala variety resulted in a control effect up to 21 and 29% respectively. In physiological studies, enhanced activities of PR-proteins (chitinase and β-1, 3-glucanase) were detected, which are well known as biochemical markers for systemic acquired resistance. Application of ASM to apple shoots caused the highest chitinase activity followed by strain Ra39. The enzyme activity was increased after 2, 4 and 6 days from application. In addition, ASM-treatment caused the higher β-1, 3-glucanase activity than strain Ra39. Maximum enzyme activity was recorded after 6 days from application and then decreased after 8 and 10 days from application.

Direct Antimicrobial Activity and Induction of Systemic Resistance in Potato Plants Against Bacterial Wilt Disease by Plant Extracts

        Hassan, M.A.E.,Bereika, M.F.F.,Abo-Elnaga, H.I.G.,Sallam, M.A.A. The Korean Society of Plant Pathology 2009 Plant Pathology Journal Vol.25 No.4

The potential of three plants extracts, to protect potato plants against bacterial wilt caused by Ralstonia solanacearum was determined under greenhouse and field conditions. All soil drenching treatments of aqueous plant extracts of Hibsicus sabdariffa, Punica granatum and Eucalyptus globulus significantly reduced the disease severity compared with inoculated control. Although the applications of all three plant extracts resulted in similar reductions of disease severity in field up 63.23 to 68.39%, treatment of E. globulus leaf extract was found greater in restricting the symptom development than other the two plant extracts in the greenhouse. More than 94% reduction in the bacterial wilt symptom was observed in potato plants. All tested plant extracts were effective in inhibiting the growth of bacterial pathogen, not only in vitro, but also in stem of potato plants as compared with the inoculated control Potato plants treated with extract of H. sabdariffa reduced bacterial growth more effectively than treatment with P. granatum and E. globulus. Activity of defence-related enzymes, including peroxidase, polyphenoloxidase and phenylalanine ammonia lyase, were significantly increased in plants treated with the plant extracts compared to the control during the experimental period. In general, the higher enzymes activities were determined in both inoculated and non-inoculated treated potato plants after 8 days from plant extracts treatment. These results suggested that these plant extracts may be play an important role in controlling the potato bacterial wilt disease, through they have antimicrobial activity and induction of systemic resistance in potato plants.

Synthesis, molecular docking studies of coumarinyl-pyrazolinyl substituted thiazoles as non-competitive inhibitors of mushroom tyrosinase

        Saeed, A.,Mahesar, P.A.,Channar, P.A.,Abbas, Q.,Larik, F.A.,Hassan, M.,Raza, H.,Seo, S.Y. Academic Press 2017 Bioorganic chemistry Vol.74 No.-

A series of coumarinyl-pyrazolinyl substituted thiazoles derivatives were synthesized and their inhibitory effects on the DPPH and mushroom tyrosinase were evaluated. The results showed that all of the synthesized compounds exhibited significant mushroom tyrosinase inhibitory activities. In particular, 3-(5-(4-(benzyloxy)-3-methoxyphenyl)-1-(4-(4-bromophenyl)thiazol-2-yl)-4,5-dihydro-1H-pyrazol-3-yl)-2H-chromen-2-one (7j) exhibited the most potent tyrosinase inhibitory activity with IC50 value 0.00458+/-0.00022μM compared with the IC50 value of kojic acid is 16.84+/-0.052μM. The inhibition mechanism analyzed by Lineweaver-Burk plots revealed that the type of inhibition of compound 7j on tyrosinase was noncompetitive. The docking study against tyrosinase enzyme was also performed to determine the binding affinity of the compounds. The compound 7a showed the highest binding affinity (-10.20kcal/mol) with active binding site of tyrosinase. The initial structure activity relationships (SARs) analysis suggested that further development of such compounds might be of interest. The statistics of our results endorses that compound 7j may serve asa structural template for the design and development of novel tyrosinase inhibitors.

Management of Charcoal Rot of Sesame by Seed Soaking in Medicinal Plant Extracts and Hot Water

        Hoda A. M. Ahmed,A. A. Abdel-Razik,M. H. A. Hassan,S. A. Khaled 한국식물병리학회 2010 Plant Pathology Journal Vol.26 No.4

Macrophomina phaseolina causing charcoal rot was isolated from sesame seeds (cvs. Giza 32 and Shandawel-3)collected from different localities of Assiut, Sohage and El-Minia Governorates. The fungus was found in the highest frequently in samples collected from Assiut Governorate followed by Sohag and finally EL Minia Governorate. The obtained isolates were different in their virulence on the tested sesame cvs. Also, they differed in their growth nature including colony color and sclerotial production. The color of colonies of the pathogen seem to be correlated with density of sclerotial formation. Aqueous extracts of Majorna, Wild chamomile,Geranium oil and Nees plants were highly toxic to tested isolates of M. phaseolina, in vitro. On the other hand the rest of the tested aqueous extracts had no effect. Under greenhouse conditions in 2005 and 2006seasons, soaking seeds of sesame before sowing in aqueous extracts of Eucalyptus, Nerium, Ocimum and Roesmary plants decreased the disease incidence. Aqueous extracts of Eucalyptus and Ocimum were the most effective treatment. Dipping sesame seeds in hot water at 60oC for 5 minutes increased seed germination of Giza 32 and Shandawel-3 cvs. followed by 55 oC, 50oC and 45oC, while 40oC treatment resulted the lowest seed germination rate. Dipping sesame seeds in hot water at different temperature before planting decreased seed, seedling and charcoal rots. Soaking seeds in hot water at 60oC increased greatly plant height and decreased seed, seedling rot and charcoal rot followed by 55oC and 50oC, under greenhouse condition.

Electricity generation from rice straw using a microbial fuel cell

        Hassan, S.H.A.,Gad El-Rab, S.M.F.,Rahimnejad, M.,Ghasemi, M.,Joo, J.H.,Sik-Ok, Y.,Kim, I.S.,Oh, S.E. Pergamon Press ; Elsevier Science Ltd 2014 International journal of hydrogen energy Vol.39 No.17

This study demonstrated electricity generation from rice straw without pretreatment in a two-chambered microbial fuel cell (MFC) inoculated with a mixed culture of cellulose-degrading bacteria (CDB). The power density reached 145 mW/m2 with an initial rice straw concentration of 1 g/L; while the coulombic efficiencies (CEs) ranged from 54.3 to 45.3%, corresponding to initial rice straw concentrations of 0.5-1 g/L. Stackable MFCs in series and parallel produced an open circuit voltage of 2.17 and 0.723 V, respectively, using hexacyanoferrate as the catholyte. The maximum power for serial connection of three stacked MFCs was 490 mW/m2 (0.5 mA). In parallelly stacked MFCs, the current levels were approximately 3-fold (1.5 mA) higher than those produced from the serial connection. These results demonstrated that electricity can be produced from rice straw by exploiting CDB as the biocatalyst. Thus, this method provides a promising way to utilize rice straw for bioenergy production.

Behavior of gusset plate-T0-CCFT connections with different configurations

        M.M. Hassan,H.M. Ramadan,M. Naeem,S.A. Mourad 국제구조공학회 2014 Steel and Composite Structures, An International J Vol.17 No.5

Concrete-filled steel tube (CFT) composite columns, either circular (CCFT) or rectangular (RCFT), have many economical and aesthetic advantages but the behavior of their connections are complicated. This study aims to investigate, through an experimental program, the performance and behavior of different connections configurations between circular concrete filled steel tube columns (CCFT) and gusset plates subjected to shear and axial compression loadings. The study included seventeen connection subassemblies consisting of a fixed length steel tube and gusset plate connected to the tube end with different details tested under half cyclic loading. A notable effect was observed on the behavior of the connections due to its detailing changes with respect to capacity, failure mode, ductility, and stress distribution.
